Summary
Background The neuropeptide calcitonin gene‐related peptide (CGRP) is released in the lung by sensory nerves during allergic airway responses. Pulmonary dendritic cells (DC) orchestrating the ...allergic inflammation could be affected by CGRP.
Objective To determine the immunomodulatory effects of CGRP on DC function and its impact on the induction of allergic airway inflammation.
Methods CGRP receptor expression on lung DC was determined by RT‐PCR and immunofluorescence staining. The functional consequences of CGRP receptor triggering were evaluated in vitro using bone marrow‐derived DC. DC maturation and the induction of ovalbumin (OVA)‐specific T cell responses were analysed by flow cytometry. The in vivo relevance of the observed DC modulation was assessed in a DC‐transfer model of experimental asthma. Mice were sensitized by an intrapharyngeal transfer of OVA‐pulsed DC and challenged with OVA aerosol. The impact of CGRP pretreatment of DC on airway inflammation was characterized by analysing differential cell counts and cytokines in bronchoalveolar lavage fluid (BALF), lung histology and cytokine responses in mediastinal lymph nodes.
Results RT‐PCR, immunofluorescence and cAMP assay demonstrated the expression of functionally active CGRP receptors in lung DC. RT‐PCR revealed a transcriptional CGRP receptor down‐regulation during airway inflammation. CGRP specifically inhibited the maturation of in vitro generated DC. Maturation was restored by blocking with the specific antagonist CGRP8−37. Consequently, CGRP‐pretreated DC reduced the activation and proliferation of antigen‐specific T cells and induced increased the numbers of T regulatory cells. The transfer of CGRP‐pretreated DC diminished allergic airway inflammation in vivo, shown by reduced eosinophil numbers and increased levels of IL‐10 in BALF.
Conclusions and Clinical Relevance CGRP inhibits DC maturation and allergen‐specific T cell responses, which affects the outcome of the allergic airway inflammation in vivo. This suggests an additional mechanism by which nerve‐derived mediators interfere with local immune responses. Thus, CGRP as an anti‐inflammatory mediator could represent a new therapeutic tool in asthma therapy.

Inhalation is a non-invasive approach for both local and systemic drug delivery. This study aimed to define the therapeutic window for solid lipid nanoparticles (SLNs) as a drug delivery system by ...inhalation from a toxicological point of view.
To estimate the toxic dose of SLNs
in vitro, A549 cells and murine precision-cut lung slices (PCLS) were exposed to increasing concentrations of SLNs. The cytotoxic effect of SLNs on A549 cells was evaluated by MTT and NRU assays. Viability of lung tissue was determined with WST assay and by life/dead staining using calcein AM/EthD-1 for confocal microscopy (CLSM) followed by quantitative analysis with IMARIS. Inflammation was assessed by measuring chemokine KC and TNF-α levels. The
in vivo effects were determined in a 16-day repeated-dose inhalation toxicity study using female BALB/c mice, which were daily exposed to different concentrations of SLN30 aerosols (1–200
μg deposit dose). Local inflammatory effects in the respiratory tract were evaluated by determination of total protein content, LDH, chemokine KC, IL-6, and differential cell counts, performed on days 4, 8, 12, and 16 in bronchoalveolar lavage fluid. Additionally, a histopathological evaluation of toxicologically relevant organs was accomplished.
The
in vitro and
ex vivo dose finding experiments showed toxic effects beginning at concentrations of about 500
μg/ml. Therefore, we used 1–200
μg deposit doses/animal for the
in vivo experiments. Even after 16
days of challenge with a 200-μg deposit dose, SLNs induced no significant signs of inflammation. We observed no consistent increase in LDH release, protein levels, or other signs of inflammation such as chemokine KC, IL-6, or neutrophilia. In contrast, the particle control (carbon black) caused inflammatory and cytotoxic effects at corresponding concentrations.
These results confirm that repeated inhalation exposure to SLN30 at concentrations lower than a 200-μg deposit dose is safe in a murine inhalation model.
Bronchoconstriction is a characteristic symptom of various chronic obstructive respiratory diseases such as chronic obstructive pulmonary disease and asthma. Precision-cut lung slices (PCLS) are a ...suitable ex vivo model to study physiological mechanisms of bronchoconstriction in different species. In the present study, we established an ex vivo model of bronchoconstriction in nonhuman primates (NHPs). PCLS prepared from common marmosets, cynomolgus macaques, rhesus macaques, and anubis baboons were stimulated with increasing concentrations of representative bronchoconstrictors: methacholine, histamine, serotonin, leukotriene D₄ (LTD₄), U46619, and endothelin-1. Alterations in the airway caliber were measured and compared with previously published data from rodents, guinea pigs, and humans. Methacholine induced maximal airway constriction, varying between 74 and 88% in all NHP species, whereas serotonin was ineffective. Histamine induced maximal bronchoconstriction of 77 to 90% in rhesus macaques, cynomolgus macaques, and baboons and a lesser constriction of 53% in marmosets. LTD₄ was ineffective in marmosets and rhesus macaques but induced a maximum constriction of 44 to 49% in cynomolgus macaques and baboons. U46619 and endothelin-1 caused airway constriction in all NHP species, with maximum constrictions of 65 to 91% and 70 to 81%, respectively. In conclusion, PCLS from NHPs represent a valuable ex vivo model for studying bronchoconstriction. All NHPs respond to mediators relevant to human airway disorders such as methacholine, histamine, U46619, and endothelin-1 and are insensitive to the rodent mast cell product serotonin. Only PCLS from cynomolgus macaques and baboons, however, responded also to leukotrienes, suggesting that among all compared species, these two NHPs resemble the human airway mechanisms best.

