The lower portion of Taquari River is influenced by compounds from anthropic activities causing concern about the drinking water supplied to cities in the region. The study objective was to ...investigate the presence of contaminants at drinking water abstraction sites, defining the mutagenic effects of these stressors as an ecosystem quality parameter and its possible effects on human health. Geographic Information System techniques were used to investigate sources of contamination and it was found that agricultural activities predominated with a few medium and high potential pollutant agricultural activities, besides a soil area that was contaminated and undergoing an intervention process. Mutagenic effects were evaluated by Salmonella/microsome assay using TA98, TA97a, TA100, YG1041 and YG1042 strains in the presence and absence of metabolic activation (S9). Mutagenesis found in organic sediment extracts and surface water samples showed the prevalence of direct-acting mutagens at the drinking water abstraction sites. Taquari (Ta032, the sampling points were named according to the initial letters of the river (Ta), followed by the number of kilometers from the mouth) showed the highest mutagenic potency in sediment, while Ta063, at Bom Retiro do Sul, presented it in the water sample. In the Triunfo region (Ta011) there were significant responses in sediment and in water samples. The samples at General Câmara (Ta006) showed the least presence of contaminants. The Allium cepa test applied to sediments in natura showed significant micronucleus induction in Ta032 in accordance with the Salmonella/microssome assay. The test performed on Danio rerio embryos (FET) in the in natura water samples did not present significant responses. Chemical analyses of polycyclic aromatic hydrocarbons and metals already identified as chemical markers in the area indicated a small contribution to the mutagenic potency, calling attention to the fact that other direct-acting pollutants may be present at the drinking water abstraction sites.

This is a scoping review limited to the period between 2015 and 2022.The results of this scoping review are discussed and presented separately in 3 articles. This second paper synthesizes research evidence on the risk factors, diagnostic methods and treatment of infection in adult patients undergoing left ventricular assist device implantation.
The initial searches identified 771 studies. Sixty-nine patients met the eligibility criteria and were included in the scoping review. Forty-three articles addressing the risk factors, diagnosis and treatment of infection were included to answer the questions of this review.
Obesity has been shown to be the most common risk factor for the described process of infection by left ventricular assist devices.18F-fluorodeoxyglucose positron emission tomography showed high sensitivity in detecting cardiac device infection, and labeled leukocyte or gallium citrate-67 scintigraphy showed high specificity for left ventricular assist device infections; therefore, it can help differentiate infection from inflammation, particularly in patients with equivocal fluorodeoxyglucose positron emission tomography. Also, this review brings and discusses the limitations and strengths of diagnostic tests, the knowledge regarding the risk factors for left ventricular assist device infection, the therapeutic heterogeneity, the methodological issues of the studies, and the vast opportunity for future research on left ventricular assist device.
Ventricular assist device professionals should evaluate risk factors prior to device implantation and periodically.18F-fluorodeoxyglucose positron emission tomography should be considered as diagnostic tool in detecting superficial and deep driveline infections. Early treatment, including chronic suppressive therapy and serial surgical debridement, combined with driveline exteriorization and delayed driveline relocation may constitute a potential therapeutic strategy for deep driveline infections.
ABSTRACT Integrative and Complementary Practices in Health include different health care forms and have been present for more than decades in health services in the Federal District, where the ...implementation assessment of your policy is not completely defined. To assist in understanding reality, an Evaluability Study was carried out collaboratively, involving discussion with stakeholders, questionnaires and consultation of documents. The policy components were identified, organized and modeled. A simplified-logical-model and one of the operational-logical-models, validated by the participants, are presented in this article. For policy evaluation, the initial aspects of the modeled logical implementation chain were prioritized. It was identified as fundamental the promotion of the actors’ knowledge about the set of resources to the implementation of the Integrative Practices services and about the related intersectoral responsibilities. Formative, qualitative and collaborative assessments are suggested. The modeling carried out in the studies of this policy, shed light on the study of similar ones.
RESUMO As Práticas Integrativas e Complementares em Saúde incluem diversas formas de cuidado de saúde e estão presentes há mais de três décadas nos serviços de saúde no Distrito Federal, onde a avaliação de implantação da sua política não está completamente definida. Para auxiliar na compreensão da realidade, foi realizado Estudo de Avaliabilidade, de forma colaborativa, envolvendo discussão com stakeholders, questionários e consulta a documentos. Foram identificados, organizados e modelizados os componentes da política. Um Modelo-lógico simplificado e um dos modelos-lógico-operacionais, com conteúdo validado pelos participantes, são apresentados neste artigo. Para avaliação da política, foram priorizados aspectos iniciais da cadeia lógica de implementação modelizada. Identificou-se como fundamental o fomento ao conhecimento dos atores sobre o conjunto de recursos necessários para implementação de serviços de práticas integrativas e sobre as responsabilidades intersetoriais relacionadas. Sugerem-se avaliações formativas, qualitativas e colaborativas. A modelização realizada possibilita continuidade nos estudos desta política, lançando luz ao estudo de políticas semelhantes.
ABSTRACT Objective: To estimate the prevalence of common mental disorders and their associated factors in the elderly in a municipality in the countryside of the Brazilian Midwest. Methods: ...Quantitative, observational, cross-sectional approach research, conducted with 218 elderly people. Results: The prevalence of the common mental disorder among the elderly was 25.1%; in the univariate analysis, it was higher in females (35.4%), in the age group of 80 years or older (46.1%), brown skin color (30.2%), widowed (42.6%), illiterate (47.6%), and retired (27.6%). In the multiple analysis, the variables female gender (p<0.006), age 80 years or older (p<0.036), dissatisfaction with life (p<0.009), lack of social interaction (p=0.017), and dysfunctional family (p=0.021) remained associated with CMD. Conclusion: The results revealed are helpful and contribute to the reinforcement of the need for mental health care in this population extract so growing worldwide.

