ABSTRACT The recent detections of two transit events attributed to the super-Earth candidate K2-18b have provided the unprecedented prospect of spectroscopically studying a habitable-zone planet ...outside the solar system. Orbiting a nearby M2.5 dwarf and receiving virtually the same stellar insolation as Earth, K2-18b would be a prime candidate for the first detailed atmospheric characterization of a habitable-zone exoplanet using the Hubble Space Telescope (HST)and James Webb Space Telescope (JWST). Here, we report the detection of a third transit of K2-18b near the predicted transit time using the Spitzer Space Telescope. The Spitzer detection demonstrates the periodic nature of the two transit events discovered by K2, confirming that K2-18 is indeed orbited by a super-Earth in a 33 day orbit, ruling out the alternative scenario of two similarly sized, long-period planets transiting only once within the 75 day Kepler Space Telescope (K2) observation. We also find, however, that the transit event detected by Spitzer occurred 1.85 hr ( 7 ) before the predicted transit time. Our joint analysis of the Spitzer and K2 photometry reveals that this early occurrence of the transit is not caused by transit timing variations, but the result of an inaccurate ephemeris due to a previously undetected data anomaly in the K2 photometry. We refit the ephemeris and find that K2-18b would have been lost for future atmospheric characterizations with HST and JWST if we had not secured its ephemeris shortly after the discovery. We caution that immediate follow-up observations as presented here will also be critical for confirming and securing future planets discovered by the Transiting Exoplanet Survey Satellite (TESS), in particular if only two transit events are covered by the relatively short 27-day TESS campaigns.
Since 2014, NASA's K2 mission has observed large portions of the ecliptic plane in search of transiting planets and has detected hundreds of planet candidates. With observations planned until at ...least early 2018, K2 will continue to identify more planet candidates. We present here 275 planet candidates observed during Campaigns 0-10 of the K2 mission that are orbiting stars brighter than 13 mag (in Kepler band) and for which we have obtained high-resolution spectra (R = 44,000). These candidates are analyzed using the vespa package in order to calculate their false-positive probabilities (FPP). We find that 149 candidates are validated with an FPP lower than 0.1%, 39 of which were previously only candidates and 56 of which were previously undetected. The processes of data reduction, candidate identification, and statistical validation are described, and the demographics of the candidates and newly validated planets are explored. We show tentative evidence of a gap in the planet radius distribution of our candidate sample. Comparing our sample to the Kepler candidate sample investigated by Fulton et al., we conclude that more planets are required to quantitatively confirm the gap with K2 candidates or validated planets. This work, in addition to increasing the population of validated K2 planets by nearly 50% and providing new targets for follow-up observations, will also serve as a framework for validating candidates from upcoming K2 campaigns and the Transiting Exoplanet Survey Satellite, expected to launch in 2018.
Adenosine deaminases acting on RNA (ADARs) catalyze the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA) and thereby potentially alter the information content and ...structure of cellular RNAs. Notably, although the overwhelming majority of such editing events occur in transcripts derived from Alu repeat elements, the biological function of non-coding RNA editing remains uncertain. Here, we show that mutations in ADAR1 (also known as ADAR) cause the autoimmune disorder Aicardi-Goutières syndrome (AGS). As in Adar1-null mice, the human disease state is associated with upregulation of interferon-stimulated genes, indicating a possible role for ADAR1 as a suppressor of type I interferon signaling. Considering recent insights derived from the study of other AGS-related proteins, we speculate that ADAR1 may limit the cytoplasmic accumulation of the dsRNA generated from genomic repetitive elements.

