The basic helix-loop-helix transcription factor (TF) Bhlhe40 is emerging as a key regulator of immunity during infection, autoimmunity, and inflammatory conditions. We describe the roles of Bhlhe40 ...in the circulating and tissue-resident arms of the immune system, with emphasis on recent work on the regulation of cytokine production and proliferation. We explore the mechanisms behind these functions in mouse models and human cells, including interactions with other TFs, and propose that Bhlhe40 is a central mediator of both inflammation and pathogen control, as well as a crucial regulator of a growing number of tissue-resident leukocyte populations. Finally, we suggest areas for further study that may advance our understanding of immunity and disease.
Bhlhe40 is a key regulator of cytokine production by human and mouse T cells.Bhlhe40 can drive inflammation and enables typical immune responses in autoimmunity, transplantation, cancer, and infection.Bhlhe40 is a tissue-specific regulator of murine tissue-resident macrophage proliferation.Bhlhe40 and Bhlhe41 are required by mouse B1a cells for normal proliferation, survival, and BCR repertoire.Bhlhe40 is essential for mitochondrial metabolism in mouse tissue-resident memory CD8+ T cells and tumor-infiltrating lymphocytes.Bhlhe40 contributes to controlling the balance between inflammation and disease tolerance, and promotes highly inflammatory responses.
Summary
Scour monitoring is crucial for providing early warning of bridge safety and extending the knowledge of scour process. Traditional methods that rely on underwater instruments encounter ...difficulties in installation and operation in harsh fluvial environment. Time domain reflectometry (TDR)‐based scour sensing wire was recently introduced as a promising technique to improve field deployment and durability. This study focused on further improving the concept of TDR sensing wire for better sensor package and performance. An innovative bundled bottom‐up sensing cable was proposed to improve sensor durability and avoid the adverse effect of sensor fouling. The sensor durability is enhanced by twisting two sets of steel strands (as two opposing electrodes for the waveguide) around a coaxial cable into a composite sensing cable. The inner coaxial cable is introduced and armored by the sensing steel strands to direct TDR pulse to the bottom end before connecting to the sensing waveguide formed by steel strands for bottom‐up sensing. Furthermore, taking advantage of a new data reduction method by using time‐lapse differential waveform, robustness of system calibration and scour estimation is further enhanced. Experimental results revealed the optimal design of bundled TDR sensing cable, effectiveness of the bottom‐up sensing approach, and better performance of the differential waveform method.

LIBSVM ...has gained wide popularity in machine learning and many other areas. In this article, we present all implementation details of LIBSVM. Issues such as solving SVM optimization problems theoretical convergence multiclass classification probability estimates and parameter selection are discussed in detail.
•A comprehensive framework consists of TDR installation, data acquisition, post-processing, and warning index.•The old colluvium slope at Lushan was characterized by TDR as a buckling fold.•Creep ...found by TDR contributed to the large surface displacement at Lushan.•Rebounded deformation was captured by TDR’s high temporal and spatial resolution at Jufen.•Simplified index for early warning was substantiated by field implementation.
Due to the climate change and hazardous geological environment in Taiwan, real-time landslide monitoring for safety of adjacent tribes or infrastructures is much conspicuous. Time domain reflectometry (TDR) is considered as valuable technology for continuously capturing dynamics and locations of shear deformations with high temporal and spatial coverages. In this study, a comprehensive framework was proposed by integrating up-to-date procedure of installation guide, data acquisition, sliding deformation identification, and early warning processing. The advantages of the proposed framework were then examined and substantiated via field cases. The first case is at a large colluvium slope in Lushan, central Taiwan. The colluvium is still active after heavy rainfall events, but limited evidence of sliding characteristic was collected previously by manual inclinometer logging. Therefore, the TDR framework was deployed and multiple landslide surfaces at great depths were timely observed. Movement of colluvium mass was thereby interpreted as a buckling fold. Additionally, creep occurred among the colluvium layer during the monsoon season because of strength weakening influenced by pore pressure fluctuations. Monitoring results of the second landslide case in Jufan, northern Taiwan, reported the dynamics of a thin sliding plane captured with TDR high temporal resolution. This unique behavior was further applied for devising of a novel early-warning strategy in terms of time-variation of TDR response, validating that the comprehensive framework of TDR landslide can effectively contribute to early warning.
For promoting the successful aging of elderly residents of Chang Gung Silver Village in Taiwan, five interactive exergames were developed to promote the well-being of the elderly. The exergames ...included both physical games and cognitive games, and were implemented using various computer-based technologies in the Chang Gung Silver Village. The exergames were trialed by 39 elderly residents (15 male, 24 female; mean age 79.5 ± 17.5 years) of Chang Gung Silver Village. Following the trials, the participants were requested to complete a Technology Acceptance Model 2 (TAM2) questionnaire. The results showed that the perceived playfulness and perceived usefulness of the exergames were significantly related to the users' usage behavior and intention to use for both the physical games and the cognitive games. However, a relationship between the output quality of the game and the usage behavior was apparent only in the case of the cognitive exergames. Finally, the impact of social influence on the intention to use and the usage behavior was more pronounced for the physical exergames. Overall, the results revealed that the acceptance of exergames by the elderly depends not so much on the awareness of fun in using the game, but the perceived usefulness of the related physical and cognitive abilities.

