Triple-negative breast cancer (TNBC) puts a great threat to women’s health. GLIS family zinc finger 3 (GLIS3) belongs to the GLI transcription factor family and acts as a critical factor in cancer ...progression. Nevertheless, the part of GLIS3 played in TNBC is not known. Immunohistochemical (IHC) staining analysis displayed that GLIS3 was highly expressed in TNBC tissues. The effect of GLIS3 on the malignant phenotype of TNBC was tested in two different cell lines according to GLIS3 regulation. Upregulation of GLIS3 promoted the proliferation, migration, and invasion of TNBC cell lines, whereas the knockdown of GLIS3 suppressed these tumor activities. Inhibition of GLIS3 induced TNBC cell apoptosis. Furthermore, study as immunofluorescence and electrophoretic mobility shift assay confirmed that the nuclear factor-κB (NF-κB) signaling pathway activated by GLIS3 played an important role in TNBC cells’ malignant phenotype. In conclusion, the present work demonstrated that GLIS3 acts as a crucial element in TNBC progression via activating the NF-κB signaling pathway. Accordingly, above mentioned findings indicated that modulation of GLIS3 expression is a potential tactic to interfere with the progression of TNBC.
Bacterial blight of rice is an important disease in Asia and Africa. The pathogen, Xanthomonas oryzae pv. oryzae (Xoo), secretes one or more of six known transcription-activator-like effectors ...(TALes) that bind specific promoter sequences and induce, at minimum, one of the three host sucrose transporter genes SWEET11, SWEET13 and SWEET14, the expression of which is required for disease susceptibility. We used CRISPR-Cas9-mediated genome editing to introduce mutations in all three SWEET gene promoters. Editing was further informed by sequence analyses of TALe genes in 63 Xoo strains, which revealed multiple TALe variants for SWEET13 alleles. Mutations were also created in SWEET14, which is also targeted by two TALes from an African Xoo lineage. A total of five promoter mutations were simultaneously introduced into the rice line Kitaake and the elite mega varieties IR64 and Ciherang-Sub1. Paddy trials showed that genome-edited SWEET promoters endow rice lines with robust, broad-spectrum resistance.
Dispersion engineering is essential to the performance of most modern optical systems including fiber-optic devices. Even though the chromatic dispersion of a meter-scale single-mode fiber used for ...endoscopic applications is negligible, optical lenses located on the fiber end face for optical focusing and imaging suffer from strong chromatic aberration. Here we present the design and nanoprinting of a 3D achromatic diffractive metalens on the end face of a single-mode fiber, capable of performing achromatic and polarization-insensitive focusing across the entire near-infrared telecommunication wavelength band ranging from 1.25 to 1.65 µm. This represents the whole single-mode domain of commercially used fibers. The unlocked height degree of freedom in a 3D nanopillar meta-atom largely increases the upper bound of the time-bandwidth product of an achromatic metalens up to 21.34, leading to a wide group delay modulation range spanning from -8 to 14 fs. Furthermore, we demonstrate the use of our compact and flexible achromatic metafiber for fiber-optic confocal imaging, capable of creating in-focus sharp images under broadband light illumination. These results may unleash the full potential of fiber meta-optics for widespread applications including hyperspectral endoscopic imaging, femtosecond laser-assisted treatment, deep tissue imaging, wavelength-multiplexing fiber-optic communications, fiber sensing, and fiber lasers.
The need for a high‐frequency and high‐efficiency gallium nitride (GaN)‐based buck converter that can be controlled directly by a low swing pulse width‐modulated (PWM) signal, without the need for ...additional buffers or preamplifiers, poses a significant challenge in the field of power conversion and dynamically power supply applications. To solve this problem, this letter presents a monolithic two‐phase synchronous buck converter, fabricated using a 0.25‐um GaN‐on‐Si process, integrated both the drivers and power stage transistors, without requiring additional buffers or preamplifiers. So, the proposed converter can be directly controlled by a PWM signal with a swing of only 1 V (−0.5 to 0.5 V). At a 100‐MHz switching frequency, the converter achieves a maximum output power of 12 W with a power stage efficiency of 82%. The converter includes two identical half‐bridge Monolithic Microwave Integrated Circuit (MMICs), and with the frequency multiplication property of multi‐phase topology, the equivalent switching frequency can reach 200 MHz in the open‐loop operating mode. The experimental results show that the converter can track a 40‐MHz bandwidth envelope signal (256 QAM, 6 dB PAPR) with 80.1% efficiency.

