Robert Hoddle reserved land for customs at the northern end of Bellarine Street in his 1838 Geelong town plan. In 1895 Harding Park was built across this reserve including its deep natural gully used ...in earlier decades as a local tip. The park was converted into a council car park and in 2004 became a construction site for apartments. The archaeology of Harding Park and its gully were investigated through the monitoring of construction excavations. Two artefact assemblages of mostly imported mid-19th-century domestic wares were collected from deeply buried deposits. The provenance of these artefacts is discussed with reference to the historical development of the Corio Street neighbourhood. Over six decades, the Harding Park gully changed from being a customs reserve en route to Geelong's hinterland and town, to a storm water drain and local rubbish tip.

J Strength ...Cond Res 28(10)2845–2851, 2014—Cutting ability is an important aspect of many team sports, however, the biomechanical determinants of cutting performance are not well understood. This study aimed to address this issue by identifying the kinetic and kinematic factors correlated with the time to complete a cutting maneuver. In addition, an analysis of the test-retest reliability of all biomechanical measures was performed. Fifteen (n = 15) elite multidirectional sports players (Gaelic hurling) were recruited, and a 3-dimensional motion capture analysis of a 75° cut was undertaken. The factors associated with cutting time were determined using bivariate Pearsonʼs correlations. Intraclass correlation coefficients (ICCs) were used to examine the test-retest reliability of biomechanical measures. Five biomechanical factors were associated with cutting time (2.28 ± 0.11 seconds)peak ankle power (r = 0.77), peak ankle plantar flexor moment (r = 0.65), range of pelvis lateral tilt (r = −0.54), maximum thorax lateral rotation angle (r = 0.51), and total ground contact time (r = −0.48). Intraclass correlation coefficient scores for these 5 factors, and indeed for the majority of the other biomechanical measures, ranged from good to excellent (ICC >0.60). Explosive force production about the ankle, pelvic control during single-limb support, and torso rotation toward the desired direction of travel were all key factors associated with cutting time. These findings should assist in the development of more effective training programs aimed at improving similar cutting performances. In addition, test-retest reliability scores were generally strong, therefore, motion capture techniques seem well placed to further investigate the determinants of cutting ability.
Our previous studies have shown that systemic neonatal murine cytomegalovirus (MCMV) infection of BALB/c mice spread to the eye with subsequent establishment of latency in choroid/RPE. In this study, ...RNA sequencing (RNA-Seq) analysis was used to determine the molecular genetic changes and pathways affected by ocular MCMV latency. MCMV (50 pfu per mouse) or medium as control were injected intra-peritoneally (i.p.) into BALB/c mice at <3 days after birth. At 18 months post injection, the mice were euthanized, and the eyes were collected and prepared for RNA-Seq. Compared to three uninfected control eyes, we identified 321 differentially expressed genes (DEGs) in six infected eyes. Using the QIAGEN Ingenuity Pathway Analysis (QIAGEN IPA), we identified 17 affected canonical pathways, 10 of which function in neuroretinal signaling, with the majority of DEGs being downregulated, while 7 pathways function in upregulated immune/inflammatory responses. Retinal and epithelial cell death pathways involving both apoptosis and necroptosis were also activated. MCMV ocular latency is associated with upregulation of immune and inflammatory responses and downregulation of multiple neuroretinal signaling pathways. Cell death signaling pathways are also activated and contribute to the degeneration of photoreceptors, RPE, and choroidal capillaries.
Abstract The aim of this study was to assess and compare the ability of discrete point analysis (DPA), functional principal component analysis (fPCA) and analysis of characterizing phases (ACP) to ...describe a dependent variable (jump height) using vertical ground reaction force curves captured during the propulsion phase of a countermovement jump. FPCA and ACP are continuous data analysis techniques that reduce the dimensionality of a data set by identifying phases of variation (key phases), which are used to generate subject scores that describe a subject׳s behavior. A stepwise multiple regression analysis was used to measure the ability to describe jump height of each data analysis technique. Findings indicated that the order of effectiveness (high to low) across the examined techniques was: ACP (99%), fPCA (78%) and DPA (21%). DPA was outperformed by fPCA and ACP because it can inadvertently compare unrelated features, does not analyze the whole data set and cannot examine important features that occur solely as a phase. ACP outperformed fPCA because it utilizes information within the combined magnitude-time domain, and identifies and examines key phases separately without the deleterious interaction of other key phases.

