Identification of the CD1a+ moDC as an inflammatory subset with specialized activation of RLR and associated cytokine and functional activities.
Cytosolic RIG‐I‐like helicases (RLR) are PRRs involved ...in type I IFN production and antiviral immunity. This study focuses to the comparison of the expression, function, and signaling cascades associated to RLR in the previously identified CD14−DC‐SIGN+PPARγlowCD1a+ and CD14lowDC‐SIGN+PPARγhighCD1a− human moDC subsets. Our results revealed that the expression of RLR genes and proteins as well as the activity of the coupled signaling pathways are significantly higher in the CD1a+ subset than in its phenotypically and functionally distinct counterpart. Specific activation of RLR in moDCs by poly(I:C) or influenza virus was shown to induce the secretion of IFN‐β via IRF3, whereas induction of proinflammatory cytokine responses were predominantly controlled by TLR3. The requirement of RLR‐mediated signaling in CD1a+ moDCs for priming naïve CD8+ T lymphocytes and inducing influenza virus‐specific cellular immune responses was confirmed by RIG‐I/MDA5 silencing, which abrogated these functions. Our results demonstrate the subset‐specific activation of RLR and the underlying mechanisms behind its cytokine secretion profile and identify CD1a+ moDCs as an inflammatory subset with specialized functional activities. We also provide evidence that this migratory DC subset can be detected in human tonsil and reactive LNs.
More aggressive prostate cancer cells (PCCs) are often resistant to chemotherapy. Differences exist in redox status and mitochondrial metabolism that may help explain this phenomenon. Two human PCC ...lines, PC-3 cells (more aggressive) and LNCaP cells (less aggressive), were compared with regard to cellular glutathione (GSH) levels, susceptibility to either oxidants or GSH depletors, and expression of several proteins involved in apoptosis and stress response to test the hypothesis that more aggressive PCCs exhibit higher GSH concentrations and are relatively resistant to cytotoxicity. PC-3 cells exhibited 4.2-fold higher GSH concentration than LNCaP cells but only modest differences in acute cytotoxicity were observed at certain time points. However, only LNCaP cells underwent diamide-induced apoptosis. PC-3 cells exhibited higher levels of Bax and caspase-8 cleavage product but lower levels of Bcl-2 than LNCaP cells. However, LNCaP cells exhibited higher expression of Fas receptor (FasR) but also higher levels of several stress response and antioxidant proteins than PC-3 cells. LNCaP cells also exhibited higher levels of several mitochondrial antioxidant systems, suggesting a compensatory response. Thus, significant differences in redox status and expression of proteins involved in apoptosis and stress response may contribute to PCC aggressiveness.
Although frequent and open parent-child sexual communication (PCSC) has been shown to yield positive effects on adolescent sexual health outcomes, most PCSC is one-sided and parent dominated rather ...than ideal communication characterized by openness. Adolescent children's disclosure of sexual feelings and behaviors to parents can prompt parent-child sexual communication (PCSC) and help parents tailor PCSC to children's needs, increasing the effectiveness of PCSC in promoting positive sexual outcomes. However, very little work has been done exploring correlates of adolescent disclosure about sexuality. Thus, the current study explored the associations between frequent PCSC and child disclosure and secret keeping, and whether open PCSC moderated the relationships. The sample included 603 mother-child dyads from the United States. Adolescent children were 12-17 years old (M = 14.55, SD = 1.68), White (55.6%) and cisgender female (49.8%). A common-fate SEM model revealed that PCSC frequency was positively associated with adolescent disclosure and adolescent secret keeping. However, when PCSC was open, PCSC frequency was associated with a decrease in adolescent secret keeping and an increase in adolescent disclosure. We discuss the importance of encouraging both open and frequent PCSC, as their interaction is related to increased adolescent disclosure and less adolescent secret keeping.
