MicroRNAs (miRNAs) have recently emerged as a new class of biomarkers for disease diagnosis and play vital roles in physiological and pathological processes. Here, a novel fiber-optic prototype that ...allows synchronized optical transmittance measurements at different wavelengths has been designed and fabricated for selective and sensitive miRNAs detection based on a ratiometric colorimetric strategy. The proposed measurement method automatically adjusts the output light power through the LEDs, minimizing eventual differences in brightness and potential losses through the optical fiber without the need expensive optical components. The prototype has been validated for the detection of miRNAs identified as biomarkers of bovine mastitis in dairy cattle. Observation of the color changes from red to blue of the AuNP solution, and therefore on the transmittance signal, caused by miRNA-induced gold nanoparticles (AuNPs), aggregation has been used for rapid detection of such genetic biomarker. The equipment designed and constructed is portable, being the main parameters of the measurement managed with a smartphone, and it could be potentially employed with any other biosensor whose detection strategy is based on changes in the distance between AuNPs.
Covalent conjugation of allergens to toll-like receptor (TLR) agonists appears to be a powerful strategy for the development of safety compounds for allergen-specific immunomodulatory response toward ...tolerance in allergy. In this work, we have synthesized two family of ligands, an 8-oxoadenine derivative as a ligand for TLR7 and a pyrimido5,4-bindole as a ligand for TLR4, both conjugated with a T-cell peptide of Pru p 3 allergen, the lipid transfer protein (LTP) responsible for LTP-dependent food allergy. These conjugates interact with dendritic cells, inducing their specific maturation, T-cell proliferation, and cytokine production in peach allergic patients. Moreover, they increased the Treg-cell frequencies in these patients and could induce the IL-10 production. These outcomes were remarkable in the case of the TLR7 ligand conjugated with Pru p 3, opening the door for the potential application of these allergen–adjuvant systems in food allergy immunotherapy.
To investigate food allergy-tolerance mechanisms induced through allergen-specific immunotherapy we used RNA-Sequencing to measure gene expression in lymph-node-derived dendritic cells from Pru p ...3-anaphylactic mice after immunotherapy with glycodendropeptides at 2 nM and 5 nM, leading to permanent tolerance and short-term desensitization, respectively. Gene expression was also measured in mice receiving no immunotherapy (anaphylaxis); and in which anaphylaxis could never occur (antigen-only). Compared to anaphylaxis, the antigen-only group showed the greatest number of expression-changes (411), followed by tolerant (186) and desensitized (119). Only 29 genes changed in all groups, including Il12b, Cebpb and Ifngr1. The desensitized group showed enrichment for genes related to chronic inflammatory response, secretory granule, and regulation of interleukin-12 production; the tolerant group showed genes related to cytokine receptor activity and glucocorticoid receptor binding, suggesting distinct pathways for similar outcomes. We identified genes and processes potentially involved in the restoration of long-term tolerance via allergen-specific immunotherapy, representing potential prognostic biomarkers.

The discretization error is not always calculated, even though it is essential for the studies of computational solid mechanics. However, it is well known that an error committed by the mesh used can ...be as large as the measured variable, which greatly invalidates the results obtained. The grid convergence index (GCI) method makes possible to determine on a solid basis, the order of convergence and the asymptotic solution. This method seems to be a suitable estimator despite further research is needed in the context of blast situations and finite element (FE) calculations. For this purpose, field trials were performed consisting in the detonation of a spherical hanging load of homemade explosive. The pressure generated by the shock wave was measured in different positions at two distances. With these data, a TNT equivalent has been obtained and used to calculate the shock propagation with the solvers LS-DYNA and ProsAir. This work aims to verify the GCI method by comparing its results with field data along with the simulations carried out. The comparison also seeks to validate the methodology used to obtain the TNT equivalent.This research shows that the GCI gives good results for both solvers despite the complexity of the physical problem. Besides, LS-DYNA displays better correlation with the experimental data than the ProsAir results, with an error of less than 10% in all values.

In this paper we ensure that for some class of impulsive differential equations with delay the zero solution is asymptotically stable by means of fixed point theory.
We introduce boundary value conditions involving antiperiodic and nonlocal three-point boundary conditions. We solve a nonlinear fractional differential equation supplemented with those conditions. ...We obtain some existence results for the given problem by applying some standard tools of fixed point theory. These results are well illustrated with the aid of examples.
Este texto profundiza en cinco interiores, cinco locales comerciales, proyectados por Alejandro de la Sota entre 1952 y 1953. Durante estos dos años de supuesta inactividad el joven Sota fundó su ...corpus conceptual. Aunque en su mayoría frustrados, y a pesar de la distancia formal y su entrega a la expresión y la sensualidad, estos proyectos comparten la raíz poética--el vocabulario sotiano--con las obras maestras que produjo inmediatamente después. El maestro gallego evolucionó desde estrategias fondo-figura en los primeros proyectos hasta llegar a la anulación del entorno, la nada, como espacio expositivo ideal. La nada, el silencio, el vacío, la ausencia o la desaparición, que tanto resonarán en la obra posterior, fue literal y artificiosa en estas primeras tentativas. El morbo de explorar territorio prohibido, las obras posteriores, la habilidad para escapar del entorno y el esfuerzo de Sota en crearse una personalidad artística propia justifican esta investigación.
Introducción. Las escalas de autorreporte se han considerado útiles para evaluar y vigilar los síntomas de la enfermedad maníaco-depresiva. Uno de estos instrumentos, la escala de medición de la ...enfermedad maníaco-depresiva mediante autorreporte (EMUN-AR), no se ha validado aún.Objetivo. Validar la escala EMUN-AR usando el modelo de Rasch.Materiales y métodos. La escala EMUN-AR mide 26 ítems que evalúan por dimensiones los síntomas de la enfermedad maníaco-depresiva en tres categorías: frecuencia, intensidad y grado de molestia causada. Para evaluar las propiedades psicométricas de la escala, se efectuaron análisis factoriales exploratorios con el fin de tener una aproximación de su estructura latente, y se usaron modelos de Rasch que incluyeron la 'unidimensionalidad', la confiabilidad, el ajuste de ítems, el ordenamiento de umbrales, y los mapas de personas e ítems.Resultados. Se seleccionó una muestra de 267 pacientes hospitalizados, 204 de los cuales había sufrido un episodio maníaco (76,4 %) y estaba recibiendo tratamiento farmacológico. Se encontró una estructura factorial que puede resumirse en cuatro dominios: activación, inhibición, ideas depresivas e inadaptación. El análisis de Rasch evidenció la unidimensionalidad en los factores, adecuados valores de confiabilidad, buen ajuste de los 26 ítems en general (excepto en el ítem que mide las ideas de muerte o suicidio), un funcionamiento deficiente del sistema de calificación de los ítems y un cubrimiento limitado de los síntomas de la enfermedad correspondientes a las formas atenuadas.Conclusión. Se modificó la escala original en dos aspectos: se ajustó un ítem cuyos indicadores marginales tenían mal ajuste y se redujo el número de categorías. En su forma actual, el instrumento es apropiado para medir las formas graves de la enfermedad maníaco-depresiva, mas no así las atenuadas.
