Hepatitis B virus (HBV) affects approximately 68 million people in China, and 10–15% of adults infected with HBV develop chronic hepatitis B, liver cirrhosis, liver failure or hepatocellular ...carcinoma (HCC). HLA-DPB1 gene polymorphism and expression have been shown to be associated with HBV infection susceptibility and spontaneous clearance. The aim of this study is to evaluate the role of HLA-DPB1 gene polymorphism in HBV infection. HLA-DPB1 and rs9277535 polymorphisms were investigated in 259 patients with HBV infection and 442 healthy controls (HCs) using sequence-based typing. The mRNA of HLA-DPB1 was measured by real-time polymerase chain reaction. HLA-DPB1 genes and rs9277535 polymorphisms were all associated with HBV infection in the Sichuan Han population. rs9277535A and HLA-DPB1*04:02 played a protective role against HBV infection. rs9277535G and DPB1*05:01 were associated with susceptibility to HBV infection. rs9277535GG had significantly higher HLA-DPB1 mRNA expression in the HBV infection group compared with the HC group. HLA-DPB1*05:01 and HLA-DPB1*21:01 had significantly lower mRNA expression in the HBV infection group compared with the HC group. The meta-analysis revealed that HLA-DPB1*02:01, HLA-DPB1*02:02, HAL-DPB1*04:01 and HLA-DPB1*04:02 protected against HBV infection, while HLA-DPB1*05:01, HLA-DPB1*09:01, and HLA-DPB1*13:01 were risk factors for susceptibility to HBV infection. HLA-DPB1*02:01, HLA-DPB1*02:02, and HLA-DPB1*04:01 were associated with HBV spontaneous clearance, while HLA-DPB1*05:01 was associated with chronic HBV infection. HLA-DPB1 alleles and rs9277535 have a major effect on the risk of HBV infection, and HBV infection is associated with lower HLA-DPB1 expression. HLA-DPB1 alleles have an important role in HBV susceptibility and spontaneous clearance.

This work provides an in-depth insight into the relationship between structure and performance, which can provide positive feedback on the design of novel adsorbents for this field.
Abstract Background Extensive evidence indicates that both lifestyle factors and air pollution are strongly associated with all-cause mortality. However, little studies in this field have integrated ...these two factors in order to examine their relationship with mortality and explore potential interactions. Methods A cohort of 271,075 participants from the UK Biobank underwent analysis. Lifestyles in terms of five modifiable factors, namely smoking, alcohol consumption, physical activity, diet, and sleep quality, were classified as unhealthy (0–1 score), general (2–3 score), and healthy (4–5 score). Air pollution, including particle matter with a diameter ≤ 2.5 μm (PM 2.5 ), particulate matter with a diameter ≤ 10 μm (PM 10 ), particulate matter with a diameter 2.5–10 μm (PM 2.5−10 ), nitrogen dioxide (NO 2 ), and nitrogen oxides (NO x ), was divided into three levels (high, moderate, and low) using Latent Profile Analysis (LPA). Cox proportional hazard regression analysis was performed to examine the links between lifestyle, air pollution, and all-cause mortality before and after adjustment for potential confounders. Restricted cubic spline curves featuring three knots were incorporated to determine nonlinear relationships. The robustness of the findings was assessed via subgroup and sensitivity analyses. Results With unhealthy lifestyles have a significantly enhanced risk of death compared to people with general lifestyles (HR = 1.315, 95% CI, 1.277–1.355), while people with healthy lifestyles have a significantly lower risk of death (HR = 0.821, 95% CI, 0.785–0.858). Notably, the difference in risk between moderate air pollution and mortality risk remained insignificant (HR = 0.993, 95% CI, 0.945–1.044). High air pollution, on the other hand, was independently linked to increased mortality risk as compared to low air pollution (HR = 1.162, 95% CI, 1.124–1.201). The relationship between NO x , PM 10 , and PM 2.5−10 and all-cause mortality was found to be nonlinear ( p for nonlinearity < 0.05). Furthermore, no significant interaction was identified between lifestyle and air pollution with respect to all-cause mortality. Conclusions Exposure to ambient air pollution elevated the likelihood of mortality from any cause, which was impacted by individual lifestyles. To alleviate this hazard, it is crucial for authorities to escalate environmental interventions, while individuals should proactively embrace and sustain healthy lifestyles.

