Additive manufacturing (AM) techniques and so-called 2D materials have undergone an explosive growth in the past decade. The former opens multiple possibilities in the manufacturing of ...multifunctional complex structures, and the latter on a wide range of applications from energy to water purification. Extrusion-based 3D printing, also known as Direct Ink Writing (DIW), robocasting, and often simply 3D printing, provides a unique approach to introduce advanced and high-added-value materials with limited availability into lab-scale manufacturing. On the other hand, 2D colloids of graphene oxide (GO) exhibit a fascinating rheology and can aid the processing of different materials to develop 'printable' formulations. This work provides an in-depth rheological study of GO suspensions with a wide range of behaviours from Newtonian-like to viscoelastic 'printable' soft solids. The combination of extensional and shear rheology reveals the network formation process as GO concentration increases from <0.1 vol% to 3 vol%. The Omicron variant is rapidly becoming the dominant SARS-CoV-2 virus circulating globally. It is important to define reductions in virus neutralizing activity in the serum of convalescent or ...vaccinated individuals to understand potential loss of protection against infection by Omicron. We previously established that a 50% plaque reduction neutralization antibody titer (PRNT
) ≥25.6 in our live virus assay corresponded to the threshold for 50% protection from infection against wild-type (WT) SARS-CoV-2. Here we show markedly reduced serum antibody titers against the Omicron variant (geometric mean titer (GMT) < 10) compared to WT virus 3-5 weeks after two doses of BNT162b2 (GMT = 218.8) or CoronaVac vaccine (GMT = 32.5). A BNT162b2 booster dose elicited Omicron PRNT
titers ≥25.6 in 88% of individuals (22 of 25) who previously received 2 doses of BNT162b2 and 80% of individuals (24 of 30) who previously received CoronaVac. However, few (3%) previously infected individuals (1 of 30) or those vaccinated with three doses of CoronaVac (1 of 30) met this threshold. Our findings suggest that countries primarily using CoronaVac vaccines should consider messenger RNA vaccine boosters in response to the spread of Omicron. Studies evaluating the effectiveness of different vaccines against the Omicron variant are urgently needed.
The extent or existence of similarities between fully developed turbulent pipes and channels, and in zero-pressure-gradient turbulent boundary layers has come into question in recent years. This is ...in contrast to the traditionally accepted view that, upon appropriate normalization, all three flows can be regarded as the same in the near-wall region. In this paper, the authors aim to provide clarification of this issue through streamwise velocity measurements in these three flows with carefully matched Reynolds number and measurement resolution. Results show that mean statistics in the near-wall region collapse well. However, the premultiplied energy spectra of streamwise velocity fluctuations show marked structural differences that cannot be explained by scaling arguments. It is concluded that, while similarities exist at these Reynolds numbers, one should exercise caution when drawing comparisons between the three shear flows, even near the wall.
An array of surface hot-film shear-stress sensors together with a traversing hot-wire probe is used to identify the conditional structure associated with a large-scale skin-friction event in a ...high-Reynolds-number turbulent boundary layer. It is found that the large-scale skin-friction events convect at a velocity that is much faster than the local mean in the near-wall region (the convection velocity for large-scale skin-friction fluctuations is found to be close to the local mean at the midpoint of the logarithmic region). Instantaneous shear-stress data indicate the presence of large-scale structures at the wall that are comparable in scale and arrangement to the superstructure events that have been previously observed to populate the logarithmic regions of turbulent boundary layers. Conditional averages of streamwise velocity computed based on a low skin-friction footprint at the wall offer a wider three-dimensional view of the average superstructure event. These events consist of highly elongated forward-leaning low-speed structures, flanked on either side by high-speed events of similar general form. An analysis of small-scale energy associated with these large-scale events reveals that the small-scale velocity fluctuations are attenuated near the wall and upstream of a low skin-friction event, while downstream and above the low skin-friction event, the fluctuations are significantly amplified. In general, it is observed that the attenuation and amplification of the small-scale energy seems to approximately align with large-scale regions of streamwise acceleration and deceleration, respectively. Further conditional averaging based on streamwise skin-friction gradients confirms this observation. A conditioning scheme to detect the presence of meandering large-scale structures is also proposed. The large-scale meandering events are shown to be a possible source of the strong streamwise velocity gradients, and as such play a significant role in modulating the small-scale motions.
The objective was to investigate the changes in urology practice during coronavirus disease 2019 (COVID-19) pandemic with a perspective from our experience with severe acute respiratory syndrome ...(SARS) in 2003.
Institutional data from all urology centres in the Hong Kong public sector during the COVID-19 pandemic (1 Feb 2020-31 Mar 2020) and a non-COVID-19 control period (1 Feb 2019-31 Mar 2019) were acquired. An online anonymous questionnaire was used to gauge the impact of COVID-19 on resident training. The clinical output of tertiary centres was compared with data from the SARS period.
The numbers of operating sessions, clinic attendance, cystoscopy sessions, prostate biopsy, and shockwave lithotripsy sessions were reduced by 40.5%, 28.5%, 49.6%, 44.8%, and 38.5%, respectively, across all the centres reviewed. The mean numbers of operating sessions before and during the COVID-19 pandemic were 85.1±30.3 and 50.6±25.7, respectively (P=0.005). All centres gave priority to cancer-related surgeries. Benign prostatic hyperplasia-related surgery (39.1%) and ureteric stone surgery (25.5%) were the most commonly delayed surgeries. The degree of reduction in urology services was less than that during SARS (47.2%, 55.3%, and 70.5% for operating sessions, cystoscopy, and biopsy, respectively). The mean numbers of operations performed by residents before and during the COVID-19 pandemic were 75.4±48.0 and 34.9±17.2, respectively (P=0.002).