J Strength Cond Res 29(12)3292–3299, ...2015—Previous studies investigating the biomechanical factors associated with maximal countermovement jump height have typically used cross-sectional data. An alternative but less common approach is to use pre-to-posttraining change data, where the relationship between an improvement in jump height and a change in a factor is examined more directly. Our study compared the findings of these approaches. Such an evaluation is necessary because cross-sectional studies are currently a primary source of information for coaches when examining what factors to train to enhance performance. The countermovement jump of 44 males was analyzed before and after an 8-week training intervention. Correlations with jump height were calculated using both cross-sectional (pretraining data only) and pre-to-posttraining change data. Eight factors identified in the cross-sectional analysis were not significantly correlated with a change in jump height in the pre-to-post analysis. Additionally, only 6 of 11 factors identified in the pre-to-post analysis were identified in the cross-sectional analysis. These findings imply that (a) not all factors identified in a cross-sectional analysis may be critical to jump height improvement and (b) cross-sectional analyses alone may not provide an insight into all of the potential factors to train to enhance jump height. Coaches must be aware of these limitations when examining cross-sectional studies to identify factors to train to enhance jump ability. Additional findings highlight that although exercises prescribed to improve jump height should aim to enhance concentric power production at all joints, a particular emphasis on enhancing hip joint peak power may be warranted.

(2) Methods: caspase-12−/− (KO) or caspase-12+/+ (WT) mice were immunosup eyes were ...analyzed by plaque assay, TUNEL assay, immunohistochemical staining, western blotting, and real-time PCR. (3) Results: increased retinitis and a more extensive virus spread were detected in the retina of infected eyes of KO mice compared to WT mice at day 14 p.i. Compared to MCMV injected WT eyes, mRNA levels of interferons α, β and γ were significantly reduced in the neural retina of MCMV-infected KO eyes at day 14 p.i. Although similar numbers of MCMV infected cells, similar virus titers and similar numbers of TUNEL-staining cells were detected in injected eyes of both KO and WT mice at days 7 and 10 p.i., significantly lower amounts of cleaved caspase-3 and p53 protein were detected in infected eyes of KO mice at both time points. (4) Conclusions: caspase-12 contributes to caspase-3-dependent and independent retinal bystander cell death during MCMV retinitis and may also play an important role in innate immunity against virus infection of the retina.
Chronic bone degenerative diseases represent a major threat to the health and well-being of the population, particularly those with advanced age. This study isolated exosomes (EXO), natural ...nano-particles, from dendritic cells, the "directors" of the immune response, to examine the immunobiology of DC EXO in mice, and their ability to reprogram immune cells responsible for experimental alveolar bone loss in vivo. Distinct DC EXO subtypes including immune-regulatory (regDC EXO), loaded with TGFB1 and IL10 after purification, along with immune stimulatory (stimDC EXO) and immune "null" immature (iDCs EXO) unmodified after purification, were delivered via I.V. route or locally into the soft tissues overlying the alveolar bone. Locally administrated regDC EXO showed high affinity for inflamed sites, and were taken up by both DCs and T cells in situ. RegDC EXO-encapsulated immunoregulatory cargo (TGFB1 and IL10) was protected from proteolytic degradation. Moreover, maturation of recipient DCs and induction of Th17 effectors was suppressed by regDC EXO, while T-regulatory cell recruitment was promoted, resulting in inhibition of bone resorptive cytokines and reduction in osteoclastic bone loss. This work is the first demonstration of DC exosome-based therapy for a degenerative alveolar bone disease and provides the basis for a novel treatment strategy.
The drop jump is a popular form of plyometric exercise often undertaken to enhance countermovement jump ability (jump height). Despite its popularity the effects of drop jump training on ...countermovement jump height are often inconsistent. Such inconsistencies may be as a result of differences in the drop jump technique being employed. Two recognised forms of drop jump are the "countermovement" drop jump and the "bounce" drop jump and the current study examined the effects of eight weeks of training with these drop jump techniques on countermovement jump height. Methods: A kinetic and kinematic analysis of each participant's countermovement jump, bounce- and countermovement drop jumps was undertaken prior to training. Participants were then randomly assigned to a bounce drop jump training group (n = 34), a countermovement drop jump training group (n = 35) or a control group (n = 34). Changes in jump height were examined following training. Results: The countermovement drop jump training group increased their countermovement jump height by 2.9 cm (6%), which was a significant change (P < 0.05) in comparison to that experienced by the bounce drop jump (-0.2 cm, -0.4%) and the control group (-0.1 cm, 0.2%). Conclusion: The countermovement drop jump may be more effective than the bounce drop jump at enhancing countermovement jump height.

