We present Herschel observations of water isotopologues in the atmosphere of the Jupiter-family comet 45P/Honda-Mrkos-Pajdusakova No HDO emission is detected, with a 3sigma upper limit of 2.0 x 10 ...super(-4) for the D/H ratio. This value is consistent with the earlier Herschel measurement in the Jupiter-family comet 103P/Hartley 2. The canonical value of 3 x 10 super(-4) measured pre-Herschel in a sample of Oort-cloud comets can be excluded at a 4.5sigma level. The observations presented here further confirm that a diversity of D/H ratios exists in the comet population and emphasize the need for additional measurements with future ground-based facilities, such as CCAT, in the post-Herschel era.
Context. Since its arrival at the comet 67P/Churyumov–Gerasimenko in August 2014, the Rosetta spacecraft followed the comet as it went past the perihelion and beyond until September 2016. During this ...time there were many scientific instruments operating on board Rosetta to study the comet and its evolution in unprecedented detail. In this context, our study focusses on the distribution and evolution of exposed bright features that have been observed by OSIRIS, which is the scientific imaging instrument aboard Rosetta. Aims. We envisage investigating various morphologies of exposed bright features and the mechanisms that triggered their appearance. Methods. We co-registered multi-filter observations of OSIRIS images that are available in reflectance. The Lommel–Seeliger disk function was used to correct for the illumination conditions and the resulting colour cubes were used to perform spectrophotometric analyses on regions of interest. Results. We present a catalogue of 57 exposed bright features observed on the nucleus of the comet, all of which are attributed to the presence of H2O ice on the comet. Furthermore, we categorise these patches under four different morphologies and present geometric albedos for each category. Conclusions. Although the nucleus of 67P/Churyumov–Gerasimenko appears to be dark in general, there are localised H2O ice sources on the comet. Cometary activity escalates towards the perihelion passage and reveals such volatile ices. We propose that isolated H2O ice patches found in smooth terrains in regions, such as Imhotep, Bes, and Hapi, result from frost as an aftermath of the cessation of the diurnal water cycle on the comet as it recedes from perihelion. Upon the comet’s return to perihelion, such patches are revealed when sublimation-driven erosion removes the thin dust layers that got deposited earlier. More powerful activity sources such as cometary outbursts are capable of revealing much fresher, less contaminated H2O ice that is preserved with consolidated cometary material, as observed on exposed patches resting on boulders. This is corroborated by our albedo calculations that attribute higher albedos for bright features with formations related to outbursts.
In classic Hodgkin lymphoma (HL) and posttransplantation lymphoproliferative disease (PTLD), 2 malignancies frequently associated with Epstein-Barr virus (EBV), the tumor cells often appear to derive ...from B-cell receptor (BCR)–deficient and therefore preapoptotic germinal center (GC) B cells. To test whether EBV can rescue BCR-less GC B cells, we infected human tonsillar CD77+ GC B cells in vitro with EBV. More than 60 monoclonal lymphoblastoid cell lines (LCLs) were established. Among these, 28 cell lines did not express surface immunoglobulin (sIg). Two of the sIg-negative cell lines carry obviously destructive mutations that have been introduced into originally functional VH gene rearrangements during the process of somatic hypermutation. Quantitative reverse transcriptase–polymerase chain reaction (RT-PCR) showed that in most other lines the sIg deficiency was not simply the result of transcriptional down-regulation, but it was rather due to posttranscriptional defects. These findings strongly support the idea that EBV plays a central role in the pathogenesis of classic HL and PTLD by rescuing BCR-deficient, preapoptotic GC B cells from apoptosis, and that EBV infection renders the cells independent from survival signals normally supplied by a BCR. The monoclonal LCLs represent valuable models for early stages of lymphoma development in classic HL and PTLD.
Introduction
Dexmedetomidine is the sedative agent administered in combination with remifentanil and low dose of sevoflurane in the interventional arm of the ongoing TREX trial (Trial Remifentanil ...DExmedetomidine). The TREX pilot study (published in Paediatr Anaesth 2019;29:59–67) established infusion rates higher than those initially proposed. This could be attributed to an inappropriate target concentration for sedation or incorrect initial pharmacokinetic parameter estimates.
