While the photochemistry of duplex DNA has been extensively studied, the photochemistry of nonduplex DNA structures is largely unexplored. Because the structure and stereochemistry of DNA ...photoproducts depend on the secondary structure and conformation of the DNA precursor, they can serve as intrinsic probes of DNA structure. This review focuses on the structures and stereoisomers of pyrimidine dimer photoproducts arising from adjacent and nonadjacent pyrimidines in A, B and denatured DNA, bulge loops, G‐quadruplexes and reverse Hoogsteen hairpins and methods for their detection.
While irradiation of duplex DNA produces adjacent cis,syn cyclobutane pyrimidine dimers (CPDs), irradiation of other structures can produce other isomers, such as the nonadjacent trans,anti CPD formed in G‐quadruplexes. This review investigates the various types of adjacent and nonadjacent dipyrimidine photoproducts and methods for their detection that could be used to identify the presence of various types of unusual structures in DNA.
This paper argues for a novel account of deceitful scientific communication, as “wishful speaking”. This concept is of relevance both to philosophy of science and to discussions of the ethics of ...lying and misleading. Section 1 outlines a case-study of “ghost-managed” research. Section 2 introduces the concept of “wishful speaking” and shows how it relates to other forms of misleading communication. Sections 3–5 consider some complications raised by the example of pharmaceutical research; concerning the ethics of silence; how research strategies—as well as the communication of results—may be misleading; and questions of multiple authorship. The conclusion suggests some more general conclusions.
Are lockdown measures ethically justified? This paper outlines some of the key issues relevant to answering that question, paying particular attention to how decisions are framed. Section 1 argues ...that ethical reasoning about lockdown ought to be guided by a distinction between prudential and ethical reasons, grounded in a concern to respect the separateness of persons, but also that—as public health messaging implies—it can be unclear whether measures are in individuals’ prudential interests or not. Section 2 suggests that a similar set of problems affect attempts to adopt alternative cost-benefit-analysis frameworks for assessing lockdown. Section 3 suggests an answer to these shared problems: we need a process for determining when wellbeing claims and systems of categorization are ethically apt. Section 4 argues that settling the question of aptness is our key ethical task in assessing lockdown.

. Efforts to build response predictors have not incorporated this knowledge. We collected clinical, digital pathology, genomic and transcriptomic profiles of pre-treatment biopsies of breast tumours from 168 patients treated with chemotherapy with or without HER2 (encoded by ERBB2)-targeted therapy before surgery. Pathology end points (complete response or residual disease) at surgery
were then correlated with multi-omic features in these diagnostic biopsies. Here we show that response to treatment is modulated by the pre-treated tumour ecosystem, and its multi-omics landscape can be integrated in predictive models using machine learning. The degree of residual disease following therapy is monotonically associated with pre-therapy features, including tumour mutational and copy number landscapes, tumour proliferation, immune infiltration and T cell dysfunction and exclusion. Combining these features into a multi-omic machine learning model predicted a pathological complete response in an external validation cohort (75 patients) with an area under the curve of 0.87. In conclusion, response to therapy is determined by the baseline characteristics of the totality of the tumour ecosystem captured through data integration and machine learning. This approach could be used to develop predictors for other cancers.
It has traditionally been believed that resistance training can only induce muscle growth when the exercise intensity is greater than 65% of the 1-repetition maximum (RM). However, more recently, the ...use of low-intensity resistance exercise with blood-flow restriction (BFR) has challenged this theory and consistently shown that hypertrophic adaptations can be induced with much lower exercise intensities (<50% 1-RM). Despite the potent hypertrophic effects of BFR resistance training being demonstrated by numerous studies, the underlying mechanisms responsible for such effects are not well defined. Metabolic stress has been suggested to be a primary factor responsible, and this is theorised to activate numerous other mechanisms, all of which are thought to induce muscle growth via autocrine and/or paracrine actions. However, it is noteworthy that some of these mechanisms do not appear to be mediated to any great extent by metabolic stress but rather by mechanical tension (another primary factor of muscle hypertrophy). Given that the level of mechanical tension is typically low with BFR resistance exercise (<50% 1-RM), one may question the magnitude of involvement of these mechanisms aligned to the adaptations reported with BFR resistance training. However, despite the low level of mechanical tension, it is plausible that the effects induced by the primary factors (mechanical tension and metabolic stress) are, in fact, additive, which ultimately contributes to the adaptations seen with BFR resistance training. Exercise-induced mechanical tension and metabolic stress are theorised to signal a number of mechanisms for the induction of muscle growth, including increased fast-twitch fibre recruitment, mechanotransduction, muscle damage, systemic and localised hormone production, cell swelling, and the production of reactive oxygen species and its variants, including nitric oxide and heat shock proteins. However, the relative extent to which these specific mechanisms are induced by the primary factors with BFR resistance exercise, as well as their magnitude of involvement in BFR resistance training-induced muscle hypertrophy, requires further exploration.

It has never been more important to understand how international law enables and constrains international politics. By drawing together the legal theory of Lon Fuller and the insights of ...constructivist international relations scholars, this book articulates a pragmatic view of how international obligation is created and maintained. First, legal norms can only arise in the context of social norms based on shared understandings. Second, internal features of law, or 'criteria of legality', are crucial to law's ability to promote adherence, to inspire 'fidelity'. Third, legal norms are built, maintained or destroyed through a continuing practice of legality. Through case studies of the climate change regime, the anti-torture norm, and the prohibition on the use of force, it is shown that these three elements produce a distinctive legal legitimacy and a sense of commitment among those to whom law is addressed.

Along rivers in Bali, small groups of farmers meet regularly in water temples to manage their irrigation systems. They have done so for a thousand years. Over the centuries, water temple networks ...have expanded to manage the ecology of rice terraces at the scale of whole watersheds. Although each group focuses on its own problems, a global solution nonetheless emerges that optimizes irrigation flows for everyone. Did someone have to design Bali's water temple networks, or could they have emerged from a self-organizing process?
Perfect Order--a groundbreaking work at the nexus of conservation, complexity theory, and anthropology--describes a series of fieldwork projects triggered by this question, ranging from the archaeology of the water temples to their ecological functions and their place in Balinese cosmology. Stephen Lansing shows that the temple networks are fragile, vulnerable to the cross-currents produced by competition among male descent groups. But the feminine rites of water temples mirror the farmers' awareness that when they act in unison, small miracles of order occur regularly, as the jewel-like perfection of the rice terraces produces general prosperity. Much of this is barely visible from within the horizons of Western social theory.
