Différentes études épidémiologiques ont montré, au cours des 50 dernières années, une augmentation de l'incidence des cancers du testicule, des malformations congénitales de l'appareil reproducteur ...masculin –– cryptorchidie et hypospadias ––, associée à une détérioration probable de la production spermatique dans l'espèce humaine. Le syndrome de dysgénésie testiculaire (SDT) a été proposé comme entité pathologique sous-jacente à ces trois perturbations. L'exposition à des composés chimiques présents dans notre environnement appelés perturbateurs endocriniens (PE) parce qu'ils modifient la production et/ou l'action des hormones endogènes serait à l'origine du SDT. Les PE sont présents dans notre environnement quotidien, c'est-à-dire dans les aliments et l'eau (à travers l'utilisation des herbicides et pesticides), les cosmétiques, les produits ménagers, etc. Des modèles expérimentaux ont été développés pour (i) établir un lien entre l'exposition aux PE et le SDT et (ii) identifier les mécanismes d'action qui entrent en jeu. Après un rappel sur les PE et après avoir souligné l'importance de la fenêtre d'exposition, quatre mécanismes identifiés seront décrits : (i) transmission intergénérationnelle (épigénétique), (ii) mort cellulaire programmée, (iii) altération du signal androgénique et (iv) rôle des cellules de soutien des cellules germinales. Pour conclure, nous tenterons de proposer quelques biomarqueurs qui pourraient être utilisés pour identifier, en pathologie testiculaire humaine, un lien avec une exposition intra-utérine aux antiandrogènes.
Several epidemiologic studies have demonstrated during the last 50 years an increased incidence in testis cancer, male genital tract malformations (cryptorchidism and hypospadias) and a decrease in sperm quality in men. These three pathologies seem to be linked and to belong to the testicular dysgenesis syndrome (TDS). It was suggested that TDS is a consequence of intra-uterine exposure to environmental compounds that disrupt the metabolism of native hormones. Such substances are so called endocrine disruptors (EDs). EDs are present in our daily environment such as food and water (through the use of pesticides), cosmetics, house-care products etc. Experimental models have been carried out to (i) establish a link between EDs exposure and SDT and (ii) identify the mechanisms that are involved in. After a brief definition of EDs and having underlined the importance of the window of exposure to EDs, several mechanisms will be described such as (i) intergenerational transmission (epigenetic), (ii) programmed cell death of testicular cells, (iii) modification of the androgenic signal and (iv) role of the germ cells-nourishing cells. To conclude, we will try to propose some biomarkers that would be useful to identify the potential link between fetal exposure to anti-androgenic EDs and male testicular pathology.
Peripheral blood lymphocytes frm 16 patients with sarcoidosis (9 patients with skin sarcoids, 7 patients with erythema nodosum and bilateral hilar adenopathy) and cutaneous anergy and from 23 ...age-matched healthy controls were characterized by reactivity with monoclonal antibodies OKT3, OKT4, OKT8 directed to surface antigens of T lymphocytes, helper-inducer and suppressor-cytotoxic T cell subsets, respectively. In contrast to healthy controls, patients with sarcoidosis had reduced percentages of OKT3+, OKT4+ and OKT8+ cells and a major decrease in the OKT8+ (suppressor) subset. However, these changes were significant only in the group of patients with acute sarcoidosis (erythema nodosum). This abnormal T cell distribution correlates with the alterations in cell-mediated immunity previously observed and suggests the presence of a defective circulating suppressor T cell activity in acute sarcoidosis.
