The immunostimulatory potential of the marine yeast Yarrowia lipolytica (D1 and N6 strains) administered orally was evaluated in the white shrimp Litopenaeus vannamei. Yeasts and commercial glucans ...were mixed with a commercial feed to formulate diets with a 1.1% concentration of immunostimulants. The shrimp were fed daily for a period of 21 days. Weekly determinations were performed for immunological parameters in hemolymph, such as total hemocyte count (THC), lysozyme activity (LYZ), prophenoloxidase activity, antioxidant enzymatic activities (superoxide dismutase SOD, catalase CAT, and peroxidases), and bactericidal activity against Vibrio parahaemolyticus. Expression profiles of penaeidin (PEN), lysozyme (LYZ), and prophenoloxidase (proPO) immune genes were evaluated in hemocytes. In general, an increase in the immune parameters was observed in shrimp fed yeast diet compared to glucan and the control diets. Yarrowia lipolytica, especially strain N6, provided maximum immunostimulatory effects evidenced by the increase of immune parameters (THC, LYZ, SOD, CAT) and gene expression profile. In conclusion, this study demonstrated that Y. lipolytica had immunostimulatory effects and increased bactericidal activity in L. vannamei hemocytes against V. parahaemolyticus. These findings open the path for the potential application of Y. lipolytica-based immunostimulant for shrimp aquaculture.

The spread of non‐indigenous and invasive seaweeds has increased worldwide, and their potential effects on native seaweeds have raised concern. Undaria pinnatifida is considered among the most ...prolific non‐indigenous species. This species has expanded rapidly in the Northeast Pacific, overlapping with native communities such as the iconic giant kelp forests (Macrocystis pyrifera). Canopy shading by giant kelp has been argued to be a limiting factor for the presence of U. pinnatifida in the understory, thus its invasiveness capacity. However, its physiological plasticity under light limitation remains unclear. In this work, we compared the physiology and growth of juvenile U. pinnatifida and M. pyrifera sporophytes transplanted to the understory of a giant kelp forest, to juveniles growing outside of the forest. Extreme low light availability compared to that outside (~0.2 and ~4.4 mol photon ⋅ m−2 ⋅ d−1, respectively) likely caused a “metabolic energy crisis” in U. pinnatifida, thus restricting its photoacclimation plasticity and nitrogen acquisition, ultimately reducing its growth. Despite M. pyrifera juveniles showing photoacclimatory responses (e.g., increases in photosynthetic efficiency and lower compensation irradiance, Ec), their physiological/vegetative status deteriorated similarly to U. pinnatifida, which explains the low recruitment inside the forest. Generally, our results revealed the ecophysiological basis behind the limited growth and survival of juvenile U. pinnatifida sporophytes in the understory.
Vibrio parahaemolyticus, V. campbellii, V. owensii and V. harveyi are causative agents of Acute Hepatopancreatic Necrosis Disease (AHPND) in shrimp. AHPND is recently one of the most important ...diseases worldwide because of the high mortality and huge economic losses in shrimp farms. Antibiotic resistance has caused concern into finding other options to fight against pathogenic bacterial diseases. Currently, the use of lytic phages has reemerged as a promising strategy to combat bacterial pathogens. Likewise, modern molecular tools have been applied to modify phages by genetic engineering to express specific antigens and antibodies. This study reviewed recent research reports in the development of phages to control Vibrio spp. infections in shrimp and discusses different specific strategies, placing them in perspective to combat AHPND.
The RNA subunit of telomerase is an essential component whose primary sequence and length are poorly conserved among eukaryotic organisms. The phytopathogen Ustilago maydis is a dimorphic fungus of ...the order Ustilaginales. We analyzed several species of Ustilaginales to computationally identify the TElomere RNA (TER) gene ter1. To confirm the identity of the TER gene, we disrupted the gene and characterized telomerase-negative mutants. Similar to catalytic TERT mutants, ter1Δ mutants exhibit phenotypes of growth delay, telomere shortening and low replicative potential. ter1-disrupted mutants were unable to infect maize seedlings in heterozygous crosses and showed defects such as cell cycle arrest and segregation failure. We concluded that ter1, which encodes the TER subunit of the telomerase of U. maydis, have similar and perhaps more extensive functions than trt1.