Forging strong links between traits and performance is essential for understanding and predicting community assembly and dynamics. Functional trait analyses of trees that have correlated single-trait ...values with measures of performance such as growth and mortality have generally found weak relationships. A reason for these weak relationships is the failure to use individual-level trait data while simultaneously putting that data into the context of the abiotic setting, neighborhood composition, and the remaining axes constituting the overall phenotype. Here, utilizing detailed growth and trait data for 59 species of trees in a subtropical forest, we demonstrate that the individual-level functional trait values are strongly related to individual growth rates, and that the strength of these relationships critically depends on the context of that individual. We argue that our understanding of trait-performance relationships can be greatly improved with individuallevel data so long as that data is put into the proper context.
Macroalgae host various symbionts on their surface, which play a critical role in their growth and development processes. However, there is still incomplete understanding of this epiphytic ...bacteria-host algae interactions. This study comprehensively analysed variation of the epiphytic bacterial communities (EBC) composition of red macroalga Gracilariopsis lemaneiformis at different geographic locations and environmental factors (i.e., nitrogen and phosphorus), which shape the EBC composition of G. lemaneiformis. The composition and structure of EBC were characterized using high throughput sequencing of the V3-V4 hypervariable region of the 16S rRNA gene. The results revealed that epiphytic bacteria varied significantly among three different geographic locations in China, i.e., Nan'ao Island (NA), Lianjiang County (LJ), and Nanri Island (NR). Redundancy analysis (RDA) showed that the relative abundance of Bacteroidetes, Firmicutes, Verrucomicrobia, and Epsilonbacteraeota at NR were strongly positively correlated with total nitrogen (TN), total phosphorus (TP), nitrate nitrogen (NO
-N), and dissolved inorganic nitrogen (DIN), but negatively correlated with nitrite nitrogen (NO
-N). The relative abundance of Cyanobacteria at NA and LJ were strongly positively correlated with NO
-N, but negatively correlated with TN, TP, NO
-N, and DIN. Besides, the Mantel test results indicated that the EBC composition was significantly correlated with these environmental factors, which was also confirmed by Spearman correlation analysis. Thus, environmental factors such as NO
-N and DIN play a key role in the community composition of epiphytic bacteria on G. lemaneiformis. This study provides important baseline knowledge on the community composition of epiphytic bacteria on G. lemaneiformis and shows correlation between different epiphytic bacteria and their surrounding environmental factors.

We proposed, for the first time, a ultrasensitive amplification-free, label-free, and enzyme-free homogeneous electrochemical cytosensing platform based on the target-induced displacement reaction for tumor cell “signal-on” assay. To realize this proposal, the electroactive probe, Fe(CN)63−, was entrapped in the pores of positively charged MSN (PMSN), and then the negatively charged aptamer of the tumor cell was adsorbed on the surface of PMSN as a biogate. Once the tumor cells were recognized and captured by the aptamer, the biogate could be efficiently opened and the entrapped Fe(CN)63− released, which can be ascribed to the decreased adhesion between the aptamer-cell complex and PMSN. Consequently, the electrochemical signal dramatically increased. Overall, the “signal-on” strategy was conveniently realized. Notably, the detection limit for tumor cells assay (13 cells/mL) is comparable to those of the reported methods Furthermore, based on the similar proposal, this biosensor also can be extended for the ultrasensitive detection of microRNA (38 aM, S/N = 3) and Hg2+ (0.47 pM, S/N = 3). Therefore, this biosensing platform has the great potential to be a powerful tool in the applications of disease diagnostics and environmental monitoring.
