This study attempted to evaluate the transmission dynamics of Trypanosoma cruzi in four indigenous communities of Sierra Nevada de Santa Marta (SNSM), Colombia. Low-stringency single ...primer-polymerase chain reaction (LSSP-PCR) of the minicircles and Southern blot analyses were used to characterize samples from patients, vectors, and reservoirs in these communities. The LSSP-PCR profiles revealed a high genetic variability but with similarities among the parasites present in the samples of vectors, patients, and reservoirs of the same and different communities. Cluster and analysis of molecular variance (AMOVA) analyses of data derived from LSSP-PCR and Southern blot suggest a gene flux among populations of T. cruzi circulating in patients, vectors, and reservoirs. The results support the idea that the domestic and wild transmission cycles overlap in the SNSM, with Rhodnius prolixus as the main vector and Triatoma dimidiata playing an important role in the transmission of Chagas disease in this zone, making the vector control strategy by spraying unsuccessful.

Geochronological databases are powerful tools for characterizing the crustal evolution and the age spectra of a region and allow comparison with other areas at a regional scale. In this contribution, ...we present the Colombian Geochronological Database (CGD), which contains a curated compilation of ca. 67,406 individual published U-Th-Pb, K-Ar, Ar-Ar, Rb-Sr, Sm-Nd, Lu-Hf, Fission-track, U-Th-He, and Re-Os mineral and whole rock ages that are reported in the published literature. Each date includes geographic coordinates, geological setting, petrologic and chemical information extracted from the respective publications. The structure of the database provides a powerful interface for constructing queries and allows searching and extracting information on geographic domains, provinces, stratigraphic units, isotopic systems, date interpretations, references, etc. This information establishes a framework for regional and global geological interpretations with geochronological, stratigraphic, structural and palaeogeographic implications.
With the present effort we present to the geoscience community a clear insight, from a regional perspective, to the geology and tectonics of Colombia since Precambrian times. The comparison of all (detrital and magmatic) single zircon U-Pb dates from the Colombian (Gondwana sourced) geochronological database with the Global and North American (Laurentia sourced) databases provides a temporal constraint on the evolution of the South American continent. U-Pb zircon ages in Colombia define 13 peak clusters centred at 1767, 1530, 1325, 1178, 1007, 605,540, 468, 271, 237, 182, 76 and 10 Ma but of those, only few have a good correlation in all three databases: 1007 (Grenvillian/Orinoquian/Putumayo Orogeny), 605 (Braziliano/Pan-African Orogeny), 468 (Famatinian/Taconic Orogeny), and 182 Ma (Break-up of Pangea) zircon peaks. This correlation suggests that some tectonic events in Colombia are global and might represent crustal production and preservation while the other peaks might just represent local arc magmatic events.
BACKGROUND: Chagas disease is a neglected illness, with limited treatments, caused by the parasite Trypanosoma cruzi. Two drugs are prescribed to treat the disease, nifurtimox and benznidazole, which ...have been previously reported to have limited efficacy and the appearance of resistance by T. cruzi. Acquisition of drug-resistant phenotypes is a complex physiological process based on single or multiple changes of the genes involved, probably in its mechanisms of action. RESULTS: The differential genes expression of a sensitive Trypanosoma cruzi strain and its induced in vitro benznidazole-resistant phenotypes was studied. The stepwise increasing concentration of BZ in the parental strain generated five different resistant populations assessed by the IC₅₀ ranging from 10.49 to 93.7 μM. The resistant populations maintained their phenotype when the BZ was depleted from the culture for many passages. Additionally, the benznidazole-resistant phenotypes presented a cross-resistance to nifurtimox but not to G418 sulfate. On the other hand, four of the five phenotypes resistant to different concentrations of drugs had different expression levels for the 12 genes evaluated by real-time PCR. However, in the most resistant phenotype (TcR5x), the levels of mRNA from these 12 genes and seven more were similar to the parental strain but not for NTR and OYE genes, which were down-regulated and over-expressed, respectively. The number of copies for these two genes was evaluated for the parental strain and the TcR5x phenotype, revealing that the NTR gene had lost a copy in this last phenotype. No changes were found in the enzyme activity of CPR and SOD in the most resistant population. Finally, there was no variability of genetic profiles among all the parasite populations evaluated by performing low-stringency single-specific primer PCR (LSSP-PCR) and random amplified polymorphic DNA RAPD techniques, indicating that no clonal selection or drastic genetic changes had occurred for the exposure to BZ. CONCLUSION: Here, we propose NTR as the major marker of the appearance of resistance to BZ.
COVID-19 pandemic has challenged public health systems worldwide, particularly affecting developing countries in Latin America like Ecuador. In this report, we exposed the fundamental role of the ...Ecuadorian universities to improve COVID-19 surveillance in the country, with an overall contribution over 15% of the total SARS-CoV-2 RT-PCR tests done. We highlight the role of our university during the first semester of the COVID-19 pandemic, contributing to a massive free SARS-CoV-2 testing up to almost 10% of the total diagnosis completed in the country, mainly focus on underserved urban, rural and indigenous communities. Finally, we described our contribution to a high quality and low-cost SARS-CoV-2 RT-PCR diagnostic in Ecuador.