Métodos: Pesquisa de abordagem quantitativa, observacional, transversal, realizada com 218 idosos. Resultados: A prevalência de transtorno mental comum entre os idosos foi de 25,1%; na análise univariada, foi superior no sexo feminino (35,4%), na faixa etária igual ou superior a 80 anos (46,1%), cor da pele parda (30,2%), viúvos (42,6%), analfabetos (47,6%) e aposentados (27,6%). Na análise múltipla, mantiveram-se associadas à TMC as variáveis sexo feminino (p<0,006), idade igual ou superior a 80 anos (p<0,036), insatisfação com a vida (p<0,009), falta de interação social (p=0,017) e família disfuncional (p=0,021). Conclusão: Os resultados revelados são úteis e contribuem no reforço da necessidade de cuidados visando à saúde mental desse extrato populacional tão crescente mundialmente.

Resultados: La prevalencia de trastorno mental común entre los ancianos fue de 25,1%; en el análisis univariado, fue superior en el sexo femenino (35,4%), en la franja etaria igual o superior a 80 años (46,1%), color de piel parda (30,2%), viudos (42,6%), analfabetos (47,6%) y jubilados (27,6%). En el análisis múltiple, se mantuvieron relacionados a la TMC las variables sexo femenino (p<0,006), edad igual o superior a 80 años (p<0,036), insatisfacción con la vida (p<0,009), falta de interacción social (p=0,017) y familia disfuncional (p=0,021). Conclusión: Los resultados revelados son útiles y contribuyen en el refuerzo de la necesidad de cuidados visando a la salud mental de ese extracto poblacional tan creciente mundialmente.
Systemic lupus erythematosus (SLE) is an autoimmune disease with a strong genetic component and etiology characterized by chronic inflammation and autoantibody production. The purpose of this study ...was to ascertain copy number variation (CNV) in SLE using a case-control design in an admixed Brazilian population. The whole-genome detection of CNV was performed using Cytoscan HD array in SLE patients and healthy controls. The best CNV candidates were then evaluated by quantitative real-time PCR in a larger cohort or validated using droplet digital PCR. Logistic regression models adjusted for sex and ancestry covariates was applied to evaluate the association between CNV with SLE susceptibility. The data showed a synergistic effect between the FCGR3B and ADAM3A loci with the presence of deletions in both loci significantly increasing the risk to SLE (5.9-fold) compared to the deletion in the single FCGR3B locus (3.6-fold). In addition, duplications in these genes were indeed more frequent in healthy subjects, suggesting that high FCGR3B/ADAM3A gene copy numbers are protective factors against to disease development. Overall, 21 rare CNVs were identified in SLE patients using a four-step pipeline created for identification of rare variants. Furthermore, heterozygous deletions overlapping the CFHR4, CFHR5 and HLA-DPB2 genes were described for the first time in SLE patients. Here we present the first genome-wide CNV study of SLE patients in a tri-hybrid population. The results show that novel susceptibility loci to SLE can be found once the distribution of structural variants is analyzed throughout the whole genome.

