Objective of the present work was to evaluate the presence of phytochemical constituents and pharmacological activities (antimicrobial and anti-nociceptive) of crude extract isolated from Iris ...albicans and its corresponding fractions. Extraction was made with methanol and extract was evaluated for the presence of different bioactive constituents, as per standard protocols. Extract and its corresponding fractions were evaluated for their antimicrobial and anti-nociceptive potential. Well diffusion method was used to evaluate the antibacterial effects while anti-nociceptive effect was studied using in-vivo models (acetic acid induced writhing, hotplate and tail immersion tests) at different dose level (100, 200 and 300 mg/kg of body weight) and compared with diclofenac sodium (dose = 10 mg/kg body weight). Results showed that the I. albicans extracts contained secondary metabolites including alkaloids, phytosterols, flavonoids, saponins, terpenoids, tannins, phenols, steroids, fixed oil, glycosides and carbohydrates. Furthermore, it was observed that different fractions in decreasing polarity order such as chloroform >n-Hexane > Ethyl acetate > Crude Methanol > Aqueous extract exhibited effective antimicrobial response against all test organisms. Results of the study showed that the extracts have significant antimicrobial and analgesic activity, providing evidence for its folklore use.

The GSTs is one of the most important multifunctional protein families which has been playing a crucial role in the different aspects of plant growth. This extensive study about GSTs may establish a ...solid foundation for the brief functional analysis of BraGSTs in future. In this study, a total of 75 genes were identified in B. rapa. Phylogenetic analysis characterized them into eight different subclasses, while Tau and Phi subclasses were the most numerous. The exon-intron structure and the motif composition of BraGSTs were exhibited accordingly to their subclasses. Notably, we also investigated 15 tandem paralogous pairs of genes, which highlighted that all the pairs were purifying in nature as their synonymous values were lower than 1.00. Duplication analysis indicated that about 45.33% of genes mainly occurred through tandem duplication in B. rapa. Predominately, the tandem cluster of genes in subclass Tau was greater than the other subclasses. Furthermore, among eight multiple hormonal treatments (ABA, GA, BR, ETH, IAA, IBA, NPA, and JA), most number of BraGSTs was activated by NPA, BR, and ABA treatments. This analysis has provided comprehensive information about GSTs family which may assist in elucidating their exact functions in B. rapa.
COVID‐19 has impacted the world in many ways, including loss of lives, economic downturn and social isolation. COVID‐19 was emerged due to the SARS‐CoV‐2 that is highly infectious pandemic. Every ...country tried to control the COVID‐19 spread by imposing different types of lockdowns. Therefore, there is an urgent need to forecast the daily confirmed infected cases and deaths in different types of lockdown to select the most appropriate lockdown strategies to control the intensity of this pandemic and reduce the burden in hospitals. Currently are imposed three types of lockdown (partial, herd, complete) in different countries. In this study, three countries from every type of lockdown were studied by applying time‐series and machine learning models, named as random forests, K‐nearest neighbors, SVM, decision trees (DTs), polynomial regression, Holt winter, ARIMA, and SARIMA to forecast daily confirm infected cases and deaths due to COVID‐19. The models' accuracy and effectiveness were evaluated by error based on three performance criteria. Actually, a single forecasting model could not capture all data sets' trends due to the varying nature of data sets and lockdown types. Three top‐ranked models were used to predict the confirmed infected cases and deaths, the outperformed models were also adopted for the out‐of‐sample prediction and obtained very close results to the actual values of cumulative infected cases and deaths due to COVID‐19. This study has proposed the auspicious models for forecasting and the best lockdown strategy to mitigate the causalities of COVID‐19.
Optimized data set model is proposed to predict 10 thinspace
days ahead of lung infection and death cases due to COVID‐19. The model predicted results very close to the actual values. The best policy to control infection and death cases was the Herd lockdown strategy.