Methods
The TREX study is a Phase III, randomized, active controlled, parallel group, blinded evaluator, multicenter, superiority trial comparing neurological outcome after standard sevoflurane anesthesia with dexmedetomidine/remifentanil, and low dose sevoflurane anesthesia in children aged less than 2 years undergoing anesthesia of 2 h or longer. In this report, dexmedetomidine pharmacokinetics were analyzed in the interventional arm of the Italian population.
Results
There were 162 blood samples from 32 infants (22 male and 10 female). The median (IQR) age was 12 (5.2–15.5) months, weight 9.9 (7.3–10.8) kg. Duration of anesthesia ranged from 2 to 6 h. None of the children were born premature (median postnatal age 39 weeks, IQR 38–40 weeks). A 3‐compartment PK model that incorporated allometric scaling and a maturation function demonstrated plasma concentration observations from the current Italian arm of the TREX study were consistent with those predicted by a “universal” model using pooled data obtained from neonates to adults.
Conclusions
This current PK analysis from the Italian arm of the TREX study confirms that plasma concentration of dexmedetomidine is predictable using known covariates such as age and size. The initial target concentration (0.6 μg.L−1) used to sedate children cared for in the intensive care after cardiac surgery was inadequate for infants in the current TREX study. A target concentration 1 mcg.L−1, corresponding to a loading dose of 1 mcg.kg−1 followed by an infusion of 1 mcg.kg−1.h−1, provided adequate sedation.
Anaplastic large cell lymphoma (ALCL) is a main type of T-cell lymphomas and comprises three distinct entities: systemic anaplastic lymphoma kinase (ALK) positive, systemic ALK(-) and cutaneous ...ALK(-) ALCL (cALCL). Little is known about their pathogenesis and their cellular origin, and morphological and immunophenotypical overlap exists between ALK(-) ALCL and classical Hodgkin lymphoma (cHL). We conducted gene expression profiling of microdissected lymphoma cells of five ALK(+) and four ALK(-) systemic ALCL, seven cALCL and sixteen cHL, and of eight subsets of normal T and NK cells. The analysis supports a derivation of ALCL from activated T cells, but the lymphoma cells acquired a gene expression pattern hampering an assignment to a CD4(+), CD8(+) or CD30(+) T-cell origin. Indeed, ALCL display a down-modulation of many T-cell characteristic molecules. All ALCL types show significant expression of NFkappaB target genes and upregulation of genes involved in oncogenesis (e.g. EZH2). Surprisingly, few genes are differentially expressed between systemic and cALCL despite their different clinical behaviour, and between ALK(-) ALCL and cHL despite their different cellular origin. ALK(+) ALCL are characterized by expression of genes regulated by pathways constitutively activated by ALK. This study provides multiple novel insights into the molecular biology and pathogenesis of ALCL.
Ba0.5Sr0.5Co0.8Fe0.2O3-delta (BSCF) exhibits high oxygen permeability, which is why it has potential for gas separation (oxygen transport membrane) in zero-emission power plants using oxyfuel ...technology when the membrane is operated in a clean environment, i.e. no flue gas contact. The influence of membrane processing on microstructure and oxygen permeation was studied. Pure-phase BSCF powder was synthesised using a modified Pechini method. For comparison, commercially available powder was also used, synthesised by a solid-state reaction. Disc-shaped membranes of various microstructures, i.e. closed porosities and grain sizes, were prepared by uniaxial pressing and sintering of the powders processed in different ways. The powders and membranes were characterised by BET, SEM, XRD, and DSC. The microstructures obtained by different sintering conditions were investigated by SEM and TEM. Sintering at 1150 C led to incongruent melting of BSCF, as indicated by DSC. The liquid phase appeared at three-phase boundaries grain-grain-air and consisted of nearly pure cobalt oxide with small impurities of barium and strontium detected by TEM/EDX analysis. Oxygen permeation of the membranes was measured in an air/Ar gradient depending on temperature and membrane microstructure. The closed porosity of different processed membranes was varied between 2 and 15% with uniform grain sizes in the range of about 10 micron. The average grain size was increased from 10 to 45 micron by increasing the sintering temperature. Neither porosity nor grain size significantly influenced the oxygen permeation rate of 1-mm-thick discs in the investigated parameter range.
