Hybridization has many and varied impacts on the process of speciation. Hybridization may slow or reverse differentiation by allowing gene flow and recombination. It may accelerate speciation via ...adaptive introgression or cause near‐instantaneous speciation by allopolyploidization. It may have multiple effects at different stages and in different spatial contexts within a single speciation event. We offer a perspective on the context and evolutionary significance of hybridization during speciation, highlighting issues of current interest and debate. In secondary contact zones, it is uncertain if barriers to gene flow will be strengthened or broken down due to recombination and gene flow. Theory and empirical evidence suggest the latter is more likely, except within and around strongly selected genomic regions. Hybridization may contribute to speciation through the formation of new hybrid taxa, whereas introgression of a few loci may promote adaptive divergence and so facilitate speciation. Gene regulatory networks, epigenetic effects and the evolution of selfish genetic material in the genome suggest that the Dobzhansky–Muller model of hybrid incompatibilities requires a broader interpretation. Finally, although the incidence of reinforcement remains uncertain, this and other interactions in areas of sympatry may have knock‐on effects on speciation both within and outside regions of hybridization.

The eigenvalues and eigenvectors of the molecular dynamics propagator (or transfer operator) contain the essential information about the molecular thermodynamics and kinetics. This includes the ...stationary distribution, the metastable states, and state-to-state transition rates. Here, we present a variational approach for computing these dominant eigenvalues and eigenvectors. This approach is analogous to the variational approach used for computing stationary states in quantum mechanics. A corresponding method of linear variation is formulated. It is shown that the matrices needed for the linear variation method are correlation matrices that can be estimated from simple MD simulations for a given basis set. The method proposed here is thus to first define a basis set able to capture the relevant conformational transitions, then compute the respective correlation matrices, and then to compute their dominant eigenvalues and eigenvectors, thus obtaining the key ingredients of the slow kinetics.

The gut microbiome plays an important role in immune modulation. Specifically, presence or absence of certain gut bacterial taxa has been associated with better antitumor immune responses. ...Furthermore, in trials using fecal microbiota transplantation (FMT) to treat melanoma patients unresponsive to immune checkpoint inhibitors (ICI), complete responses (CR), partial responses (PR), and durable stable disease (SD) have been observed. However, the underlying mechanism determining which patients will or will not respond and what the optimal FMT composition is, has not been fully elucidated, and a discrepancy in microbial taxa associated with clinical response has been observed between studies. Furthermore, it is unknown whether a change in the microbiome itself, irrespective of its origin, or FMT from ICI responding donors, is required for reversion of ICI-unresponsiveness. To address this, we will transfer microbiota of either ICI responder or nonresponder metastatic melanoma patients via FMT.
In this randomized, double-blinded phase Ib/IIa trial, 24 anti-PD1-refractory patients with advanced stage cutaneous melanoma will receive an FMT from either an ICI responding or nonresponding donor, while continuing anti-PD-1 treatment. Donors will be selected from patients with metastatic melanoma treated with anti-PD-1 therapy. Two patients with a good response (≥ 30% decrease according to RECIST 1.1 within the past 24 months) and two patients with progression (≥ 20% increase according to RECIST 1.1 within the past 3 months) will be selected as ICI responding or nonresponding donors, respectively. The primary endpoint is clinical benefit (SD, PR or CR) at 12 weeks, confirmed on a CT scan at 16 weeks. The secondary endpoint is safety, defined as the occurrence of grade ≥ 3 toxicity. Exploratory endpoints are progression-free survival and changes in the gut microbiome, metabolome, and immune cells.
Transplanting fecal microbiota to restore the patients' perturbed microbiome has proven successful in several indications. However, less is known about the potential role of FMT to improve antitumor immune response. In this trial, we aim to investigate whether administration of FMT can reverse resistance to anti-PD-1 treatment in patients with advanced stage melanoma, and whether the ICI-responsiveness of the feces donor is associated with its effectiveness.

