Summary
Background
Strict avoidance is the only accepted management for cow's milk (CM) allergy. CM oral immunotherapy (CM‐OIT) is under investigation.
Objectives
To evaluate long‐term safety of ...CM‐OIT. To identify clinical/immunological predictors of adverse events.
Methods
Prospective longitudinal epidemiological intervention study. CM‐allergic children aged 5–18 underwent a Spanish‐approved CM‐OIT protocol without premedication. Clinical data, skin prick test (SPT) and specific IgE (sIgE) at baseline and 1 year after OIT were registered. All dose‐related reactions, treatments needed and cofactors involved were recorded. Through survival analysis, we studied the cumulative probability of reactions resolution over time and clinical/immunological risk factors of reactions persistence.
Results
81 children were recruited. Mean follow‐up was 25 months. 95% of children suffered reactions, 91% of which affected a single organ. Reactions were heterogeneously distributed: (a) 60 children (75%) had occasional symptoms which ceased over time. 86% of them reached complete desensitization (200 mL). (b) 20 children (25%) suffered frequent (78% of total reactions), more severe and unpredictable reactions, which persisted during follow‐up or led to withdrawal (6 cases). Reactions persistence was associated with a higher frequency and severity. Kaplan–Meier estimate revealed a cumulative probability of reactions resolution of 25% at 3 months (95% CI: 1.9–4.1) and 50% (95% CI: 6.1–9.9) at 8 months based on all patients. Cox proportional hazards multivariate regression model identified 3 variables (CM‐sIgE ≥ 50 KU L−1, CM‐SPT ≥ 9 mm and Sampson's severity grades 2, 3 and 4 at baseline food challenge) as independent risk factors of reactions persistence. The combination of 2 or 3 of these factors involved hazard ratios to develop persistent reactions of 2.26 (95% CI: 1.14–4.46; P = 0.019) and 6.06 (95% CI: 2.7–13.7; P < 0.001), respectively.
Clinical implications
CM‐OIT was insufficiently safe in 25% of children. The above‐mentioned clinical and immunological parameters would help clinicians to identify highly reactive patients before CM‐OIT. In them, individualized schedules and premedication should be considered.
Disrupted hippocampal performance underlies psychiatric comorbidities and cognitive impairments in patients with neurodegenerative disorders. To understand the contribution of adult hippocampal ...neurogenesis (AHN) to amyotrophic lateral sclerosis, Huntington’s disease, Parkinson’s disease, dementia with Lewy bodies, and frontotemporal dementia, we studied postmortem human samples. We found that adult-born dentate granule cells showed abnormal morphological development and changes in the expression of differentiation markers. The ratio of quiescent to proliferating hippocampal neural stem cells shifted, and the homeostasis of the neurogenic niche was altered. Aging and neurodegenerative diseases reduced the phagocytic capacity of microglia, triggered astrogliosis, and altered the microvasculature of the dentate gyrus. Thus, enhanced vulnerability of AHN to neurodegeneration might underlie hippocampal dysfunction during physiological and pathological aging in humans.

We describe cyanobacterial diversity in Mediterranean semiarid soil crusts along an aridity gradient by using next-generation sequencing and bioinformatics analyses, and detect clear shifts along it in cyanobacterial dominance.
Statistical analyses show that temperature and precipitation were major parameters determining cyanobacterial composition, suggesting the presence of differentiated climatic niches for distinct cyanobacteria. The responses to temperature of a set of cultivated, pedigreed strains representative of the field populations lend direct support to that contention, with psychrotolerant vs thermotolerant physiology being strain dependent, and consistent with their dominance along the natural gradient.
Our results suggest a possible replacement, as global warming proceeds, of cool-adapted by warm-adapted nitrogen-fixing cyanobacteria (such as Scytonema) and a switch in the dominance of Microcoleus vaginatus by thermotolerant, novel phylotypes of bundle-forming cyanobacteria. These differential sensitivities of cyanobacteria to rising temperatures and decreasing precipitation, their ubiquity, and their low generation time point to their potential as bioindicators of global change.
