In humans, many diseases are associated with the accumulation of free radicals. Antioxidants can scavenge free radicals and minimize their impact. Therefore, the search for naturally occurring ...antioxidants of plant origin is imperative. Here, we aimed to investigate the antioxidant and free radical scavenging properties of methanolic extracts from Tabebuia pallida (T. pallida) stem bark (TPSB), root bark (TPRB), leaves (TPL), and flowers (TPF).
The antioxidant and free radical scavenging activity were determined by several standard methods using spectrophotomer. Total phenolic and flavonoid contents were estimated using Folin-Ciocalteu reagent and aluminum chloride colorimetric assay methods, respectively.
Among the extracts, TPL showed the highest total antioxidant capacity followed by TPRB, TPF, and TPSB. Based on DPPH and hydroxyl radical scavenging activity, TPL showed strong scavenging activity (91.05 ± 1.10 and 62.00 ± 0.57) with IC50 of 9.20 ± 0.28 and 46.00 ± 2.84 μg/mL, respectively when compared with standard BHT (IC50 of 7.00 ± 0.25 μg/mL) and CA (75.00 ± 0.14 μg/mL). These results suggest that TPL had the highest radical scavenging activity among the extractives that closely resembled the standard's. In lipid peroxidation inhibition assay, TPL exhibited the most potent inhibitory activity (83.18 ± 2.12 %) with IC50 of 12.00 ± 2.12 μg/mL, which closely resembled standard CA (IC50 of 10.50 ± 0.28 μg/mL). Also, the reducing capacity on ferrous ion was in the following order: TPL > TPRB > TF > TPSB. The phenolic and flavonoid contents of TPL were higher than other extractives. A positive correlation (p value <0.001) was observed between phenolic content and free radical (DPPH(·) and (·)OH) scavenging efficiencies and lipid peroxidation inhibition activity.
Methanolic extract of T. pallida leaf is a potential source of natural antioxidants and serves as an effective free radical scavenger and/or inhibitor. Hence, T. pallida might be a good plant-based pharmaceutical product for several diseases caused by free radicals.
Multidrug-resistant (MDR) pathogens have created a fatal problem for human health and antimicrobial treatment. Among the currently available antibiotics, many are inactive against MDR pathogens. In ...this context, heterocyclic compounds/drugs play a vital role. Thus, it is very much essential to explore new research to combat the issue. Of the available nitrogen-bearing heterocyclic compounds/drugs, pyridine derivatives are of special interest due to their solubility. Encouragingly, some of the newly synthesized pyridine compounds/drugs are found to inhibit multidrug-resistant S. aureus (MRSA). Pyridine scaffold bearing poor basicity generally improves water solubility in pharmaceutically potential molecules and has led to the discovery of numerous broad-spectrum therapeutic agents. Keeping these in mind, we have reviewed the chemistry, recent synthetic techniques, and bacterial preventative activity of pyridine derivatives since 2015. This will facilitate the development of pyridine-based novel antibiotic/drug design in the near future as a versatile scaffold with limited side effects for the next-generation therapeutics.
Platinum‐induced peripheral neurotoxicity (PIPN) is a common side effect of platinum‐based chemotherapy that may cause dose reduction and discontinuation, with oxaliplatin being more neurotoxic. PIPN ...includes acute neurotoxicity restricted to oxaliplatin, and chronic non‐length‐dependent sensory neuronopathy with positive and negative sensory symptoms and neuropathic pain in both upper and lower limbs. Chronic sensory axonal neuropathy manifesting as stocking‐and‐glove distribution is also frequent. Worsening of neuropathic symptoms after completing the last chemotherapy course may occur. Motor and autonomic involvement is uncommon. Ototoxicity is frequent in children and more commonly to cisplatin. Platinum‐based compounds result in more prolonged neuropathic symptoms in comparison to other chemotherapy agents. Patient reported outcomes questionnaires, clinical evaluation and instrumental tools offer complementary information in PIPN. Electrodiagnostic features include diffusely reduced/abolished sensory action potentials, in keeping with a sensory neuronopathy. PIPN is dependent on cumulative dose but there is a large variability in its occurrence. The search for additional risk factors for PIPN has thus far yielded no consistent findings. There are currently no neuroprotective strategies to reduce the risk of PIPN, and symptomatic treatment is limited to duloxetine that was found effective in a single phase III intervention study. This review critically examines the pathogenesis, incidence, risk factors (both clinical and pharmacogenetic), clinical phenotype and management of PIPN.