K2 greatly extended Kepler's ability to find new planets, but it was typically limited to identifying transiting planets with orbital periods below 40 days. While analyzing K2 data through the ...Exoplanet Explorers project, citizen scientists helped discover one super-Earth and four sub-Neptune sized planets in the relatively bright (V = 12.21, K = 10.3) K2-138 system, all which orbit near 3:2 mean-motion resonances. The K2 light curve showed two additional transit events consistent with a sixth planet. Using Spitzer photometry, we validate the sixth planet's orbital period of 41.966 ± 0.006 days and measure a radius of 3.44 (+0.32,-.031)Rꚛ, solidifying K2-138 as the K2 system with the most currently known planets. There is a sizeable gap between the outer two planets, since the fifth planet in the system, K2-138 f, orbits at 12.76 days. We explore the possibility of additional nontransiting planets in the gap between f and g. Due to the relative brightness of the K2-138 host star, and the near resonance of the inner planets, K2-138 could be a key benchmark system for both radial velocity and transit-timing variation mass measurements, and indeed radial velocity masses for the inner four planets have already been obtained. With its five sub-Neptunes and one super-Earth, the K2-138 system provides a unique test bed for comparative atmospheric studies of warm to temperate planets of similar size, dynamical studies of near-resonant planets, and models of planet formation and migration.
Aicardi‐Goutières syndrome (AGS) is a rare, genetically determined encephalopathy whose importance from a clinical viewpoint is magnified because of the risk of misdiagnosis as the sequelae of ...congenital infection. Recent molecular advances have shown that AGS can be caused by mutations in any one of at least five genes (four of which have so far been identified), most commonly on a recessive basis but occasionally as a dominant trait. Additionally, a recent genotype–phenotype correlation has shown that two clinical presentations can be delineated; an early onset neonatal form highly reminiscent of congenital infection seen particularly with TREX1 mutations, and a later‐onset presentation, sometimes occurring after several months of normal development and occasionally associated with remarkably preserved neurological function, most frequently due to RNASEH2B mutations. Evidence is emerging to show that the nucleases defective in AGS are involved in removing endogenous nucleic acid species produced during normal cellular processing, and that a failure of this removal results in inappropriate activation of the innate immune system. This hypothesis explains the phenotypic overlap of AGS with congenital infection and some aspects of systemic lupus erythematosus, where a similar interferon alpha‐mediated innate immune response is triggered by viral and host nucleic acids respectively.

Given that Campaign 16 of the K2 mission is one of just two K2 campaigns observed so far in "forward-facing" mode, which enables immediate follow-up observations from the ground, we present a catalog ...of interesting targets identified through photometry alone. Our catalog includes 30 high-quality planet candidates (showing no signs of being non-planetary in nature), 48 more ambiguous events that may be either planets or false positives, 164 eclipsing binaries, and 231 other regularly periodic variable sources. We have released light curves for all targets in C16 and have also released system parameters and transit vetting plots for all interesting candidates identified in this paper. Of particular interest is a candidate planet orbiting the bright F dwarf HD 73344 (V = 6.9, K = 5.6) with an orbital period of 15 days. If confirmed, this object would correspond to a 2.56 0.18 R⊕ planet and would likely be a favorable target for radial velocity characterization. This paper is intended as a rapid release of planet candidates, eclipsing binaries, and other interesting periodic variables to maximize the scientific yield of this campaign, and as a test run for the upcoming TESS mission, whose frequent data releases call for similarly rapid candidate identification and efficient follow up.
Abstract
We present a transmission spectrum for the Neptune-sized exoplanet HD 106315c from optical to infrared wavelengths based on transit observations from the Hubble Space Telescope/Wide Field ...Camera 3, K2, and Spitzer. The spectrum shows tentative evidence for a water absorption feature in the 1.1–1.7
μ
m wavelength range with a small amplitude of 30 ppm (corresponding to just 0.8 ± 0.04 atmospheric scale heights). Based on an atmospheric retrieval analysis, the presence of water vapor is tentatively favored with a Bayes factor of 1.7–2.6 (depending on prior assumptions). The spectrum is most consistent with either an enhanced metallicity or high-altitude condensates, or both. Cloud-free solar composition atmospheres are ruled out at >5
σ
confidence. We compare the spectrum to grids of cloudy and hazy forward models and find that the spectrum is fit well by models with moderate cloud lofting or haze formation efficiency over a wide range of metallicities (1–100× solar). We combine the constraints on the envelope composition with an interior structure model and estimate that the core mass fraction is ≳0.3. With a bulk composition reminiscent of that of Neptune and an orbital distance of 0.15 au, HD 106315c hints that planets may form out of broadly similar material and arrive at vastly different orbits later in their evolution.
