We develop a new physical model for the broadband spectral energy distribution (SED) of X-ray illuminated accretion discs, that takes into account the mutual interaction of the accretion disc and the ...X-ray corona, including all relativistic effects. We assume a Keplerian, optically thick and geometrically thin accretion disc and an X-ray source in the lamp-post geometry that emits an isotropic power-law spectrum with a high-energy cut-off. We assume that all the energy that would be released by thermal radiation in the standard disc model in its innermost part, is transported to the corona, effectively cooling the disc in this region. We include the disc heating due to thermalisation of the absorbed part of the disc illumination by X-ray corona. The X-ray reflection from the disc is also included. We compute the X-ray luminosity and the low-energy X-ray cut-off through an iterative process, taking full account of the interplay between the X-ray illumination of the disc and the resulting accretion disc spectrum which enters the corona so that the energy balance is preserved. The corona radius is also computed from the conservation of the photon's number during Comptonization. We discuss the model SEDs and their dependence on system parameters. The disc-corona interaction has profound effects - it constrains the X-ray luminosity and changes the shape and normalisation of the UV/optical blue bump. We use the new code to fit the broad-band SED of a typical Seyfert 1 galaxy, NGC 5548. We infer a high black-hole spin, an intermediate system inclination, and an accretion rate below 10% of Eddington. The X-ray luminosity in this source could be supported by 45-70% of the accretion energy dissipated in the disc. The new model, named KYNSED, is publicly available to be used for fitting AGN SEDs inside the XSPEC spectral analysis tool.
Group 3 innate lymphoid cells (ILC3s) sense environmental signals and are critical for tissue integrity in the intestine. Yet, which signals are sensed and what receptors control ILC3 function remain ...poorly understood. Here, we show that ILC3s with a lymphoid-tissue-inducer (LTi) phenotype expressed G-protein-coupled receptor 183 (GPR183) and migrated to its oxysterol ligand 7α,25-hydroxycholesterol (7α,25-OHC). In mice lacking Gpr183 or 7α,25-OHC, ILC3s failed to localize to cryptopatches (CPs) and isolated lymphoid follicles (ILFs). Gpr183 deficiency in ILC3s caused a defect in CP and ILF formation in the colon, but not in the small intestine. Localized oxysterol production by fibroblastic stromal cells provided an essential signal for colonic lymphoid tissue development, and inflammation-induced increased oxysterol production caused colitis through GPR183-mediated cell recruitment. Our findings show that GPR183 promotes lymphoid organ development and indicate that oxysterol-GPR183-dependent positioning within tissues controls ILC3 activity and intestinal homeostasis.

Thermal reverberation in accretion discs of active galactic nuclei is thought to be the reason of the continuum UV/optical time lags seen in these sources. Recently, we studied thermal reverberation ...of a standard Novikov-Thorne accretion disc illuminated by an X-ray point-like source, and we derived an analytic prescription for the time lags as function of wavelength. In this work, we use this analytic function to fit the time-lags spectra of seven Seyferts, that have been intensively monitored, in many wave-bands, in the last few years. We find that thermal reverberation can explain the observed UV/optical time lags in all these sources. Contrary to previous claims, the magnitude of the observed UV/optical time-lags is exactly as expected in the case of a standard accretion disc in the lamp-post geometry, given the black hole mass and the accretion rate estimates for the objects we study. We derive estimates of the disc accretion rates and corona height for a non-spinning and a maximally spinning black hole scenarios. We also find that the modelling of the continuum optical/UV time-lags can be used to estimate the black hole spin, when combined with additional information. We also find that the model under-predicts the observed X-ray to UV time-lags, but this difference is probably due to the broad X-ray auto-correlation function of these sources.
Platelets play a critical role in atherogenesis and thrombosis-mediated myocardial ischemia, processes that are accelerated in diabetes. Whether hyperglycemia promotes platelet production and whether ...enhanced platelet production contributes to enhanced atherothrombosis remains unknown. Here we found that in response to hyperglycemia, neutrophil-derived S100 calcium-binding proteins A8/A9 (S100A8/A9) interact with the receptor for advanced glycation end products (RAGE) on hepatic Kupffer cells, resulting in increased production of IL-6, a pleiotropic cytokine that is implicated in inflammatory thrombocytosis. IL-6 acts on hepatocytes to enhance the production of thrombopoietin, which in turn interacts with its cognate receptor c-MPL on megakaryocytes and bone marrow progenitor cells to promote their expansion and proliferation, resulting in reticulated thrombocytosis. Lowering blood glucose using a sodium-glucose cotransporter 2 inhibitor (dapagliflozin), depleting neutrophils or Kupffer cells, or inhibiting S100A8/A9 binding to RAGE (using paquinimod), all reduced diabetes-induced thrombocytosis. Inhibiting S100A8/A9 also decreased atherogenesis in diabetic mice. Finally, we found that patients with type 2 diabetes have reticulated thrombocytosis that correlates with glycated hemoglobin as well as increased plasma S100A8/A9 levels. These studies provide insights into the mechanisms that regulate platelet production and may aid in the development of strategies to improve on current antiplatelet therapies and to reduce cardiovascular disease risk in diabetes.
