Smart charging of electric vehicles (EVs) is an essential approach to reduce the costs and maximise the benefits of increasing numbers of EVs being connected to the power grid. This article analyses ...139 tariffs and services for smart EV charging available in Europe. It finds that while the market for smart EV charging services is growing, there is a lack of consumer information on the savings and broader environmental benefits it offers, such as integration of renewables. Offers are also unevenly distributed across the continent, resulting in unequal access to smart EV charging. The article outlines six strategies to establish framework conditions in energy markets that would address these gaps and establish smart charging as a standard means of charging, to ensure the beneficial integration of EVs into power grids.
Machine learning (ML) is a key technology in smart manufacturing as it provides insights into complex processes without requiring deep domain expertise. This work deals with deep learning algorithms ...to determine a 3D reconstruction from a single 2D grayscale image. The potential of 3D reconstruction can be used for quality control because the height values contain relevant information that is not visible in 2D data. Instead of 3D scans, estimated depth maps based on a 2D input image can be used with the advantage of a simple setup and a short recording time. Determining a 3D reconstruction from a single input image is a difficult task for which many algorithms and methods have been proposed in the past decades. In this work, three deep learning methods, namely stacked autoencoder (SAE), generative adversarial networks (GANs) and U-Nets are investigated, evaluated and compared for 3D reconstruction from a 2D grayscale image of laser-welded components. In this work, different variants of GANs are tested, with the conclusion that Wasserstein GANs (WGANs) are the most robust approach among them. To the best of our knowledge, the present paper considers for the first time the U-Net, which achieves outstanding results in semantic segmentation, in the context of 3D reconstruction tasks. Unlike the U-Net, which uses standard convolutions, the stacked dilated U-Net (SDU-Net) applies stacked dilated convolutions. Of all the 3D reconstruction approaches considered in this work, the SDU-Net shows the best performance, not only in terms of evaluation metrics but also in terms of computation time. Due to the comparably small number of trainable parameters and the suitability of the architecture for strong data augmentation, a robust model can be generated with only a few training data.
We investigate the formation of unilamellar lipid vesicles (liposomes) with diameters of tens of nanometers by controlled microfluidic mixing and nanoparticle determination (COMMAND). Our study ...includes liposome synthesis experiments and numerical modeling of our microfluidic implementation of the batch solvent injection method. We consider microfluidic liposome formation from the perspective of fluid interfaces and convective-diffusive mixing, as we find that bulk fluid flow parameters including hydrodynamically focused alcohol stream width, final alcohol concentration, and shear stress do not primarily determine the vesicle formation process. Microfluidic device geometry in conjunction with hydrodynamic flow focusing strongly influences vesicle size distributions, providing a coarse method to control liposome size, while total flow rate allows fine-tuning the vesicle size in certain focusing regimes. Although microfluidic liposome synthesis is relatively simple to implement experimentally, numerical simulations of the mixing process reveal a complex system of fluid flow and mass transfer determining the formation of nonequilibrium vesicles. These results expand our understanding of the microfluidic environment that controls liposome self-assembly and yield several technological advances for the on-chip synthesis of nanoscale lipid vesicles.
Summary Given the dual epidemics of HIV and tuberculosis in sub-Saharan Africa and evidence suggesting a disproportionate burden of these diseases among detainees in the region, we aimed to ...investigate the epidemiology of HIV and tuberculosis in prison populations, describe services available and challenges to service delivery, and identify priority areas for programmatically relevant research in sub-Saharan African prisons. To this end, we reviewed literature on HIV and tuberculosis in sub-Saharan African prisons published between 2011 and 2015, and identified data from only 24 of the 49 countries in the region. Where data were available, they were frequently of poor quality and rarely nationally representative. Prevalence of HIV infection ranged from 2·3% to 34·9%, and of tuberculosis from 0·4 to 16·3%; detainees nearly always had a higher prevalence of both diseases than did the non-incarcerated population in the same country. We identified barriers to prevention, treatment, and care services in published work and through five case studies of prison health policies and services in Zambia, South Africa, Malawi, Nigeria, and Benin. These barriers included severe financial and human-resource limitations and fragmented referral systems that prevent continuity of care when detainees cycle into and out of prison, or move between prisons. These challenges are set against the backdrop of weak health and criminal-justice systems, high rates of pre-trial detention, and overcrowding. A few examples of promising practices exist, including routine voluntary testing for HIV and screening for tuberculosis upon entry to South African and the largest Zambian prisons, reforms to pre-trial detention in South Africa, integration of mental health services into a health package in selected Malawian prisons, and task sharing to include detainees in care provision through peer-educator programmes in Rwanda, Zimbabwe, Zambia, and South Africa. However, substantial additional investments are required throughout sub-Saharan Africa to develop country-level policy guidance, build human-resource capacity, and strengthen prison health systems to ensure universal access to HIV and tuberculsosis prevention, treatment, and care of a standard that meets international goals and human rights obligations.