Antibiotic usage to control infectious diseases in shrimp aquaculture has led to serious problems on antimicrobial resistance. An alternative to mitigate this issue is the use of probiotics, which ...can be easily administered by feed and water. This study examines immunomodulatory and protective effects of the marine yeasts Debaryomyces hansenii CBS8339 (Dh) and Yarrowia lipolytica Yl-N6 (Yl) -alone and mixed-in white shrimp Penaeus vannamei post-larvae. Administration routes (fed and water alone or in combination), supplementation frequency and time elapsed after the last dietary supplement were tested on growth and gene expression of penaeidin, lectin, lysozyme, superoxide dismutase, catalase, and peroxidase, as well as survival upon Vibrio parahaemolyticus IPNGS16 challenge. Penaeidin and lectin genes were upregulated in post-larvae fed orally with Yl or combined Dh + Yl. Higher growth and survival for yeast supplementation treatments were observed compared to the control group, mainly when yeasts (Dh + Yl) and administration routes (feed and water) were combined. In conclusion, mixed yeast and combined administration routes improved growth and immunity against V. parahaemolyticus.
•Individually or combined yeast administration increase growth of shrimp postlarvae.•Combined yeast routes induce higher survival in shrimp postlarvae after infection.•Oral yeast supplementation was the most efficient route on immune response in shrimp postlarvae.•Daily administration of combined yeast and route protect shirmp against bacterial infection at least for seven days.
Due to climate change, the incidence of marine heat waves (MHWs) has increased, yet their effects on seaweeds are still not well understood. Adult sporophytes of Macrocystis pyrifera, the species ...forming the iconic giant kelp forests, can be negatively affected by thermal stress and associated environmental factors (e.g., nutrient depletion, light deprivation); however, little is known about the tolerance/vulnerability of juvenile sporophytes. Simultaneously to MHWs, juveniles can be subjected to light limitation for extended periods of time (days–weeks) due to factors causing turbidity, or even because of shading by understory canopy‐forming seaweeds. This study evaluated the effects of a simulated MHW (24°C, 7 d) in combination (or not) with light deprivation, on the photosynthetic capacities, nutrient uptake, and tissue composition, as well as oxidative stress descriptors of M. pyrifera juvenile sporophytes (single blade stage, up to 20 cm length). Maximum quantum yield (Fv/Fm) decreased in juveniles under light at 24°C, likely reflecting some damage on the photosynthetic apparatus or dynamic photoinhibition; however, no other sign of physiological alteration was found in this treatment (i.e., pigments, nutrient reserves and uptake, oxidative stress). Photosynthetic capacities were maintained or even enhanced in plants under light deprivation, likely supported by photoacclimation (pigments increment); by contrast, nitrate uptake and internal storage of carbohydrates were strongly reduced, regardless of temperature. This study indicated that light limitation can be more detrimental to juvenile survival, and therefore recruitment success of M. pyrifera forests, than episodic thermal stress from MHWs.
The widespread overuse of antibiotics in aquaculture has led to the emergence of antibiotic-resistance shrimp pathogens, the negative impact on shrimp gut microbiota, and the presence of ...antimicrobial residues in aquaculture products, with negative consequences on human health. Alternatively, probiotics have positive effects on immunological responses and productive performance of aquatic animals. In this study, three probiotic bacteria, (Bacillus licheniformis MAt32, B. subtilis MAt43 and B. subtilis subsp. subtilis GAtB1), isolated from the Anadara tuberculosa were included in diets for juvenile shrimp, Litopenaeus vannamei, to evaluate their effects on growth, survival, disease prevalence, and immune-related gene expression. Shrimp naturally infected with WSSV and IHHNV were fed with the basal diet (control, T1) and diets supplemented with four levels of bacilli probiotic mix (1:1:1) at final concentration of (T2) 1 × 106, (T3) 2 × 106, (T4) 4 × 106, and (T5) 6 × 106 CFU g−1 of feed. The specific growth rate of shrimp was significantly higher in T2 than in T1 (control) treatment, and the final growth as well as the survival were similar among treated groups. The prevalence of WSSV and IHHNV infected shrimp was reduced in T2 and T4 treatments, respectively, compared with control. The mRNA expression of proPO gene was higher in treatment T4 than control. The LvToll1 gene was significantly up-regulated in treatments T4 and T5 compared to control. The SOD gene was up-regulated in treatment T5 compared to control. In contrast, the mRNA expression of the Hsp70 gene was down-regulated in treatments T4 and T5 respect to control, and the TGase gene remained unaffected by the level of bacillus probiotic mix. As conclusion, the bacilli probiotic mix (Bacillus spp.) enhanced immune-related gene expression in WSSV and IHHNV naturally infected shrimp. This is the first report of probiotic potential of bacteria isolated from A. tuberculosa on the immune response and viral prevalence in shrimp Litopenaeus vannamei.