Abstract
GRS 1915+105 is a stellar-mass black hole that is well known for exhibiting at least 12 distinct classes of X-ray variability and correlated multi-wavelength behavior. Despite such ...extraordinary variability, GRS 1915+105 remained one of the brightest sources in the X-ray sky. However, in early 2019, the source became much fainter, apparently entering a new accretion state. Here, we report the results of an extensive, year-long monitoring campaign of GRS 1915+105 with the Neil Gehrels Swift Observatory. During this interval, the flux of GRS 1915+105 gradually diminished; the observed count rate eventually dropped by two orders of magnitude. Simple but robust spectral fits to these monitoring observations show that this new state results from the combination of a dramatic and persistent increase in internal obscuration, and a reduced mass accretion rate. The internal obscuration is the dominant effect, with a median value of

. In a number of observations, the source appears to be Compton-thick. We suggest that this state should be identified as the “obscured state,” and discuss the implications of this new (or rarely observed) accretion mode for black holes across the mass scale.
Quasi-periodic eruptions (QPEs) are repeating thermal X-ray bursts associated with accreting massive black holes, the precise underlying physical mechanisms of which are still unclear. We present a ...new candidate QPE source, AT 2019vcb (nicknamed Tormund by the ZTF collaboration), which was found during an archival search for QPEs in the XMM-Newton archive. It was first discovered in 2019 as an optical tidal disruption event (TDE) at \(z=0.088\), and its X-ray follow-up exhibited QPE-like properties. Our goals are to verify its robustness as QPE candidate and to investigate its properties to improve our understanding of QPEs. We performed a detailed study of the X-ray spectral behaviour of this source over the course of the XMM-Newton archival observation. We also report on recent Swift and NICER follow-up observations to constrain the source's current activity and overall lifetime, as well as an optical spectral follow-up. The first two Swift detections and the first half of the 30 ks XMM-Newton exposure of Tormund displayed a decaying thermal emission typical of an X-ray TDE. However, the second half of the exposure showed a dramatic rise in temperature (from 53 to 114 eV) and 0.2-2 keV luminosity (from \(3.2\times10^{42}\) to \(1.2\times10^{44}\) erg s\(^{-1}\)). The late-time NICER follow-up indicates that the source is still X-ray bright more than three years after the initial optical TDE. Although only a rise phase was observed, Tormund's strong similarities with a known QPE source (eRO-QPE1) and the impossibility to simultaneously account for all observational features with alternative interpretations allow us to classify Tormund as a candidate QPE. If confirmed as a QPE, it would further strengthen the observational link between TDEs and QPEs. It is also the first QPE candidate for which an associated optical TDE was directly observed, constraining the formation time of QPEs.
Nitrate contamination still remains one of the main groundwater quality issues in several aquifers worldwide, despite the perduring efforts of the international scientific community to effectively ...tackle this problem. The classical hydrogeological and isotopic investigations are obviously of paramount importance for the characterization of contaminant sources, but are clearly not sufficient for the correct and long-term protection of groundwater resources. This paper aims at demonstrating the effectiveness of the socio-hydrogeological approach as the best tool to tackle groundwater quality issues, while contributing bridging the gap between science and society. An integrated survey, including land use, hydrochemical (physicochemical parameters and major ions) and isotopic (δ15NNO3 and δ18ONO3) analyses, coupled to capacity building and participatory activities was carried out to correctly attribute the nitrate origin in groundwater from the Grombalia Basin (North Tunisia), a region where only synthetic fertilizers have been generally identified as the main source of such pollution. Results demonstrates that the basin is characterized by high nitrate concentrations, often exceeding the statutory limits for drinking water, in both the shallow and deep aquifers, whereas sources are associated to both agricultural and urban activities.
The public participation of local actors proved to be a fundamental element for the development of the hydrogeological investigation, as it permitted to obtain relevant information to support data interpretation, and eventually guaranteed the correct assessment of contaminant sources in the studied area. In addition, such activity, if adequately transferred to regulators, will ensure the effective adoption of management practices based on the research outcomes and tailored on the real needs of the local population, proving the added value to include it in any integrated investigation.