Purpose - The objective of this paper is to analyse whether individuals' socioeconomic characteristics - age, gender and income - influence their online shopping behaviour. The individuals analysed ...are experienced e-shoppers i.e. individuals who often make purchases on the internet.Design methodology approach - The technology acceptance model was broadened to include previous use of the internet and perceived self-efficacy. The perceptions and behaviour of e-shoppers are based on their own experiences. The information obtained has been tested using causal and multi-sample analyses.Findings - The results show that socioeconomic variables moderate neither the influence of previous use of the internet nor the perceptions of e-commerce; in short, they do not condition the behaviour of the experienced e-shopper.Practical implications - The results obtained help to determine that once individuals attain the status of experienced e-shoppers their behaviour is similar, independently of their socioeconomic characteristics. The internet has become a marketplace suitable for all ages and incomes and both genders, and thus the prejudices linked to the advisability of selling certain products should be revised.Originality value - Previous research related to the socioeconomic variables affecting e-commerce has been aimed at forecasting who is likely to make an initial online purchase. In contrast to the majority of existing studies, it is considered that the current development of the online environment should lead to analysis of a new kind of e-shopper (experienced purchaser), whose behaviour differs from that studied at the outset of this research field. The experience acquired with online shopping nullifies the importance of socioeconomic characteristics.
Direct numerical simulation data of a Mach 2.9, 24○ compression ramp configuration are used to analyse the shock motion. The motion can be observed from the animated DNS data available with the ...online version of the paper and from wall-pressure and mass-flux signals measured in the free stream. The characteristic low frequency is in the range of (0.007–0.013) U∞/δ, as found previously. The shock motion also exhibits high-frequency, of O(U∞/δ), small-amplitude spanwise wrinkling, which is mainly caused by the spanwise non-uniformity of turbulent structures in the incoming boundary layer. In studying the low-frequency streamwise oscillation, conditional statistics show that there is no significant difference in the properties of the incoming boundary layer when the shock location is upstream or downstream. The spanwise-mean separation point also undergoes a low-frequency motion and is found to be highly correlated with the shock motion. A small correlation is found between the low-momentum structures in the incoming boundary layer and the separation point. Correlations among the spanwise-mean separation point, reattachment point and the shock location indicate that the low-frequency shock unsteadiness is influenced by the downstream flow. The potential of near infrared spectroscopy (NIR) with remote reflectance fiber-optic probes for determining the mineral composition of propolis was evaluated. This technology allows direct ...measurements without prior sample treatment. Ninety one samples of propolis were collected in Chile (Bio-Bio region) and Spain (Castilla-León and Galicia regions). The minerals measured were aluminum, calcium, iron, potassium, magnesium, phosphorus, and some potentially toxic trace elements such as zinc, chromium, nickel, copper and lead. The modified partial least squares (MPLS) regression method was used to develop the NIR calibration model. The determination coefficient (R2) and root mean square error of prediction (RMSEP) obtained for aluminum (0.79, 53), calcium (0.83, 94), iron (0.69, 134) potassium (0.95, 117), magnesium (0.70, 99), phosphorus (0.94, 24) zinc (0.87, 10) chromium (0.48, 0.6) nickel (0.52, 0.7) copper (0.64, 0.9) and lead (0.70, 2) in ppm. The results demonstrated that the capacity for prediction can be considered good for wide ranges of potassium, phosphorus and zinc concentrations, and acceptable for aluminum, calcium, magnesium, iron and lead. This indicated that the NIR method is comparable to chemical methods. The method is of interest in the rapid prediction of potentially toxic elements in propolis before consumption.