NAC transcription factors are widely distributed in the plant kingdom and play an important role in the response to various abiotic stresses in plant species. Tritipyrum, an octoploid derived from ...hybridization of Triticum aestivum (AABBDD) and Thinopyrum elongatum (EE), is an important genetic resource for integrating the desirable traits of Th. elongatum into wheat. In this study, we investigated the tissue distribution and expression of Tritipyrum NAC genes in the whole genomes of T. aestivum and Th. elongatum after obtaining their complete genome sequences. Based on phylogenetic relationships, conserved motifs, gene synthesis, evolutionary analysis, and expression patterns, we identified and characterized 732 Tritipyrum NAC genes. These genes were divided into six main groups (A, B, C, D, E, and G) based on phylogenetic relationships and evolutionary studies, with members of these groups sharing the same motif composition. The 732 TtNAC genes are widely distributed across 28 chromosomes and include 110 duplicated genes. Gene synthesis analysis indicated that the NAC gene family may have a common ancestor. Transcriptome data and quantitative polymerase chain reaction (qPCR) expression profiles showed 68 TtNAC genes to be highly expressed in response to various salt stress and recovery treatments. Tel3E01T644900 (TtNAC477) was particularly sensitive to salt stress and belongs to the same clade as the salt tolerance genes ANAC019 and ANAC055 in Arabidopsis. Pearson correlation analysis identified 751 genes that correlated positively with expression of TtNAC477, and these genes are enriched in metabolic activities, cellular processes, stimulus responses, and biological regulation. TtNAC477 was found to be highly expressed in roots, stems, and leaves in response to salt stress, as confirmed by real-time PCR. These findings suggest that TtNAC477 is associated with salt tolerance in plants and might serve as a valuable exogenous gene for enhancing salt tolerance in wheat.

Paclitaxel is a widely used and potent chemotherapeutic agent for the treatment of cancer. However, patients receiving paclitaxel often develop an acute pain syndrome for which there are few ...treatment options. Astrocytes play an important role in the pathogenesis of pain in multiple preclinical models, as well as in paclitaxel-treated rodents. However, it is still unclear what the exact contribution of astrocytes may be in paclitaxel-associated acute pain syndrome (P-APS).
P-APS was modeled by a single systemic or intrathecal injection of paclitaxel and astrocyte contribution tested by immunohistochemical, pharmacological, and behavioral approaches. Cell cultures were also prepared to assess whether paclitaxel treatment directly activates astrocytes and whether intrathecal injection of paclitaxel-treated astrocytes produces pain that is reminiscent of P-APS.
Systemic injection of paclitaxel resulted in increased expression of glial fibrillary acidic protein (a common marker of astrocytic activation), as well as both systemic or intrathecal injection of paclitaxel induced pain hypersensitivity indicated by the development of mechanical allodynia, which was significantly reversed by the astrocytic inhibitor L-α-AA. Cultured astrocytes were activated by paclitaxel with significant increases in protein levels for tumor necrosis factor-α (TNF-α) and stromal-derived cell factor 1 (SDF-1). Importantly, intrathecal injection of paclitaxel-activated astrocytes produced mechanical allodynia that was reversed by TNF-α and SDF-1 neutralizing antibodies.
Our results suggest for the first time that paclitaxel can directly activate astrocytes, which are sufficient to produce acute pain by releasing TNF-α and SDF-1. Targeting astrocytes and these cytokines may offer new treatments for P-APS.
Cadmium (Cd) is a heavy metal nonessential for plants; this toxic metal accumulation in crops has significant adverse effects on human health. The crosstalk between copper (Cu) and Cd has been ...reported; however, the molecular mechanisms remain unknown. The present study investigated the function of wheat Cu transporter 3D (TaCOPT3D) in Cd tolerance. The TaCOPT3D transcripts significantly accumulated in wheat roots under Cd stress. Furthermore, TaCOPT3D-overexpressing lines were compared to wildtype (WT) plants to test the role of TaCOPT3D in Cd stress response. Under 20 mM Cd treatment, TaCOPT3D-overexpressing lines exhibited more biomass and lower root, shoot, and grain Cd accumulation than the WT plants. In addition, overexpression of TaCOPT3D decreased the reactive oxygen species (ROS) levels and increased the active antioxidant enzymes under Cd conditions. Moreover, the transcription factor (TF) TaWRKY22, which targeted the TaCOPT3D promoter, was identified in the regulatory pathway of TaCOPT3D under Cd stress. Taken together, these results show that TaCOPT3D plays an important role in regulating plant adaptation to cadmium stress through bound by TaWRKY22. These findings suggest that TaCOPT3D is a potential candidate for decreasing Cd accumulation in wheat through genetic engineering.
