The supramolecular architectures of amide-containing compounds are highly dependent on the side-chain substituents, although the potential impact of isoxazole substituents on polymorph formation has ...not been thoroughly explored. Hence, three distinct forms of N 1 , N 3 -bis(5-methylisoxazol-3-yl)malonamide (1) were obtained and characterized: two polymorphic forms and one solvate. An in-depth analysis of the interactions and energy content of the crystals based on supramolecular clusters allowed us to propose crystallization mechanisms (crystal retrosynthesis). Specifically, the energy similarities between the interaction of the first sites NH amide ⋯OC amide (form 1I) and the symmetric sites NH amide ⋯N isox (form 1II) were found to contribute to their formation. Nonetheless, the presence of DMSO resulted in the formation of form 1III, where the solvent molecule disrupted amide-amide interactions. The first nuclei are more stable than forms 1I and 1II. The compound of N 1 , N 2 -bis(5-methylisoxazol-3-yl)oxalamide (2) was used as a comparison, and through the absence of polymorphs, revealed that the central carbon in molecule 1 allows a flexible adaptation that leads to the three forms. These findings suggest that variations in solvents, flexibility, and the presence/absence of amide–amide interactions can modulate the competition between amide-containing isoxazole compounds.
The objective of this work was to find the ideal pretreatment conditions with high efficiency to obtain a hydrolyzate rich in fermentable sugars and low possible inhibitors levels. Thus, it was ...applied diluted phosphoric acid to pretreat the sugarcane biomass. Through a Central Composite Design, it was evaluated the influence of temperature, operating time and acid concentration. The pretreatment efficiency was verified by the concentration of total monosaccharides in the liquid fraction after the reaction. The phosphoric acid concentration of 4.95% at 80 °C, during 375 min, resulted in a hemicellulosic hydrolyzate with the highest concentration of fermentable sugars (saccharification greater than 99%), with the absence of HMF and furfural, and relatively low amounts of acetic acid.
Molecular detection and classification of the bacterial groups in a sample are relevant in several areas, including medical research and forensics. Sanger sequencing of the 16S rRNA gene is ...considered the gold standard for microbial phylogenetic analysis. However, the development of massively parallel sequencing (MPS) offers enhanced sensitivity and specificity for microbiological analyses. In addition, 16S rRNA target amplification followed by MPS facilitates the combined use of multiple markers/regions, better discrimination of sample background, and higher sample throughput. We designed a novel set of 16S rRNA gene primers for detection of bacterial species associated with clinical, bioweapon, and biohazards microorganisms via alignment of 364 sequences representing 19 bacterial species and strains relevant to medical and forensics applications. In silico results indicated that the hypervariable regions (V1V2), (V4V5), and (V6V7V8) support the resolution of a selected group of bacteria. Interspecies and intraspecies comparisons showed 74.23%–85.51% and 94.48%–99.98% sequencing variation among species and strains, respectively. Sequence reads from a simulated scenario of bacterial species mapped to each of the three hypervariable regions of the respective species with different affinities. The minimum limit of detection was achieved using two different MPS platforms. This protocol can be used to detect or monitor as low as 2,000 genome equivalents of bacterial species associated with clinical, bioweapon, and biohazard microorganisms and potentially can distinguish natural outbreaks of pathogenic microorganisms from those occurring by intentional release.
Climate changes have become undisputed, as have their consequences for global ecosystems and mankind. The coastal areas are among the most affected areas on the planet due to their geographical ...location. The effects suffered by coastal areas can render the residing populations homeless, as well as compromise the continuity of the history and culture of these environments. The Marine Extractive Reserve of the city of Soure (coastal area of eastern Amazonia) stands out for housing populations that have developed an intimate relationship with nature and have knowledge that can explain people's perception of climate changes. In this context, this study investigated how local residents perceive climate change and its consequences considering different temporal and spatial scales. To this end, questionnaires were developed and applied using a 5-point Likert scale. Our results indicate that perception is shaped by socioeconomic and demographic factors, and that they are perceived on different time scales and geographic space. These findings reflect the awareness-raising efforts of the management body of this Conservation Unit and the local knowledge, derived from the relationship of the residents with the natural environment, which, together, provided the population with assertive information that favor a better understanding of this phenomenon.
•The environmental perceptions are shaped by community age and geographic distance.•Climate change perceptions are tied to expectations of the future.•Climate change is understood at different temporal and spatial scales.
