A highly substituted family of imidazoles is effectively obtained through the Debus-Japp-Radziszewski reaction. In this process, benzil, ammonium acetate, and various benzaldehydes react in a ...particularly notable solvent: ethyl lactate (EL). This bio-based solvent, derived from biomass fermentation, stands out not only for its sustainable origin but also for its remarkable properties. Ethyl lactate is biodegradable, health-risk-free, is easily recyclable, and is non-corrosive, categorizing it as an exemplary green solvent. The multicomponent reaction, carried out under these conditions, eliminates the need for a catalyst, resulting in products with good yields. The isolation of the products is very simple, requiring only filtration, as they are insoluble in the solvent. In this way, this methodology aligns with various principles of green chemistry, emphasizing the strategic choice of ethyl lactate. This choice has a positive impact on the synthesis of imidazoles, well-known for their pharmacological properties.
This study aimed to assess consumers' uses of and opinions on the current Brazilian food label and their reaction to the introduction of a front-of-package warning label. We conducted 12 focus groups ...among a diverse sample of adult consumers, to broadly assess: (1) uses of and perceptions about the current food label, and (2) opinions about implementing a front-of-package warning label to guide food purchases. Data was analyzed with a triangulation of researchers using an exploratory content analysis, which allowed codes to emerge from the data. The frequency of codes across focus groups was compared by gender and socioeconomic status to explore differences by these sociodemographic factors. Codes were divided into six main themes: (1) "Reasons for using food labels"; (2) "Barriers to using food labels"; (3) "Requirements for a new label"; (4) "Perceived influence on consumption behaviors"; (5) "Perceived influence on child behaviors"; and (6) "Perceptions of the food manufacturers using of warning labels". Participants used food labels to check nutrient content and ingredient information but the format of these labels and the technicality of the content displayed often made the information inaccessible, particularly for those with low socioeconomic status. Most participants were supportive of the display of front-of-package warning labels on products and considered them useful to inform purchases. Women believed that they and their children would reduce the consumption of foods with front-of-package warning labels, while men reported more polarity in their intentions. For men and their children, front-of-package warning labels would result in either stopping food intake entirely or continued consumption without changes to the amount. The study results highlight the potential of front-of-package warning labels to support healthier behaviors in both consumers and their children.

Abstract
Background
Dialysis patients are typically inactive and their physical activity (PA) decreases over time. Uremic toxicity has been suggested as a potential causal factor of low PA in ...dialysis patients. Post-dilution high-volume online hemodiafiltration (HDF) provides greater higher molecular weight removal and studies suggest better clinical/patient-reported outcomes compared with hemodialysis (HD).
Methods
HDFIT was a randomized controlled trial at 13 clinics in Brazil that aimed to investigate the effects of HDF on measured PA (step counts) as a primary outcome. Stable HD patients (vintage 3–24 months) were randomized to receive HDF or high-flux HD. Treatment effect of HDF on the primary outcome from baseline to 3 and 6 months was estimated using a linear mixed-effects model.
Results
We randomized 195 patients (HDF 97; HD 98) between August 2016 and October 2017. Despite the achievement of a high convective volume in the majority of sessions and a positive impact on solute removal, the treatment effect HDF on the primary outcome was +538 95% confidence interval (CI) −330 to 1407 steps/24 h after dialysis compared with HD, and was not statistically significant. Despite a lack of statistical significance, the observed size of the treatment effect was modest and driven by steps taken between 1.5 and 24.0 h after dialysis, in particular between 20 and 24 h (+197 steps; 95% CI −95 to 488).
Conclusions
HDF did not have a statistically significant treatment effect on PA 24 h following dialysis, albeit effect sizes may be clinically meaningful and deserve further investigation.

