Aims
This study aimed to monitor development of spoilage‐associated microbiota on high‐oxygen modified atmosphere packaged (MAP) minced beef, assess diversity of Pseudomonas sp. therein employing a ...polyphasic approach and probe their ability to grow anaerobically in the presence of carbon dioxide.
Methods and Results
Headspace atmosphere and total viable count of MAP minced beef were monitored, and spoilage‐associated microbiota was identified using matrix‐assisted laser desorption/ionization time of flight mass spectrometry (MALDI‐TOF MS). Pseudomonas spp. represented a major part of the spoilage‐associated microbiota throughout the spoilage process and were characterized with a polyphasic approach including MALDI‐TOF, randomly amplified polymorphic DNA biotyping, 16S rDNA and rpoD sequence analysis, and carA multiplex polymerase chain reaction. Pseudomonas isolates displayed high diversity and varying assertiveness under conditions employed in MAP minced beef with P. fragi, P. lundensis and P. weihenstephanensis as dominant species.
Conclusions
The polyphasic approach enabled high‐throughput characterization of Pseudomonas sp. Their adapted capability to grow anaerobically and resistance to high levels of CO2 is suggested to be a general feature within the genus, which is hitherto underexplored.
Significance and Impact of the Study
This study shows that diverse Pseudomonas generally regarded as strict aerobes and CO2‐sensitive appear well adapted to grow under MAP conditions, leading to high cell counts in minced beef and ultimately contribute to spoilage of the product.

transcriptional reporters revealed reduced transcription of the prodigiosin biosynthetic operon at 37°C compared to 30°C indicating transcriptional control of pigment production. Moreover, induced expression of the prodigiosin biosynthetic operon at 37°C was able to produce pigmented colonies and cultures demonstrating that physiological conditions at 37°C allow prodigiosin production and indicating that post-transcriptional control is not a major contributor to the thermoregulation of prodigiosin pigmentation. Genetic experiments support the model that the HexS transcription factor is a key contributor to thermoregulation of pigmentation, whereas CRP plays a minor role, and a clear role for EepR and PigP was not observed. Together, these data indicate that thermoregulation of prodigiosin production at elevated temperatures is controlled largely, if not exclusively, at the transcriptional level.

was recently demonstrated to influence ocular pathogenesis in a rabbit model of keratitis, with Rcs-defective mutants causing greater pathology and Rcs-activated strains demonstrating reduced inflammation. The Rcs system is activated by a variety of insults, including β-lactam antibiotics and polymyxin B. In this study, we developed three luminescence-based transcriptional reporters for Rcs system activity and used them to test whether antibiotics used for empiric treatment of ocular infections influence Rcs system activity in a keratitis isolate of
. These included antibiotics to which the bacteria were susceptible and resistant. Results indicate that cefazolin, ceftazidime, polymyxin B, and vancomycin activate the Rcs system to varying degrees in an RcsB-dependent manner, whereas ciprofloxacin and tobramycin activated the promoter fusions, but in an Rcs-independent manner. Although minimum inhibitory concentration (MIC) analysis demonstrated resistance of the test bacteria to polymyxin B and vancomycin, the Rcs system was activated by sub-inhibitory concentrations of these antibiotics. Together, these data indicate that a bacterial stress system that influences numerous pathogenic phenotypes and drug-tolerance is influenced by different classes of antibiotics despite the susceptibility status of the bacterium.
We have analyzed the first 3.75 years of data from the Taiwanese American Occultation Survey (TAOS). TAOS monitors bright stars to search for occultations by Kuiper Belt objects (KBOs). This data set ...comprises 5 X 105 star hours of multi-telescope photometric data taken at 4 or 5 Hz. No events consistent with KBO occultations were found in this data set. We compute the number of events expected for the Kuiper Belt formation and evolution models of Pan & Sari, Kenyon & Bromley, Benavidez & Campo Bagatin, and Fraser. A comparison with the upper limits we derive from our data constrains the parameter space of these models. This is the first detailed comparison of models of the KBO size distribution with data from an occultation survey. Our results suggest that the KBO population is composed of objects with low internal strength and that planetary migration played a role in the shaping of the size distribution.
