This study utilized a colorimeter to determine the color values of 23 beauty tea (BT) samples, the color and the taste characteristics were also quantitatively described through ultraviolet–visible (UV–Vis) spectroscopy and taste equivalent quantification. Furthermore, metabolomic analysis was conducted by using ultra-high-performance liquid chromatography-mass spectrometry (UPLC-MS). Correlation analysis was employed to preliminarily identify the compounds that contribute to the color and taste of BT infusion. Finally, the contributing compounds were further determined through verification experiment. The results showed that within a certain range, as the color of BT infusion deepened, the taste became stronger, more bitter and astringent, while on the contrary, it became sweeter and mellower. Theaflavins, kaempferol, astragalin, and 5-p-coumaroylquinic acid influenced both the color and taste of the BT infusion. Gallic acid was also determined as a contributor to the color. This study provides new insights into research on tea quality in infusion color and taste aspects.
Seismic damage of spatial structures of rectangular plan with RC substructures was observed in several earthquakes, especially in the RC substructures. In order to reduce the seismic damage ...potential, a new structural configuration of spatial structure of rectangular plan is proposed, the substructures of which are composed of the steel substructure and the RC substructure. The latter only bears the vertical load of roof by the arrangement of horizontal sliding bearings between the roof and the RC substructure. The pushover analyses are performed on a steel braced frame and an RC frame with similar lateral stiffness, and the results show that the lateral capacity of the steel structure is much larger than those of the RC structures. A spatial structure of rectangular plan with two different substructures is designed according to Chinese structural designing codes. Seismic time history analyses are carried out for the spatial structure under five ground motions. The results show that the damage mainly concentrates on the substructures, and the seismic performance of the structure with steel and RC substructures is much better than that of the structure with RC substructures.

We report an experimental demonstration of optical single-sideband (SSB) modulation in 60-GHz radio-over-fiber systems based on an injection-locked distributed-feedback (DFB) laser. Two heterodyning ...optical modes with 60-GHz spacing are injected into the DFB laser and one of them is used to lock the DFB laser. When the DFB laser is directly modulated, the modulation index of the locked mode is 15 dB larger than that of the unlocked one. The 2.5-Gb/s data transmission at 60 GHz is successfully achieved over 50-km standard single-mode fiber using the proposed SSB scheme.
Phenylketonuria (PKU), a deficiency in the activity of the enzyme phenylalanine hydroxylase, leads to toxic levels of phenylalanine (Phe) in the blood and brain. Pegvaliase (recombinant Anabaena ...variabilis phenylalanine ammonia lyase conjugated with polyethylene glycol) is approved to manage PKU in patients aged greater than or equal to 18 years in the United States and in patients aged greater than or equal to 16 years in the European Union. Pharmacokinetic, pharmacodynamic, and immunogenicity results from five open‐label pegvaliase trials were assessed. Studies with induction/titration/maintenance (I/T/M) dosing regimens demonstrated pharmacokinetic stabilization and sustained efficacy associated with maintenance doses (20, 40, or 60 mg/day). Immune‐mediated pegvaliase clearance was high during induction/titration phases when the early immune response was peaking. The combination of low drug dosage and high drug clearance led to low drug exposure and minimal decreases in blood Phe levels during induction/titration. Higher drug exposure and substantial reductions in blood Phe levels were observed later in treatment as drug clearance was reduced due to the maturation of the immune response, which allowed for increased dosing to target levels. The incidence of hypersensitivity reactions was temporally associated with the peaking of the early antidrug immune response and decreased with time as immune response matured after the first 6 months of treatment. These results support an I/T/M dosing regimen and suggest a strategy for administration of other nonhuman biologics to achieve efficacy and improve tolerability.
