A series of B-ring, halo-substituted chalcones and azachalcones were synthesized to evaluate and compare their anti-inflammatory activity. Mouse BALB/c macrophage RAW 264.7 were pre-treated with 10 ...μg/mL of each compound for one hour before induction of inflammation by lipopolysaccharide (1 μg/mL) for 6 h. Some halo-chalcones and -azachalcones suppressed expression of pro-inflammatory factors toll-like receptor 4 (TLR4), IκB-α, transcription factor p65, interleukine 1β (IL-1β), IL-6, tumor necrosis factor α (TNF-α), and cyclooxygenase 2 (COX-2). The present results showed that the synthetic halo-azachalcones exhibited more significant inhibition than halo-chalcones. Therefore, the nitrogen atom in this series of azachalcones must play a more crucial role than the corresponding C-2 hydroxyl group of chalcones in biological activity. Our findings will lay the background for the future development of anti-inflammatory nutraceuticals.
The aim of this study was to apply deep excavation behavior prediction models in the geotechnical field to establish localized soil parameters for gravel layers. Common software tools, including ...PLAXIS and SoilWorks, were used extensively. Monitoring data from deep excavation cases related to gravel layers in the Xindian area of Taiwan were collected. In the background analysis, the deformation of the retaining walls was used instead of parameters typically used in deep excavation analysis. This was performed to provide the ideal range recommendations for the input parameters when conducting a numerical simulation analysis of the Xindian District stratum or similar strata. The assessment results show that when setting the fifth layer of gravel to SPT-N = 100, PLAXIS suggested a soil elastic modulus range of 7840 N to 9800 N per square meter (kN/m2), while SoilWorks recommended a range of 2450 N to 3430 N per square meter (kN/m2). These ranges allow for a reasonable estimation of the maximum wall deformation during the final excavation stage. Based on the research findings, it is recommended that when conducting an excavation analysis in gravel layers in the Xindian area of Taiwan or in similar strata, engineers should refer to the abovementioned recommended ranges when selecting the soil elastic modulus for different software programs. This will enhance the accuracy of the deformation predictions during the final excavation stage.
The ability of organic photovoltaics (OPVs) to be deposited on flexible substrates by roll-to-roll (R2R) processes is highly attractive for rapid mass production. Many research teams have ...demonstrated the great potential of flexible OPVs. However, the fabrication of R2R-coated OPVs is quite limited. There is still a performance gap between the R2R flexible OPVs and the rigid OPVs. In this study, we demonstrate the promising photovoltaic characteristics of flexible OPVs fabricated from blends of low bandgap polymer poly(2,6-(4,8-bis(5-(2-ethylhexyl)thiophen-2-yl)-benzo1,2-b:4,5-b′dithiophene))-alt-(5,5-(1′,3′-di-2-thienyl-5′,7′-bis(2-ethylhexyl)benzo1′,2′-c:4′,5′-c′dithiophene-4,8-dione) (PBDB-T) and non-fullerene 3,9-bis(2-methylene-(3-(1,1-dicyanomethylene)-indanone))-5,5,11,11-tetrakis(4-hexylphenyl)-dithieno2,3-d:2′,3′-d′-s-indaceno1,2-b:5,6-b′dithiophene (ITIC). We successfully R2R slot-die coated the flexible OPVs with high power conversion efficiency (PCE) of over 8.9% under irradiation of simulated sunlight. Our results indicate that the processing parameters significantly affect the PCE of R2R flexible OPVs. By adjusting the amount of solvent additive and processing temperature, as well as optimizing thermal annealing conditions, the high PCE of R2R slot-die coated OPVs can be obtained. These results provide significant insights into the fundamentals of highly efficient OPVs for the R2R slot-die coating process.
This manuscript delineates an innovative artificial intelligence-based methodology for forecasting the displacement of retaining walls due to extensive deep excavation processes. In our selection of ...17 training cases, we strategically chose a wall configuration that was not influenced by the corner effects. This careful selection was conducted with the intention of ensuring that each deep excavation instance included in our study was supported symmetrically, thereby streamlining the analysis in the ensuing phases. Our proposed multilayer functional-link network demonstrates superior performance over the traditional backpropagation neural network (BPNN), excelling in the precise prediction of displacements at predetermined observation points, peak wall displacements, and their respective locations. Notably, the predictive accuracy of our advanced model surpassed that of the conventional BPNN and RIDO assessment tools by a substantial 5%. The network process model formulated through this research offers a valuable reference for future implementations in diverse geographical settings. Furthermore, by utilizing local datasets for the training, testing, and validation phases, our system ensures the effective and accurate execution of displacement predictions.