Accumulating evidence indicated that leukocyte telomere length (LTL) was related to sarcopenia. However, it is still not clear whether the association of changes in LTL with sarcopenia is likely to ...be causal, or could be explained by reverse causality. Thus, we carried on bidirectional Mendelian randomization (MR) and multivariable MR analyses to identify the causal relationship between LTL and sarcopenia-related traits. Summary-level data and independent variants used as instruments came from large genome-wide association studies of LTL (472,174 participants), appendicular lean mass (450,243 participants), low grip strength (256,523 participants), and walking pace (450,967 participants). We identified suggestive association of longer LTL with larger appendicular lean mass odds ratio (OR) = 1.053; 95% confidence interval (CI), 1.009-1.099; P = 0.018, and causal association of longer LTL with a lower risk of low grip strength (OR = 0.915; 95% CI, 0.860-0.974; P = 0.005). In the reverse MR analysis, we also observed a positive causal association between walking pace and LTL (OR = 1.252; 95% CI, 1.121-1.397; P < 0.001). Similar results can be repeated in sensitivity analyses. While in the multivariable MR analysis, the estimate of the impact of walking pace on LTL underwent a transformation after adjusting for T2DM (OR = 1.141; 95%CI: 0.989-1.317; P = 0.070). The current MR analysis supported a causal relationship between shorter telomere length and both low muscle mass and strength. Additionally, walking pace may affect LTL through T2DM.

Testicular Leydig cells are the primary source of testosterone in males. Adult Leydig cells have been shown to arise from stem cells present in the neonatal testis. Once established, adult Leydig ...cells turn over only slowly during adult life, but when these cells are eliminated experimentally from the adult testis, new Leydig cells rapidly reappear. As in the neonatal testis, stem cells in the adult testis are presumed to be the source of the new Leydig cells. As yet, the mechanisms involved in regulating the proliferation and differentiation of these stem cells remain unknown. We developed a unique in vitro system of cultured seminiferous tubules to assess the ability of factors from the seminiferous tubules to regulate the proliferation of the tubule-associated stem cells, and their subsequent entry into the Leydig cell lineage. The proliferation of the stem Leydig cells was stimulated by paracrine factors including Desert hedgehog (DHH), basic fibroblast growth factor (FGF2), platelet-derived growth factor (PDGF), and activin. Suppression of proliferation occurred with transforming growth factor β (TGF-β). The differentiation of the stem cells was regulated positively by DHH, lithium- induced signaling, and activin, and negatively by TGF-β, PDGFBB, and FGF2. DHH functioned as a commitment factor, inducing the transition of stem cells to the progenitor stage and thus into the Leydig cell lineage. Additionally, CD90 (Thy1) was found to be a unique stem cell surface marker that was used to obtain purified stem cells by flow cytometry.

Our study was designed to spatially assess the regulation potential of watershed water-land resources, and determine its regulation objectives for subsequent optimal allocation. We centred around the Yiluo River Basin, being a soil conservation demonstration plot in the middle course of the Yellow River Basin. In accordance with the criteria of the green development, the high efficiency and the safety, the evaluation system of water-land resources regulation potential was established. Then the fuzzy comprehensive evaluation method was applied to assess the watershed water-land resources during 2001–2020 and 2030–2050, and the evaluation results were represented by the eigenvalue T. The research findings pointed out that the assessment grade of watershed water-land resources was mainly moderate and better state, and presented significant spatial agglomeration features in the historical and forecast years. In the context of dual influences of climatic variation and water-land resources exploitation, the assessment eigenvalue T of watershed water-land resources increased by 6% in the historical periods, and decreased by 4.57 ∼ 7.74% in the future periods. Based on the assessment results, it was recommended to restrict the water-land resources exploitation in the upstream, control the soil erosion in the midstream, and coordinate the relationship between the exploitation and protection of water-land resources in the lower reaches. Our study offers scientific evidence for the elaborate regulation of watershed water-land resources.

Structured light has proven useful for numerous photonic applications. However, the current use of structured light in optical fiber science and technology is severely limited by mode mixing or by ...the lack of optical elements that can be integrated onto fiber end-faces for wavefront engineering, and hence generation of structured light is still handled outside the fiber via bulky optics in free space. We report a metafiber platform capable of creating arbitrarily structured light on the hybrid-order Poincaré sphere. Polymeric metasurfaces, with unleashed height degree of freedom and a greatly expanded 3D meta-atom library, were 3D laser nanoprinted and interfaced with polarization-maintaining single-mode fibers. Multiple metasurfaces were interfaced on the fiber end-faces, transforming the fiber output into different structured-light fields, including cylindrical vector beams, circularly polarized vortex beams, and arbitrary vector field. Our work provides a paradigm for advancing optical fiber science and technology towards fiber-integrated light shaping, which may find important applications in fiber communications, fiber lasers and sensors, endoscopic imaging, fiber lithography, and lab-on-fiber technology.