The Covid-19 pandemic has been responsible for millions of deaths worldwide. Patients with comorbidities– such as those on peritoneal dialysis (PD)– present higher morbidity and mortality than the ...general population. We prospectively evaluated all Chilean patients on PD (48 centres) and followed those who had Covid-19 from the beginning of the Covid-19 pandemic in Chile (March 2020) to January 2021 (start of vaccination campaign). We described demographic history, comorbidities, factors related to infection, need for hospitalisation and death due to Covid-19. During the study period, 106 adults on PD were infected by SARS-CoV-2, with a mean age of 53.1 (±16.3) and of which 53.9% were female. From that group, 54.8% required hospitalisation and 24.5% (n = 26) died due to Covid-19. Most of the patients (63.4%) were infected at home and 22.8% during hospitalisation for other reasons. There was a significant association for Covid-19 mortality with: being ≥60 years old, diabetes, time on PD ≥5 years, need for hospitalisation and hospital-acquired infection. At 90 days of follow-up, all deaths associated to Covid-19 occurred before 40 days. We conclude that patients on PD without Covid-19 vaccination have a high mortality and need for hospitalisation associated to Covid-19. To avoid this negative outcome, it is necessary to intensify strategies to avoid contagion, especially in those ≥60 years old, with diabetes and/or ≥5 years spent on PD.
To describe real-life experience with sensor-augmented pump therapy with predictive low-glucose management (SAPT-PLGM), in terms of hypoglycemia and glycemic control after one year of follow-up in ...T1D patients with hypoglycemia as the main indication of therapy.
Retrospective cohort study under real life conditions. Baseline and one-year follow-up variables of glycemic control, hypoglycemia and glycemic variability were compared.
Fifty patients were included, 31 on prior treatment with SAPT with low-glucose suspend (LGS) feature and 19 on multiple dose insulin injections (MDI). Mean HbA1c decreased in the MDI group (8.24%–7.08%; p = 0.0001). HbA1c change was not significant in the SAPT-LGS group. Area under the curve (AUC) below 70 mg/dl improved in both SAPT-LGS and MDI groups while AUC, %time and events below 54 mg/dl decreased in SAPT-LGS group. Glycemic variability improved in the MDI group. Less patients presented severe hypoglycemia with SAPT-PLGM in both groups, however the change was non-significant.
Under real life conditions, SAPT-PLGM reduced metrics of hypoglycemia in patients previously treaded with MDI and SAPT-LGS without deteriorating glycemic control in SAPT-LGS patients, while improving it in patients treated with MDI.

Berteo ex Speng) from Ecuador were studied to determine their phenolic acid profile, nutrient minerals and capacity to protect primary human dermal fibroblasts (HDFa) against ...oxidative-induced damage. Up to five phenolic acids were identified, with homovanillic acid as the main one. Vitamin C,
-carotene and lutein were also determined. Phosphorus and potassium were the main macrominerals, while iron was the principal micromineral. HDFa were preincubated with a crude pechiche extract (PCext) and then subjected to oxidative stress. The activity of five antioxidant enzymes, intracellular reactive oxygen species (ROS) and ATP levels and lipid peroxidation and protein oxidation were used as markers of oxidative damage. Preincubation with PCext for 24 h allowed for the significant reduction of intracellular ROS levels, improved the intracellular ATP levels and protected lipids and proteins against oxidative damage (
0.05). Additionally, preincubation with PCext was also able to significantly (
0.05) improve the activity of the antioxidant enzymes catalase, superoxide dismutase, glutathione peroxidase, glutathione reductase and glutathione transferase, compared to the stressed group without pretreatment. The results obtained in this study suggest the potential of pechiche as a source of bioactive compounds, as well as its beneficial effect against oxidative stress.
Despite using sensor-augmented pump therapy (SAPT) with predictive low-glucose management (PLGM), hypoglycemia is still an issue in patients with type 1 Diabetes (T1D). Our aim was to determine ...factors associated with clinically significant hypoglycemia (<54 mg/dl) in persons with T1D treated with PLGM-SAPT.
ology: This is a multicentric prospective real-life study performed in Colombia, Chile and Spain. Patients with T1D treated with PLGM-SAPT, using sensor ≥70% of time, were included. Data regarding pump and sensor use patterns and carbohydrate intake from 28 consecutive days were collected. A bivariate and multivariate Poisson regression analysis was carried out, to evaluate the association between the number of events of <54 mg/dl with the clinical variables and patterns of sensor and pump use.
188 subjects were included (41 ± 13.8 years-old, 23 ± 12 years disease duration, A1c 7.2% ± 0.9). The median of events <54 mg/dl was four events/patient/month (IQR 1–10), 77% of these events occurred during day time. Multivariate analysis showed that the number of events of hypoglycemia were higher in patients with previous severe hypoglycemia (IRR1.38; 95% CI 1.19–1.61; p < 0.001), high glycemic variability defined as Coefficient of Variation (CV%) > 36% (IRR 2.09; 95%CI 1.79–2.45; p < 0.001) and hypoglycemia unawareness. A protector effect was identified for adequate sensor calibration (IRR 0.77; 95%CI 0.66–0.90; p:0.001), and the use of bolus wizard >60% (IRR 0.74; 95%CI 0.58–0.95; p:0.017).
In spite of using advanced SAPT, clinically significant hypoglycemia is still a non-negligible risk. Only the identification and intervention of modifiable factors could help to prevent and reduce hypoglycemia in clinical practice.
•Despite the use of advance sensor augmented pump therapy (SAPT), hypoglycemia remains a problem in type 1 diabetes.•High glycemic variability, previous severe and unawareness hypoglycemia, were associated with hypoglycemic events.•The intervention of factors such as sensor calibration and the use of bolus wizard could help to prevent hypoglycemia.•This is the first multicenter study that describes variables associated with hypoglycemia in patients treated with SAPT.