This study used a person-centred approach to consider profiles of parent-child sex communication. We sought to determine whether profiles of sex communication were distinguishable from one another ...based on aspects of both the parent and the child. Participants included 596 US young people (51% female at birth; Mage = 14.55, SD = 1.70, 56% white), and their mother (n = 596; Mage = 41.23, SD = 8.18; 64% white) and father (n = 354; Mage = 43.44, SD = 8.56; 62% white) figures. Analyses suggested three profiles of parent-child sex communication with mothers and four with fathers. Patterns were similar across profiles, with variation being primarily the degree to which parents reported timing, frequency and quality of communication. Parents who more strongly endorsed proactive and reactive parenting, and who reported more frequent and open conversations with their children were warmer and had higher levels of self-efficacy. They also had children who were more sexually active, but who had healthier sexual outcomes. Discussion focuses on the importance of considering multiple aspects of parent-child sex communication and highlights the importance of continuing to empower parents to discuss sex and sexuality with their children.
This paper aims to present the approach to the creation and rendering of a detailed solution for cooling plants including all supporting documents. The paper presents all steps and specific sub steps ...of the presented approach and the approach has been verified on an example of a cooling plant with water-cooled chillers in the pharmaceutical industry. The authors were motivated to use such approach to find an expedient and high-quality solution for the system architecture. They believe that optimal cooling plant system architecture can be provided by an adequate selection of the required equipment and all system parameters it needs to comply with, through two phases of evaluation analysis, including all supporting technical documents. The approach is based on the designers’ engineering knowledge and years of experience in this field. It is based on a so-called Configuration and Change Management approach where the required/acceptable solution is provided by configuring the product and changing product properties through several loopback steps. The approach is quite comprehensive
and complex, however, it produces a satisfactory solution with well presented results, which makes it applicable to the creation of new cooling plants.
Elemental boron has one of the highest volumetric heats of combustion known and is therefore of interest as a high energy density fuel. The fact that boron combustion is inherently a heterogeneous ...process makes rapid efficient combustion difficult. An obvious strategy is to increase the surface area/volume ratio by decreasing the particle size. This approach is limited by the fact that boron forms a ∼0.5 nm thick native oxide layer, which not only inhibits combustion, but also consumes an increasing fraction of the particle mass as the size is decreased. Another strategy might be to coat the boron particles with a material (e.g., catalyst) to enhance combustion of either the boron itself or of a hydrocarbon carrier fuel. We present a simple, scalable, one-step process for generating air-stable boron nanoparticles that are unoxidized, soluble in hydrocarbons, and coated with a combustion catalyst. Ball milling is used to produce ∼50 nm particles, protected against room temperature oxidation by oleic acid functionalization, and optionally coated with catalyst. Scanning and transmission electron microscopy and dynamic light scattering were used to investigate size distributions, with X-ray photoelectron spectroscopy to probe the boron surface chemistry.
The aim of our study was to evaluate associations of elevated preoperative neutrophil-to-lymphocyte ratio (NLR) with testicular germ cell tumors (GCT) characteristics other than cancer specific ...survival (CSS) and progression free survival (PFS).
NLR was recently presented as a widely available and inexpensive marker of poor prognosis in several types of solid tumors. Previous study showed no predictive value of NLR for CSS and PFS in testicular GCT.
Association of high NLR with histological type of tumor, presence of metastatic disease preoperatively and worse than T1 stadium in TNM classification preoperatively was analyzed in 103 patients who underwent radical orchiectomy for testicular GCT.
No statistically significant difference in the prevalence of seminomas and non-seminomas neither in the group with NLR≥4 (p=0.6698) nor in the group with NLR<4 (p=0.9115) was detected. Similarly, no statistically significant difference in the prevalence of metastatic and non-metastatic disease in the group with NLR≥4 (p=0.2008), however statistically significant higher prevalence of non-metastatic disease in the group with NLR<4 (p=0.0001) was found. There was a statistically significant higher number of patients with worse than T1 stadium in patients with NLR≥4 (p=0.0105), but not significant difference in the group with NLR<4 (p=0.0956).