The landslide monitoring method that uses the Sentinel 1A Interferogram Synthetic Aperture Radar (InSAR) through the Stanford Method for Persistent Scatterers (StaMPS) method is a complimentary but ...complex procedure without exact guidelines. Hence, this paper delivered a parametric test by examining the optimal settings of the Sentinel 1A Persistent Scatterer (PS)- and Small Baseline Subset (SBAS)-InSAR using the StaMPS compared to the Global Navigation Satellite Systems (GNSS) in landslide cases. This study first revealed parameters with the suggested values, such as amplitude dispersion used to describe amplitude stability, ranging from 0.47 to 0.48 for PS and equal to or more than 0.6 for SBAS in WuWanZai, Ali Mt. The study further examined the suggested values for other factors, including the following: unwrap grid size to re-estimate the size of the grid; unwrap gold n win as the Goldstein filtering window to reduce the noise; and unwrap time win as the smoothing window (in days) for estimating phase noise distributions between neighboring pixels. Furthermore, the study substantiated the recommended settings in the Woda and Shadong landslide cases with the GNSS, inferring that the SBAS has adequate feasibility in practical landslide monitoring.

An increasing population of dementia patients produces substantial societal impacts. We assessed the prevalence of mild cognitive impairment (MCI) and all-cause dementia, including very mild dementia ...(VMD), in Taiwan. In a nationwide population-based cross-sectional survey, participants were selected by computerized random sampling from all 19 Taiwan counties and were enrolled between December 2011 and March 2013. Cases were identified through in-person interviews based on the National Institute on Aging-Alzheimer's Association clinical criteria. Demographic data and histories involving mental status and function in daily living were collected. The principal objective assessments were the Taiwanese Mental Status Examination and Clinical Dementia Rating. In all, 10,432 people aged 65 years or older (mean age 76.2 ± 6.7, 52.3% women) were interviewed. The age-adjusted prevalence of all-cause dementia was 8.04% (95% CI 7.47-8.61), including a 3.25% (95% CI 2.89-3.61) prevalence of VMD; that of MCI was 18.76% (95% CI 17.91-19.61). Women had a higher prevalence than men of both all-cause dementia (9.71% vs. 6.36%) and MCI (21.63% vs. 15.57%). MCI affects a considerable portion of the population aged 65 and above in Taiwan. The inclusion of VMD yields dementia prevalence rates higher than those previously reported from Taiwan. Old age, female gender, and a low educational level are significant associated factors.

Understanding the excited-state dynamics and conformational relaxation in thermally activated delayed fluorescence (TADF) molecules, including conformations that potentially support intramolecular ...through-space charge transfer, can open new avenues for TADF molecular design as well as elucidate complex photophysical pathways in structurally complex molecules. Emissive molecules comprising a donor (triphenylamine, TPA) and an acceptor (triphenyltriazine, TRZ) bridged by a second donor (9,9-dimethyl-9-10-dihydroacridin, DMAC, or phenoxazine, PXZ) are synthesized and characterized. In solution, the flexibility of the sp3-hybridized carbon atom in DMAC of DMAC–TPA–TRZ, compared to the rigid PXZ, allows significant conformational reorganization, giving rise to multiple charge-transfer excited states. As a result of such a reorganization, the TRZ and TPA moieties become cofacially aligned, driven by a strong dipole–dipole attraction between the TPA and TRZ units, forming a weakly charge-transfer dimer state, in stark contrast to the case of PXZ–TPA–TRZ where the rigid PXZ bridge only supports a single PXZ–TRZ charge transfer (CT) state. The low-energy TPA-TRZ dimer is found to have a high-energy dimer local triplet state, which quenches delayed emission because the resultant singlet CT local triplet energy gap is too large to mediate efficient reverse intersystem crossing. However, organic light-emitting diodes using PXZ–TPA–TRZ as an emitting dopant resulted in external quantum efficiency as high as 22%, more than two times higher than that of DMAC–TPA–TRZ-based device, showing the impact that such intramolecular reorganization and donor–acceptor dimerization have on TADF performance.
