The Bayesian reweighting procedure is extended to the case of multiple independent extractions of transverse momentum dependent parton distributions (TMDs). By exploiting the data on transverse ...single spin asymmetries, AN, for inclusive pion production in polarized proton-proton collisions measured at RHIC, we perform a simultaneous reweighting of the quark Sivers, transversity and Collins TMD functions extracted from semi-inclusive deep inelastic scattering (SIDIS) and e+e− annihilation into hadron pairs. The impact of the implementation of the Soffer bound, as well as the differences between older and newer AN data, are investigated. The agreement with AN data at large-xF values, a kinematical region complementary to those explored in SIDIS measurements, is enhanced, improving the knowledge of the polarized quark TMDs in the large-x region.
Infectious bursal disease (IBD) is an important immunosuppressive disease of chickens. The causative agent, infectious bursal disease virus (IBDV), consists of two serotypes, 1 and 2. Serotype 1 ...consists of classic IBDV (cIBDV) and variant IBDV (vIBDV). Both of these strains vary in antigenicity and pathogenesis. The goal of this study was to compare the immunopathogenesis of cIBDV and vIBDV. Three-week-old specific pathogen free chickens were inoculated intraocularly with standard challenge strain (STC) (cIBDV) and a variant strain Indiana (IN) (vIBDV). The cIBDV produced more pronounced bursal damage, inflammatory response and infiltration of T cells as compared to vIBDV. There were significant differences in the expression of innate (IFN-α and IFN-β), proinflammatory cytokine and mediator (IL-6 and iNOS) in cIBDV- and vIBDV-infected bursas. The expression of chemokines genes, IL-8 and MIP-α was also higher in cIBDV-infected chickens during the early phase of infection. The expression of Toll like receptor 3 (TLR3) was downregulated at post inoculation days (PIDs) 3, 5, and 7 in the bursas of vIBDV-infected chickens whereas TLR3 was upregulated at PIDs 3 and 5 in cIBDV-infected bursas. In vIBDV-infected bursa, TLR7 expression was downregulated at PIDs 3 and 5 and upregulated at PID 7. However, TLR7 was upregulated at PIDs 3 and 7 in cIBDV-infected bursas. The expression of MyD88 was downregulated whereas TRIF gene expression was upregulated in cIBDV- and vIBDV-infected bursa. These findings demonstrate the critical differences in bursal lesions, infiltration of T cells, expression of cytokines, chemokines and TLRs in the bursa of cIBDV-and vIBDV-infected chickens.
The Andromeda galaxy is the closest spiral galaxy to us and has been the subject of numerous studies. It harbors a massive dark matter halo, which may span up to ∼600 kpc across and comprises ∼90% of ...the galaxy's total mass. This halo size translates into a large diameter of 42° on the sky, for an M31-Milky Way (MW) distance of 785 kpc, but its presumably low surface brightness makes it challenging to detect with γ-ray telescopes. Using 7.6 yr of Fermi Large Area Telescope (Fermi-LAT) observations, we make a detailed study of the γ-ray emission between 1-100 GeV toward M31's outer halo, with a total field radius of 60° centered at M31, and perform an in-depth analysis of the systematic uncertainties related to the observations. We use the cosmic-ray propagation code GALPROP to construct specialized interstellar emission models to characterize the foreground γ-ray emission from the MW, including a self-consistent determination of the isotropic component. We find evidence for an extended excess that appears to be distinct from the conventional MW foreground, having a total radial extension upward of ∼120-200 kpc from the center of M31. We discuss plausible interpretations of the excess emission, but emphasize that uncertainties in the MW foreground-and in particular, modeling of the H i-related components-have not been fully explored and may impact the results.
The center of the Milky Way is predicted to be the brightest region of γ-rays generated by self-annihilating dark matter particles. Excess emission about the Galactic center above predictions made ...for standard astrophysical processes has been observed in γ-ray data collected by the Fermi Large Area Telescope. It is well described by the square of a Navarro, Frenk, and White dark matter density distribution. Although other interpretations for the excess are plausible, the possibility that it arises from annihilating dark matter is valid. In this paper, we characterize the excess emission as annihilating dark matter in the framework of an effective field theory. We consider the possibility that the annihilation process is mediated by either pseudoscalar or vector interactions and constrain the coupling strength of these interactions by fitting to the Fermi Large Area Telescope data for energies 1–100 GeV in the 15°×15° region about the Galactic center using self-consistently derived interstellar emission models and point source lists for the region. The excess persists and its spectral characteristics favor a dark matter particle with a mass in the range approximately from 50 to 190 (10 to 90) GeV and annihilation cross section approximately from 1×10−26 to 4×10−25 (6×10−27 to 2×10−25) cm3/s for pseudoscalar (vector) interactions. We map these intervals into the corresponding WIMP-neutron scattering cross sections and find that the allowed range lies well below current and projected direct detection constraints for pseudoscalar interactions, but are typically ruled out for vector interactions.

