As more and more teacher preparation programs realize the need to include courses that deal with
computer-assisted language learning, a crucial decision as to what is taught needs to be made, taking ...into consideration the
various post-graduation goals ranging from teacher or teacher-trainer to researcher. Thus, the question of whether to go beyond
teaching the potential uses of the computer and the Internet and to delve into how to create relevant computer-based tasks using
the WWW and to train teachers in the use of advanced web page development techniques, including Flash, web-enabled databases, and
the interactive possibilities of the WWW needs to be addressed. This paper tackles these issues and illustrates a sample approach
in dealing with the grooming of the new generation of CALL researchers and practitioners. What can be accomplished during the
first year of an MATESL program that highlights the importance of technology is explored through a reflective questionnaire,
a computer capabilities matrix, and an analysis of web-based course projects. Nine first-year M.A. students participated in the
study, which revealed that despite widely differing initial computer and Internet expertise among these future teachers and
researchers, their knowledge of creating and using web-based materials increased significantly, culminating in a project in
which the skills acquired throughout the semester were successfully integrated.
The major active constituents of ginseng are ginsenosides, and Rg(1) is a predominant compound of the total extract. Recent studies have demonstrated that Rg(1) can promote angiogenesis in vivo and ...in vitro. In this study, we used a DNA microarray technology to elucidate the mechanisms of action of Rg(1). We report that Rg(1) induces the proliferation of HUVECs, monitored using (3)H-thymidine incorporation and Trypan blue exclusion assays. Furthermore, Rg(1) (150-600 nM) also showed an enhanced tube forming inducing effect on the HUVEC. Rg(1) was also demonstrated to promote angiogenesis in an in vivo Matrigel plug assay, and increase endothelial sprouting in the ex vivo rat aorta ring assay. Differential gene expression profile of HUVEC following treatment with Rg(1) revealed the expression of genes related to cell adhesion, migration and cytoskeleton, including RhoA, RhoB, IQGAP1, CALM2, Vav2 and LAMA4. Our results suggest that Rg(1) can promote angiogenesis in multiple models, and this effect is partly due to the modulation of genes that are involved in the cytoskeletal dynamics, cell-cell adhesion and migration.
Monoclonal antibodies are subjected to a wide variety of post-translational modifications (PTMs) that cause structural heterogeneity. Characterization and control of these modifications or quality ...attributes are critical to ensure antibody quality and to define any potential effects on the ultimate safety and potency of antibody therapeutics. The biopharmaceutical industry currently uses numerous tools to analyze these quality attributes individually, which requires substantial time and resources. Here, we report a simple and ultrafast bottom-up liquid chromatography-mass spectrometry (uLC-MS) method with 5 min tryptic digestion to simultaneously analyze multiple modifications, including oxidation, deamidation, isomerization, glycation, glycosylation, and N-terminal pyro-glutamate formation, which can occur during antibody production in mammalian cell culture, during purification and/or on storage. Compared to commonly used preparation procedures, this uLC-MS method eliminates assay artifacts of falsely-increased Met oxidation, Asp isomerization, and Asn deamidation, a problem associated with long digestion times in conventional LC-MS methods. This simple, low artifact multi-attribute uLC-MS method can be used to quickly and accurately analyze samples at any stage of antibody drug development, in particular for clone and media selection during cell culture development.
