The cereal endosperm is a major factor determining seed size and shape. However, the molecular mechanisms of endosperm development are not fully understood. Long noncoding RNAs (lncRNAs) function in ...various biological processes. Here we show a lncRNA, MISSEN, that plays an essential role in early endosperm development in rice (Oryza sativa). MISSEN is a parent-of-origin lncRNA expressed in endosperm, and negatively regulates endosperm development, leading to a prominent dent and bulge in the seed. Mechanistically, MISSEN functions through hijacking a helicase family protein (HeFP) to regulate tubulin function during endosperm nucleus division and endosperm cellularization, resulting in abnormal cytoskeletal polymerization. Finally, we revealed that the expression of MISSEN is inhibited by histone H3 lysine 27 trimethylation (H3K27me3) modification after pollination. Therefore, MISSEN is the first lncRNA identified as a regulator in endosperm development, highlighting the potential applications in rice breeding.
Tumor associated macrophages (TAMs) are considered with the capacity to have both negative and positive effects on tumor growth. The prognostic value of TAM for survival in patients with solid tumor ...remains controversial.
We conducted a meta-analysis of 55 studies (n = 8,692 patients) that evaluated the correlation between TAM (detected by immunohistochemistry) and clinical staging, overall survival (OS) and disease free survival (DFS). The impact of M1 and M2 type TAM (n = 5) on survival was also examined.
High density of TAM was significantly associated with late clinical staging in patients with breast cancer risk ratio (RR) = 1.20 (95% confidence interval (CI), 1.14-1.28) and bladder cancer RR = 3.30 (95%CI, 1.56-6.96) and with early clinical staging in patients with ovarian cancer RR = 0.52 (95%CI, 0.35-0.77). Negative effects of TAM on OS was shown in patients with gastric cancer RR = 1.64 (95%CI, 1.24-2.16), breast cancer RR = 8.62 (95%CI, 3.10-23.95), bladder cancer RR = 5.00 (95%CI, 1.98-12.63), ovarian cancer RR = 2.55 (95%CI, 1.60-4.06), oral cancer RR = 2.03 (95%CI, 1.47-2.80) and thyroid cancer RR = 2.72 (95%CI, 1.26-5.86),and positive effects was displayed in patients with colorectal cancer RR = 0.64 (95%CI, 0.43-0.96). No significant effect was showed between TAM and DFS. There was also no significant effect of two phenotypes of TAM on survival.
Although some modest bias cannot be excluded, high density of TAM seems to be associated with worse OS in patients with gastric cancer, urogenital cancer and head and neck cancer, with better OS in patients with colorectal cancer.

) mutation-positive resected stage II-IIIA (N1-N2) non-small-cell lung cancer (NSCLC). Here, we report the final overall survival (OS) results.
From September 2011 to April 2014, 222 patients from 27 sites were randomly assigned 1:1 to adjuvant gefitinib (n = 111) or VP (n = 111). Patients with resected stage II-IIIA (N1-N2) NSCLC and
-activating mutation were enrolled, receiving gefitinib for 24 months or VP every 3 weeks for four cycles. The primary end point was DFS (intention-to-treat ITT population). Secondary end points included OS, 3-, 5-year (y) DFS rates, and 5-year OS rate. Post hoc analysis was conducted for subsequent therapy data.
Median follow-up was 80.0 months. Median OS (ITT) was 75.5 and 62.8 months with gefitinib and VP, respectively (hazard ratio HR, 0.92; 95% CI, 0.62 to 1.36;
= .674); respective 5-year OS rates were 53.2% and 51.2% (
= .784). Subsequent therapy was administered upon progression in 68.4% and 73.6% of patients receiving gefitinib and VP, respectively. Subsequent targeted therapy contributed most to OS (HR, 0.23; 95% CI, 0.14 to 0.38) compared with no subsequent therapy. Updated 3y DFS rates were 39.6% and 32. 5% with gefitinib and VP (
= .316) and 5y DFS rates were 22. 6% and 23.2% (
= .928), respectively.
