This work examines the mapping of logic simulation onto massively parallel computer architectures. We discuss alternative communication primitives for a massively parallel instruction set ...architecture and the impact of the choice of communication primitives on logic simulation. We have developed compilation tools to automatically map the simulation of an MOS transistor circuit onto a massively parallel computer. We analyze the efficiency of this mapping as a function of the available communication primitives. The compilation process is illustrated by describing our pilot implementation on a 32k processor Connection Machine.
According to Kant’s dictum, morality leads inescapably to religion. Notably, this implies two unavoidable shifts: From ‘morality’ to the ‘religion of reason’ and from the ‘religion of reason’ to ...‘positive religions’ (‘revelation’). I explain the grounds for each shift, focusing on the different kinds of necessity involved. I then analyze Fichte’s
(1792), which mainly addresses the second shift, discussed only briefly by Kant. As I show, whereas for Kant revelation is conditioned by a prior free determination of the will, for Fichte it obtains a primordial status. I explain how Fichte’s approach fills in the Kantian gap between the concept of God, which seems to exclude the possibility of revelation, and the unavoidable human need to assume such an occurrence.
At the restriction point (R), mammalian cells irreversibly commit to divide. R has been viewed as a point in G1 that is passed when growth factor signaling initiates a positive feedback loop of Cdk ...activity. However, recent studies have cast doubt on this model by claiming R occurs prior to positive feedback activation in G1 or even before completion of the previous cell cycle. Here we reconcile these results and show that whereas many commonly used cell lines do not exhibit a G1 R, primary fibroblasts have a G1 R that is defined by a precise Cdk activity threshold and the activation of cell-cycle-dependent transcription. A simple threshold model, based solely on Cdk activity, predicted with more than 95% accuracy whether individual cells had passed R. That a single measurement accurately predicted cell fate shows that the state of complex regulatory networks can be assessed using a few critical protein activities.

Schwarz et al. examine the responses of primary cells expressing a live-cell Cdk activity sensor to growth factor removal. They find that Cdk activity at the time of growth factor removal is an accurate predictor of whether a cell subsequently divides.
The term ‘miracle’ generally refers to events that are not explicable by natural causes alone. Kant’s notion of miracles is usually understood along these lines. However, Kant’s occupation with ...miracles should be understood in a practical context. Belief in miracles plays a constitutive role in Kant’s philosophy of religion concerning the need to strengthen the will both before and after departing from original evil. I demonstrate how my argument sheds new light on Kant’s claim that theoretical reason precludes the possibility of material miracles.
Despite progress in perceptual tasks such as image classification, computers still perform poorly on cognitive tasks such as image description and question answering. Cognition is core to tasks that ...involve not just recognizing, but reasoning about our visual world. However, models used to tackle the rich content in images for cognitive tasks are still being trained using the same datasets designed for perceptual tasks. To achieve success at cognitive tasks, models need to understand the interactions and relationships between objects in an image. When asked “What vehicle is the person riding?”, computers will need to identify the objects in an image as well as the relationships

to answer correctly that “the person is riding a horse-drawn carriage.” In this paper, we present the Visual Genome dataset to enable the modeling of such relationships. We collect dense annotations of objects, attributes, and relationships within each image to learn these models. Specifically, our dataset contains over 108K images where each image has an average of

objects,

attributes, and
21
pairwise relationships between objects. We canonicalize the objects, attributes, relationships, and noun phrases in region descriptions and questions answer pairs to WordNet synsets. Together, these annotations represent the densest and largest dataset of image descriptions, objects, attributes, relationships, and question answer pairs.
Sulfate geoengineering (SG) methods based on lower stratospheric tropical injection of sulfur dioxide (SO2) have been widely discussed in recent years, focusing on the direct and indirect effects ...they would have on the climate system. Here a potential alternative method is discussed, where sulfur emissions are located at the surface or in the troposphere in the form of carbonyl sulfide (COS) gas. There are two time-dependent chemistry–climate model experiments designed from the years 2021 to 2055, assuming a 40 Tg-Syr-1 artificial global flux of COS, which is geographically distributed following the present-day anthropogenic COS surface emissions (SG-COS-SRF) or a 6 Tg-Syr-1 injection of COS in the tropical upper troposphere (SG-COS-TTL). The budget of COS and sulfur species is discussed, as are the effects of both SG-COS strategies on the stratospheric sulfate aerosol optical depth (∼Δτ=0.080 in the years 2046–2055), aerosol effective radius (0.46 µm), surface SOx deposition (+8.9 % for SG-COS-SRF; +3.3 % for SG-COS-TTL), and tropopause radiative forcing (RF; ∼-1.5 W m−2 in all-sky conditions in both SG-COS experiments). Indirect effects on ozone, methane and stratospheric water vapour are also considered, along with the COS direct contribution. According to our model results, the resulting net RF is −1.3 W m−2, for SG-COS-SRF, and −1.5 W m−2, for SG-COS-TTL, and it is comparable to the corresponding RF of −1.7 W m−2 obtained with a sustained injection of 4 Tg-Syr-1 in the tropical lower stratosphere in the form of SO2 (SG-SO2, which is able to produce a comparable increase of the sulfate aerosol optical depth). Significant changes in the stratospheric ozone response are found in both SG-COS experiments with respect to SG-SO2 (∼5 DU versus +1.4 DU globally). According to the model results, the resulting ultraviolet B (UVB) perturbation at the surface accounts for −4.3 % as a global and annual average (versus −2.4 % in the SG-SO2 case), with a springtime Antarctic decrease of −2.7 % (versus a +5.8 % increase in the SG-SO2 experiment). Overall, we find that an increase in COS emissions may be feasible and produce a more latitudinally uniform forcing without the need for the deployment of stratospheric aircraft. However, our assumption that the rate of COS uptake by soils and plants does not vary with increasing COS concentrations will need to be investigated in future work, and more studies are needed on the prolonged exposure effects to higher COS values in humans and ecosystems.

Abstract Objective To raise awareness among clinicians and epidemiologists that single-patient (n-of-1) trials are potentially useful for informing personalized treatment decisions for patients with ...chronic conditions. Study Design and Setting We reviewed the clinical and statistical literature on methods and applications of single-patient trials and then critically evaluated the needs for further methodological developments. Results Existing literature reports application of 2,154 single-patient trials in 108 studies for diverse clinical conditions; various recent commentaries advocate for wider application of such trials in clinical decision making. Preliminary evidence from several recent pilot acceptability studies suggests that single-patient trials have the potential for widespread acceptance by patients and clinicians as an effective modality for increasing the therapeutic precision. Bayesian and adaptive statistical methods hold promise for increasing the informational yield of single-patient trials while reducing participant burden, but are not widely used. Personalized applications of single-patient trials can be enhanced through further development and application of methodologies on adaptive trial design, stopping rules, network meta-analysis, washout methods, and methods for communicating trial findings to patients and clinicians. Conclusions Single-patient trials may be poised to emerge as an important part of the methodological armamentarium for comparative effectiveness research and patient-centered outcomes research. By permitting direct estimation of individual treatment effects, they can facilitate finely graded individualized care, enhance therapeutic precision, improve patient outcomes, and reduce costs.