Lead halide perovskite solar cells (PVSCs) have potential toward commercialization because of their high efficiency and low cost. The hole transport layer (HTL) of p-i-n perovskite solar cell is ...usually made of NiOX. However, the NiOX needs to be processed at 300 °C for 15 min for good hole transport property. This long heating time prohibits the development of continuous commercial process. Thus, a rapid heating process for the NiOX film deposition is critical to realize the commercialization of PVSCs in the future. In this study, we develop a facile method to obtain high quality NiOX films annealed by NIR in a short time of 50 s. A short-wave NIR lamp at 2500 K was used to systematically investigate the effect of NIR intensity on the film quality of sol-gel NiOX. The PVSCs fabricated from NIR-annealed NiOX (NIR-NiOX) film show a comparable power conversion efficiency (PCE) to those fabricated from traditional hot-plate annealed-NiOX (HP-NiOX). In addition, the NIR annealed cobalt-doped NiOX (NIR-Co:NiOX) was synthesized to replace pristine NIR-NiOX. The PCE of PVSCs fabricated from this new NiOX film can be increased from 15.99% to 17.77%, which is due to the efficient hole extraction, less charge accumulation, and reducing Voc loss resulting from the improved hole mobility, reduced interface resistance and well-matched work function. Our study paves a way to fulfill the requirements of low cost and low energy consumption of large scale production of high efficiency PVSCs.

This study focuses on the hole transport layer of molybdenum trioxide (MoO3) for inverted bulk heterojunction (BHJ) organic photovoltaics (OPVs), which were fabricated using a combination of a spray ...coating and low-temperature annealing process as an alternative to the thermal evaporation process. To achieve a good coating quality of the sprayed film, the solvent used for solution-processed MoO3 (S-MoO3) should be well prepared. Isopropanol (IPA) is added to the as-prepared S-MoO3 solution to control its concentration. MoO3 solutions at concentrations of 5 mg/mL and 1 mg/mL were used for the spray coating process. The power conversion efficiency (PCE) depends on the concentration of the MoO3 solution and the spray coating process parameters of the MoO3 film, such as flow flux, spray cycles, and film thickness. The results of devices fabricated from solution-processed MoO3 with various spray fluxes show a lower PCE than that based on thermally evaporated MoO3 (T-MoO3) due to a limiting FF, which gradually increases with decreasing spray cycles. The highest PCE of 2.8% can be achieved with a 1 mg/mL concentration of MoO3 solution at the sprayed flux of 0.2 mL/min sprayed for one cycle. Additionally, S-MoO3 demonstrates excellent stability. Even without any encapsulation, OPVs can retain 90% of their initial PCE after 1300 h in a nitrogen-filled glove box and under ambient air conditions. The results represent an evaluation of the feasibility of solution-processed HTL, which could be employed for a large-area mass production method.
Small extracellular vesicles (sEVs) are cell-released vesicles ranging from 30-150nm in size. They have garnered increasing attention because of their potential for both the diagnosis and treatment ...of disease. The diversity of sEVs derives from their biological composition and cargo content. Currently, the isolation of sEV subpopulations is primarily based on bio-physical and affinity-based approaches. Since a standardized definition for sEV subpopulations is yet to be fully established, it is important to further investigate the correlation between the biomolecular composition of sEVs and their physical properties. In this study, we employed a platform combining single-vesicle surface-enhanced Raman spectroscopy (SERS) and machine learning to examine individual sEVs isolated by size-exclusion chromatography (SEC). The biomolecular composition of each vesicle examined was reflected by its corresponding SERS spectral features (biomolecular “fingerprints”), with their roots in the composition of their collective Raman-active bonds. Origins of the SERS spectral features were validated through a comparative analysis between SERS and mass spectrometry (MS). SERS fingerprinting of individual vesicles was effective in overcoming the challenges posed by EV population averaging, allowing for the possibility of analyzing the variations in biomolecular composition between the vesicles of similar and/or different sizes. Using this approach, we uncovered that each of the size-based fractions of sEVs contained particles with predominantly similar SERS spectral features. Indeed, more than 84% of the vesicles residing within a particular group were clearly distinguishable from that of the other EV sub-populations, despite some spectral variations within each sub-population. Our results suggest the possibility that size-based EV fractionation methods produce samples where similarly eluted sEVs are correlated with their respective biochemical contents, as reflected by their SERS spectra. Our findings therefore highlight the possibility that the biogenesis and respective biological functionalities of the various sEV fractions may be inherently different.