MicroRNAs (miRNAs) are 19-25-nucleotides regulatory non-protein-coding RNA molecules that regulate the expressions of a wide variety of genes, including some involved in cancer development. In this ...study, we investigated the possible role of miR-143 in colorectal cancer (CRC).
Expression levels of human mature miRNAs were examined using real-time PCR-based expression arrays on paired colorectal carcinomas and adjacent non-cancerous colonic tissues. The downregulation of miR-143 was further evaluated in colon cancer cell lines and in paired CRC and adjacent non-cancerous colonic tissues by qRT-PCR. Potential targets of miR-143 were defined. The functional effect of miR-143 and its targets was investigated in human colon cancer cell lines to confirm miRNA-target association.
Both real-time PCR-based expression arrays and qRT-PCR showed that miR-143 was frequently downregulated in 87.5% (35 of 40) of colorectal carcinoma tissues compared with their adjacent non-cancerous colonic tissues. Using in silico predictions, DNA methyltranferase 3A (DNMT3A) was defined as a potential target of miR-143. Restoration of the miR-143 expression in colon cell lines decreased tumour cell growth and soft-agar colony formation, and downregulated the DNMT3A expression in both mRNA and protein levels. DNMT3A was shown to be a direct target of miR-143 by luciferase reporter assay. Furthermore, the miR-143 expression was observed to be inversely correlated with DNMT3A mRNA and protein expression in CRC tissues.
Our findings suggest that miR-143 regulates DNMT3A in CRC. These findings elucidated a tumour-suppressive role of miR-143 in the epigenetic aberration of CRC, providing a potential development of miRNA-based targeted approaches for CRC therapy.
The spread of coronavirus disease 2019 (COVID-19) has affected both physical health and mental well-being around the world. Stress-related reactions, if prolonged, may result in mental health ...problems. We examined the consequences of the COVID-19 pandemic on mental health in a multinational study and explored the effects of government responses to the outbreak. We sampled 18,171 community adults from 35 countries/societies, stratified by age, gender, and region of residence. Across the 35 societies, 26.6% of participants reported moderate to extremely severe depression symptoms, 28.2% moderate to extremely severe anxiety symptoms, and 18.3% moderate to extremely severe stress symptoms. Coronavirus anxiety comprises two factors, namely Perceived Vulnerability and Threat Response. After controlling for age, gender, and education level, perceived vulnerability predicted higher levels of negative emotional symptoms and psychological distress, whereas threat response predicted higher levels of self-rated health and subjective well-being. People in societies with more stringent control policies had more threat response and reported better subjective health. Coronavirus anxiety exerts detrimental effects on subjective health and well-being, but also has the adaptive function in mobilizing safety behaviors, providing support for an evolutionary perspective on psychological adaptation.

In light of the Industry 5.0 trend towards human-centric and resilient industries, human-robot collaboration (HRC) assembly lines can be used to enhance productivity and workers’ well-being, provided that the optimal allocation of tasks and available resources can be determined. This study investigates the assembly line balancing problem (ALBP), considering HRC. This problem, abbreviated ALBP-HRC, arises in advanced manufacturing systems, where humans and collaborative robots share the same workplace and can simultaneously perform tasks in parallel or in collaboration. Driven by the need to solve the more complex assembly line-balancing problems found in the automotive industry, this study aims to address the ALBP-HRC with the cycle time and the number of operators (humans and robots) as the primary and secondary objective, respectively. In addition to the traditional ALBP constraints, the human and robot characteristics, in terms of task times, allowing multiple humans and robots at stations, and their joint/collaborative tasks are formulated into a new mixed-integer linear programming (MILP) model. A neighborhood-search simulated annealing (SA) is proposed with customized solution representation and neighborhood search operators designed to fit into the problem characteristics. Furthermore, the proposed SA features an adaptive neighborhood selection mechanism that enables the SA to utilize its exploration history to dynamically choose appropriate neighborhood operators as the search evolves. The proposed MILP and SA are implemented on real cases taken from the automotive industry where stations are designed for HRC. The computational results over different problems show that the adaptive SA produces promising solutions compared to the MILP and other swarm intelligence algorithms, namely genetic algorithm, particle swarm optimization, and artificial bee colony. The comparisons of human/robot versus HRC settings in the case study indicate significant improvement in the productivity of the assembly line when multiple humans and robots with collaborative tasks are permissible at stations.
Epitheliotropic intestinal T-cell lymphoma (EITL, also known as type II enteropathy-associated T-cell lymphoma) is an aggressive intestinal disease with poor prognosis and its molecular alterations ...have not been comprehensively characterized. We aimed to identify actionable easy-to-screen alterations that would allow better diagnostics and/or treatment of this deadly disease. By performing whole-exome sequencing of four EITL tumor-normal pairs, followed by amplicon deep sequencing of 42 tumor samples, frequent alterations of the JAK-STAT and G-protein-coupled receptor (GPCR) signaling pathways were discovered in a large portion of samples. Specifically, STAT5B was mutated in a remarkable 63% of cases, JAK3 in 35% and GNAI2 in 24%, with the majority occurring at known activating hotspots in key functional domains. Moreover, STAT5B locus carried copy-neutral loss of heterozygosity resulting in the duplication of the mutant copy, suggesting the importance of mutant STAT5B dosage for the development of EITL. Dysregulation of the JAK-STAT and GPCR pathways was also supported by gene expression profiling and further verified in patient tumor samples. In vitro overexpression of GNAI2 mutants led to the upregulation of pERK1/2, a member of MEK-ERK pathway. Notably, inhibitors of both JAK-STAT and MEK-ERK pathways effectively reduced viability of patient-derived primary EITL cells, indicating potential therapeutic strategies for this neoplasm with no effective treatment currently available.