Adjuvant therapy with gefitinib in patients with early-stage NSCLC and
mutation demonstrated improved DFS over standard of care chemotherapy. Although this DFS advantage did not translate to a significant OS difference, OS with adjuvant gefitinib was one of the longest observed in this patient group compared with historic data.
The development of high-performance near-infrared organic light-emitting diodes is hindered by strong non-radiative processes as governed by the energy gap law. Here, we show that exciton ...delocalization, which serves to decouple the exciton band from highly vibrational ladders in the S0 ground state, can bring substantial enhancements in the photoluminescence quantum yield of emitters, bypassing the energy gap law. Experimental proof is provided by the design and synthesis of a series of new Pt(ii) complexes with a delocalization length of 5–9 molecules that emit at 866–960 nm with a photoluminescence quantum yield of 5–12% in solid films. The corresponding near-infrared organic light-emitting diodes emit light with a 930 nm peak wavelength and a high external quantum efficiency up to 2.14% and a radiance of 41.6 W sr−1 m−2. Both theoretical and experimental results confirm the exciton–vibration decoupling strategy, which should be broadly applicable to other well-aligned molecular solids.Pt(ii) complexes allow the fabrication of efficient near-infrared organic light-emitting diodes that operate beyond the 900 nm region.
The successful joint observation of the gravitational wave (GW) event GW170817 and its multiwavelength electromagnetic counterparts enabled us to witness a definite merger event of two neutron stars ...(NSs) for the first time. This historical event confirms the origin of short-duration gamma-ray bursts (GRBs) and, in particular, identifies the theoretically predicted kilonova phenomenon that is powered by radioactive decays of r-process heavy elements. However, whether or not a long-lived remnant NS could be formed during this merger event remains unknown; though, such a central engine has been suggested by afterglow observations of some short-duration GRBs. By invoking this long-lived remnant NS, we propose a model of hybrid energy sources for the kilonova AT 2017gfo associated with GW170817. While the early emission of AT 2017gfo is still powered radioactively, as is usually suggested, its late emission is primarily caused by delayed energy injection from the remnant NS. In our model, only one single opacity is required and an intermediate value of κ 0.97 cm2 g−1 is revealed, which could be naturally provided by lanthanide-rich ejecta that are deeply ionized by the emission from a wind of the NS. These self-consistent results indicate that a long-lived remnant NS, which must have a very stiff equation of state, was formed during the merger event of GW170817. This provides a very stringent constraint on the strong interaction in nuclear-quark matter. It is further implied that such GW events could provide a probe of the early spin and magnetic evolutions of NSs, e.g., the burying of surface magnetic fields.
Emerging evidences demonstrate that circular RNAs (circRNAs) are abnormally expressed in tumors and could serve as prognostic markers for cancers. However, the expression patterns and clinical ...implications of circRNAs in non-small cell lung cancer (NSCLC) remain obscure. In this study, we profiled circRNA expressions in 10 pairs of lung adenocarcinoma (LUAD) and squamous cell carcinoma (LUSC) after ribosomal RNA-depletion and RNase R digestion to enrich circRNAs. Combining five circRNA computational programs, we found that LUAD and LUSC not only share common expression patterns, but also exhibit distinct circRNA expression signatures. Moreover, the Receiver Operating Characteristic (ROC) curve analysis indicated that hsa_circ_0077837 and hsa_circ_0001821 could serve as potential biomarkers for both LUAD and LUSC, while hsa_circ_0001073 and hsa_circ_0001495 could be diagnostic/subtyping marker for LUAD and LUSC, respectively. Therefore, our findings highlight the important diagnostic potential of circRNAs in NSCLC.