Guillain-Barré syndrome (GBS) is a rare, but potentially fatal, immune-mediated disease of the peripheral nerves and nerve roots that is usually triggered by infections. The incidence of GBS can ...therefore increase during outbreaks of infectious diseases, as was seen during the Zika virus epidemics in 2013 in French Polynesia and 2015 in Latin America. Diagnosis and management of GBS can be complicated as its clinical presentation and disease course are heterogeneous, and no international clinical guidelines are currently available. To support clinicians, especially in the context of an outbreak, we have developed a globally applicable guideline for the diagnosis and management of GBS. The guideline is based on current literature and expert consensus, and has a ten-step structure to facilitate its use in clinical practice. We first provide an introduction to the diagnostic criteria, clinical variants and differential diagnoses of GBS. The ten steps then cover early recognition and diagnosis of GBS, admission to the intensive care unit, treatment indication and selection, monitoring and treatment of disease progression, prediction of clinical course and outcome, and management of complications and sequelae.
Complement activation plays a critical role in the pathogenesis of Guillain-Barré syndrome (GBS), a debilitating immune-mediated neuropathy. Mannose-binding lectin (MBL) is a complement activation ...factor of lectin pathway which as genetic host factor may influence the susceptibility or severity of GBS. We investigated the frequency of MBL2 promoter (- 550H/L and - 221X/Y) and functional region (exon 1 A/O) polymorphisms and their association with disease susceptibility, clinical features and serum MBL among GBS patients (n = 300) and healthy controls (n = 300) in Bangladesh. The median patient age was 30 years (IQR: 18-42; males, 68%). MBL2 polymorphisms were not significantly associated with GBS susceptibility compared to healthy controls. HL heterozygosity in GBS patients was significantly associated with mild functional disability at enrolment (P = 0.0145, OR, 95% CI 2.1, 1.17-3.82). The HY, YA, HA and HYA heterozygous haplotypes were more common among mildly affected (P = 0.0067, P = 0.0086, P = 0.0075, P = 0.0032, respectively) than severely affected patients with GBS. Reduced serum MBL was significantly associated with the LL, OO and no HYA variants and GBS disease severity. No significant association was observed between MBL2 polymorphisms and electrophysiological variants, recent Campylobacter jejuni infection or anti-ganglioside (GM1) antibody responses in GBS. In conclusion, MBL2 gene polymorphisms are related to reduced serum MBL and associated with the severity of GBS.
The kinetics of total organic carbon (TOC) destruction during supercritical water gasification (SCWG) of glucose were studied at 400–500°C and 25 MPa in a 600 mL batch reactor. Both TOC and water ...concentrations are critical for the conversion of TOC in supercritical water, especially at longer residence times. Initially, it was assumed that the TOC destruction reaction followed first‐order kinetics ignoring the water concentration. However, experimental results showed that the feed‐to‐water ratio had a significant effect on TOC decomposition. Considering the water concentration in the reaction, the reaction orders of TOC (2.35) and water (1.45) were calculated using nonlinear regression analysis (the Runge‐Kutta method). The estimated pre‐exponential factor (k’) and activation energy (E) were calculated to be 8.1 ± 2/min and 90.37 ± 9.38 kJ/mol respectively.
Sugar-fatty acid esters (SFAEs, especially sucrose- and glucose-based ones) have dominated the chemical industries for more than 50 years. In comparison to other carbohydrate products, SFAEs serve ...essential roles in a variety of industries, including the food, pharmaceutical, and cosmetic industries. In this context, the 6-O-hexanoyl ester of 3-O-butyl-1,2-O-isopropylidene-α-D-glucofuranose was synthesized from commercially available D-glucose in a few steps. For a comparative biological study, the 5,6-di-O-benzyl ether of 3-O-butyl-1,2-O-isopropylidene-α-D-glucofuranose was also prepared and characterized. An in vitro antimicrobial test of all the ester and ether compounds indicated that these compounds are more susceptible to fungi than bacteria. Also, they have more potential for A. niger than A. flavus. According to Prediction of Spectra for Substances (PASS), the chemicals found in the current investigation have a variety of potential biological functions.
