Alzheimer's disease (AD) is an aging-related neurological disorder characterized by synaptic loss and dementia. Wnt/β-catenin signaling is an essential signal transduction pathway that regulates ...numerous cellular processes including cell survival. In brain, Wnt/β-catenin signaling is not only crucial for neuronal survival and neurogenesis, but it plays important roles in regulating synaptic plasticity and blood-brain barrier integrity and function. Moreover, activation of Wnt/β-catenin signaling inhibits amyloid-β production and tau protein hyperphosphorylation in the brain. Critically, Wnt/β-catenin signaling is greatly suppressed in AD brain via multiple pathogenic mechanisms. As such, restoring Wnt/β-catenin signaling represents a unique opportunity for the rational design of novel AD therapies.
In the present work, we synthesized and characterized an electrocatalyst consisting of sub-nanometric Pt clusters uniformly dispersed on a TiO 2 support. X-ray photoelectron spectra (XPS) and X-ray ...adsorption fine structure (XAFS) data demonstrate that these sub-nanometric Pt clusters are in a highly oxidized state and possess two localized Pt–O coordination structures. The Pt–O bonds between the oxidized Pt clusters and the TiO 2 give rise to a strong metal–support interaction (SMSI). When applied to the hydrogen evolution reaction (HER), this catalyst exhibits significantly enhanced catalytic activity (increased by a factor of up to 8.4) and enhanced stability compared with the state-of-the-art commercial Pt/C catalysts. Particularly, the additional XPS and XAFS characterizations of the catalyst after long-term electrolysis demonstrate the absence of metallic Pt species, confirming that the catalytic active site comes from the oxidized Pt clusters rather than from the the metallic Pt species. This improved performance is considered to be induced by the unique electronic structure of the oxidized Pt clusters and by the SMSI. Based on the results of density functional theory calculations, the 5d orbital of the oxidized Pt cluster atoms appears to hybridize with the H 1s orbital to form weak Pt–H valence bonds, leading to a Δ G (relative free energy) value of approximately zero eV for H* absorption. This effect explains the mechanism responsible for the excellent catalytic activity of these oxidized Pt clusters for the HER. This work therefore provides important insights into the role of oxidized Pt clusters as an HER electrocatalyst. The evident stabilization of the oxidized Pt clusters on TiO 2 supports via the charge-transfer mechanism provides a useful approach for improving the durability of electrocatalysts that may be applicable to other noble metal/support systems.
Abstract The rapid development of new anticancer drugs that are safe and effective is a common goal shared by basic scientists, clinicians and patients. The current review discusses one such agent, ...namely niclosamide, which has been used in the clinic for the treatment of intestinal parasite infections. Recent studies repeatedly identified niclosamide as a potential anticancer agent by various high-throughput screening campaigns. Niclosamide not only inhibits the Wnt/β-catenin, mTORC1, STAT3, NF-κB and Notch signaling pathways, but also targets mitochondria in cancer cells to induce cell cycle arrest, growth inhibition and apoptosis. A number of studies have established the anticancer activities of niclosamide in both in vitro and in vivo models. Moreover, the inhibitory effects of niclosamide on cancer stem cells provide further evidence for its consideration as a promising drug for cancer therapy. This article reviews various aspects of niclosamide as they relate to its efficacy against cancer and associated molecular mechanisms.
Volatile organic compounds (VOCs) and flavor characteristics of
Tratt. (RR) and
(RS) were analyzed using headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry ...(HS-SPME-GC-MS). The flavor network was constructed by combining relative odor activity values (ROAVs), and the signature differential flavor components were screened using orthogonal partial least squares discriminant analysis (OPLS-DA) and random forest (RF). The results showed that 61 VOCs were detected in both RR and RS: 48 in RR, and 26 in RS. There were six key flavor components (ROAVs ≥ 1) in RR, namely nonanal, ethyl butanoate, ethyl hexanoate, (
)-3-hexen-1-yl acetate, ethyl caprylate, and styrene, among which ethyl butanoate had the highest contribution, whereas there were eight key flavor components (ROAVs ≥ 1) in RS, namely 2-nonanol, (
)-2-hexenal, nonanal, methyl salicylate, β-ocimene, caryophyllene, α-ionone, and styrene, among which nonanal contributed the most to RS. The flavor of RR is primarily fruity, sweet, green banana, and waxy, while the flavor of RS is primarily sweet and floral. In addition, OPLS-DA and RF suggested that (
)-2-hexenal, ethyl caprylate, β-ocimene, and ethyl butanoate could be the signature differential flavor components for distinguishing between RR and RS. In this study, the differences in VOCs between RR and RS were analyzed to provide a basis for further development and utilization.