This paper presents comparative research on marine shell from four 19th century historical archaeological sites in Melbourne. The shell derives predominantly from Mud Oyster (Ostrea angasi) ...and Sydney Rock Oyster (Saccostrea glomerata) commercially harvested from natural reefs along the south‐east Australian coastline. The research collects quantitative data that informs on the 19th century oyster industry and investigates inter‐site shell variability and its implications for processing, consumption and discard. Dredging of subtidal reefs provides an explanation for the numerical dominance of oyster, the presence of subfossil cultch (Anadara) and the wide range of minor shellfish. Mud oyster and Sydney rock oyster comparisons in valve size, sided counts and preservation record significant differences within and between sites due to the origins, depositional conditions and the processing of the shell. These data form the basis of two models. The first predicts the archaeological representation of reef dredging and ranks shellfish according to categories, from live oysters to dead shell sampled from the reef substrate. Based on oyster shell anatomy and the separate uses of the right (lid) and left (dish) valves, the second model considers how oyster processing and consumption is characterised archaeologically in differential valve counts and pairing rates.

Cet article présente une étude comparative sur les coquillages marins provenant de quatre sites archéologiques historiques du XIXe siècle à Melbourne. Les coquilles proviennent principalement d'huîtres plates (Ostrea angasi) et d'huitres rocheuses de Sydney (Saccostrea glomerata) récoltées à but commercial sur les récifs naturels le long de la côte sud‐est de l'Australie. La recherche récolte des données quantitatives renseignant sur l'industrie ostréicole au 19e siècle et étudie la variabilité des coquilles entre les sites et ses conséquences sur la transformation, la consommation et les rejets. Le dragage des récifs infra littoraux explique la prédominance numérique des huîtres, la présence de culches subfossiles (Anadara) et la large gamme de crustacés mineurs. Les comparaisons entre les huitres plates et les huitres rocheuses de Sydney en terme de taille des valves, du nombre de cotés et de préservation, indiquent d'importantes différences au sein et entre les sites en raison des conditions de dépôt et le traitement des coquilles. Ces données constituent la base de deux modèles. La première prédit la représentation archéologique du dragage du récif et classe les coquillages selon des catégories, depuis les huîtres vivantes jusqu'aux coquilles mortes échantillonnées dans le substrat récifal. Basé sur l'anatomie des coquilles d'huître et les utilisations distinctes des valves droite (couvercle) et gauche (plat), le deuxième modèle examine comment le traitement et la consommation des huîtres sont caractérisées archéologiquement par les différents nombres de valves et taux d'appariement.
