An effective colonization of the host plant tissue by the necrotrophic fungus Sclerotinia sclerotiorum requires the secretion of the non-host specific toxin oxalic acid (OA), which is known to ...suppress the generation of reactive oxygen intermediates (ROI). A full-length cDNA coding for an oxalate decarboxylase (TOXDC), which converts OA into CO₂ and formate, was isolated from the basidiomycete Trametes versicolor. It was overexpressed in tobacco plants to study the role of ROI and OA in the interaction between tobacco and S. sclerotiorum. The transgenic plants contained less OA and showed a delayed colonization of S. sclerotiorum; furthermore a strong ROI accumulation and nearly no catalase activity compared to the wild type (WT) plants could be detected. In addition, inoculation experiments with transgenic catalase-deficient plants (CAT1AS) and in vitro studies showed that S. sclerotiorum copes with strong ROI stress. Our results indicate that OA supports the infection process caused by S. sclerotiorum and the fungus itself is able to tolerate high ROI concentrations.
Rhizopus rot, caused by the fungus Rhizopus oryzae (Mucorales, Zygomycota) is the most economically important post-harvest disease of flue-cured tobacco and is also known to cause diseases on ...vegetables and fruits. In case of high infection rates, losses in can reach up to 100% during curing of tobacco leaves. The primary sites of entry of this pathogen are wounds on leaves which are inflicted during harvest and handling. Several compounds were first tested in vitro to determine their antifungal capacities, then applied to racks filled with freshly harvested leaves to prevent colonization by R. oryzae during the subsequent curing process. Calcium propionate, hydrogen peroxide, and azoxystrobin reduced mycelial growth, spore production, and germination of R. oryzae in in vitro tests, whereas calcium chloride mainly influenced the production of fungal biomass. As calcium propionate was phytotoxic when applied to leaves, only calcium chloride, hydrogen peroxide, and azoxystrobin were selected for rack treatments. In addition, fluorescent pseudomonads were tested against natural infections of leaves by the fungus. Hydrogen peroxide had no effect on post-harvest decay. However, disease severity in flue-cured tobacco was significantly reduced by calcium chloride and fluorescent pseudomonads used alone or in combination, and nearly repressed by azoxystrobin. Therefore, an application of antagonistic bacteria alone or in combination with calcium chloride is a valuable alternative in pest management of flue-cured tobacco where residual fungicides have to be avoided.

A field experiment was conducted in order to analyse the effects of the herbicide Basta with glufosinate-ammonium as the active ingredient (that inhibits specifically the enzyme glutamine-synthetase) ...on the grapevine plant and the infestation with Plasmopara viticola, the downy mildew pathogen. The herbicide was applied at sublethal concentrations and the toxic effects on leaves and berries based on different quality parameters were estimated. Low concentrations of glufosinate-ammonium (< 0,625 mM) did not affect the chlorophyll and amino acid concentrations in leaves. This was also the case for the content of salicylic acid and phenolic compounds. Furthermore, must parameters such as sugars (glucose and fructose), acids (tartaric acid, malic acid, total acids and volatile acids), and alcohol (ethanol and glycerine) also remained unchanged compared to the fungicide-treated or untreated control, whereas the concentrations of nitrogenous components metabolically available to yeasts increased with increasing concentrations of glufosinate- ammonium. The amount of berries harvested at the end of the experiment depended on the glufosinate-ammonium concentration applied. The harvest was highest when glufosinate-ammonium was applied as a 0.1 mM solution and differed not significantly from the fungicide-treated control, but decreased with increasing herbicide doses. However, the infestation of leaves with P. viticola was significantly reduced by the application of the herbicide, even though not each infection was eradicated due to the non- systemic mode of action of the herbicide. The particular influence of the herbicide onto this host-parasite interaction still remains unclear. Vitis riparia
selection ‘Gloire de Montpellier’ and susceptible cv. ‘Riesling’. Controls were treated with sterile water. Messenger RNAs extracted 12 h post infection were subjected to differential display. Seven transcripts appeared specifically induced during the incompatible interaction. Sequencing showed that they build three classes. One of them, named

in vitro-plants confirm that enhanced disease resistance is associated with an expression of distinct reactions in a chronological order: An increased production of reactive oxygen species (superoxide radicals, 4-6 hours post infection, hpi) was followed by a hypersensitive response (6-8 hpi), an increased activity of peroxidase in cells flanking the infection area and in the vascular tissue (10-12 hpi) and an increased production, accumulation or conversion of phenolic compounds (12-15 hpi). These mechanisms seem also to be present in susceptible varieties as shown after an inoculation with non-host oomycetic pathogens on the basis of peroxidase activity, but they do not become activated after
P. viticola infection.
The investigation of the peroxidase activity in leaves at several time points after an infection with
P. viticola indicated that there is a strong correlation between the POX activity in leaves of

L.) nach Niederschlägen verursacht Verluste und Qualitätseinbußen im Weinbau. Häufig ist dies in der Spätphase der ...Fruchtentwicklung mit einem Befall der Trauben mit Fäulnispilzen verbunden. In der vorliegenden Studie wird die Rolle des Grauschimmels (
Botrytis cinerea
Pers.:Fr.), der als dominierender Fäulniserreger auftritt, beim Platzen von Beeren der Sorte ‘Riesling’ untersucht. Hierzu wurden Weinbeeren mit charakteristischen Befallssymptomen (braun-violette Verfärbung der Fruchthaut) des Pilzes nach natürlicher Infektion am Standort ausgewählt. Nach Niederschlag platzten diese ausschließlich in einem Bereich, in dem Befallssymptome vorlagen. Die Risse waren mehrheitlich, d. h. bei 55 % der Früchte, quer zur Fruchtachse zwischen Stiel- und Griffelansatz orientiert. Auf der Fruchthaut von befallenen Weinbeeren traten gehäuft in der Griffelregion und z. T. an Lentizellen mikroskopische Risse auf, die als Mikrorisse bezeichnet werden. Diese Risse zeigten nach Inkubation mit Acridinorange Infiltrationshöfe unterschiedlicher Größe. Die infiltrierte Fläche korrelierte positiv mit der Risslänge (r
2
=#x003D; 0,43*). Die Wasseraufnahme von abgeschnittenen Beeren stieg mit der Zeit an. Die Wasseraufnahmerate variierte stark, war für infizierte Beeren höher als für solche ohne Symptome und korrelierte bei infizierten Beeren mit der kumulativen Risslänge pro Beere (Produkt aus Anzahl der Risse pro Beere und mittlerer Risslänge; r

=#x003D; 0,45*). Durch Anwesenheit von Wasser auf der gespannten Fruchthaut wurden Mikrorisse induziert, deren Zahl in befallenen Beeren gegenüber solchen ohne Symptome um 55 % anstieg. Die Ergebnisse belegen, dass
Botrytis
-Befall die Platzempfindlichkeit von Weinbeeren der Sorte Riesling erhöht, insbesondere durch Schwächung der Fruchthaut, einen Anstieg der Risslänge pro Beere, wodurch sich eine erhöhte Wasseraufnahme ergibt.