BCOR is a component of a variant Polycomb repressive complex 1 (PRC1.1). PRC1 and PRC2 complexes together constitute a major gene regulatory system critical for appropriate cellular differentiation. ...The gene is upregulated in germinal center (GC) B cells and mutated in a number of hematologic malignancies. We report BCOR inactivating alterations in 4/7 classic Hodgkin lymphoma (cHL) cell lines, subclonal somatic mutations in Hodgkin and Reed-Sternberg (HRS) cells of 4/10 cHL cases, and deletions in HRS cells of 7/17 primary cHL cases. In mice, conditional loss of Bcor driven by AID-Cre in GC B cells resulted in gene expression changes of 46 genes (>2-fold) including upregulated Lef1 that encodes a transcription factor responsible for establishing T-cell identity and Il9r (interleukin-9 receptor), an important member of the cytokine network in cHL. Our findings suggest a role for BCOR loss in cHL pathogenesis and GC-B cell homeostasis.

We provide a three-dimensional model of the inner layered structure of comet 67P based on the hypothesis of an extended layering independently wrapping each lobe. A large set of terrace orientations ...was collected on the latest shape model and then used as a proxy for the local orientation of the surfaces of discontinuity which defines the layers. We modelled the terraces as a family of concentric ellipsoidal shells with fixed axis ratios, producing a model that is completely defined by just eight free parameters. Each lobe of 67P has been modelled independently, and the two sets of parameters have been estimated by means of non-linear optimization of the measured terrace orientations. The proposed model is able to predict the orientation of terraces, the elongation of cliffs, the linear traces observed in the Wosret and Hathor regions and the peculiar alignment of boulder-like features which has been observed in the Hapi region, which appears to be related to the inner layering of the big lobe. Our analysis allowed us to identify a plane of junction between the two lobes, further confirming the independent nature of the lobes. Our layering models differ from the best-fitting topographic ellipsoids of the surface, demonstrating that the terraces are aligned to an internal structure of discontinuities, which is unevenly exposed on the surface, suggesting a complex history of localized material removal from the nucleus.
Context. On 12 March 2015 the OSIRIS WAC camera onboard the ESA Rosetta spacecraft orbiting comet 67P/Churyumov-Gerasimenko observed a small outburst originating from the Imhotep region at the foot ...of the big lobe of the comet. These measurements are unique since it was the first time that the initial phase of a transient outburst event could be directly observed. Aims. We investigate the evolution of the dust jet in order to derive clues about the outburst source mechanism and the ejected dust particles, in particular the dust mass, dust-to-gas ratio and the particle size distribution. Methods. Analysis of the images and of the observation geometry using comet shape models in combination with gasdynamic modeling of the transient dust jet were the main tools used in this study. Synthetic images were computed for comparison with the observations. Results. Analysis of the geometry revealed that the source region was not illuminated until 1.5 h after the event implying true nightside activity was observed. The outburst lasted for less than one hour and the average dust production rate during the initial four minutes was of the order of 1 kg/s. During this time the outburst dust production rate was approximately constant, no sign for an initial explosion could be detected. For dust grains between 0.01−1 mm a power law size distribution characterized by an index of about 2.6 provides the best fit to the observed radiance profiles. The dust-to-gas ratio of the outburst jet is in the range 0.6−1.8.
Abstract
Cometary outbursts on several comets have been observed both by ground-based telescopes and by in situ instruments on spacecraft. However, the mechanism behind these phenomena and their ...physical properties are still unclear. The optical, spectrocopic and infrared remote imaging system (OSIRIS) onboard the Rosetta spacecraft provided first-hand information on the outbursts from comet 67P/Churyumov–Gerasimenko during its perihelion passage in 2015. The physical properties of the outbursts can be investigated by examining the time series of these high-resolution images. An analysis is made of the wide- and narrow-angle images obtained during the monitoring of the outburst sequences, which occurred between July and September in 2015. A ring-masking technique is used to calculate the excess brightness of the outbursts. The ejected mass and expansion velocity of the outbursts is estimated from differences in images made with the same filter (orange filter). The calculated excess brightness from these outburst plumes ranges from a few per cent to 28 per cent. In some major outbursts, the brightness contribution from the outburst plume can be one or two times higher than that of the typical coma jet activities. The strongest event was the perihelion outburst detected just a few hours before perihelion. The mass ejection rate during a generic outburst could reach a few per cent of the steady-state value of the dust coma. Transient events are detected by studying the brightness slope of the outburst plume with continuous streams of outflowing gas and dust triggered by driving mechanisms, as yet not understood, which remain active for several minutes to less than a few hours.