Mutations in PNPO are a known cause of neonatal onset seizures that are resistant to pyridoxine but responsive to pyridoxal phosphate (PLP). Mills et al. show that PNPO mutations can also cause ...neonatal onset seizures that respond to pyridoxine but worsen with PLP, as well as PLP-responsive infantile spasms.
The first described patients with pyridox(am)ine 5’-phosphate oxidase deficiency all had neonatal onset seizures that did not respond to treatment with pyridoxine but responded to treatment with pyridoxal 5’-phosphate. Our data suggest, however, that the clinical spectrum of pyridox(am)ine 5’-phosphate oxidase deficiency is much broader than has been reported in the literature. Sequencing of the PNPO gene was undertaken for a cohort of 82 individuals who had shown a reduction in frequency and severity of seizures in response to pyridoxine or pyridoxal 5’-phosphate. Novel sequence changes were studied using a new cell-free expression system and a mass spectrometry-based assay for pyridoxamine phosphate oxidase. Three groups of patients with PNPO mutations that had reduced enzyme activity were identified: (i) patients with neonatal onset seizures responding to pyridoxal 5’-phosphate (n = 6); (ii) a patient with infantile spasms (onset 5 months) responsive to pyridoxal 5’-phosphate (n = 1); and (iii) patients with seizures starting under 3 months of age responding to pyridoxine (n = 8). Data suggest that certain genotypes (R225H/C and D33V) are more likely to result in seizures that to respond to treatment with pyridoxine. Other mutations seem to be associated with infertility, miscarriage and prematurity. However, the situation is clearly complex with the same combination of mutations being seen in patients who responded and did not respond to pyridoxine. It is possible that pyridoxine responsiveness in PNPO deficiency is affected by prematurity and age at the time of the therapeutic trial. Other additional factors that are likely to influence treatment response and outcome include riboflavin status and how well the foetus has been supplied with vitamin B6 by the mother. For some patients there was a worsening of symptoms on changing from pyridoxine to pyridoxal 5’-phosphate. Many of the mutations in PNPO affected residues involved in binding flavin mononucleotide or pyridoxal 5’-phosphate and many of them showed residual enzyme activity. One sequence change (R116Q), predicted to affect flavin mononucleotide binding and binding of the two PNPO dimers, and with high residual activity was found in Groups (ii) and (iii). This sequence change has been reported in the 1000 Genomes project suggesting it could be a polymorphism but alternatively it could be a common mutation, perhaps responsible for the susceptibility locus for genetic generalized epilepsy on 17q21.32 (close to rs72823592). We believe the reduction in PNPO activity and B6-responsive epilepsy in the patients reported here indicates that it contributes to the pathogenesis of epilepsy.
We report the discovery of a new ultra-short-period planet and summarize the properties of all such planets for which the mass and radius have been measured. The new planet, K2-131b, was discovered ...in K2 Campaign 10. It has a radius of and orbits a G dwarf with a period of 8.9 hr. Radial velocities obtained with Magellan/PFS and TNG/HARPS-N show evidence for stellar activity along with orbital motion. We determined the planetary mass using two different methods: (1) the "floating chunk offset" method, based only on changes in velocity observed on the same night; and (2) a Gaussian process regression based on both the radial velocity and photometric time series. The results are consistent and lead to a mass measurement of and a mean density of g cm−3.
Abstract
The alignment of planetary orbits with respect to the stellar rotation preserves information on their dynamical histories. Measuring this angle for young planets helps illuminate the ...mechanisms that create misaligned orbits for older planets, as different processes could operate over timescales ranging from a few megayears to a gigayear. We present spectroscopic transit observations of the young exoplanet V1298 Tau b; we update the age of V1298 Tau to be 28 ± 4 Myr based on Gaia EDR3 measurements. We observed a partial transit with Keck/HIRES and LBT/PEPSI, and detected the radial velocity anomaly due to the Rossiter–McLaughlin effect. V1298 Tau b has a prograde, well-aligned orbit, with