Due to the excellent theoretical capacity, silicon-based electrodes have been extensively studied to develop high energy-density lithium-ion batteries (LIBs). Nevertheless, the large volume expansion ...and unfavorable interface seriously hamper its application, and the underlying physics behind are still unclear. In this work, using in-situ environment scanning electron microscopy (ESEM), the morphological evolution of composite silicon-graphite electrodes with different ionic liquids as electrolytes are compared in the range of 20 °C–60 °C. Surprisingly, combining the microstructure and surface reaction products from transmission electron microscope (TEM) and X-ray photoelectron spectroscopy (XPS), it finds out that the fully reaction of lithium-silicon does not generate huge volume expansion, while the side reaction between graphite and electrolyte decomposition products causes dramatic volume expansion and hinders the further lithiation of silicon. On the other hand, the electrolyte which is capable of rapidly releasing F− to form LiF-rich solid electrolyte interphase (SEI) is favorable to maintain the structure integrity and cycling performance. This work casts new understanding from the perspective of intrinsic reaction between electrode and ionic liquid electrolyte, which suggests that the practical application of silicon-based electrodes with appropriate electrolyte is a promising route for high energy-density LIBs.

The Wnt/β-catenin signaling pathway is important for tumor initiation and progression. The low density lipoprotein receptor-related protein-6 (LRP6) is an essential Wnt co-receptor for Wnt/β-catenin ...signaling and represents a promising anticancer target. Recently, the antihelminthic drug, niclosamide was found to inhibit Wnt/β-catenin signaling, although the mechanism was not well defined. We found that niclosamide was able to suppress LRP6 expression and phosphorylation, block Wnt3A-induced β-catenin accumulation, and inhibit Wnt/β-catenin signaling in HEK293 cells. Furthermore, the inhibitory effects of niclosamide on LRP6 expression/phosphorylation and Wnt/β-catenin signaling were conformed in human prostate PC-3 and DU145 and breast MDA-MB-231 and T-47D cancer cells. Moreover, we showed that the mechanism by which niclosamide suppressed LRP6 resulted from increased degradation as evident by a shorter half-life. Finally, we demonstrated that niclosamide was able to induce cancer cell apoptosis, and displayed excellent anticancer activity with IC(50) values less than 1 µM for prostate PC-3 and DU145 and breast MDA-MB-231 and T-47D cancer cells. The IC(50) values are comparable to those shown to suppress the activities of Wnt/β-catenin signaling in prostate and breast cancer cells. Our data indicate that niclosamide is a unique small molecule Wnt/β-catenin signaling inhibitor targeting the Wnt co-receptor LRP6 on the cell surface, and that niclosamide has a potential to be developed a novel chemopreventive or therapeutic agent for human prostate and breast cancer.

The low-density lipoprotein receptor-related protein 1 (LRP1) is a multifunctional endocytic receptor involved in the metabolism of various extracellular ligands, including proteinases, that play ...critical roles in tumor invasion. Although several studies have shown an increased expression of LRP1 in cancer cells, its function in tumor development and progression remains largely unclear. Here, we reveal a novel mechanism by which LRP1 induces the expression of matrix metalloproteinase 2 (MMP2) and MMP9 and thereby promotes the migration and invasion of human glioblastoma U87 cells. Knockdown of LRP1 expression greatly decreased U87 cell migration and invasion, which was rescued by the forced expression of a functional LRP1 minireceptor. Inhibition of ligand binding to LRP1 by a specific antagonist, receptor-associated protein, also led to reduced cancer cell migration and invasion. Because MMPs play critical roles in cancer cell migration and invasion, we examined the expression of several MMPs and found that the expression of functional MMP2 and MMP9 was selectively decreased in LRP1 knockdown cells. More importantly, decreased cell migration and invasion of LRP1 knockdown cells were completely rescued by exogenous expression of MMP2 or MMP9, suggesting that these MMPs are likely downstream targets of LRP1-mediated signaling. We further show that the level of phosphorylated extracellular signal-regulated kinase (ERK) was significantly decreased in LRP1-silenced cells, suggesting that ERK is a potential mediator of LRP1-regulated MMP2 and MMP9 expression in U87 cells. Together, our data strongly suggest that LRP1 promotes glioblastoma cell migration and invasion by regulating the expression and function of MMP2 and MMP9 perhaps via an ERK-dependent signaling pathway.