Farfantepenaeus californiensis). Presumptive lactic acid bacteria were characterized for hemolytic and enzymatic activity, hydrophobicity, growth, and molecular identification. Two mixtures of four isolates were tested and their effect measured on the hemocyte number, survival, and prevalence of WSSV and IHHNV. Each mixture was applied at two different concentrations in a 15-day bioassay with shrimp naturally infected with WSSV and IHHNV as determined by single and/or nested PCR. In the treated animals total hemocyte count and survival were similar to control group. All shrimp fed with bacterial mixtures showed a decrease in the prevalence of WSSV but not IHHNV. The results obtained in this preliminary study revealed a protective effect of the two bacterial mixtures against WSSV latent infections.

Aim of study: The effect of microencapsulated phenolic compound extracts of Maclura tinctoria (MTBE) on growth performance and humoral immunity markers of the white leg shrimp Litopenaeus vannamei ...juveniles (0.5 ± 0.2 g initial weight) was studied.Area of study: M. tinctoria was collected from Hampolol, Campeche, and Arroyo del Agua, Culiacan, Sinaloa, Mexico.Material and methods: Three MTBE inclusions (0.5, 1, and 2.5 g MTBE/kg, Purina®) were compared with a control commercial feed (Purina®) during 30 days. Nine phenolic acids, nine flavonols, four dihydro-flavonoids, four flavones, and seven unidentified phenolic compounds were determined fin the MTBE using a Perkin Elmer® HPLC chromatograph and diode array-detection.Main results: The mean concentrations of total phenolic compounds, total flavonoid compounds, and condensed tannins were 198.05 ± 5.59 mg gallic acid equivalent (GAE) g-1 dw, 78.57 ± 1.80 quercetin equivalent g-1, and 28.32 ± 0.33 mg epicatechin equivalent g-1, respectively. The ferric reducing antioxidant power and the total antioxidant capacity, respectively, averaged 28.32 mg GAE mL-1 and 10.9 mg ascorbic acid equivalent mL-1. Survival, weight gain, and specific growth rate of L. vannamei were similar among the experimental diets. The dietary inclusion of MTBE at 0.5 g/kg of food showed significant higher (p < 0.05) plasma hemocyte lysate protein (1.35 ± 0.055 µg mL-1), prophenoloxidase (0.47 ± 0.15, Abs. 492 nm), and superoxide anion (O2.-) activity (0.21 ± 0.07, Abs. 630 nm).Research highlights: The supplementation of MTBE at 0.5 g/kg of food could be considered as a potential alternative additive for L. vannamei diet in the juvenile production, since it improved the response of the humoral immunity markers at post larval life stages, when cultivated shrimp are more susceptible to be infected by pathogens.
Caesalpinia platyloba was evaluated as an alternative for the retention of atmospheric carbon and as a feasible and viable economic activity in terms of income for tropical deciduous forest (TDF) ...peasants in the carbon markets. A total of 110 trees of C. platyloba from plantations and a TDF in the Northwest of Mexico were sampled. Growth (increase in height, diameter, and volume curves) was adjusted to assess their growth. Growth of individuals (height, diameter at breast height DBH, age, and tree crown cover) was recorded. The Schumacher model (H = β(0)e(β1 • E-1)), by means of the guided curve method, was used to adjust growth models. Information analysis was made through the non-linear procedure with the multivariate secant or false position (DUD) method using the SAS software. Growth and increase models revealed acceptable adjustments (pseudo R(2)>0.8). C. platyloba reaches >8m of height with 12 cm in diameter and 550 cm(3) of volume, presenting the highest increase at 11 years considered as basal age. Highest significant density of wood was in good quality sites (0.80 g • cm(-3)), with a carbon content (average of 99.15tC • ha(-1)) at the highest density of 2500 trees • ha(-1) (without thinning). Average incomes of US$483.33tC • ha(-1) are expected. The profitability values (NPW = US$81,646.65, IRR = 472%, and B/C = 0.82) for C. platyloba make its cultivation a viable and profitable activity, considering a management scheme of the income derived from wood selling and from carbon credits.