Chronic inflammation is a hallmark of obesity and is linked to the development of numerous diseases. The activation of toll-like receptor 4 (TLR4) by long-chain saturated fatty acids (lcSFAs) is an ...important process in understanding how obesity initiates inflammation. While experimental evidence supports an important role for TLR4 in obesity-induced inflammation in vivo, via a mechanism thought to involve direct binding to and activation of TLR4 by lcSFAs, several lines of evidence argue against lcSFAs being direct TLR4 agonists. Using multiple orthogonal approaches, we herein provide evidence that while loss-of-function models confirm that TLR4 does, indeed, regulate lcSFA-induced inflammation, TLR4 is not a receptor for lcSFAs. Rather, we show that TLR4-dependent priming alters cellular metabolism, gene expression, lipid metabolic pathways, and membrane lipid composition, changes that are necessary for lcSFA-induced inflammation. These results reconcile previous discordant observations and challenge the prevailing view of TLR4's role in initiating obesity-induced inflammation.

Saturated fatty acids are believed to be TLR4 agonists, promoting macrophage activation and obesity-associated inflammation. Lancaster et al. demonstrate that the inflammatory fatty acid palmitate is actually not a TLR4 agonist. TLR4 is, however, required for palmitate-induced inflammation through TLR4-dependent priming and altered cellular metabolism, thus reconciling discordant observations.
The X-ray Integral Field Unit (X-IFU) is the high resolution X-ray spectrometer of ESA's Athena X-ray observatory. It will deliver X-ray data in the 0.2-12 keV band with an unprecedented spectral ...resolution of 2.5 eV up to 7 keV. During the observation of very bright X-ray sources, the X-IFU detectors will receive high photon rates. The count rate capability of the X-IFU will be improved by using the defocusing option, which will enable the observations of extremely bright sources with fluxes up to \(\simeq 1\) Crab. In the defocused mode, the point spread function (PSF) of the telescope will be spread over a large number of pixels. In this case, each pixel receives a small fraction of the overall flux. Due to the energy dependence of the PSF, this mode will generate energy dependent artefacts increasing with count rate if not analysed properly. To account for the degradation of the energy resolution with pulse separation in a pixel, a grading scheme (here four grades) will be defined to affect the proper energy response to each event. This will create selection effects preventing the use of the nominal Auxiliary Response File (ARF) for all events. We present a new method for the reconstruction of the spectra obtained from observations performed with a PSF that varies as a function of energy. We apply our method to the case of the X-IFU spectra obtained during the defocused observations. We use the end-to-end SIXTE simulator to model defocused X-IFU observations. Then we estimate new ARF for each of the grades by calculating the effective area at the level of each pixel. Our method allows us to successfully reconstruct the spectra of bright sources when employed in the defocused mode, without any bias. Finally, we address how various sources of uncertainty related to our knowledge of the PSF as a function of energy affect our results.
Context. Recently, nearby bright galaxies have been the subject of long monitoring surveys in the X-rays, UV, and optical. All of these campaigns revealed a strong correlation between the various UV ...and optical bands, with time lags that increase with wavelength. In a series of papers, we have demonstrated that a scenario in which an X-ray source located on the rotation axis of the central black hole illuminating the accretion disc is a viable explanation for the observed correlations. However, some of the monitored sources showed low or moderate correlation between the X-rays and the UV, which could challenge this scenario. Aims. In this paper, we analyse the time-averaged and the variable broadband X-ray/UV/optical spectral energy distributions (SEDs) of NGC 5548, one of the most intensely monitored Seyfert 1 galaxies, using Swift , HST, and ground-based telescopes. The aim of this paper is to test whether the broadband spectral behaviour of the source could be explained with the X-ray illumination hypothesis, despite the apparently moderate correlation between the X-rays and longer wavelength. Methods. We modelled the broadband time-averaged SED, from the Space Telescope and Optical Reverberation Mapping (STORM) monitoring campaign of the source performed using the KYNSED model, which assumes X-ray illumination of the disc. We also modelled 15 time-resolved SEDs extracted from the same campaign to check whether this model can account for the spectral variability seen in the various wavebands during the monitoring. We assumed in our modelling that the X-ray corona is powered via the accretion process. Results. Our results show that the proposed scenario could describe the time-averaged and the time-resolved SEDs of NGC 5548 perfectly well. In this scenario, the height of the corona, the X-ray photon index, and the power that is transferred to the corona all vary. This would explain the variability behaviour at the various wavelengths. The best-fit model is obtained for a non-spinning black hole accreting at a constant rate of 5% of its Eddington limit. Since each of the variable parameters will affect the observed flux in a particular way, the combined variability of all of these parameters will then account for the moderate correlation between the X-rays and UV/optical. Conclusions. We demonstrate in this paper that X-ray illumination of the accretion disc can actually explain the observed properties of NGC 5548. In fact, this model not only fits the broadband spectra of the source well, but it also explains the time-lag behaviour as a function of wavelength as well as the power spectral distribution, providing a complete description of the behaviour of this source.
