Tralokinumab is a human monoclonal antibody targeting interleukin-13 that is approved for the treatment of moderate-severe atopic dermatitis. Studies analyzing the efficacy and safety of tralokinumab ...in a real-world setting are scarce.
A European, multicentric, real-world, retrospective cohort study was defined to assess the effectiveness and safeness profile of tralokinumab, investigating the achievement of pre-specified treatment goals; and to detect potential differences in terms of effectiveness and safeness across some selected patient subcohorts.
A total of 194 adult patients were included in this study. A significant improvement in physician-assessed disease severity was detected at each follow-up visit as compared with baseline and similar trend was observed for patient-reported outcomes and quality of life. No meaningful difference in effectiveness was found when considering patient age (<65 versus ≥65 years), neither dissecting patient cohort in dupilumab-naive vs dupilumab-treated subjects. Among tralokinumab-treated patients, 88% achieved at least one currently identified real-world therapeutic goal at week 16.
This retrospective multicenter study confirmed the effectiveness and safeness of tralokinumab throughout 32 weeks of observation, showing the achievement of therapeutic goals identified in both trial and real-world settings in a large proportion of tralokinumab-treated patients.
We present the results of a point-cloud-based survey deriving from the use of image-based techniques, in particular with multi-image monoscopic digital photogrammetry systems and software, the ...so-called “structure-from-motion” technique. The aim is to evaluate the advantages and limitations of such procedures in architectural surveying, particularly in conditions that are “at the limit”. A particular case study was chosen: the courtyard of Palazzo dei Capitani del Popolo in Ascoli Piceno, Italy, which can be considered the ideal example due to its notable vertical, rather than horizontal, layout. In this context, by comparing and evaluating the different results, we present experimentation regarding this single case study with the aim of identifying the best workflow to realise a complex, articulated set of representations—using 3D modelling and 2D processing—necessary to correctly document the particular characteristics of such an architectural object.
Chloride anions permeate the bacterial NanC porin in physiological processes. Here we present a DFT-based QM/MM study of this porin in the presence of these anions. Comparison is made with classical ...MD simulations on the same system. In both QM/MM and classical approaches, the anions are almost entirely solvated by water molecules. However, the average water-Cl(-) distance is significantly larger in the first approach. Polarization effects of protein groups close to Cl(-) anion are sizeable. These effects might modulate the anion-protein electrostatic interactions, which in turn play a central role for selectivity mechanisms of the channel.

The survey of historical façades contains several bottlenecks, mainly related to the geometrical structure, the decorative framework, the presence of natural or artificial obstacles, the environment ...limitations. Urban context presents additional restrictions, binding by ground acquisition activity and leading to building data loss. The integration of TLS and close-range photogrammetry allows to go over such stuff, not overcoming the shadows effect due to the ground point of view. In the last year the massive use of UAVs in survey activity has permitted to enlarge survey capabilities, reaching a deeper knowledge in the architecture analysis. In the meanwhile, several behaviour rules have been introduced in different countries, regulating the UAVs use in different field, strongly restricting their application in urban areas. Recently very small and light platforms have been presented, which can partially overcome these rules restrictions, opening to very interesting future scenarios. This article presents the application of one of these very small RPAS (less than 300 g), equipped with a low-cost camera, in a close range photogrammetric survey of an historical building façade in Bologna (Italy). The suggested analysis tries to point out the system accuracy and details acquisition capacity. The final aim of the paper is to validate the application of this new platform in an architectonic survey pipeline, widening the future application of close-range photogrammetry in the architecture acquisition process.
This paper presents some steps in experimentation aimed at describing urban spaces made following the series of earthquakes that affected a vast area of central Italy starting on 24 August 2016. More ...specifically, these spaces pertain to historical centres of limited size and case studies that can be called “problematic” (due to complex morphological and settlement conditions, because they are difficult to access, or because they have been affected by calamitous events, etc.). The main objectives were to verify the use of sets of heterogeneous data that are already largely available to define a workflow and develop procedures that would allow some of the steps to be automated as much as possible. The most general goal was to use the experimentation to define a methodology to approach the problem aimed at developing descriptive responsive models of the urban space, that is, morphological and computer-based models capable of being modified in relation to the constantly updated flow of input data.