In approximately 15% of patients with acute myeloid leukemia (AML), total and phosphorylated EGFR proteins have been reported to be increased compared to healthy CD34
+
samples. However, it is ...unclear if this subset of patients would benefit from EGFR signaling pharmacological inhibition. Pre-clinical studies on AML cells provided evidence on the pro-differentiation benefits of EGFR inhibitors when combined with ATRA or ATO
in vitro
. Despite the success of ATRA and ATO in the treatment of patients with acute promyelocytic leukemia (APL), therapy-associated resistance is observed in 5-10% of the cases, pointing to a clear need for new therapeutic strategies for those patients. In this context, the functional role of EGFR tyrosine-kinase inhibitors has never been evaluated in APL. Here, we investigated the EGFR pathway in primary samples along with functional
in vitro
and
in vivo
studies using several APL models. We observed that total and phosphorylated EGFR (Tyr992) was expressed in 28% and 19% of blast cells from APL patients, respectively, but not in healthy CD34
+
samples. Interestingly, the expression of the EGF was lower in APL plasma samples than in healthy controls. The EGFR ligand AREG was detected in 29% of APL patients at diagnosis, but not in control samples.
In vitro
, treatment with the EGFR inhibitor gefitinib (ZD1839) reduced cell proliferation and survival of NB4 (ATRA-sensitive) and NB4-R2 (ATRA-resistant) cells. Moreover, the combination of gefitinib with ATRA and ATO promoted myeloid cell differentiation in ATRA- and ATO-resistant APL cells.
In vivo
, the combination of gefitinib and ATRA prolonged survival compared to gefitinib- or vehicle-treated leukemic mice in a syngeneic transplantation model, while the gain in survival did not reach statistical difference compared to treatment with ATRA alone. Our results suggest that gefitinib is a potential adjuvant agent that can mitigate ATRA and ATO resistance in APL cells. Therefore, our data indicate that repurposing FDA-approved tyrosine-kinase inhibitors could provide new perspectives into combination therapy to overcome drug resistance in APL patients.
Bees are typically diurnal but around 1% of described species have nocturnal activity. Nocturnal bees are still poorly studied due to bias towards studying diurnal insects. However, knowledge ...concerning their biology and role as crop pollinators has increased. We review the literature on nocturnal bees’ traits and their host plants, and assess the crop pollination effectiveness of this neglected group. Nocturnal bees have visual adaptations to cope with low light intensities, and floral scents are a key sensory cue used to find their host flowers. Nocturnal bees generally show high flower constancy, the ability to vibrate flowers, and high transfer rates of pollen grains to stigmas. The flowers visited by nocturnal bees range from small radial and zygomorphic flowers to large brush blossoms; moreover, they visit plants with different flowering strategies. Nocturnal bees are effective pollinators of regional fruit crops in Brazil, such as cambuci (Campomanesia phaea), guaraná (Paullinia cupana), cajá (Spondias mombin), and in North America of cultivated pumpkins (Cucurbita species). However, they most likely are pollinators of several other crops. Strategies to host high numbers of nocturnal bees around cropping areas should be taken, such as preserving adjacent native forests, restricting soil management, providing food resources beyond crop flowers, and avoiding light pollution.
Systemic sclerosis (SSc) is a rare chronic autoimmune disease characterized by vascular abnormalities and connective tissue disorders. In 2021, the multidisciplinary team of the university hospital ...recognized a demand for oral health information among SSc patients. This study aims to describe the development of an SSc oral health booklet and its validation by people with SSc.
For the development of the booklet, the project creators employed the action research methodology involving ten stages. SSc patients evaluated the material by filling out a digital form.
One hundred and thirty-one people with SSc evaluated the booklet. It received an average score of 9.73 (SD: 0.80) in relevance, 9.82 (SD: 0.47) in explicitness, and 9.49 (SD: 0.93) in aesthetics. The final version was emailed to the participants and posted on social media.
This is the first booklet that deeply addresses these issues in Portuguese and constitutes a soft technology to instruct and assist patients with SSc.
The production of high-quality educational materials on oral health for patients with SSc is necessary. The online distribution increased its reach and had great importance in the context of the COVID-19 pandemic.
