To elucidate the mammalian system for synthesis of cobalamin coenzymes, microsomal NADPH-linked aquacobalamin reductase was purified and characterized. The enzyme was purified about 534-fold over rat ...liver microsomal fraction in a yield of about 32%. The purified enzyme was homogeneous in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and had a monomeric molecular weight of 79000. The purified aquacobalamin reductase showed a high specific activity (about 55 micromol/min per mg protein) of NADPH-cytochrome c (P-450) reductase. About 33% of the NADPH-cytochrome c reductase activity found in the microsomal fraction was recovered in the final purified preparation. The activity ratio of NADPH-cytochrome c reductase/NADPH-linked aquacobalamin reductase was about 5.0 through the purification steps, indicating that the rat liver microsomal NADPH-linked aquacobalamin reductase is the NADPH-cytochrome c reductase.

Magnetic resonance images of two cases of sellar spine are reported. The sellar spine is an anatomical variant characterized by an osseous spine that arises in the midline from the dorsum sellae and ...protrudes into the pituitary fossa. On short repetition time/short echo time (T1-weighted) images the sellar spine is demonstrated as a hypointense structure within the high intensity posterior pituitary. We suggest that the sellar spine is the ossified remnant of the cephalic tip of the notochord.
Bischofia javanica leaf extract exhibited inhibitory activities against hyaluronidase, collagenase, tyrosinase and urease, suggesting that it is useful as an active ingredient for functional foods ...and cosmeceutical products. In particular, its 50% inhibitory concentration for hyaluronidase was comparable to that of disodium cromoglycate, which is a well-known hyaluronidase inhibitor used as an anti-inflammation and anti-allergy agent. In addition, the extract also inhibited urease with the almost the same potential as acetohydroxamic acid, which is reported to suppress Helicobacter pylori-induced gastritis by inhibiting urease.
Diagnostic capability was compared prospectively between ultrasound and CT using 25 consecutive cases of clinically suspected ovarian mass, which were found postoperatively to be comprised of 12 ...cases of ovarian neoplasm (of which 7 were malignant). 5 cases of endometriotic cyst, and 8 cases of nonovarian tumors including 4 cases of abscess. Accuracy for the histologic diagnosis with ultrasound and CT was 56 and 84%, respectively (P less than 0.05). With respect to discrimination of cystadenoma from cystadenocarcinoma, detection of intraperitoneal seedings, of paraaortic lymph node metastasis, and of ascites, either of the two imaging modalities was comparable to the other in their proficiency. In contrast to ultrasound, which permitted evaluation of adhesions to the uterus only in selected cases and hence was of limited use in this respect, CT was proficient in predicting the presence or absence of adhesions around the mass involving the uterus as well as other adjoining structures. The fact that CT is competent in detecting adhesions adds further value to CT as a powerful tool for the preoperative investigation of pelvic mass.
The present study was to determine the effects of dietary vitamin B12 (B12) deficiency on the reproductive system in mammals. Male rats were fed a vitamin B12-deficienctdiet for 14 weeks (group 1) or ...23 weeks (group 2). In the B12-deficient groups 1 and 2, the body weight and the wet weight of testisand epididymis were significantly depressed, as compared with the control group. The diameter of seminiferous tubules and the thickness of spermatogenic epithelium were decreased to about 50% of the control. In the spermatogenic epithelium of seminiferous tubules, the relative incidence of Sertoli cells in the B12-deficient groups was higher than the control, and aplasia of spermatids and sperms were detected. Also numerous TUNEL positive cells were located in the spermatogenic epithelium from B12-deficient rats in the group 1. The ultrastratural features by TEM can be considered to be apoptosis. However, the mechanism underlying which the induction of apoptosis by B12 deficiency are unclear. In the connective tissue between the seminiferous tubules, many interstitial cells of Leydig and blood vessel were observed in the B12-deficient groups, as compared with the control. In the present study B12 deficiency affected the male reproductive system and inhibited the maturation of spermatogenic cells, which indicate that B12 may be an essential constituent for spermatogenesis in rats.
A pilot PACS project, named KIDS, has been running in Kyoto University Hospital. The purpose of the system is to establish a small PACS that includes all digital imaging modalities and to evaluate ...it. The project has been continued from the first phase (KIDS-1) to the second phase (KIDS-2). In the first phase, a small-scale PACS was developed. In the second phase, the expansion of coverage of modalities and completion of the image database was intended. At present, the database contains image data of 16264 patients amounting to 150 Gbytes. Simulation of the retrieval process to the database shows that 154.3 s per patient is required for retrieving his/her entire image data. This calculated value is close to the actual time.
Resemblances and differences on photosynthesis of C3 plant and cyanobacterium Synechococcus PCC7942 were discussed with a simplified model of photosynthesis and photorespiration. Then the ...transformation of the cyanobacterium was shown with Chromatium vinosum (Cv) RuBisCO. Total RuBisCO activity and photosynthesis were compared with wild type S. 7942 and RuBisCO high expression mutant. Results were explained with RuBisCO environment of S. 7942 mutant. For excluding the influence of inorganic carbon concentrating mechanism (CCM) on the effect of RuBisCO, carboxysome less mutant was created with homologous recombination of ccm operon to be replaced with kanamycine resistant gene. And the deletion of the operon was confirmed by genomic PCR on the transformant. The confirmation was performed with three primers. Electron micrograms of S. 7942 show the existence of carboxysome in wild type and its absence in the mutant. Growth rate of CL mutant on different CO2 concentration shows that there is a possibility to increase a photosynthetic ability of a cyanobacterium with increases the RuBisCO activity.
