The highly interactive nature of predator-prey relationship is essential for ecosystem conservation; predators have been extirpated, however, from entire ecosystems all over the Earth. ...Reintroductions comprise a management technique to reverse this trend. Species Distribution Models (SDM) are preemptive tools for release-site selection, and can define levels of habitat quality over the species distribution. The Atlantic Forest of South America has lost most of its apex predators, and Harpy Eagles Harpia harpyja-Earth's largest eagle-are now limited to few forest pockets in this domain. Harpy Eagles are supposedly widespread in the Amazon Forest, however, where habitat loss and degradation is advancing at a rapid pace. We aim to describe the suitability of threatened Amazonian landscapes for this eagle. We also aim to assess the suitability of remaining Atlantic Forest sites for Harpy Eagle reintroductions. Here we show that that considerable eagle habitat has already been lost in Amazonia due to the expansion of the "Arc of Deforestation", and that Amazonian forests currently represent 93% of the current distribution of the species. We also show that the Serra do Mar protected areas in southeastern Brazil is the most promising region for Harpy Eagle reintroductions in the Atlantic Forest. Reintroduction and captive breeding programs have been undertaken for Harpy Eagles, building the technical and biological basis for a successful restoration framework. Our distribution range for this species represents a 41% reduction of what is currently proposed by IUCN. Furthermore, habitat loss in Amazonia, combined with industrial logging and hunting suggest that the conservation status of this species should be reassessed. We suggest researchers and conservation practitioners can use this work to help expand efforts to conserve Harpy Eagles and their natural habitats.

A Tricistronic vector utilizing internal ribosome entry site (IRES) elements to express the light chain (LC), heavy chain (HC), and a neomycin phosphotransferase (NPT) selection marker from one ...transcript is designed for generation of mAb expressing CHO cell lines. As compared to the commonly used vectors, benefits of this design include: (1) minimized non-expressing clones, (2) enhanced stable mAb productivity without gene amplification, (3) control of LC and HC expression at defined ratios, and (4) consistent product quality. After optimization of the LC and HC arrangement and increasing selection stringency by weakening the NPT selection marker, this Tricistronic vector is able to generate stably transfected pools with specific productivity (qmAb) greater than 5pg/cell/day (pcd) and titers over 150mg/L. 5% of clones from these pools have qmAb greater than 20pcd and titers ranging from 300 to more than 500mg/L under non-optimized shake flask batch cultures using commercially available protein-free medium. The mAb produced by these clones have low aggregation and consistent glycosylation profiles. The entire process of transfection to high-expressing clones requires only 6 months. The IRES-mediated Tricistronic vector provides an attractive alternative to commonly used vectors for fast generation of mAb CHO cell lines with high productivity.
Atopic dermatitis (AD) is a common allergic skin disease, characterized by dryness, itchiness, thickening and inflammation of the skin. Infiltration of eosinophils into the dermal layer and presence ...of edema are typical characteristics in the skin biopsy of AD patients. Previous in vitro and clinical studies showed that the Pentaherbs formula (PHF) consisting of five traditional Chinese herbal medicines, Flos Lonicerae, Herba Menthae, Cortex Phellodendri, Cortex Moutan and Rhizoma Atractylodis at w/w ratio of 2:1:2:2:2 exhibited therapeutic potential in treating AD. In this study, an in vivo murine model with oxazolone (OXA)-mediated dermatitis was used to elucidate the efficacy of PHF. Active ingredients of PHF water extract were also identified and quantified, and their in vitro anti-inflammatory activities on pruritogenic cytokine IL-31- and alarmin IL-33-activated human eosinophils and dermal fibroblasts were evaluated. Ear swelling, epidermis thickening and eosinophils infiltration in epidermal and dermal layers, and the release of serum IL-12 of the murine OXA-mediated dermatitis were significantly reduced upon oral or topical treatment with PHF (all p < 0.05). Gallic acid, chlorogenic acid and berberine contents (w/w) in PHF were found to be 0.479%, 1.201% and 0.022%, respectively. Gallic acid and chlorogenic acid could suppress the release of pro-inflammatory cytokine IL-6 and chemokine CCL7 and CXCL8, respectively, in IL-31- and IL-33-treated eosinophils-dermal fibroblasts co-culture; while berberine could suppress the release of IL-6, CXCL8, CCL2 and CCL7 in the eosinophil culture and eosinophils-dermal fibroblasts co-culture (all p < 0.05). These findings suggest that PHF can ameliorate allergic inflammation and attenuate the activation of eosinophils.