Este estudio realiza una caracterización en salud mental y calidad de vida de habitantes del barrio La Cruz de Medellín, entendiendo la salud como concepto íntegro que abarca niveles psicológicos, ...físicos y sociales. Se tuvo una muestra de 83 personas del sector, 59% mujeres y 41% hombres, a quienes se les aplicaron los siguientes instrumentos: Cuestionario de salud PHQ-9, Cuestionario de ansiedad GAD-7, Escala GENCAT de calidad de vida para adultos, Cuestionario para niños y adolescentes Kiddo-KINDL y la Escala de valoración del barrio para adolescentes. Se encontraron altos niveles de depresión y ansiedad en niños, adolescentes y adultos; así como también relaciones inversas entre la ansiedad con: el desarrollo personal (en adultos), el bienestar emocional (en niños y adolescentes) y el bienestar físico (en adolescentes). Se concluye la pertinencia de una contextualización sobre un barrio con índices de calidad de vida bajos, según antecedentes; además se conocen particularidades del sector y se obtienen resultados significativos con miras a desarrollar programas de promoción y prevención en salud que abarquen factores de riesgo y protección de la comunidad.

Grape juices made using small‐scale production techniques are widely consumed. The extraction procedures employed to produce them, however, can affect bioactive compounds and antioxidant ...activity in the final product. In this study, juices prepared using four extraction methods (steam, extractor, juicer and blender) were evaluated for soluble and hydrolysable polyphenol content, total anthocyanin content, antioxidant capacity, physicochemical characteristics and colour. Acceptance of steam‐extracted juices and their stability during storage were also evaluated. Steam extraction resulted in a higher soluble phenolic (1073 ± 58 mg gallic acid L−1) and anthocyanin content (138 ± 22 mg cyanidin L−1), as well as a higher antioxidant capacity when compared to juices prepared using other methods. Although steam‐extracted juice remained microbiologically stable during 24 months of storage, changes in phytochemical compounds and antioxidant capacity did occur. Our results indicate that steam‐extracted grape juices have high commercial potential.
Juices prepared using four extraction methods were evaluated. Steam extraction resulted in higher soluble phenolic and anthocyanin contents and higher antioxidant capacity when compared to juices prepared using other methods. Although steam extracted juice have remained microbiologically stable during storage for 24 months, changes in phytochemical compounds and antioxidant capacity occurred mainly throughout the first five months.
PIP4K2A is an insufficiently studied type II lipid kinase that catalyzes the conversion of phosphatidylinositol-5-phosphate (PI5P) into phosphatidylinositol 4,5-bisphosphate (PI4,5P2). The ...involvement of PIP4K2A/B in cancer has been suggested, particularly in the context of p53 mutant/null tumors. PIP4K2A/B depletion has been shown to induce tumor growth inhibition, possibly due to hyperactivation of AKT and reactive oxygen species-mediated apoptosis. Herein, we report the identification of the novel potent and highly selective inhibitors BAY-091 and BAY-297 of the kinase PIP4K2A by high-throughput screening and subsequent structure-based optimization. Cellular target engagement of BAY-091 and BAY-297 was demonstrated using cellular thermal shift assay technology. However, inhibition of PIP4K2A with BAY-091 or BAY-297 did not translate into the hypothesized mode of action and antiproliferative activity in p53-deficient tumor cells. Therefore, BAY-091 and BAY-297 serve as valuable chemical probes to study PIP4K2A signaling and its involvement in pathophysiological conditions such as cancer.