Fast radio bursts (FRBs) are one of the most tantalizing mysteries of the radio sky; their progenitors and origins remain unknown and until now no rapid multiwavelength follow-up of an FRB has been ...possible. New instrumentation has decreased the time between observation and discovery from years to seconds, and enables polarimetry to be performed on FRBs for the first time. We have discovered an FRB (FRB 140514) in real-time on 2014 May 14 at 17:14:11.06 UTC at the Parkes radio telescope and triggered follow-up at other wavelengths within hours of the event. FRB 140514 was found with a dispersion measure (DM) of 562.7(6) cm−3 pc, giving an upper limit on source redshift of z ≲ 0.5. FRB 140514 was found to be 21 ± 7 per cent (3σ) circularly polarized on the leading edge with a 1σ upper limit on linear polarization <10 per cent. We conclude that this polarization is intrinsic to the FRB. If there was any intrinsic linear polarization, as might be expected from coherent emission, then it may have been depolarized by Faraday rotation caused by passing through strong magnetic fields and/or high-density environments. FRB 140514 was discovered during a campaign to re-observe known FRB fields, and lies close to a previous discovery, FRB 110220; based on the difference in DMs of these bursts and time-on-sky arguments, we attribute the proximity to sampling bias and conclude that they are distinct objects. Follow-up conducted by 12 telescopes observing from X-ray to radio wavelengths was unable to identify a variable multiwavelength counterpart, allowing us to rule out models in which FRBs originate from nearby (z < 0.3) supernovae and long duration gamma-ray bursts.
This contribution concerns the effect of the Ag content in wide‐gap AgwCu1‐wIn1‐xGaxSe2 (ACIGS) absorber films and its impact on solar cell performance. First‐principles calculations are conducted, ...predicting trends in absorber band gap energy (Eg) and band structure across the entire compositional range (w and x). It is revealed that a detrimental negative conduction band offset (CBO) with a CdS buffer can be avoided for all possible absorber band gap values (Eg = 1.0–1.8 eV) by adjusting the Ag alloying level. This opens a new path to reduce interface recombination in wide‐gap chalcopyrite solar cells. Indeed, corresponding samples show a clear increase in open‐circuit voltage (VOC) if a positive CBO is created by sufficient Ag addition. A further extension of the beneficial compositional range (positive CBO at buffer/ACIGS interface) is possible when exchanging CdS with Zn1‐ySnyOz, because of its lower electron affinity (χ). Nevertheless, the experimental results strongly suggest that at present, residual interface recombination still limits the performance of solar cells with optimized CBO, which show an efficiency of up to 15.1% for an absorber band gap of Eg = 1.45 eV.
Wide‐gap (Ag,Cu)(In,Ga)Se2 solar cells with varying absorber composition and different buffer layers were processed and characterized. As predicted by the presented first‐principles calculations, a detrimental negative conduction band offset at the interface between the CdS buffer and the absorber can be avoided by adjusting the Ag addition level to balance the Ga concentration. Interface recombination is further reduced when changing to an alternative (Zn,Sn)O buffer layer, thereby paving the way to narrow the efficiency gap between wide‐gap and low‐gap chalcopyrite solar cells.
The safety and immunogenecity of a booster dose of live attenuated varicella-zoster virus (VZV) vaccine was evaluated in 196 healthy subjects, ⩾60 years old, who had already received a VZV vaccine >5 ...years before. This repeat booster dose was well tolerated. Cell-mediated immunity (CMI) to VZV was measured by an interferon-γ (IFN-γ) enzyme-linked immunosorbent spot-forming cell (ELISPOT) assay and a limiting dilution responder cell frequency (RCF) assay. Prevaccination responses decreased as a function of increasing age but were detectable in all subjects by use of the IFN-γ ELISPOT assay. In most subjects, VZV-specific CMI was increased at 6 weeks postvaccination. The magnitude of the vaccine-induced IFN-γ ELISPOT response was inversely related to prevaccination values. Although there was a significant correlation between the IFN-γ ELISPOT and RCF assays, the ELISPOT assay had greater sensitivity and a wider dynamic range. A live attenuated VZV vaccine is safe and immunogenic in an elderly population, and the vaccine-induced immunity may be monitored by the IFN-γ ELISPOT assay
The Centre for Ecology & Hydrology – Gridded Estimates of Areal Rainfall (CEH-GEAR) data set was developed to provide reliable 1 km gridded estimates of daily and monthly rainfall for Great Britain ...(GB) and Northern Ireland (NI) (together with approximately 3500 km2 of catchment in the Republic of Ireland) from 1890 onwards. The data set was primarily required to support hydrological modelling. The rainfall estimates are derived from the Met Office collated historical weather observations for the UK which include a national database of rain gauge observations. The natural neighbour interpolation methodology, including a normalisation step based on average annual rainfall (AAR), was used to generate the daily and monthly rainfall grids. To derive the monthly estimates, rainfall totals from monthly and daily (when complete month available) rain gauges were used in order to obtain maximum information from the rain gauge network. The daily grids were adjusted so that the monthly grids are fully consistent with the daily grids. The CEH-GEAR data set was developed according to the guidance provided by the British Standards Institution. The CEH-GEAR data set contains 1 km grids of daily and monthly rainfall estimates for GB and NI for the period 1890–2012. For each day and month, CEH-GEAR includes a secondary grid of distance to the nearest operational rain gauge. This may be used as an indicator of the quality of the estimates. When this distance is greater than 100 km, the estimates are not calculated due to high uncertainty. We present stacking polarized intensity as a means to study the polarization of sources that are too faint to be detected individually in surveys of polarized radio sources. Stacking offers not only ...high sensitivity to the median signal of a class of radio sources, but also avoids a detection threshold in polarized intensity, and therefore an arbitrary exclusion of sources with a low percentage of polarization. Correction for polarization bias is done through a Monte Carlo analysis and tested on a simulated survey. We show that the nonlinear relation between the real polarized signal and the detected signal requires knowledge of the shape of the distribution of fractional polarization, which we constrain using the ratio of the upper quartile to the lower quartile of the distribution of stacked polarized intensities. Stacking polarized intensity for NRAO VLA Sky Survey (NVSS) sources down to the detection limit in Stokes I, we find a gradual increase in median fractional polarization that is consistent with a trend that was noticed before for bright NVSS sources, but is much more gradual than found by previous deep surveys of radio polarization. Consequently, the polarized radio source counts derived from our stacking experiment predict fewer polarized radio sources for future surveys with the Square Kilometre Array and its pathfinders.