Subjects submitted to intensive rehabilitation after first-ever stroke, from 1st January 2009 to 31st December 2010.
Participation was investigated through the Frenchay Activity Index (FAI) and the Functional Status Questionnaire (FSQ) at 2.4±0.5 years after the event. Basic activities of daily living (ADL) and mood were also assessed through the Modified Barthel Index (MBI) and the Beck Depression Inventory (BDI). A retrospective search of the medical records looked for: type/side of brain lesion, stroke clinical syndromes, comorbidities and functional condition at discharge from intensive rehabilitation (upper limb motricity index-ULMI-, Functional Ambulation Category-FAC, MBI, cognitive deficits).
Forty-five subjects (17 female, age 70.1±11.5 years) were enrolled. They showed a striking restriction in their Participation, mainly for FAI-outdoor activities (median FAI score was <50% of the theoretical maximum). A poor gait function (FAC) and an impaired mood (BDI) were the only independent predictors of FAI indoor (F=6.1; p=,005; R^2= 64%) and outdoor activities (F=4.1; P=0.01; R^2=48%), respectively. The univariate analysis showed a strong dependence of all FSQ scores from global disability (MBI), motor function impairment (ULMI and FAC) and cognitive deficits. Depression influenced "psychological function" score, whereas gait capacity was the only factor significantly associated with the "work performance" score. The gait function level, achieved after intensive rehabilitation, was extrapolated by the multivariate analysis, as the most powerful independent predictor of the chronic activity limitations, as measured by MBI (F=33.8, P<0.0001, R2=0.539).
Gait dysfunction is the main factor of Activity limitations and Participation restriction in chronic stroke. Participation is restricted by global disability, depression, older age and dementia. More than 50% variance of Participation measures cannot be explained by the quoted factors.
The study results support the need to integrate the standard rehabilitation approach with vocational rehabilitation in order to reduce Participation restriction.
Enhancement of cultural heritage is not simply a matter of preserving material objects but comes full circle only when the heritage can be enjoyed and used by the community. This is the rationale ...behind this presentation: an urban Virtual Tour to explore the 1937 design of the Fascist Headquarters in Littoria, now part of Latina, by the architect Oriolo Frezzotti. Although the application is deliberately "simple", it was part of a much broader framework of goals. One such goal was to create "friendly and perceptively meaningful" interfaces by integrating different "3D models" and so enriching. In fact, by exploiting the activation of natural mechanisms of visual perception and the ensuing emotional emphasis associated with vision, the illusionistic simulation of the scene facilitates access to the data even for "amateur" users. A second goal was to "contextualise the information" on which the concept of cultural heritage is based. In the application, communication of the heritage is linked to its physical and linguistic context; the latter is then used as a basis from which to set out to explore and understand the historical evidence. A third goal was to foster the widespread dissemination and sharing of this heritage of knowledge. On the one hand we worked to make the application usable from the Web, on the other, we established a reliable, rapid operational procedure with high quality processed data and ensuing contents. The procedure was also repeatable on a large scale.

We review here some of the most reliable and efficient computational theoretical ab initio techniques for the prediction of optical and electronic spectroscopic properties and show some important applications to molecules, surfaces, and solids. We investigate the role of the solvent in the optical absorption spectrum of indole molecule. We study the excited-state properties of a photo-active minimal model molecule for the retinal of rhodopsin, responsible for vision mechanism in animals. We then show a study about spectroscopic properties of Si(111) surface. Finally we simulate a bulk system: paper, that is mainly made of cellulose, a pseudo-crystalline material representing 40% of annual biomass production in the Earth.
Binding of proteins and small molecules to RNA involves many electrostatic interactions, which may alter the distribution of ions around the RNA molecule. Here, we use molecular dynamics simulations ...to investigate how binding of a cyclic peptide mimic of the HIV-1 Tat protein affects the ionic distribution around the HIV-1 TAR RNA element. The calculations reproduce the structural properties observed in NMR studies of TAR and its complex. They also provide insight into the rearrangement of counterions during the molecular recognition events leading to the formation of the protein/RNA complex.
We analyze the recently proposed mirror superposition experiment of Marshall, Simon, Penrose, and Bouwmeester, assuming that the mirror's dynamics contains a nonunitary term of the Lindblad-type ...proportional to -q,q,rho, with q the position operator for the center of mass of the mirror, and rho the statistical operator. We derive an exact formula for the fringe visibility for this system. We discuss the consequences of our result for tests of environmental decoherence and of collapse models. In particular, we find that with the conventional parameters for the continuous spontaneous localization model of state vector collapse, maintenance of coherence is expected to within an accuracy of at least 1 part in 10(8). Increasing the apparatus coupling to environmental decoherence may lead to observable modifications of the fringe visibility, with time dependence given by our exact result.
