The global surge in multi-drug resistant bacteria, including extended-spectrum β-lactamase (ESBL)-producing Escherichia coli has led to a growing need for new antibacterial compounds. Despite being ...promising, the potential of fish-derived antimicrobial peptides (AMPs) in combating ESBL-producing E. coli is largely unexplored. In this study, native African catfish antimicrobial peptides (NACAPs) were extracted from the skin mucus of farmed African catfish, Clarias gariepinus, using a combination of 10% acetic acid solvent hydrolysis, 5 kDa ultrafiltration, and C18 hydrophobic interaction chromatography. Peptides were then sequenced using Orbitrap Fusion Lumos Tribrid Mass Spectrometry. The identified peptides were screened for potential antibacterial activity using Random Forest and AdaBoost machine learning algorithms. The most promising peptide was chemically synthesized and evaluated in vitro for safety on rabbit red blood cells and activity against ESBL-producing E. coli (ATCC 35218) utilizing spot-on-lawn and broth dilution methods. Eight peptides ranging from 13 to 22 amino acids with molecular weights between 968.42 and 2434.11 Da were identified. Peptide NACAP-II was non-hemolytic to rabbit erythrocytes (p > 0.05) with a zone of inhibition (ZOI) of 22.7 ± 0.9 mm and a minimum inhibitory concentration (MIC) of 91.3 ± 1.2 μg/mL. The peptide is thus a candidate antibacterial compound with enormous potential applications in the pharmaceutical industry. However, further studies are still required to establish an upscale production strategy and optimize its activity and safety in vivo.
Early detection of Mycoplasmal mastitis is greatly hampered by late seroconversion, slow growth of Mycoplasma organisms, intermittent shedding, and the high cost of diagnostic tests. To improve ...future diagnostic development, examining the available techniques is necessary. Accordingly, the present study systematically reviewed M. bovis diagnostic studies published between January 2000 and April 2023 utilizing the Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) protocol. The protocol registration was performed according to the Open Science Framework (osf.io/ug79h), and the electronic search was conducted in the World Catalog, Mendeley, ProQuest, ScienceDirect, Semantic Scholar, PubMed, Google Scholar, Prime Scholar, and PubMed Central databases using a Boolean operator and inclusion and exclusion criteria. Of the 1194 pieces of literature retrieved, 67 studies were included. Four broad categories of up to 16 diagnostic approaches were reported: microbial culture, serological, DNA-based, and mass spectrometry. Overall, DNA-based techniques were the most published (48.0%), with recombinase polymerase amplification (RPA) and loop-mediated isothermal amplification (LAMP) as the most promising user-friendly, equipment-free techniques. On the other hand, mass spectrometry was reported as the least utilized (2.9%) given the high equipment cost. Though costly and laboratory-allied, DNA-based techniques, particularly PCRs, were reported as the most rapid and specific approach.
Fascioliasis (liver fluke infestation) is one of the most important parasitic diseases affecting cattle, other ruminant animals and humans. Fascioliasis causes large, but usually neglected, economic ...losses to cattle farmers and traders. The objectives of this study were to assess the prevalence and associated risks for fascioliasis in slaughter cattle and estimate the financial losses due to liver condemnation at the Lira Municipal abattoir in Uganda. A total of 216 cattle were sampled during the study period. Animal breed and sex were determined by observing the phenotypic characteristics of the animals. Age was determined by assessing the eruption and wearing of permanent teeth. After slaughter, the liver was examined for presence of
spp. (liver flukes) by visual inspection, palpation, and incisions. The bile ducts and gall bladder were similarly examined for presence of mature
spp. The gross weight and amount of liver trimmed-off due to fluke infestation were determined. Of the 216 liver examined, 65.7% (
= 142) were infested with
spp. Cattle that were aged 4-5 years old at the time of slaughter had significantly greater odds (OR = 5.84; CI 2.79-12.22) of being infested with
spp. compared to those that were younger than 3.5 years old. In contrast, cattle that had a body condition score of 3.5 or 4 had lower odds (OR= 0.42; CI 0.21-0.88 and OR = 0.22; CI 0.04-1.10) of fascioliasis than those with a BCS of 3. Other tested variables including animal origin, breed, sex, and gross weight of the liver had no significant effect on the prevalence of fascioliasis. This study also revealed that the abattoir loses an estimated 38 million UGX annually due to condemnation of
-infested liver (one UGX= 0.00027 USD; July 2016). Our study showed that the prevalence of fascioliasis was high in Lira District, Uganda, which results in a large amount of liver being condemned and destroyed, leading to financial losses for affected farmers in the area. Therefore, there is a need to take the necessary preventive measures to control the disease and increase awareness among farmers and medical personnel in the area due to the zoonotic nature of fascioliasis.