Objective: To investigate the comparative effects of Diospyros blancoi(Ebenaceae)leaves(DBL), root bark(DBRB) and stem bark(DBSB) on free radicals and cancer.Methods: The polyphenol contents, ...antioxidant and free radical scavenging properties were determined using standard spectrophotometric methods. Cytotoxicity and anticancer activities were performed on brine shrimp nauplii and Ehrlich ascite carcinoma cells, respectively.Results: Among the extracts, DBSB showed the highest total antioxidant capacity and reducing capacity on ferrous ion. Based on 1,1-diphenyl-2-picrylhydrazyl and hydroxyl radical scavenging activities, DBSB showed(95.760 ± 0.343)% and(67.460 ± 2.641)%scavenging with IC50of(3.10 ± 0.17) and(50.00 ± 3.11) mg/m L, respectively. The IC50 values of standard butylated hydroxytoluene and catechin(CA) for 1,1-diphenyl-2-picrylhydrazyl and hydroxyl radicals were(8.50 ± 0.25) and(75.00 ± 0.14) mg/m L,respectively suggesting that DBSB had a significant(P < 0.05) radical scavenging activity than standards. In lipid peroxidation inhibition assay, the inhibitory activity of the extracts and the standard was in the following order: DBSB > DBRB > CA > DBL.Also, the phenolic (139.91 ± 3.924) mg gallic acid equivalent/g and flavonoid contents(412.00 ± 16.70) mg catechin equivalent/g) of DBSB were higher than that of other extracts. In addition, the DBSB showed the moderate cytotoxic and anticancer properties.Conclusions: Our results indicate that Diospyros blancoi stem bark had the significant highest antioxidant and free radical scavenging properties as well as moderate anticancer activity.Hence, we assume that the anticancer activity of this plant can be, at least in part, attributed to its content in phenolic compounds as well as its significant free radical scavenging properties.
Originating from the base of southern foothill of the Shillong Plateau, Jadukata fan is one of the largest fans situated in the mid-northern edge of Bangladesh. The area in and around Jadukata fan ...lies in the active and complex Himalayan tectonic zone as envisaged by the high seismicity in the area. The fan was studied with the main objective of understanding the formation and development of this tropical, humid alluvial fan in relation to Holocene tectonics. The geological and geomorphological inferences are based on multi-temporal remote sensing data. Information extracted from RS data were checked in the field and compared with other relevant data and information. Identified evidences are topographic expression, anomalous and preferred channel migration, aligned bends in consecutive channels, change in sediments distribution pattern etc. on the fan. These geological and geomorphological evidences as well as past and recent seismic events of the area strongly indicate that the alluvial fan building processes is being controlled by the neotectonic activities. The article also highlights the reasons for the occurrences of the natural hazard based on on-going tectonic activities.
SARS-CoV2 virus could be potentially myopathic. Serum creatinine phosphokinase (CPK) is frequently found elevated in severe SARS-CoV2 infection, which indicates skeletal muscle damage precipitating ...limb weakness or even ventilatory failure.
We addressed such a patient in his forties presented with features of severe SARS-CoV2 pneumonia and high serum CPK. He developed severe sepsis and acute respiratory distress syndrome (ARDS) and received intravenous high dose corticosteroid and tocilizumab to counter SARS-CoV2 associated cytokine surge. After 10 days of mechanical ventilation (MV), weaning was unsuccessful albeit apparently clear lung fields, having additionally severe and symmetric limb muscle weakness. Ancillary investigations in addition with serum CPK, including electromyogram, muscle biopsy, and muscle magnetic resonance imaging (MRI) suggested acute myopathy possibly due to skeletal myositis.
We wish to stress that myopathogenic medication in SARS-CoV2 pneumonia should be used with caution. Additionally, serum CPK could be a potential marker to predict respiratory failure in SARS-CoV2 pneumonia as skeletal myopathy affecting chest muscles may contribute ventilatory failure on top of oxygenation failure due to SARS-CoV2 pneumonia.