Plastic pollution is one of the most acute environmental topics of our time. While there is a great scientific effort to tackle this problem, it has not always been well-coordinated or properly ...targeted. In this short review, we call for scientists to get involved in three crucial topics (threats) underestimated—or ignored—in freshwater systems: (i) plastic-species entanglement, (ii) plastic as nesting material, and (iii) macroplastic debris coming from mismanaged household solid waste. Reducing the knowledge gaps between marine and freshwater environments will be crucial to solute the plastic pollution problem effectively and globally. Therefore, we make a plea here to reinforce research activities on these three issues in freshwater environments worldwide.
The role of microglia during neurodegeneration remains controversial. We investigated whether microglial cells have a neurotoxic or neuroprotective function in the retina. Retinal explants from ...10-day-old mice were treated in vitro with minocycline to inhibit microglial activation, with LPS to increase microglial activation, or with liposomes loaded with clodronate (Lip-Clo) to deplete microglial cells. Flow cytometry was used to assess the viability of retinal cells in the explants and the TUNEL method to show the distribution of dead cells. The immunophenotypic and morphological features of microglia and their distribution were analyzed with flow cytometry and immunocytochemistry. Treatment of retinal explants with minocycline reduced microglial activation and simultaneously significantly decreased cell viability and increased the presence of TUNEL-labeled cell profiles. This treatment also prevented the migration of microglial cells towards the outer nuclear layer, where cell death was most abundant. The LPS treatment increased microglial activation but had no effect on cell viability or microglial distribution. Finally, partial microglial removal with Lip-Clo diminished the cell viability in the retinal explants, showing a similar effect to that of minocycline. Hence, cell viability is diminished in retinal explants cultured in vitro when microglial cells are removed or their activation is inhibited, indicating a neurotrophic role for microglia in this system.

Bacterial symbioses allow annelids to colonise extreme ecological niches, such as hydrothermal vents and whale falls. Yet, the genetic principles sustaining these symbioses remain unclear. Here, we ...show that different genomic adaptations underpin the symbioses of phylogenetically related annelids with distinct nutritional strategies. Genome compaction and extensive gene losses distinguish the heterotrophic symbiosis of the bone-eating worm Osedax frankpressi from the chemoautotrophic symbiosis of deep-sea Vestimentifera. Osedax's endosymbionts complement many of the host's metabolic deficiencies, including the loss of pathways to recycle nitrogen and synthesise some amino acids. Osedax's endosymbionts possess the glyoxylate cycle, which could allow more efficient catabolism of bone-derived nutrients and the production of carbohydrates from fatty acids. Unlike in most Vestimentifera, innate immunity genes are reduced in O. frankpressi, which, however, has an expansion of matrix metalloproteases to digest collagen. Our study supports that distinct nutritional interactions influence host genome evolution differently in highly specialised symbioses.

Microplastics can adsorb chemical pollutants such as metals or pharmaceuticals, and transferred them along the food chain. In this work, an investigation of the adsorption of Cd, Co, Cr, Cu, Ni, Pb ...and Zn by five different types of microplastics was performed in Milli-Q water and natural waters (seawater, urban wastewater and irrigation water) via a series of batch adsorption experiments. The effects of concentration of metals and physicochemical characteristics of polymers were particularly studied. Results revealed a significant adsorption of lead, chromium and zinc on microplastics, especially on polyethylene and polyvinyl chloride. In the case of polyethylene terephthalate, it showed little adsorption capacity. Specific surface, porosity and morphology are characteristics that affect the molecular interactions. The adsorption isotherms were better described by Langmuir model, which indicates that the main adsorption mechanism might be chemical adsorption. Finally, results obtained in natural waters indicated that dissolved organic matter may play a major role on metal adsorption on microplastics. Results showed an enhancement of metal adsorption in waters with high chemical and biological oxygen demands as urban wastewater and irrigation water.

Main finding of the work: PE, PP, PS and PVC showed high potential as metal carriers. Combination of all properties of polymers influences the adsorption behavior. Higher adsorption was found in natural water with high organic matter content.