The results of our study showed that NLR lower than 4 predicts non-metastatic disease and NLR higher or equal 4 predicts worse than T1 stadium (Tab. 3, Ref. 12).
Irritable Bowel Syndrome (IBS) and endometriosis are little researched co-morbidities. Symptoms overlap and the lack of a suitable biological marker or a non-invasive diagnostic test, in both cases, ...make diagnosis complex and confusing.1 IBS is a gastrointestinal condition diagnosed by the absence of organic disease and the presence of recurrent abdominal pain and alteration in bowel habit.2

Background: Irritable bowel syndrome (IBS) is a chronic debilitating functional gastrointestinal disorder. Diet and lifestyle changes are important management strategies. The aim of these guidelines is to systematically review key aspects of the dietary management of IBS, with the aim of providing evidence‐based guidelines for use by registered dietitians.
Methods: Questions relating to diet and IBS symptom management were developed by a guideline development group. These included the role of milk and lactose, nonstarch polysaccharides (NSP), fermentable carbohydrates in abdominal bloating, probiotics and empirical or elimination diets. A comprehensive literature search was conducted and relevant studies from January 1985 to November 2009 were identified using the electronic database search engines: Cinahl, Cochrane Library, Embase, Medline, Scopus and Web of Science. Evidence statements, recommendations, good practice points and research recommendations were developed.
Results: Thirty studies were critically appraised. A dietetic care pathway was produced following a logical sequence of treatment and formed the basis of these guidelines. Three lines of dietary management were identified. First line: Clinical and dietary assessment, healthy eating and lifestyle management with some general advice on lactose and NSP. Second line: Advanced dietary interventions to improve symptoms based on NSP, fermentable carbohydrates and probiotics. Third line: Elimination and empirical diets. Research recommendations were also identified relating to the need for adequately powered and well designed randomised controlled trials.
Conclusions: These guidelines provide evidence‐based details of how to achieve the successful dietary management of IBS.
In this study, we used atomic force microscopy (AFM) to quantify the size of surface pore apertures of enamel white spot lesions and then demonstrated the penetration of fluorapatite nanocrystals ...(nFA) into the subsurface of these lesions.
For the porosity study, enamel lesions were created on three sound human teeth using a demineralizing gel for 8 days. The interface between sound enamel and the artificial lesion was analyzed by AFM. To visualize the penetration of nFA tagged with a calcium-binding fluorophore (Fluo-4) into the subsurface of white spot lesions, we used two-photon microscopy. Sixteen extracted human teeth with either active, natural, or in vitro-created carious lesions in enamel were randomly divided into three groups. The teeth were treated for 2 min with either a suspension of tagged nFA crystals, Fluo-4 alone, or deionized water, and left for 30 min before being washed with distilled water and examined microscopically.
A greater concentration of surface pores with larger areas was observed on the in vitro demineralized enamel (29 % of pores greater than 1.0 µm2) when compared with the adjacent sound enamel (8 % of pores greater than 1.0 µm2) (p=0.012, Fisher exact test). In vitro and natural lesions treated with tagged nFA showed fluorescence at depths ranging from 50 to 170 µm, demonstrating penetration of the nFA into the lesion subsurface. The lesions treated with Fluo-4 alone with no crystals showed mostly surface fluorescence (restricted to the outer 25 µm), while those treated with deionized water showed minimal (restricted to the outer 20 µm) to no fluorescence.
We have demonstrated the use of AFM to quantify the surface pore apertures and two-photon microscopy to visualize nFA crystals in the subsurface of non-cavitated enamel lesions.
The restoration of the subsurface of non-cavitated caries lesions is a clinical challenge. This study demonstrated that a 2 min application of nFA could penetrate through the surface apertures of non-cavitated enamel lesions into their subsurface.