CoronaVac, an inactivated SARS-CoV-2 vaccine, has been approved for emergency use in several countries. However, its immunogenicity in immunocompromised individuals has not been well established. We ...initiated a prospective phase 4 controlled trial (no. NCT04754698, CoronavRheum) in 910 adults with autoimmune rheumatic diseases (ARD) and 182 age- and sex-frequency-matched healthy adults (control group, CG), who received two doses of CoronaVac. The primary outcomes were reduction of ≥15% in both anti-SARS-CoV-2 IgG seroconversion (SC) and neutralizing antibody (NAb) positivity 6 weeks (day 69 (D69)) after the second dose in the ARD group compared with that in the CG. Secondary outcomes were IgG SC and NAb positivity at D28, IgG titers and neutralizing activity at D28 and D69 and vaccine safety. Prespecified endpoints were met, with lower anti-SARS-Cov-2 IgG SC (70.4 versus 95.5%, P < 0.001) and NAb positivity (56.3 versus 79.3%, P < 0.001) at D69 in the ARD group than in the CG. Moreover, IgG titers (12.1 versus 29.7, P < 0.001) and median neutralization activity (58.7 versus 64.5%, P = 0.013) were also lower at D69 in patients with ARD. At D28, patients with ARD presented with lower IgG frequency (18.7 versus 34.6%, P < 0.001) and NAb positivity (20.6 versus 36.3%, P < 0.001) than that of the CG. There were no moderate/severe adverse events. These data support the use of CoronaVac in patients with ARD, suggesting reduced but acceptable short-term immunogenicity. The trial is still ongoing to evaluate the long-term effectiveness/immunogenicity.
Several diseases are associated with excess of adipose tissue, and obesity is considered an independent risk factor for the development of cardiac remodeling and heart failure. Dietary aspects have ...been studied to elucidate the mechanisms involved in these processes. Thus, the purpose was the development and characterization of an obesity experimental model from hypercaloric diets, which resulted in cardiac remodeling and predisposition to heart failure. Thirty- day-old male Wistar rats (n = 52) were randomized into four groups: control (C), high sucrose (HS), high-fat (HF) and high-fat and sucrose (HFHS) for 20 weeks. General characteristics, comorbidities, weights of the heart, left (LV) and right ventricles, atrium, and relationships with the tibia length were evaluated. The LV myocyte cross sectional area and fraction of interstitial collagen were assayed. Cardiac function was determined by hemodynamic analysis and the contractility by cardiomyocyte contractile function. Heart failure was analyzed by pulmonary congestion, right ventricular hypertrophy, and hemodynamic parameters. HF and HFHS models led to obesity by increase in adiposity index (C = 8.3 ± 0.2% vs. HF = 10.9 ± 0.5%, HFHS = 10.2 ± 0.3%). There was no change in the morphological parameters and heart failure signals. HF and HFHS caused a reduction in times to 50% relaxation without cardiomyocyte contractile damage. The HS model presented cardiomyocyte contractile dysfunction visualized by lower shortening (C: 8.34 ± 0.32% vs. HS: 6.91 ± 0.28), as well as the Ca2+ transient amplitude was also increased when compared to HFHS. In conclusion, the experimental diets based on high amounts of sugar, lard or a combination of both did not promote cardiac remodeling with predisposition to heart failure under conditions of obesity or excess sucrose. Nevertheless, excess sucrose causes cardiomyocyte contractility dysfunction associated with alterations in the myocyte sensitivity to intracellular Ca2+.

Nature has been a source of medicinal treatments for thousands of years, with the use of plants as prototypes for drug development and for the extraction of active compounds. Skin injuries occur ...regularly in everyday life, and the human skin has the ability to promote repair spontaneously under healthy conditions. However, some intrinsic and external factors may interfere with skins' natural ability, leading to nonhealing lesions and chronic wounds, which directly affect health and quality of life. Thus, attention should be given to this health problem, using an appropriated management when necessary. In this scenario, phytotherapy may be an option for cutaneous wound treatment, although further high‐quality studies are needed to firmly establish the clinical efficacy of plants. This article reviews traditionally used natural actives for wound healing, highlighting their characteristics and mode of action.
This paper proposes the problematization of generic and mass-produced forms of architecture through the oppositional relationship with indigenous self-constructed spaces. Starting from an ...ethnographic study about an unique indigenous horticulture project in the center of Rio de Janeiro, The Dja Guata Porã Garden, we demonstrate the effort to build a territory that organizes itself based on multispecies relationships. The project is contrasted with the formal urban space where it takes place, within a housing complex "Minha Casa, Minha Vida", the largest public housing policy in Brazil, where the architecture is designed with generic spaces that cannot be modified according to the residents. From this spatial contrast, we propose that this indigenous horticulture project, with pedagogical purposes, is a gap in the formal urban space.