The receptor for advanced glycation end products (RAGE), a pattern recognition receptor signaling event, has been associated with several human illnesses, including neurodegenerative diseases, ...particularly in Alzheimer's disease (AD). Vanillic acid (V.A), a flavoring agent, is a benzoic acid derivative having a broad range of biological activities, including antioxidant, anti-inflammatory, and neuroprotective effects. However, the underlying molecular mechanisms of V.A in exerting neuroprotection are not well investigated. The present study aims to explore the neuroprotective effects of V.A against lipopolysaccharides (LPS)-induced neuroinflammation, amyloidogenesis, synaptic/memory dysfunction, and neurodegeneration in mice brain. Behavioral tests and biochemical and immunofluorescence assays were applied. Our results indicated increased expression of RAGE and its downstream phospho-c-Jun n-terminal kinase (p-JNK) in the LPS-alone treated group, which was significantly reduced in the V.A + LPS co-treated group. We also found that systemic administration of LPS-injection induced glial cells (microglia and astrocytes) activation and significantly increased expression level of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-KB) and secretion of proinflammatory cytokines including tumor necrosis factor alpha (TNF-α), interleukin-1 β (IL1-β), and cyclooxygenase (COX-2). However, V.A + LPS co-treatment significantly inhibited the LPS-induced activation of glial cells and neuroinflammatory mediators. Moreover, we also noted that V.A treatment significantly attenuated LPS-induced increases in the expression of AD markers, such as β-site amyloid precursor protein (APP)-cleaving enzyme 1 (BACE1) and amyloid-β (Aβ). Furthermore, V.A treatment significantly reversed LPS-induced synaptic loss via enhancing the expression level of pre- and post-synaptic markers (PSD-95 and SYP), and improved memory performance in LPS-alone treated group. Taken together; we suggest that neuroprotective effects of V.A against LPS-induced neurotoxicity might be via inhibition of LPS/RAGE mediated JNK signaling pathway; and encourage future studies that V.A would be a potential neuroprotective and neurotherapeutic candidate in various neurological disorders.
The aim of the current study was to explore the underlying neuroprotective mechanisms of curcumin (50 mg/kg, for six weeks) against ethanol (5 mg/kg i.p., for six weeks) induced oxidative stress and ...inflammation-mediated cognitive dysfunction in mice. According to our findings, ethanol triggered reactive oxygen species (ROS), apoptosis, neuroinflammation, and memory impairment, which were significantly inhibited with the administration of curcumin, as assessed by ROS, lipid peroxidation (LPO), and Nrf2/HO-1 (nuclear factor erythroid 2-related factor 2/Heme-oxygenase-1) expression in the experimental mice brains. Moreover, curcumin regulated the expression of the glial cell markers in ethanol-treated mice brains, as analyzed by the relative expression TLR4 (Toll like Receptor 4), RAGE (Receptor for Advanced Glycations End products), GFAP (Glial fibrillary acidic protein), and Iba-1 (Ionized calcium binding adaptor molecule 1), through Western blot and confocal microscopic analysis. Moreover, our results showed that curcumin downregulated the expression of p-JNK (Phospo c-Jun N-Terminal Kinase), p-NF-kB (nuclear factor kappa-light-chain-enhancer of activated B cells), and its downstream targets, as assessed by Western blot and confocal microscopic analysis. Finally, the expression of synaptic proteins and the behavioral results also supported the hypothesis that curcumin may inhibit memory dysfunction and behavioral alterations associated with ethanol intoxication. Altogether, to the best of our knowledge, we believe that curcumin may serve as a potential, promising, and cheaply available neuroprotective compound against ethanol-associated neurodegenerative diseases.