Compared with straight tunnels, over-excavation occurs on the inner side of the curved section during shield construction of small radius curved tunnels, and the disturbance to the ground surface and ...mechanical properties of surrounding rock and segment are more severe. This paper establishes the numerical models of small radius curve tunnels and straight tunnels to study the characteristics of surface deformation caused by the shield excavation of small radius curved tunnels and the influence of shield construction parameters on ground settlement, surrounding rock deformation, and segment force. The maximum error between the numerical simulation results and the measured surface settlement curve is 7.3%, which is in good agreement. The results show that: (1) The maximum value of the surface settlement of the small radius curve tunnel appears inside the curve section, and with the decrease in the curve radius, the surface settlement increases, and the distance between the peak settlement point and the tunnel center is larger. (2) When the curve radius of the tunnel is smaller, the lateral displacement of the ground surface moves farther to the inner side, and the range of soil mass with lateral displacement in the inner side is also wider. (3) Increasing the heading face pressure and grouting pressure can reduce surface settlement, but the heading face pressure should not exceed 350 kPa, and the grouting pressure should not exceed 250 kPa. (4) When the curve radius is smaller, the deformation of surrounding rock and the segment stress is larger.
Hyperlipidemia is a prevalent chronic metabolic disease that severely affects human health. Currently, commonly used clinical therapeutic drugs are prone to drug dependence and toxic side effects. ...Dietary intervention for treating chronic metabolic diseases has received widespread attention. Rosa sterilis is a characteristic fruit tree in China whose fruits are rich in flavonoids, which have been shown to have a therapeutic effect on hyperlipidemia; however, their exact molecular mechanism of action remains unclear. Therefore, this study aimed to investigate the therapeutic effects of R. sterilis total flavonoid extract (RS) on hyperlipidemia and its possible mechanisms. A hyperlipidemic zebrafish model was established using egg yolk powder and then treated with RS to observe changes in the integral optical density in the tail vessels. Network pharmacology and molecular docking were used to investigate the potential mechanism of action of RS for the treatment of hyperlipidemia. The results showed that RS exhibited favorable hypolipidemic effects on zebrafish in the concentration range of 3.0–30.0 μg/mL in a dose-dependent manner. Topological and molecular docking analyses identified HSP90AA1, PPARA, and MMP9 as key targets for hypolipidemic effects, which were exerted mainly through lipolytic regulation of adipocytes and lipids; pathway analysis revealed enrichment in atherosclerosis, chemical carcinogenic-receptor activation pathways in cancers, and proteoglycans in prostate cancer and other cancers. Mover, chinensinaphthol possessed higher content and better target binding ability, which suggested that chinensinaphthol might be an important component of RS with hypolipidemic active function. These findings provide a direction for further research on RS interventions for the treatment of hyperlipidemia.
This study compared the clinical efficacy and safety of laparoscopic versus open resection for hilar cholangiocarcinoma (HCCA) and analyzed potential prognostic factors.
The study included patients ...who underwent HCCA resection at our center from March 2012 to February 2022. Perioperative complications and postoperative prognosis were compared between the laparoscopic surgery (LS) and open surgery (OS) groups.
After screening 313 HCCA patients, 68 patients were eligible for the study in the LS group (n = 40) and OS group (n = 28). Kaplan-Meier survival curve analysis revealed that overall survival > 2 years and 3-year disease-free survival (DFS) were more common in the LS than OS group, but the rate of 2-year DFS was lower in the LS group than OS group. Cox multivariate regression analysis revealed age (< 65 years), radical resection, and postoperative adjuvant therapy were associated with reduced risk of death (hazard ratio HR = 0.380, 95% confidence interval CI = 0.150-0.940, P = 0.036; HR = 0.080, 95% CI = 0.010-0.710, P = 0.024 and HR = 0.380, 95% CI = 0.150-0.960, P = 0.040), whereas preoperative biliary drainage was an independent factor associated with increased risk of death (HR = 2.810, 95% CI = 1.130-6.950, P = 0.026). Perineuronal invasion was identified as an independent risk factor affecting DFS (HR = 5.180, 95% CI = 1.170-22.960, P = 0.030).
Compared with OS, laparoscopic HCCA resection does not significantly differ in terms of clinical efficacy. Age (<65 years), radical resection, and postoperative adjuvant therapy reduce the risk of death, and preoperative biliary drainage increases the risk of death.