PurposeWith the help of “self-learning” algorithms and high computing power, companies are transforming Big Data into artificial intelligence (AI)-powered information and gaining economic benefits. ...AI-powered information and Big Data (simply data henceforth) have quickly become some of the most important strategic resources in the global economy. However, their value is not (yet) formally recognized in financial statements, which leads to a growing gap between book and market values and thus limited decision usefulness of the underlying financial statements. The objective of this paper is to identify ways in which the value of data can be reported to improve decision usefulness.Design/methodology/approachBased on the authors' experience as both long-term practitioners and theoretical accounting scholars, the authors conceptualize and draw up a potential data value chain and show the transformation from raw Big Data to business-relevant AI-powered information during its process.FindingsAnalyzing current International Financial Reporting Standards (IFRS) regulations and their applicability, the authors show that current regulations are insufficient to provide useful information on the value of data. Following this, the authors propose a Framework for AI-powered Information and Big Data (FAIIBD) Reporting. This framework also provides insights on the (good) governance of data with the purpose of increasing decision usefulness and connecting to existing frameworks even further. In the conclusion, the authors raise questions concerning this framework that may be worthy of discussion in the scholarly community.Research limitations/implicationsScholars and practitioners alike are invited to follow up on the conceptual framework from many perspectives.Practical implicationsThe framework can serve as a guide towards a better understanding of how to recognize and report AI-powered information and by that (a) limit the valuation gap between book and market value and (b) enhance decision usefulness of financial reporting.Originality/valueThis article proposes a conceptual framework in IFRS to regulators to better deal with the value of AI-powered information and improve the good governance of (Big)data.
Using 7 yr of MACHO survey data, we present a new determination of the optical depth to microlensing toward the Galactic bulge. We select the sample of 62 microlensing events (60 unique) on clump ...giant sources and perform a detailed efficiency analysis. We use only the clump giant sources because these are bright bulge stars and are not as strongly affected by blending as other events. Using a subsample of 42 clump events concentrated in an area of 4.5 deg super(2) with 739,000 clump giant stars, we find t = 2.17 super(+) sub(-) super(0) sub(0) super(.) sub(.) super(4) sub(3) super(7) sub(8) x 10 super(-6) at (l,b) = (1.50, - 2.68), somewhat smaller than found in most previous MACHO studies but in excellent agreement with recent theoretical predictions. We also present the optical depth in each of the 19 fields in which we detected events and find limits on optical depth for fields with no events. The errors in optical depth in individual fields are dominated by Poisson noise. We measure optical depth gradients of (1.06 c 0.71) x 10 super(-6) deg super(-1) and (0.29 c 0.43) x 10 super(-6) deg super(-1) in the Galactic latitude b and longitude l directions, respectively. Finally, we discuss the possibility of anomalous duration distribution of events in the field 104 centered on (l,b) = (3.11, - 3.01), as well as investigate spatial clustering of events in all fields.
The nature of dark matter remains mysterious, with luminous material accounting for at most ∼25 per cent of the baryons in the Universe. We accordingly undertook a survey looking for the microlensing ...of stars in the Large Magellanic Cloud (LMC) to determine the fraction of Galactic dark matter contained in massive compact halo objects (MACHOs). The presence of the dark matter would be revealed by gravitational lensing of the light from an LMC star as the foreground dark matter moves across the line of sight. The duration of the lensing event is the key observable parameter, but gives non-unique solutions when attempting to estimate the mass, distance and transverse velocity of the lens. The survey results to date indicate that between 8 and 50 per cent of the baryonic mass of the Galactic halo is in the form of MACHOs (ref. 3), but removing the degeneracy by identifying a lensing object would tighten the constraints on the mass in MACHOs. Here we report a direct image of a microlens, revealing it to be a nearby low-mass star in the disk of the Milky Way. This is consistent with the expected frequency of nearby stars acting as lenses, and demonstrates a direct determination of a lens mass from a microlensing event. Complete solutions such as this for halo microlensing events will probe directly the nature of the MACHOs.
