The DØ experiment enjoyed a very successful data-collection run at the Fermilab Tevatron collider between 1992 and 1996. Since then, the detector has been upgraded to take advantage of improvements ...to the Tevatron and to enhance its physics capabilities. We describe the new elements of the detector, including the silicon microstrip tracker, central fiber tracker, solenoidal magnet, preshower detectors, forward muon detector, and forward proton detector. The uranium/liquid-argon calorimeters and central muon detector, remaining from Run I, are discussed briefly. We also present the associated electronics, triggering, and data acquisition systems, along with the design and implementation of software specific to DØ.
TFIIS is a transcription elongation factor that stimulates transcript cleavage activity of arrested RNA polymerase II (Pol II). Recent studies revealed that TFIIS has also a role in Pol II ...transcription initiation. To improve our understanding of TFIIS function in vivo, we performed genome-wide location analysis of this factor. Under normal growth conditions, TFIIS was detected on Pol II-transcribed genes, and TFIIS occupancy was well correlated with that of Pol II, indicating that TFIIS recruitment is not restricted to NTP-depleted cells. Unexpectedly, TFIIS was also detected on almost all Pol III-transcribed genes. TFIIS and Pol III occupancies correlated well genome-wide on this novel class of targets. In vivo, some dst1 mutants were partly defective in tRNA synthesis and showed a reduced Pol III occupancy at the restrictive temperature. In vitro transcription assays suggested that TFIIS may affect Pol III start site selection. These data provide strong in vivo and in vitro evidence in favor of a role of TFIIS as a general Pol III transcription factor.
The D0 Collaboration presents first evidence for the production of single top quarks at the Fermilab Tevatron ppover collider. Using a 0.9 fb(-1) dataset, we apply a multivariate analysis to ...separate signal from background and measure sigma(ppover -->tb+X,tqb+X)=4.9+/-1.4 pb. The probability to measure a cross section at this value or higher in the absence of a signal is 0.035%, corresponding to a 3.4 standard deviation significance. We use the cross section measurement to directly determine the Cabibbo-Kobayashi-Maskawa matrix element that describes the Wtb coupling and find 0.68<|V(tb)|</=1 at 95% C.L. within the standard model.
The acrosome reaction is a unique type of regulated exocytosis. The single secretory granule of the sperm fuses at multiple points with the overlying plasma membrane. In the past few years we have ...characterized several aspects of this process using streptolysin O-permeabilized human spermatozoa. Here we show that Rab3A triggers acrosomal exocytosis in the virtual absence of calcium in the cytosolic compartment. Interestingly, exocytosis is blocked when calcium is depleted from intracellular stores. By using a membrane-permeant fluorescent calcium probe, we observed that the acrosome actually behaves as a calcium store. Depleting calcium from this compartment by using a light-sensitive chelator prevents secretion promoted by Rab3A. UV inactivation of the chelator restores exocytosis. Rab3A-triggered exocytosis is blocked by calcium pump and inositol 1,4,5-trisphosphate (IP3)-sensitive calcium channel inhibitors. Calcium measurements inside and outside the acrosome showed that Rab3A promotes a calcium efflux from the granule. Interestingly, release of calcium through IP3-sensitive calcium channels was necessary even when exocytosis was initiated by increasing free calcium in the extraacrosomal compartment in both permeabilized and intact spermatozoa. Our results show that a calcium efflux from the acrosome through IP3-sensitive channels is necessary downstream Rab3A activation during the membrane fusion process leading to acrosomal exocytosis.
We report the first direct observation of the strange b baryon Xi_b^{-}. We reconstruct the decay Xi_b^{-} to J/psi Xi^{-}, with J/psi to dimuons and Xi^{-} to Lambda pion, in ppbar collisions at ...sqrt(s) = 1.96 TeV. Using 1.3 fb^{-1} of data collected by the D0 detector, we observe 15.2 +/- 4.4(stat.)+ 1.9/-0.4(syst.) Xi_b^{-} candidates at a mass of 5.774 +/- 0.011(stat.) +/- 0.015 (syst.) GeV. The significance of the observed signal is 5.5 sigmas, equivalent to a probability of 3.3 X 10^{-8} of it arising from a background fluctuation. Normalizing to the decay Lambda_b to J/psi Lambda, we measure the relative rate to be 0.28 +/- 0.09(stat.)+ 0.09/-0.08 (syst.).
We search for the production of single top quarks via flavor-changing-neutral-current couplings of a gluon to the top quark and a charm (c) or up (u) quark. We analyze 230 pb{-1} of lepton+jets data ...from ppover collisions at a center of mass energy of 1.96 TeV collected by the D0 detector at the Fermilab Tevatron Collider. We observe no significant deviation from standard model predictions, and hence set upper limits on the anomalous coupling parameters kappa{g}{c}/Lambda and kappa{g}{u}/Lambda, where kappa{g} define the strength of tcg and tug couplings, and Lambda defines the scale of new physics. The limits at 95% C.L. are kappa{g}{c}/Lambda<0.15 TeV-1 and kappa{g}{u}/Lambda<0.037 TeV-1.
We present a measurement of the muon charge asymmetry from W boson decays using 0.3 fb(-1) of data collected at root s =1.96 GeV between 2002 and 2004 with the D0 detector at the Fermilab Tevatron ...(p) over bar Collider. We compare our findings with expectations from next-to-leading-order calculations performed using the CTEQ6.1M and MRST04 NLO parton distribution functions. Our findings can be used to constrain future parton distribution function fits.