Background
Neurotrophins such as nerve growth factor or brain‐derived neurotrophic factor influence neuronal proliferation and differentiation via the low‐affinity pan‐neurotrophin receptor ...p75NTR that may play a pivotal role in linking the immune with the nervous system. Because the precise regulation of p75NTR gene transcription in mast cells under states of allergic inflammation has not been investigated in detail so far, the present studies assessed the gene regulation and expression of this receptor.
Methods
Transcriptional expression of p75NTR in human skin was studied in isolated cutaneous cells by means of RT‐PCR. In situ lesional mast cell p75NTR expression was analysed by immunohistochemistry.
Results
The p75NTR mRNA expression was found in isolated human skin mast cells and keratinocytes. Lower mRNA levels were present in fibroblasts and melanocytes but no transcripts were found in endothelial cells. The p75NTR protein expression was found in situ in lesional and non‐lesional mast cells. A significantly increased expression of p75NTR protein was found in atopic dermatitis lesional mast cells when compared with control mast cell expression (P<0.05).
Conclusion
The demonstration of an increased level of p75NTR gene transcription in lesional mast cells points to an induction of low‐affinity neurotrophin receptor sensitivity of mast cells under states of allergic inflammation. Topically administered neurotrophin receptor‐modulating compounds may act as anti‐inflammatory mediators in cutaneous allergic inflammation.

Summary
Background
Bronchial asthma is characterized by airway inflammation and reversible obstruction. Since the gold standard of therapy, a combination of anti‐inflammatory corticosteroids and bronchodilatory β2 agonists, has recently been discussed to be related to an increased mortality, there is a need for novel therapeutic pathways.
Objective
A new experimental concept that encompasses the vasoactive intestinal peptide/pituitary adenylate cyclase activating peptide (PACAP) family of receptors by demonstrating the anti‐inflammatory effects of the PACAP receptor 1 (PAC1R) in a murine model of allergic asthma is described.
Methods
PAC1R expression was investigated in lung tissue and isolated dendritic cells (DCs) via real‐time PCR. Ovalbumin (OVA)‐induced asthma models were used in PAC1R‐deficient mice and BALB/c mice treated with PAC1R agonist maxadilan (MAX). Bronchoalveolar lavages have been performed and investigated at the cellular and cytokine levels. Fluorescence staining of a frozen lung section has been performed to detect eosinophil granulocytes in lung tissue. Plasma IgE levels have been quantified via the ELISA technique. Lung function was determined using head‐out body plethysmography or whole‐body plethysmography.
Results
Increased PAC1R mRNA expression in lung tissue was present under inflammatory conditions. PAC1R expression was detected on DCs. In OVA‐induced asthma models, which were applied to PAC1R‐deficient mice (PAC1R−/−) and to BALB/c mice treated with the specific PAC1R agonist MAX, PAC1R deficiency resulted in inflammatory effects, while agonistic stimulation resulted in anti‐inflammatory effects. No effects on lung function were detected both in the gene‐depletion and in the pharmacologic studies. In summary, here, we demonstrate that anti‐inflammatory effects can be achieved via PAC1R.
Conclusion
PAC1R agonists may represent a promising target for an anti‐inflammatory therapy in airway diseases such as bronchial asthma.
Experimental evidence and Monte-Carlo simulations for several technologies show that accurate SEE response predictions depend on a detailed description of the variability of radiation events (e.g., ...nuclear reactions), as opposed to the classical single-valued LET parameter. Rate predictions conducted with this simulation framework exhibit excellent agreement with the average observed SEU rate on NASA's MESSENGER mission to Mercury, while a prediction from the traditional IRPP method, which does not include the contribution from ion-ion reactions, falls well below the observed rate. While rate predictions depend on availability of technology information, the approach described here is sufficiently flexible that reasonably accurate results describing the response to irradiation can be obtained even in the absence of detailed information about the device geometry and fabrication process.