The technology of hairpin welding, which is frequently used in the automotive industry, entails high-quality requirements in the welding process. It can be difficult to trace the defect back to the ...affected weld if a non-functioning stator is detected during the final inspection. Often, a visual assessment of a cooled weld seam does not provide any information about its strength. However, based on the behavior during welding, especially about spattering, conclusions can be made about the quality of the weld. In addition, spatter on the component can have serious consequences. In this paper, we present in-process monitoring of laser-based hairpin welding. Using an in-process image analyzed by a neural network, we present a spatter detection method that allows conclusions to be drawn about the quality of the weld. In this way, faults caused by spattering can be detected at an early stage and the affected components sorted out. The implementation is based on a small data set and under consideration of a fast process time on hardware with limited computing power. With a network architecture that uses dilated convolutions, we obtain a large receptive field and can therefore consider feature interrelation in the image. As a result, we obtain a pixel-wise classifier, which allows us to infer the spatter areas directly on the production lines.
The bacterial Rcs phosphorelay signals perturbations of the bacterial cell envelope to its response regulator RcsB, which regulates transcription of multiple loci related to motility, biofilm ...formation and various stress responses. RcsB is unique, as its set of target loci is modulated by interaction with auxiliary regulators including BglJ. The BglJ-RcsB heteromer is known to activate the HNS repressed leuO and bgl loci independent of RcsB phosphorylation. Here, we show that BglJ-RcsB activates the promoters of 10 additional loci (chiA, molR, sfsB, yecT, yqhG, ygiZ, yidL, ykiA, ynbA and ynjI). Furthermore, we mapped the BglJ-RcsB binding site at seven loci and propose a consensus sequence motif. The data suggest that activation by BglJ-RcsB is DNA phasing dependent at some loci, a feature reminiscent of canonical transcriptional activators, while at other loci BglJ-RcsB activation may be indirect by inhibition of HNS-mediated repression. In addition, we show that BglJ-RcsB activates transcription of bgl synergistically with CRP where it shifts the transcription start by 20 bp from a position typical for class I CRP-dependent promoters to a position typical for class II CRP-dependent promoters. Thus, BglJ-RcsB is a pleiotropic transcriptional activator that coordinates regulation with global regulators including CRP, LeuO and HNS.

Studies of Malawi's option B+ programme for HIV-positive pregnant and breastfeeding women have reported high loss to follow-up during pregnancy and at the start of antiretroviral therapy (ART), but ...few data exist about retention during breastfeeding and after weaning. We examined loss to follow-up and retention in care in patients in the option B+ programme during their first 3 years on ART.
We analysed two data sources: aggregated facility-level data about patients in option B+ who started ART between Oct 1, 2011, and June 30, 2012, at 546 health facilities; and patient-level data from 20 large facilities with electronic medical record system for HIV-positive women who started ART between Sept 1, 2011, and Dec 31, 2013, under option B+ or because they had WHO clinical stages 3 or 4 disease or had CD4 counts of less than 350 cells per μL. We used facility-level data to calculate representative estimates of retention and loss to follow-up. We used patient-level data to study temporal trends in retention, timing of loss to follow-up, and predictors of no follow-up and loss to follow-up. We defined patients who were more than 60 days late for their first follow-up visit as having no follow-up and patients who were more than 60 days late for a subsequent visit as being lost to follow-up. We calculated proportions and cumulative probabilities of patients who had died, stopped ART, had no follow-up, were lost to follow-up, or were retained alive on ART for 36 months. We calculated odds ratios and hazard ratios to examine predictors of no follow-up and loss to follow-up.
Analysis of facility-level data about patients in option B+ who had not transferred to a different facility showed retention in care to be 76·8% (20 475 of 26,658 patients) after 12 months, 70·8% (18,306 of 25,849 patients) after 24 months, and 69·7% (17,787 of 25,535 patients) after 36 months. Patient-level data included 29,145 patients. 14,630 (50·2%) began treatment under option B+. Patients in option B+ had a higher risk of having no follow-up and, for the first 2 years of ART, higher risk of loss to follow-up than did patients who started ART because they had CD4 counts less than 350 cells per μL or WHO clinical stage 3 or 4 disease. Risk of loss to follow-up during the third year was low and similar for patients retained for 2 years. Retention rates did not change as the option B+ programme matured.
Our data suggest that pregnant and breastfeeding women who start ART immediately after they are diagnosed with HIV can be retained on ART through the option B+ programme, even after many have stopped breastfeeding. Interventions might be needed to improve retention in the first year on ART in option B+.

COPD is a chronic lung disease that affects millions of people, declining their lung function and impairing their life quality. Despite years of research and drug approvals, we are still not capable ...of halting progression or restoring normal lung function. Mesenchymal stem cells (MSC) are cells with extraordinary repair capacity, and MSC-based therapy brings future hope for COPD treatment, although the best source and route of administration are unclear. MSC from adipose tissue (AD-MSC) represents an option for autologous treatment; however, they could be less effective than donor MSC. We compared in vitro behavior of AD-MSC from COPD and non-COPD individuals by migration/proliferation assay, and tested their therapeutic potential in an elastase mouse model. In addition, we tested intravenous versus intratracheal routes, inoculating umbilical cord (UC) MSC and analyzed molecular changes by protein array. Although COPD AD-MSC have impaired migratory response to VEGF and cigarette smoke, they were as efficient as non-COPD in reducing elastase-induced lung emphysema. UC-MSC reduced lung emphysema regardless of the administration route and modified the inflammatory profile in elastase-treated mice. Our data demonstrate equal therapeutic potential of AD-MSC from COPD and non-COPD subjects in the pre-clinical model, thus supporting their autologous use in disease.