Early diagnosis has been proved to improve survival rate of lung cancer patients. The availability of blood-based screening could increase early lung cancer patient uptake. Our present study attempted to discover Chinese patients’ plasma metabolites as diagnostic biomarkers for lung cancer. In this work, we use a pioneering interdisciplinary mechanism, which is firstly applied to lung cancer, to detect early lung cancer diagnostic biomarkers by combining metabolomics and machine learning methods. We collected total 110 lung cancer patients and 43 healthy individuals in our study. Levels of 61 plasma metabolites were from targeted metabolomic study using LC-MS/MS. A specific combination of six metabolic biomarkers note-worthily enabling the discrimination between stage I lung cancer patients and healthy individuals (AUC = 0.989, Sensitivity = 98.1%, Specificity = 100.0%). And the top 5 relative importance metabolic biomarkers developed by FCBF algorithm also could be potential screening biomarkers for early detection of lung cancer. Naïve Bayes is recommended as an exploitable tool for early lung tumor prediction. This research will provide strong support for the feasibility of blood-based screening, and bring a more accurate, quick and integrated application tool for early lung cancer diagnostic. The proposed interdisciplinary method could be adapted to other cancer beyond lung cancer.
Lung adenocarcinoma (ADC) is the predominant histological type of lung cancer, and radiotherapy is one of the current therapeutic strategies for lung cancer treatment. Unfortunately, biological ...complexity and cancer heterogeneity contribute to radioresistance development. Karyopherin α2 (KPNA2) is a member of the importin α family that mediates the nucleocytoplasmic transport of cargo proteins. KPNA2 overexpression is observed across cancer tissues of diverse origins. However, the role of KPNA2 in lung cancer radioresistance is unclear. Herein, we demonstrated that high expression of KPNA2 is positively correlated with radioresistance and cancer stem cell (CSC) properties in lung ADC cells. Radioresistant cells exhibited nuclear accumulation of KPNA2 and its cargos (OCT4 and c‐MYC). Additionally, KPNA2 knockdown regulated CSC‐related gene expression in radioresistant cells. Next‐generation sequencing and bioinformatic analysis revealed that STAT1 activation and nuclear phospholipid scramblase 1 (PLSCR1) are involved in KPNA2‐mediated radioresistance. Endogenous PLSCR1 interacting with KPNA2 and PLSCR1 knockdown suppressed the radioresistance induced by KPNA2 expression. Both STAT1 and PLSCR1 were found to be positively correlated with dysregulated KPNA2 in radioresistant cells and ADC tissues. We further demonstrated a potential positive feedback loop between PLSCR1 and STAT1 in radioresistant cells, and this PLSCR1‐STAT1 loop modulates CSC characteristics. In addition, AKT1 knockdown attenuated the nuclear accumulation of KPNA2 in radioresistant lung cancer cells. Our results collectively support a mechanistic understanding of a novel role for KPNA2 in promoting radioresistance in lung ADC cells.

A facile modulation strategy was adopted to fabricate hierarchically porous HP-UOH- X (HP, UOH and X represented the hierarchical pores, UiO-66-(OH) 2 and the dosage of benzoic acid, respectively) ...via introducing benzoic acid with different dosages into the precursor solution of UiO-66-(OH) 2 . The formation of hierarchical pores boosted the exposure of –OH groups and the Cr( vi ) mass transfer in HP-UOH- X , which vastly enhanced its sorption capacities and sorption rates. The optimal adsorbent (HP-UOH-80) displayed better sorption capacity (266.74 mg g −1 ) toward Cr( vi ) ( T = 308 K, pH = 2.0) and faster diffusion rate ( k 1 = 14.21 mg g −1 min 0.5 , k 2 = 6.25 mg g −1 min 0.5 ) than those of the pristine UiO-66-(OH) 2 and other HP-UOH- X adsorbents. Interestingly, HP-UOH-80 exhibited good selective uptake ability toward Cr( vi ) in different simulated water samples containing various competing anions. The corresponding mechanism was proposed that the –OH groups and the defect sites played the dominant contribution to Cr( vi ) adsorption, which could be affirmed by Fourier-transform infrared (FTIR) spectroscopy and X-ray photoelectron spectroscopy (XPS). In addition, the strategy of photocatalytic Cr( vi ) reduction for desorption was introduced to replace traditional chemical desorption. In all, this work presented an effective adsorbent and a sustainable approach for Cr( vi ) elimination, which can regenerate the adsorbent via a photocatalytic process rather than chemical washing.