•Systemic sclerosis orofacial abnormalities impose challenges to health professionals.•Health education enables patients to self-manage systemic sclerosis.•Educational booklets must use simple language and illustrations to improve patients' understanding.•Patients' suggestions are essential to improve educational booklets.•The online distribution of educational materials improves their reach.
Purpose
– Interfirm relationships create value, but buyers and suppliers can appropriate this value in different amounts. Using the relational-view of strategy, the purpose of this paper is to ...explain value creation and determine the portion of that value appropriated by each organization.
Design/methodology/approach
– The data source was a survey with 166 respondents covering two industries. The authors used confirmatory factor analysis to validate construct measurement and structural equation modeling to test the hypotheses. A parallel qualitative investigation composed of 31 interviews assisted in interpreting the findings.
Findings
– Based on the relational view of strategy, the authors found support for only two of the four hypotheses that sought to explain value creation. This result calls into question the applicability of this theory to contexts other than the automotive industry, in which it was developed. Only a joint construct combining Relational Governance and Resource Complementarity had a significant effect on relational value creation. With respect to value appropriation, although both buyers and suppliers captured part of the relational value created, the buyers tended to receive the great majority of this value.
Research limitations/implications
– The focussed context of this study (Brazilian companies in the personal care/cosmetics and food/beverage industries) limits its generalizability but provides deeper insight into the interpretation of its results.
Practical implications
– Both buyers and suppliers can benefit from collaborative relationships, but buyers appear to capture a larger share, forcing suppliers to continuously seek new sources of value.
Originality/value
– This paper bridges the gap between the buyer-supplier literature and the definition of competitive advantage as value creation found in the strategic management literature. This study proposes and tests an integrative definition of the relational value that is created and appropriated in a dyad.

A total of 3109 crustaceans belonging to 50 taxa distributed in 42 families were found in 117 analysed stomachs of flying gurnard (Dactylopterus volitans). Samples were obtained in April 2008 by the ...R/V Gyre using a bottom trawl towed in 12 stations at 14-100 m depth on the continental shelf of the Campos Basin, Brazil. The carcinofauna was analysed and the order Calanoida (Copepoda) found to be the most important item in terms of relative abundance and frequency of occurrence, followed by the order Amphipoda (Peracarida), the infraorder Brachyura (Decapoda), the order Stomatopoda and the subclass Myodocopa (Ostracoda). In the order Calanoida, the species Pontellopsis cf. villosa (Pontellidae) represented 98.04% of total crustacean abundance. The diet of Dactylopterus volitans varied according to fish size, with higher diversity of Crustacea at smaller size classes, decreasing in larger fishes. A similar pattern regarding depth was obtained, with greater diversity of taxa in gurnard stomachs caught at shallower depths. Flying gurnard is considered a generalized carnivore of invertebrates, eating mobile macrobenthic organisms, such as crustaceans, and its diet varies with its life stage, without any specific group as its main food source.
Here, we evaluated whether the overexpression of transcriptionally inactive ΔNp73 cooperates with PML/RARA fusion protein in the induction of an APL-leukemic phenotype, as well as its role in vitro ...in proliferation, myeloid differentiation, and drug-induced apoptosis. Using lentiviral gene transfer, we showed in vitro that ΔNp73 overexpression resulted in increased proliferation in murine bone marrow (BM) cells from hCG-PML/RARA transgenic mice and their wild-type (WT) counterpart, with no accumulation of cells at G2/M or S phases; instead, ΔNp73-expressing cells had a lower rate of induced apoptosis. Next, we evaluated the effect of ΔNp73 on stem-cell self-renewal and myeloid differentiation. Primary BM cells lentivirally infected with human ΔNp73 were not immortalized in culture and did not present significant changes in the percentage of CD11b. Finally, we assessed the impact of ΔNp73 on leukemogenesis or its possible cooperation with PML/RARA fusion protein in the induction of an APL-leukemic phenotype. After 120 days of follow-up, all transplanted mice were clinically healthy and, no evidence of leukemia/myelodysplasia was apparent. Taken together, our data suggest that ΔNp73 had no leukemic transformation capacity by itself and apparently did not cooperate with the PML/RARA fusion protein to induce a leukemic phenotype in a murine BM transplantation model. In addition, the forced expression of ΔNp73 in murine BM progenitors did not alter the ATRA-induced differentiation rate in vitro or induce aberrant cell proliferation, but exerted an important role in cell survival, providing resistance to drug-induced apoptosis.