Starting enzyme replacement therapy (ERT) before severe irreversible muscular damage occurs is important in infantile-onset Pompe disease (IOPD). This long-term follow-up study demonstrates our ...diagnostic and treatment strategies for IOPD and compares our clinical outcomes with those of other medical centres.
In this long-term follow-up study, we analysed the outcomes of very early ERT with premedication hydrocortisone in patients with IOPD. Out of 1 228 539 infants screened between 1 January 2010 and 28 February 2021, 33 newborns had confirmed IOPD in Taipei Veterans General Hospital. Twenty-six were regularly treated and monitored at Taipei Veterans General Hospital. Echocardiographic parameters, biomarkers, IgG antibodies against alglucosidase alpha, pulmonary function variables and developmental status were all assessed regularly over an average follow-up duration of 6.18±3.14 years. We compared the long-term treatment outcomes of our patients with those of other research groups.
The average age at ERT initiation was 9.75±3.17 days for patients with classic IOPD. The average of the latest antialglucosidase alpha IgG titre was 669.23±1159.23. All enrolled patients had normal heart sizes, motor milestones, cognitive function and pulmonary function that were near-normal to normal. Compared with patients in other studies, our patients had better outcomes in all aspects.
Very early ERT using our rapid diagnostic and treatment strategy enabled our patients with IOPD to have better outcomes than patients in other medical centres.

This study aimed to investigate the absolute bioavailability, tissue distribution, and excretion of Erinacine S in
mycelia in eight-week old Sprague-Dawley rats. After oral administration and intravenous administration of 2.395 g/kg body weight of the
mycelia extract (equivalent to 50 mg/kg body weight Erinacine S) and 5 mg/kg of Erinacine S, respectively, the absolute bioavailability was estimated as 15.13%. In addition, Erinacine S was extensively distributed in organs such as brain, heart, lung, liver, kidney, stomach, small intestine, and large intestine. The maximum concentration of Erinacine S was observed in the stomach, 2 h after the oral administration of
mycelia extract, whereas the maximum amount of Erinacine S found in other tissues were seen after 8 h. Total amount of unconverted Erinacine S eliminated in feces and urine in 24 h was 0.1% of the oral dosage administrated. This study is the first to show that Erinacine S can penetrate the blood-brain barrier of rats and thus support the development of
mycelia, for the treatment of neurological diseases.
The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused the pandemic of the coronavirus induced disease 2019 (COVID-19) with evolving variants of concern. It remains urgent to ...identify novel approaches against broad strains of SARS-CoV-2, which infect host cells via the entry receptor angiotensin-converting enzyme 2 (ACE2). Herein, we report an increase in circulating extracellular vesicles (EVs) that express ACE2 (evACE2) in plasma of COVID-19 patients, which levels are associated with severe pathogenesis. Importantly, evACE2 isolated from human plasma or cells neutralizes SARS-CoV-2 infection by competing with cellular ACE2. Compared to vesicle-free recombinant human ACE2 (rhACE2), evACE2 shows a 135-fold higher potency in blocking the binding of the viral spike protein RBD, and a 60- to 80-fold higher efficacy in preventing infections by both pseudotyped and authentic SARS-CoV-2. Consistently, evACE2 protects the hACE2 transgenic mice from SARS-CoV-2-induced lung injury and mortality. Furthermore, evACE2 inhibits the infection of SARS-CoV-2 variants (α, β, and δ) with equal or higher potency than for the wildtype strain, supporting a broad-spectrum antiviral mechanism of evACE2 for therapeutic development to block the infection of existing and future coronaviruses that use the ACE2 receptor.