Mind Wandering (MW) has been associated with ADHD in a very small number of studies with adults and children. However, anxiety and depression have also been associated with MW and both are often comorbid with ADHD. The aim of this study was to investigate the role of anxiety and depression in MW in patients with ADHD.
The Mind Excessively Wandering Scale (MEWS) compared the levels of MW controlling for the presence of anxiety and depression symptoms in 78 adolescents (53 males and 25 females) comprising ADHD, clinical controls and typically developing individuals. Correlational analysis between MEWS score, demographic variables, ADHD, anxiety and depressive symptoms was performed using simple and multiple linear regression analysis demonstrating that only anxiety predicted MW scores. On a second analysis, we compared Anxiety and Non-Anxiety as well as ADHD and non-ADHD groups.
Levels of MW were significantly correlated with anxiety symptoms, but not with depression. In addition, there were no differences in ADHD and non-ADHD groups regarding MW levels.
Our results suggest MW is associated with anxiety levels, independently of an ADHD diagnosis.
Faculty members face demands such as research, outreach programs, and management activities. Such demands may expose faculty to burnout. Burnout affects the physical, psychological and social health ...of faculty members, but it is still unclear how it affects their quality of life. We aimed to assess the impact of burnout on the quality of life (QoL) of faculty members from different fields of knowledge.
Cross-sectional study using validated tools for measuring burnout and QoL (Oldenburg Burnout Inventory-OLBI and World Health Organization Quality of Life-Abbreviated version-WHOQOL-Bref) in a sample of 366 faculty members from a public university. Scores were analyzed using Student's t-test, analysis of variance (ANOVA), binary logistic regression, and structural equation modeling (SEM).
More than a third of the faculty members (n = 127; 36.6%) suffered from burnout. Men had higher scores of quality of life than women in the physical health (p = 0.001; d<0.5), psychological (p = 0.001; d<0.5) and social relationships (p = 0.048; d<0.5) domains. Women were more exhausted than men (p = 0.001; d<0.5). Faculty members' perception of quality of life and burnout did not differ according to their field of knowledge (p>0.05). Participants who felt tired before arriving at work were less likely to report good quality of life (OR = 0.46; 95% CI = 0.21-0.99). Faculty members who stated they needed more time to relax after work were less likely to be satisfied with their health (OR = 0.20; 95% CI = 0.10-0.40). Burnout showed a negative association with quality of life (λ = 0.87; p < 0,001; df = 8).
Burnout negatively affects faculty members' quality of life, regardless of their field of knowledge. Our results suggest the implementation of programs and actions to prevent burnout to faculty members, especially to women, as their quality of life may affect the quality of the education provided.

Protein tyrosine phosphatases (PTPs) are key virulence factors in pathogenic bacteria, consequently, they have become important targets for new approaches against these pathogens, especially in the ...fight against antibiotic resistance. Among these targets of interest YopH (Yersinia outer protein H) from virulent species of Yersinia is an example. PTPs can be reversibly inhibited by nitric oxide (NO) since the oxidative modification of cysteine residues may influence the protein structure and catalytic activity. We therefore investigated the effects of NO on the structure and enzymatic activity of Yersinia enterocolitica YopH in vitro. Through phosphatase activity assays, we observe that in the presence of NO YopH activity was inhibited by 50%, and that this oxidative modification is partially reversible in the presence of DTT. Furthermore, YopH S-nitrosylation was clearly confirmed by a biotin switch assay, high resolution mass spectrometry (MS) and X-ray crystallography approaches. The crystal structure confirmed the S-nitrosylation of the catalytic cysteine residue, Cys403, while the MS data provide evidence that Cys221 and Cys234 might also be modified by NO. Interestingly, circular dichroism spectroscopy shows that the S-nitrosylation affects secondary structure of wild type YopH, though to a lesser extent on the catalytic cysteine to serine YopH mutant. The data obtained demonstrate that S-nitrosylation inhibits the catalytic activity of YopH, with effects beyond the catalytic cysteine. These findings are helpful for designing effective YopH inhibitors and potential therapeutic strategies to fight this pathogen or others that use similar mechanisms to interfere in the signal transduction pathways of their hosts.
