Cerebral organoids are 3D stem cell-derived models that can be utilized to study the human brain. The current consensus is that cerebral organoids consist of cells derived from the neuroectodermal ...lineage. This limits their value and applicability, as mesodermal-derived microglia are important players in neural development and disease. Remarkably, here we show that microglia can innately develop within a cerebral organoid model and display their characteristic ramified morphology. The transcriptome and response to inflammatory stimulation of these organoid-grown microglia closely mimic the transcriptome and response of adult microglia acutely isolated from post mortem human brain tissue. In addition, organoid-grown microglia mediate phagocytosis and synaptic material is detected inside them. In all, our study characterizes a microglia-containing organoid model that represents a valuable tool for studying the interplay between microglia, macroglia, and neurons in human brain development and disease.
Objective
We investigated the pathogenicity of immunoglobulin M (IgM) anti‐GM1 antibodies in serum from patients with multifocal motor neuropathy (MMN) using human induced pluripotent stem cell ...(iPSC)‐derived motor neurons (MNs).
Methods
iPSCs were generated from fibroblasts and differentiated into MNs. We studied the binding of IgM to MNs, their complement‐activating properties, and effects on structural integrity using fluorescence and electron microscopy. Live cell imaging was used to study effects of antibody binding on MNs in the presence and absence of complement.
Results
IgM antibody binding to MNs was detected using sera from MMN patients with and without detectable anti‐GM1 IgM antibody titers in enzyme‐linked immunosorbent assay, but not with sera from (disease) controls. Competition and depletion experiments showed that antibodies specifically bound to GM1 on iPSC‐derived MNs. Binding of these antibodies disrupted calcium homeostasis by both complement‐dependent and complement‐independent pathways. MNs showed marked axonal damage after complement activation, and reduced antibody pathogenicity following treatment with immunoglobulin preparations.
Interpretation
Our data provide evidence for the pathogenicity of anti‐GM1 IgM antibodies in MMN patients and link their presence to the clinical characteristics of axonal damage and immunoglobulin responsiveness. This iPSC‐derived disease model will facilitate diagnosis, studies on autoantibody pathogenicity, drug development, and screening in immune‐mediated neuropathies. Ann Neurol 2016;80:71–88
Spinal muscular atrophy (SMA) is an autosomal recessive neurodegenerative disorder characterized by loss of motor neurons (MN) in the spinal cord leading to progressive muscle atrophy and weakness. ...SMA is caused by mutations in the survival motor neuron 1 (SMN1) gene, resulting in reduced levels of survival motor neuron (SMN) protein. The mechanisms that link SMN deficiency to selective motor neuron dysfunction in SMA remain largely unknown. We present here, for the first time, a comprehensive quantitative TMT-10plex proteomics analysis that covers the development of induced pluripotent stem cell-derived MNs from both healthy individuals and SMA patients. We show that the proteomes of SMA samples segregate from controls already at early stages of neuronal differentiation. The altered proteomic signature in SMA MNs is associated with mRNA splicing, ribonucleoprotein biogenesis, organelle organization, cellular biogenesis, and metabolic processes. We highlight several known SMN-binding partners and evaluate their expression changes during MN differentiation. In addition, we compared our study to human and mouse in vivo proteomic studies revealing distinct and similar signatures. Altogether, our work provides a comprehensive resource of molecular events during early stages of MN differentiation, containing potentially therapeutically interesting protein expression profiles for SMA.

Resultaten visade att de mest förekommande aktiviteterna som de gör är spela spel, se på film, lyssna på musik, kommunicera och informationssökning. Varför de besöker Internet är till största delen för att det är underhållande och för att det kan råda bot på tristess, samt även som komplement till andra aktiviteter. Studien kan också ur ett större perspektiv ses som ett exempel på att den generation som växer upp idag ger liv åt en ny kultur i samhället.
Den psykosociala arbetsmiljön är en viktig del i en hållbar och hälsosam organisation och handlar bland annat om att skapa balans i förhållanden mellan krav och resurser. Det är också av stor vikt ...att medarbetarna, oavsett roll i organisationen, känner delaktighet och meningsfullhet samt att det finns möjlighet till personlig utveckling. Syftet med studien var att undersöka hur kommunala vård- och omsorgschefer ser på sina förutsättningar när det kommer till att skapa en god psykosocial arbetsmiljö. Detta syfte uppnås bäst genom en semistrukturerad intervjustudie som sedan analyseras utifrån en kvalitativ innehållsanalys. Genom denna analys kom vi fram till fyra huvudkategorier; utbildningsnivå enhetschef, skapa förutsättningar, enhetschefers närvaro samt schemamässiga förutsättningar, planering och struktur. Utifrån kategorierna framkom att det är viktigt för respondenterna att få adekvat utbildning för att på bästa sätt kunna arbeta med den psykosociala arbetsmiljön. Det är också viktigt för respondenterna att det finns ett väl etablerat kontaktnät till stödfunktioner runt verksamheten att vända sig till vid behov. Att vara lyhörd i sin kommunikation med medarbetarna anses viktigt men också att få medarbetarna att trivas och känna glädje i sitt arbete och på sin arbetsplats. Genom att få medarbetarna att arbeta på samma sätt samt se till att arbetsgruppen har en god kompetensnivå, vilket i sin tur fördelar arbetsbördan väl, skapas förutsättningar för en god psykosocial arbetsmiljö. En närvarande chef är viktigt för alla medarbetare och största utmaningen här är att tillgodose de medarbetare som tjänstgör nattetid. En del i det förebyggande arbetsmiljöarbetet är att upptäcka riskfaktorer i tid samt att skapa mötesforum där den psykosociala arbetsmiljön arbetas med.
The purpose of this study is to increase the understanding for how home care services interpret and handle situations with and around caretakers alcohol use. Interviews have been conducted with two ...managers and six basic staff in two different home care service-groups. The interviews were individual with the managers and in group form with the staff. The interviews have been analyzed using Hasenfeld’s theory on Human Service Organizations and moral practice together with Beronius’ interpretation of disciplinary exercise of power. Our result showed that the handling of caretakers alcohol use differed both within and between the home care service-groups and there was a lack of guidelines on how to deal with this. Consequently it seemed to be a lot of individual assessments carried out by the staff, which sometimes led to a moral practice. Another important result was the fact that the interviewees quickly became very fixed on abuse and addiction, even though the interviews had focus on alcohol use in general. It is obvious that when a substance abuse problem exists, difficulties arise in home care services work.

In this study the authors compare the patterns of referral to alcoholism treatment from two neighborhoods adjacent to a public mental health facility. The areas are alike in terms of socioeconomic ...status, but they differ in their racial composition and social organization. They hypothesized that referrals to alcoholism treatment originate from self, family, and friends in the organized neighborhood and from police and social service agencies in the unorganized area. Results support the following conclusions: 1) self, family, and friends account for the majority of referrals to alcoholism treatment from both an organized neighborhood and an unorganized area; 2) race and sex explain more variation in referral to alcoholism treatment than does neighborhood social organization; and 3) white female patients are more likely to be institutionally referred than any other group.
