Carbon Criminals, Climate Crimes analyzes the looming threats posed by climate change from a criminological perspective. It advances the field of green criminology through a examination of the ...criminal nature of catastrophic environmental harms resulting from the release of greenhouse gases. The book describes and explains what corporations in the fossil fuel industry, the U.S. government, and the international political community did, or failed to do, in relation to global warming. Carbon Criminals, Climate Crimes integrates research and theory from a wide variety of disciplines, to analyze four specific state-corporate climate crimes: continued extraction of fossil fuels and rising carbon emissions; political omission (failure) related to the mitigation of these emissions; socially organized climate change denial; and climate crimes of empire, which include militaristic forms of adaptation to climate disruption. The final chapter reviews policies that could mitigate greenhouse gas emissions, adapt to a warming world, and achieve climate justice.
Some recombinant adeno-associated viruses (rAAVs) can cross the neonatal blood–brain barrier (BBB) and efficiently transduce cells of the central nervous system (CNS). However, in the adult CNS, ...transduction levels by systemically delivered rAAVs are significantly reduced, limiting their potential for CNS gene therapy. Here, we characterized 12 different rAAVEGFPs in the adult mouse CNS following intravenous delivery. We show that the capability of crossing the adult BBB and achieving widespread CNS transduction is a common character of AAV serotypes tested. Of note, rAAVrh.8 is the leading vector for robust global transduction of glial and neuronal cell types in regions of clinical importance such as cortex, caudate-putamen, hippocampus, corpus callosum, and substantia nigra. It also displays reduced peripheral tissue tropism compared to other leading vectors. Additionally, we evaluated rAAVrh.10 with and without microRNA (miRNA)-regulated expressional detargeting from peripheral tissues for systemic gene delivery to the CNS in marmosets. Our results indicate that rAAVrh.8, along with rh.10 and 9, hold the best promise for developing novel therapeutic strategies to treat neurological diseases in the adult patient population. Additionally, systemically delivered rAAVrh.10 can transduce the CNS efficiently, and its transgene expression can be limited in the periphery by endogenous miRNAs in adult marmosets.
State crimes are, by far, the most destructive of all crimes. The use and threat to use nuclear weapons, the aerial bombardment of civilians, wars of aggression, torture, the failure to mitigate ...global warming and adapt to climate change ecocide, along with myriad other state-corporate crimes, fill the world with death and devastation, misery and want. This article argues that criminologists have a responsibility to act as public criminologists by speaking in the “prophetic voice” concerning these crimes and their victims, and then acting in the political arena in an attempt to control and prevent these harms. The paper briefly describes three approaches to engaging in what Belknap (Criminology 53:1–23,
2015
) calls “criminology activism” on these issues. The first approach is for criminologists to counter the cultures of denial and normalization that usually cover state crimes. The second involves contesting the global corporate capitalist system and the power of the American capitalist state in an effort to achieve specific progressive policy reforms and structural changes in the global political economy. Finally, criminologists can work to enhance the democratization of the international political community and strengthen the ability of specific international legal institutions to control state crimes.
The Donald Trump administration has engaged in a number of crimes related to climate change. This article examines these climate crimes, in particular, the administration’s organized denial of global ...warming and its political omissions concerning the mitigation of greenhouse gas emissions that result in the rollback of existing regulatory policies related to the climate crisis. This criminality is explored through the lens of the concept of state–corporate crime, a concept utilized by a number of green criminologists to analyze environmental harms. The Trump administration’s rollback of climate change regulations is first located within its historical, political, and social contexts. Then, the specific actions and political omissions that constitute these rollbacks are described and analyzed as state–corporate environmental crimes.
Climate change caused by anthropogenic global warming is the most serious social problem, most important political issue, and greatest moral challenge the world community faces today. This article ...contends that the political failure of the US government to act to mitigate greenhouse gas emissions, and the socially organized denial of climate change that shapes that failure, can be conceptualized as state-corporate crimes. After a brief description of the nature and effects of climate change and a consideration of how these harms might be conceptualized as a criminological topic, a political economy theoretical analysis of the failure to reduce carbon emissions and the denial of climate change that impacts that failure is presented.
An expanded hexanucleotide repeat in C9orf72 causes amyotrophic lateral sclerosis and frontotemporal dementia (c9FTD/ALS). Therapeutics are being developed to target RNAs containing the expanded ...repeat sequence (GGGGCC); however, this approach is complicated by the presence of antisense strand transcription of expanded GGCCCC repeats. We found that targeting the transcription elongation factor Spt4 selectively decreased production of both sense and antisense expanded transcripts, as well as their translated dipeptide repeat (DPR) products, and also mitigated degeneration in animal models. Knockdown of SUPT4H1, the human Spt4 ortholog, similarly decreased production of sense and antisense RNA foci, as well as DPR proteins, in patient cells. Therapeutic targeting of a single factor to eliminate c9FTD/ALS pathological features offers advantages over approaches that require targeting sense and antisense repeats separately.