In vitro, Clo-Lipo-DOTAP inhibited proliferation, reduced viability and induced apoptosis of a macrophage-like cell line in a dose- and time-dependent manner.
In proof of functionality experiments, Clo-Lipo-DOTAP depleted macrophages in a genetic mouse model of chronic hepatitis and hepatocellular carcinoma leading to a significant reduction of F4/80-positive cells in the liver and spleen of treated mice compared to PBS-treated controls. The number of granulocytes, B and T lymphocytes was not affected.
In B16/F10 subcutaneous melanoma-bearing mice, Clo-Lipo-DOTAP significantly reduced the volume of primary tumors (P<0.001). Within the tumors, the expression F4/80 and α-SMA was significantly lowered. Plasma levels of IL-10, Mo KC, TNF-α, VEGF and PDGF-bb were statistically decreased. In B16/F10 lung metastatic melanoma model, treatment with Clo-Lipo-DOTAP significantly reduced the number of pulmonary nodules (P<0.05). F4/80-positive cells and microvessel density were statistically decreased.
In conclusion, the depletion of TAMs in primary and metastatic melanoma presents anti-tumor efficacy via inhibition of angiogenesis and modulation of inflammation related cytokines.

Deferiprone was shown to reverse iron deposition in Friedreich's ataxia. This multi-center, unblinded, single-arm pilot study evaluated safety and efficacy of deferiprone for reducing cerebral iron ...accumulation in neurodegeneration with brain iron accumulation. Four patients with genetically-confirmed pantothenate kinase-associated neurodegeneration, and 2 with parkinsonism and focal dystonia, but inconclusive genetic tests, received 15 mg/kg deferiprone bid. Magnetic resonance imaging and neurological examinations were conducted at baseline, six and 12 months. Chelation treatment caused no apparent hematologic or neurological side effects. Magnetic resonance imaging revealed decreased iron accumulation in the globus pallidus of 2 patients (one with pantothenate kinase-associated neurodegeneration). Clinical rating scales and blinded video rating evaluations documented mild-to-moderate motor improvement in 3 patients (2 with pantothenate kinase-associated neurodegeneration). These results underline the safety and tolerability of deferiprone, and suggest that chelating treatment might be effective in improving neurological manifestations associated with iron accumulation. Aim of this work was to assess the predictive value of micronuclei (MN) frequency in peripheral blood lymphocytes (PBL) for the risk of cancer death in disease-free individuals. Blood samples from ...1650 subjects selected from the general population of Pisa, Italy, were collected between June 1991 and November 1993. The follow-up until January 2005 recorded a total of 111 deaths (52 for cancer). MN frequency was assessed for 49 cancer cases and 101 matched controls. A significantly higher MN frequency was found in cancer cases (4.7+/-3.4 MN/1000 BN cells) versus controls (1.5+/-1.7; p<0.0001). Donors were stratified in two classes and multivariate logistic regression analysis confirmed that individuals with high MN frequency (>2.5 MN/1000 BN cells) had a significantly increased risk of cancer death (OR=10.7; 95% CI=4.6-24.9; p<0.0001) when compared to individuals with low MN frequency (<or=2.5 MN/1000 BN cells). Ageing was associated with a 6% increased risk per year (p=0.03). No influence of other potential confounders (gender, occupation, smoking and drinking habits) was observed. Finally, subjects with a higher MN frequency showed a higher MR for CVD (Logrank test, p=0.001). These findings provide strong evidence that MN frequency assessed in PBL of disease-free subjects is a good predictor of cancer death risk, evaluated by a nested case-control study performed 14 years after the original recruitment.
Abstract African swine fever is a highly contagious, often fatal disease of swine for which there is no vaccine or other curative treatment. The macrophage marker, CD163, is a putative receptor for ...African swine fever virus (ASFV). Pigs possessing a complete knockout of CD163 on macrophages were inoculated with Georgia 2007/1, a genotype 2 isolate. Knockout and wild type pen mates became infected and showed no differences in clinical signs, mortality, pathology or viremia. There was also no difference following in vitro infection of macrophages. The results do not rule out the possibility that other ASFV strains utilize CD163, but demonstrate that CD163 is not necessary for infection with the Georgia 2007/1 isolate. This work rules out a significant role for CD163 in ASFV infection and creates opportunities to focus on alternative receptors and entry mechanisms.