Ultrafast processes in matter, such as the electron emission after light absorption, can now be studied using ultrashort light pulses of attosecond duration (10−18 seconds) in the extreme ultraviolet ...spectral range. The lack of spectral resolution due to the use of short light pulses has raised issues in the interpretation of the experimental results and the comparison with theoretical calculations. We determine photoionization time delays in neon atoms over a 40–electron volt energy range with an interferometric technique combining high temporal and spectral resolution. We spectrally disentangle direct ionization from ionization with shake-up, in which a second electron is left in an excited state, and obtain excellent agreement with theoretical calculations, thereby solving a puzzle raised by 7-year-old measurements.
•This article presents a global time evolution analysis of the Covid-19 pandemics, through which it is shown that it has unfolded globally as a succession of six multi-logistic phases or spurts, each ...with its own characteristic duration and intensity.•The spectral analysis of the residuals of the total number of cases data fitting evidenced “hidden” oscillatory mechanisms driving the pandemics, dominated by two modes: a sharper one at a higher frequency with a period of 7 days, and a broad low frequency band with a mean period of roughly 40–50 days.•It is demonstrated that the short period mode is due to a "weekend effect", while the long period mode may be associated with the time difference of the spread of the disease among the several continents.•The detailed study of the epidemics in Portugal demonstrates the positive consequences of preventive sanitary measures, as well as how disastrous it may be the absence of such measures due hesitations and/or political positions. It's clearly demonstrated that the concentrated effort in vaccination really contributes to reducing the number of daily cases and deaths.•In the final discussion it is discussed a strong hypothesis (Gelfand's cultural tightness-looseness model) explaining the different behavior of the spreading of the Covid 19 pandemics among different countries and continents, and it is also briefly discussed the forecasting capacity of the presented model.
In this article it is presented a multi-logistic model to describe the time evolution of the Covid-19 pandemics. The model is not intended as paragon for the accurate prediction of the future number of people infected, but instead as a useful phenomenological approach for a comprehensive understanding of the pandemic development, able to uncover some hidden aspects of its unfolding. Our results, using OWID data of total cases and daily cases of Covid-19 from March 12, 2020, up to September 27, 2021, brought to light that the pandemic has unfolded globally as a multi-step logistic, namely six logistic phases, each with its own characteristic duration and intensity. Moreover, it is demonstrated how differently the pandemics spread among different countries and continents. The methodology is tested regarding its ability of forecasting, and is demonstrated that it works well in the range of circa 30 days within a margin of less than 3% error while a given phase is still in development. The case study of Portugal demonstrates the benefit of preventive sanitary measures, as well as shows how disastrous it may be the absence of such measures due to hesitations and/or political positions. Completing the article, a qualitative analysis is presented to scrutinize the possible causes of the asymmetry observed in the diffusion of Covid-19 among the different continents and countries.