The hard bottom layer of a paddy field has a great influence on the driving stability and the operation quality and efficiency of intelligent farm machinery. For paddy field machinery, continuous ...improvements in the accuracy and operation efficiency of unmanned precision operations are needed to realize unmanned rice farming. In the context of unmanned farm machinery operation, the complicated hard bottom layer situation makes it difficult to quantify the local characteristics of paddy fields. In this paper, an unmanned direct rice seeding machine chassis is used to maneuver the operation field and collect the hard bottom layer information simultaneously. This information is used to design a data processing method that automatically calibrates the sensor installation error and performs abnormal value rejection and 3D sample curve denoising of the contour trajectory. A hard bottom layer surface profile evaluation method based on the local sliding surface roughness is also proposed. The local characteristics of the hard bottom layer were quantified, and the results from the test plots showed that the mean value of the local roughness was 0.0065, where 68.27% of the plots were distributed in the variation range of 0.0042~0.0087 and 99.73% were distributed in the variation range of 0~0.0133. Using the test field data, the surface roughness features were verified to describe the variability in representative working conditions, such as the transport, downfield, operation, and trapping of unmanned intelligent farm machinery. When driving intelligent farm machinery, the proposed method for quantifying local features of the hard bottom layer can provide feedback on the local environmental features at any given position of the machinery. The method also provides a reference for the design optimization of unmanned systems, which can help to realize speed adaption and improve the local path tracking control accuracy of smart farming machines.
In order to improve the soil preparation quality, a wide puddling and flatting machine for paddy field was designed with the functionality of tillage, soil crushing, puddling, burying stubble and ...flatting in one-time operation, which can be equipped on tractors with high power. This study focused on the design and analysis of key components of the machine including puddling equipment, flat shovel and balance structure. Optimal results with field trials and response surface analysis showed that the machine can operate optimally when the working speed, puddling depth and angle of flat shovel were 1.05 km/h, 16 cm and 31.30°, respectively. The surface flatness standard deviation was 3.7 cm, and slurry degree was 1.03 g/cm^sup 3^. Field validation test results were consistent with the optimal parameters, which was able to meet agronomic requirements of mechanical planting in China.

We report durable efficacy, long-term safety, and clinical and biologic results in 15 adults with severe hemophilia A (factor VIII level, ≤1 IU per deciliter) who had received a single infusion of AAV5-hFVIII-SQ at various dose levels. We evaluated the factor VIII level, annualized rate of bleeding events, use of factor VIII, safety, expression kinetics, and biologic markers of AAV transduction for up to 3 years.
Three years after infusion, two participants (one who had received 6×10
vector genomes vg per kilogram of body weight and one who had received 2×10
vg per kilogram) had factor VIII expression of less than 1 IU per deciliter, as assessed on chromogenic assay. Seven participants (who had received 6×10
vg per kilogram) had a median factor VIII expression of 20 IU per deciliter; the median number of annualized treated bleeding events was 0, and the median use of exogenous factor VIII was reduced from 138.5 infusions to 0 infusions per year. Bleeding in all target joints (major joints with ≥3 bleeding events within 6 months) in this cohort resolved (≤2 bleeding events within 12 months). Two years after infusion, six participants (who had received 4×10
vg per kilogram) had a median factor VIII expression of 13 IU per deciliter; the median annualized rate of bleeding events was 0, and the median use of factor VIII was reduced from 155.5 infusions to 0.5 infusions per year. Bleeding in target joints resolved in five of six participants. The factor VIII pharmacodynamic profiles reflected cellular turnover in the blood and molecular events leading to episomal DNA stabilization for persistent expression, findings that are consistent with previous observations in two model systems. Transgene-derived human factor VIII (hFVIII) protein activity mirrored native hFVIII in hemostatic ability. No inhibitor development, thromboses, deaths, or persistent changes in liver-function tests were observed.
Gene therapy with AAV5-hFVIII-SQ vector in participants with hemophilia A resulted in sustained, clinically relevant benefit, as measured by a substantial reduction in annualized rates of bleeding events and complete cessation of prophylactic factor VIII use in all participants who had received 4×10
vg per kilogram or 6×10