Environmental pollution of microplastics (MPs) is known to be anthropogenically mediated menace to biosphere and becoming a debatable concern globally. Large quantities of plastic fragments are left ...behind after crop cultivation. The leftover plastic debris, gradually degrade into minute fragments with a diameter of less than 5 mm, known as MPs. MPs are responsible for many changes in the soil physicochemical characteristics, including porosity, enzymatic activities, microbial activities, plant growth, and yield. Because of their ubiquitous nature, high specific surface area and strong hydrophobicity, MPs play an important role in the transportation of toxic chemicals such as plasticisers, polycyclic aromatic hydrocarbons (PAHs), antibiotics, and potentially toxic elements (PTEs). MPs may be transported deep into the soil and can pollute underground water. This review paper investigates the deleterious effects of MPs on the soil environment, enzymatic activities, soil microbes, flora, fauna and crop production, and highlights the general concept of MPs contamination as well as its possible environmental consequences. The review also converses some of the key areas for future research and for key stakeholders concerned with policymaking

Cytokines play an important role in SARS-CoV-2 infection progression and severity. A number of inflammatory cytokines have been directly associated with disease severity including IL-6 ...(interleukin-6), IL-10, TNF-α (tumor necrosis factor alpha), IFN-γ (interferon-gamma). Here, in this study, the aim was to better understand the interplay between host immune response mediated by cytokines and severity of SARS-CoV-2 infection by assessing cytokine expression. Therefore, we measured expression levels of a total of 12 genes (IFNA-1, IFN-γ, IL-1α, IL-1β, IL-4, IL-6, IL-7, IL-10, IL-11, IL-13, IL-15, and IL-27) encoding inflammatory, anti-inflammatory and regulatory cytokines using QRT-PCR in hospitalized patients with severe infection compared to mildly infected. IFN-γ was identified as a potent marker of disease severity as indicated previously. Moreover, levels of IL-7 were also found to be partially reduced in patients compared to the healthy controls and linked negatively to disease severity. Identification of these cytokines may be helpful in not only understanding disease pathogenesis but also in better management of the patients after covid infection.

Multiple, complex molecular events characterize cancer development and progression. Deciphering the molecular networks that distinguish organ-confined disease from metastatic disease may lead to the ...identification of critical biomarkers for cancer invasion and disease aggressiveness. Although gene and protein expression have been extensively profiled in human tumours, little is known about the global metabolomic alterations that characterize neoplastic progression. Using a combination of high-throughput liquid-and-gas-chromatography-based mass spectrometry, we profiled more than 1,126 metabolites across 262 clinical samples related to prostate cancer (42 tissues and 110 each of urine and plasma). These unbiased metabolomic profiles were able to distinguish benign prostate, clinically localized prostate cancer and metastatic disease. Sarcosine, an N-methyl derivative of the amino acid glycine, was identified as a differential metabolite that was highly increased during prostate cancer progression to metastasis and can be detected non-invasively in urine. Sarcosine levels were also increased in invasive prostate cancer cell lines relative to benign prostate epithelial cells. Knockdown of glycine-N-methyl transferase, the enzyme that generates sarcosine from glycine, attenuated prostate cancer invasion. Addition of exogenous sarcosine or knockdown of the enzyme that leads to sarcosine degradation, sarcosine dehydrogenase, induced an invasive phenotype in benign prostate epithelial cells. Androgen receptor and the ERG gene fusion product coordinately regulate components of the sarcosine pathway. Here, by profiling the metabolomic alterations of prostate cancer progression, we reveal sarcosine as a potentially important metabolic intermediary of cancer cell invasion and aggressivity.

Background Primary percutaneous coronary intervention (PPCI) is the recommended treatment in ST elevated myocardial infarction (STEMI). The determination of Quality of life (QoL) for various options ...of coronary revascularization is important for establishment of a comprehensive care plan. Studies of QoL in interventional cardiology are scarce. Our study has compared utility scores and quality adjusted life year (QALY) of 2.sup.nd and 3.sup.rd generation drug eluting stents (DES). Methods An observational cohort study was conducted to evaluate QoL and QALY using EQ-5D-5L questionnaire. Patients undergoing PPCI between July-Dec 2019 were evaluated after completion of one year of procedure. Results Total 334 patients were evaluated, study population consisted of a greater number of males (87.13%) than females. Mean utility value was more in 3.sup.rd G Biomatrix stents; 0.829 ± 0.11 than 2.sup.nd G Xience stents; 0.794 ± 0.11 (p < 0.05). Visual analogue scale (VAS) value was also high in 3.sup.rd G DES (81.84 ± 8.29) as compared to 2.sup.nd G DES (77.81 ± 9.01); p< 0.05. A significant association was found between utility scores/VAS and age, DM, HTN, Current smoking, family history and CAD diagnosis. There was a gain of 0.035 QALY with the use of Biomatrix DES. Conclusion Health related quality of life (HRQOL) is a leading support in the decision making of therapeutic interventions. Our study has found that Biodegradable polymer (BP) Biomatrix DES are superior to the Durable polymer (DP) Xience DES having better QoL and QALY.