Acetylcholine (ACh) plays a crucial role in physiological responses of both the central and the peripheral nervous system. Moreover, ACh was described as an anti-inflammatory mediator involved in the ...suppression of exacerbated innate response and cytokine release in various organs. However, the specific contributions of endogenous release ACh for inflammatory responses in the lung are not well understood. To address this question we have used mice with reduced levels of the vesicular acetylcholine transporter (VAChT), a protein required for ACh storage in secretory vesicles. VAChT deficiency induced airway inflammation with enhanced TNF-α and IL-4 content, but not IL-6, IL-13 and IL-10 quantified by ELISA. Mice with decreased levels of VAChT presented increased collagen and elastic fibers deposition in airway walls which was consistent with an increase in inflammatory cells positive to MMP-9 and TIMP-1 in the lung. In vivo lung function evaluation showed airway hyperresponsiveness to methacholine in mutant mice. The expression of nuclear factor-kappa B (p65-NF-kB) in lung of VAChT-deficient mice were higher than in wild-type mice, whereas a decreased expression of janus-kinase 2 (JAK2) was observed in the lung of mutant animals. Our findings show the first evidence that cholinergic deficiency impaired lung function and produce local inflammation. Our data supports the notion that cholinergic system modulates airway inflammation by modulation of JAK2 and NF-kB pathway. We proposed that intact cholinergic pathway is necessary to maintain the lung homeostasis.

Ultrafast extreme-ultraviolet (XUV) and X-ray sources are revolutionizing our ability to follow femtosecond processes with angstrom-scale resolution. The next frontier is to simultaneously control ...the direction, duration and timing of such radiation. Here, we demonstrate a fully functional opto-optical modulator for XUV light, similar to modulators available at infrared (IR) and visible wavelengths. It works by using an IR pulse to control the spatial and spectral phase of the free induction decay that results from using attosecond pulses to excite a gas. The modulator allows us to send the XUV light in a direction of our choosing at a time of our choosing. The inherent synchronization of the XUV emission to the control pulse will allow laser-pump/X-ray probe experiments with sub-femtosecond time resolution.
This study evaluated the antibacterial and antiproliferative activities of the essential oil of Psidium guajava leaves (PG-EO), traditionally used in folk medicine. The essential oil was obtained ...from fresh leaves by hydrodistillation, using a modified Clevenger apparatus. The major PG-EO chemical constituents were identified by GC-MS and GC-FID as being β-caryophyllene (16.1%), α-humulene (11.9%), aromadendrene oxide (14.7%), δ-selinene (13.6%), and selin-11-en-4α-ol (12.5%). The antibacterial activity of the essential oil of P. guajava leaves was determined in terms of its minimum inhibitory concentrations (MIC) using the broth microdilution method in 96-well microplates. PG-EO had moderate activity against Streptococcus mutans (MIC = 200 µg/mL), S. mitis (MIC = 200 µg/mL), S. sanguinis (MIC = 400 µg/mL), S. sobrinus (MIC = 100 µg/mL), and S. salivarius (MIC = 200 µg/mL). The antiproliferative activity was evaluated against different tumor cell lines: breast adenocarcinoma (MCF-7), human cervical adenocarcinoma (HeLa), and human gliobastoma (M059J). A normal human cell line (GM07492A, lung fibroblasts) was included. The antiproliferative activity was evaluated using the XTT assay and the results were expressed as IC50. The essential oil showed significantly lower IC50 values against MCF-7 and M059J lines than that obtained for the normal line, showing selectivity. Our results suggest that the essential oil of Psidium guajava L. has promising biological activities and can be considered a new source of bioactive compounds.
Electron dynamics induced by resonant absorption of light is of fundamental importance in nature and has been the subject of countless studies in many scientific areas. Above the ionization threshold ...of atomic or molecular systems, the presence of discrete states leads to autoionization, which is an interference between two quantum paths: direct ionization and excitation of the discrete state coupled to the continuum. Traditionally studied with synchrotron radiation, the probability for autoionization exhibits a universal Fano intensity profile as a function of excitation energy. However, without additional phase information, the full temporal dynamics cannot be recovered. Here we use tunable attosecond pulses combined with weak infrared radiation in an interferometric setup to measure not only the intensity but also the phase variation of the photoionization amplitude across an autoionization resonance in argon. The phase variation can be used as a fingerprint of the interactions between the discrete state and the ionization continua, indicating a new route towards monitoring electron correlations in time.