Abstract Many elderly individuals remain dementia-free throughout their life. However, some of these individuals exhibit Alzheimer disease neuropathology on autopsy, evidenced by neurofibrillary ...tangles (NFTs) in AD-specific brain regions. We conducted a genome-wide association study to identify genetic mechanisms that distinguish non-demented elderly with a heavy NFT burden from those with a low NFT burden. The study included 299 non-demented subjects with autopsy (185 subjects with low and 114 with high NFT levels). Both a genotype test, using logistic regression, and an allele test provided consistent evidence that variants in the RELN gene are associated with neuropathology in the context of cognitive health. Immunohistochemical data for reelin expression in AD-related brain regions added support for these findings. Reelin signaling pathways modulate phosphorylation of tau, the major component of NFTs, either directly or through β-amyloid pathways that influence tau phosphorylation. Our findings suggest that up-regulation of reelin may be a compensatory response to tau-related or beta-amyloid stress associated with AD even prior to the onset of dementia.
Following the trail blazed by Bill Chambliss in his 1988 Presidential Address to the American Society of Criminology, this article engages two interrelated issues concerning the concept of ...state-organized crime that he pioneered. First, the article develops Chambliss’s argument that criminologists should define state crime as behavior that violates international agreements and principles established in the courts and treaties of international bodies. Second, although Chambliss effectively argued that international law “on the books” provides a framework of substantive concepts and categories that allows criminologists to define certain state actions as a form of crime, “in action” international laws fail to provide legal accountability for states and protection for victims. This article demonstrates, however, that Chambliss’s structural contradictions theory of law can help to explain this paradox.
Background
Caregiving for persons with dementia negatively impacts physical and mental health, including increased risk for chronic loneliness and isolation. This Phase I study aimed to recruit ...former and current dementia caregivers as engaged partners in the design of a unique, technology‐assisted pairing platform to match caregivers to one another for meaningful connection and support based on characteristics chosen by the caregivers themselves.
Method
We recruited thirty‐two caregivers to participate in two semi‐structured focus groups (audio recorded for qualitative analyses) and complete a survey to understand the characteristics desired in another caregiver for the purpose of emotional support. Participants interacted with a website prototype on their personal device and completed a user experience feedback form. An iterative process was used to design the website and matching questionnaire allowing participants to validate preferences and concerns. Website analytics were captured (e.g. time to answer each question, total time spent on the website).
Result
The efforts culminated in a website ‘look and feel’ that addressed the preferences and criticisms of study participants. Figure 1 shows the home page slider inviting caregivers to join (images 1‐3) and describes the matching process (4); the caregiver answers up to 34 questions to create a profile (5‐7) and the website provides a list of caregivers that best match criteria (8). Overall, 87% of study participants rated a high level of comfort with completing the personal profile questions; 82% felt confident the questions asked would lead to a good match. An algorithm to match caregivers was created using the profile questions which asked participants to self‐select and then rank the characteristics of highest importance. Good and poor matches are scored by the algorithm, with 1 being a “perfect” match (Figure 2). In the current cohort, each participant had at least one match that scored 0.708 or higher.
Conclusion
Our study established interest in a technology assisted peer‐to‐peer caregiver matching program that is a potential low‐cost, high‐impact resource. The study resulted in a set of matching characteristics and an algorithm prototype within a user influenced website and will be further tested in Phase 2. Research supported from NIA of the NIH.
Background
Black/African Americans (B/AA) are nearly twice as likely as White/European Americans (W/EA) to have Alzheimer’s or other dementias and are more likely to provide care for persons with ...dementia (PwD). Evidence indicates that B/AA desire having someone to talk with who understands their caregiving challenges, yet are less likely than W/EA to ask for support. Further, community resources supporting caregivers’ ability to maintain health and well‐being developed with input from B/AA are lacking. We aimed to determine how a technology‐assisted caregiver matching service designed with user input could address diverse caregiver needs.
Methods
For this Phase I project, 32 caregivers of PwD (31% B/AA) participated in semi‐structured focus groups and completed surveys to provide input regarding characteristics deemed valuable in someone they would choose to be connected with for emotional support. Through an iterative process as new information emerged, a web‐based prototype and matching algorithm were developed.
Results
Age and sex distribution were similar between B/AA and W/EA (71 and 68 yrs; 90% and 86%, respectively). W/EA were most likely caring for a spouse/significant other (73%) whereas B/AA were caring for spouse/significant other (30%), a parent (30%;18% W/EA), a sibling (10%; 0% W/EA), or other (30%; 9%W/EA). Level of emotional support was rated “poor” (50% B/AA, 18% W/EA); “average” (0% B/AA, 14% W/EA); “good” (40% B/AA, 41% W/EA); and “excellent” (10%, 27% W/EA). Characteristics B/AA deemed “very important” or “absolutely essential” for a supportive match included living in close proximity, same ethnicity, similar lifestyle, and common values. Alternatively, W/EA selected similar age, living in close proximity, same sex, similar lifestyle, common values and interests, having the same caregiving relationship, and having the same diagnosis in their PwD (see chart).
Conclusion
In this cohort, B/AA reported poorer levels of emotional support than W/EA. B/AA were equally likely to be supporting a spouse/significant other as a parent or other acquaintance, whereas W/EA provided support primarily for a spouse/significant other. Characteristics B/AA and W/EA consider important in a supportive caregiver match show important differences that should be considered when optimizing caregiving resources. Phase II will explore further in a much larger sample.