Ansätze eines Lipopolysaccharid (LPS) induzierten akuten Entzündungsmodells im Weißbüschelaffen (
Callithrix jacchus
).
Vollblut und Präzisionslungenschnitte (PCLS) aus Weißbüschelaffen wurden
ex vivo
mit LPS (2,5–500 ng/ml) und dem Phosphodiesterase-4-Inhibitor Roflumilast (Daxas®, 0,6–310 nM) inkubiert. Als Endpunkt wurde die Konzentration des Tumornekrosefaktor-alpha (TNF-α) im Überstand mittels ELISA bestimmt und mit Daten humaner Versuchsansätze korreliert. In einem
in vivo
Ansatz wurden Weißbüschelaffen nach 5-tägiger oraler Behandlung mit einer humanäquivalenten Roflumilastdosis (7 µg/kg KM) intrabronchial mit 500ng LPS stimuliert. Die Instillation erfolgte dabei segmental über den linken Bronchus. Nicht behandelte, LPS-stimulierte Tiere dienten als Positivkontrolle, Dexamethason behandelte Tiere (2 mg/kg KM oral, über 5 Tage) bildeten die Behandlungskontrolle. Nach 18 Stunden erfolgte eine ipsilaterale bronchoalveoläre Lavage (BAL). Die BAL Flüssigkeit wurde anschließend auf Zell- und Zytokinebene analysiert.
Vollblut und PCLS Ansätze der Weißbüschelaffen zeigten eine signifikant dosisabhängige TNF-α Sekretion nach LPS-Stimulation (r
2
=0,96). Die TNF-α Sekretion in Weißbüschelaffen PCLS korrelierte zudem signifikant mit gleichbehandelten humanen PCLS (r
2
=0,89). Die Inkubation mit Roflumilast führte zu einer konzentrationsabhängigen Verminderung der TNF-α Sekretion, mit ähnlichen EC
50
Werten in beiden Spezies (3,5 nM vs. 4,3 nM). Wie bereits für den Menschen
in vivo
gezeigt, kann die LPS induzierte akute Entzündung auch im Marmoset durch Gabe von Roflumilast vermindert werden. Die Zahl der neutrophilien Granulozyten, sowie die TNF-α Konzentration in der BAL Flüssigkeit können mittels Roflumilast signifikant reduziert werden.
Die Korrelation zwischen Weißbüschelaffen und humanen PCLS, sowie die anti-inflammatorische Wirkung von Roflumilast zeigen das Potential des Weißbüschelaffen als Tiermodel für entzündliche Atemwegserkrankungen wie der COPD des Menschen.

Ähnlich der segmentalen Lipopolysaccharid (LPS)-Provokation beim Menschen, wurde ein Modell der LPS induzierten, akuten Lungenentzündung beim Weißbüschelaffen (
C. jacchus
) etabliert. Die LPS induzierte Entzündung spiegelt pro-inflammatorische Aspekte einer Neutrophilen-assozierten, chronischen Atemwegserkrankung des Menschen wider.
Mittels bronchoalveolärer Lavage (BAL) wurden individuelle Basalwerte von gesunden Weißbüschelaffen erhoben. Die Tiere wurden dann, je nach Gruppe, oral mit Roflumilast (7 µg/kg KM), einem kürzlich zugelassenen PDE-4 Inhibitor, oder Dexamethason (2 mg/kg KM) als Behandlungskontrolle, über 5 aufeinander folgende Tage behandelt. Scheinbehandelte Tiere dienten als Kontrolle. In Allgemeinanästhesie wurden 500 ng LPS in die linke Hälfte der Lunge eines jeden Tieres instilliert, gefolgt von ipsilateraler BAL 18 Stunden post-Provokation. Die BAL-Flüssigkeit wurde auf Zellgehalt und Zytokinlevel untersucht.
Wie erwartet folgte der intrapulmonalen LPS Applikation ein signifanter Influx von Zellen in die Atemwege (p < 0,001), hauptsächlich verursacht durch die Aggregation von Neutrophilen. Eine Behandlung mit Roflumilast resultierte in einer signifikanten Reduktion Neutrophiler Granulozyten in der BAL-Flüssigkeit (p = 0,047).