Our objectives were to evaluate the diagnostic accuracy of a rapid and novel immunochromatography-based mastitis kit that includes 3 independent tests to detect coliforms (Escherichia coli or ...Klebsiella pneumoniae), Streptococcus spp., and Staphylococcus aureus. The kit was developed to facilitate diagnostic-based mastitis treatment. Validation of the kit was based on 154 aseptically collected mastitis samples from 2 clinical herds (clinical population) and 120 milk samples from 3 nonclinical herds (nonclinical population) without clinical cases at the time of enrollment. One herd sampled at different times was common to both populations. A 3-test in 2-population Bayesian latent class model with uniform priors for all test parameters except specificity of culture, which was modeled informatively, was used to estimate sensitivity (Se) and specificity (Sp) of the test kit, culture, and PCR at the cow level. The mastitis test kit's 96.9% Sp for Streptococcus spp. had a low false positive percentage (3.1%), which, together with the kit's rapid turnaround time for results, makes it a suitable initial screening test that producers can use to identify clinical cows to treat based on Streptococcus spp. mastitis in kit-positive results. Due to the 60.4% kit Se, producers should follow up on Streptococcus spp. kit-negative cows using a confirmatory test such as PCR (Sp of 98.4%) or culture (Sp of 99.6%). In contrast, aerobic culture had Se of 76.5% and Sp of 99.6% for Streptococcus spp. Similarly, the Sp of the kit (98.2%) and culture (99.8%) for Staph. aureus were particularly high, and even though the kit's Se (61.0%) was lower than culture (88.4%; posterior probability of difference 98%), the kit could be beneficial before use of a confirmatory test for kit-negative samples due to its ease and rapid turnaround time. Mostly, quantitative real-time (q)PCR outperformed the kit's Se (37.7%) and Sp (92.9%) for coliforms, as well as the kit's Se (60.4%) for Streptococcus spp. However, qPCR may require more technical skills and turnaround time for final results. Use of the on-farm mastitis test kit evaluated in the present study could enhance sustainable antimicrobial drug use by rapidly identifying Streptococcus mastitis for targeted treatment. Furthermore, the kit may be used in a Staph. aureus outbreak where cows can be rapidly screened to identify cases for segregation or culling during an outbreak and kit-negative cows further confirmed by milk culture or qPCR. However, the cost-effectiveness of such an approach has not been investigated.
Tetramethrin (Tm) is a commonly used pesticide that has been reported to exert estrogen-antagonistic effects selectively on female rats. The present study was undertaken to assess the protective role ...of lobaric acid (La) on estrous cycle in Tm-treated female Wistar rats. Female rats were exposed to Tm (50 mg/kg b.w/day) only or in combination with La at low (50 mg/kg b.w/day) or high (100 mg/kg b.w/day) dose for 30 days. The results showed that Tm altered the estrous cycle of female rats by decreasing the levels of luteinizing hormone, follicular-stimulating hormone, progesterone, estrone, and estradiol while increasing testosterone level. The morphology of vaginal smears of Tm-treated female rats showed the presence of abnormal cells and/or structures at different phases of estrus cycle. Strikingly, in (Tm + La)-treated rats, all the observed adverse effects of Tm on the hormonal parameters, cell morphology, and the length of each phase of estrous cycle were significantly diminished in a dose-dependent manner. The docking results showed that La competes with Tm for Gonadotropin-Releasing Hormone (GnRH) receptor, thereby reducing the toxicity of Tm but did not cancel the response of GnRH receptor completely. In conclusion, our results designated that La could be used as a potential candidate in the management of insecticide-induced alterations of the reproductive cycle of rodents.

isolates were obtained from cow and calf fecal samples, tested for susceptibility to 19 antimicrobials, and classified as resistant or non-susceptible to the antimicrobials for which breakpoints were available. For
, percent of resistant isolates by antimicrobial were as follows: ampicillin 100% (244/244), sulfadimethoxine 25.4% (62/244), trimethoprim-sulfamethoxazole 4.9% (12/244), and ceftiofur 0.4% (1/244) while percent of non-susceptible isolates by antimicrobial were: tetracycline 13.1% (32/244), and florfenicol 19.3% (47/244). For
spp., percent of resistant isolates by antimicrobial were as follows: ampicillin 0.4% (1/238) while percent of non-susceptible isolates by antimicrobial were tetracycline 12.6% (30/238) and penicillin 1.7% (4/238). No animal level or farm level management practices, including antimicrobial exposures, were significantly associated with differences in isolate resistant or non-susceptible status for either

This is contrary to the suggestion that administration of antibiotics is solely responsible for development of AMR in exposed bacteria and demonstrates that there are other factors involved, either not captured in this study or not currently well understood. In addition, the overall use of antimicrobials in this cow-calf study was lower than other sectors of the livestock industry. Limited information is available on cow-calf AMR from fecal bacteria, and the results of this study serve as a reference for future studies to support a better understanding and estimation of drivers and trends for AMR in cow-calf operations.
Abstract Despite the United Nations 2030 agenda, large number of both urban and rural dwellers in low-income countries continue to lack access to improved water. Thus, increased effort is required ...towards enhancing low-cost drinking water treatment technologies especially for developing countries. Slow sand filter (SSF) is one of the most commonly used low-cost and efficient technologies for treating household drinking water. However, effectiveness of SSF is substantially affected by very high turbidity and relatively large amounts of dissolved heavy metals. To enhance removal of both turbidity and heavy metals, this study optimized sand bed depth (SBD) of SSF and investigated the potential of natural zeolite from Uganda for removal of lead, arsenite (As(III)) and fluoride ions from water. To remove lead ions, the zeolite was used in its natural form. However, to remove As(III) and fluoride, the natural zeolite was modified using hexadecyltrimethylammonium bromide solution. Removal of high turbidity was found to require a large optimal SBD. Furthermore, efficiency of treating synthetic turbid water increased with increasing initial turbidity. Variation of final turbidity with SBD was found to be best described by an exponential function. Optimal SBDs on top of an underdrain gravel layer of 0.2 m were 453, 522, 561, and 580 mm for turbidity of 60, 80, 100, and 120 NTU, respectively. Optimized SBD used achieve at least 95% efficiency in removing suspended particles from water with turbidity 120 NTU was found to save up to 35% of the total cost for acquiring sand volume required by a conventional SSF. For a particular zeolite mass, removal efficiencies of lead, As(III) and fluoride generally increased with increasing contact time. Removal efficiencies of lead, As(III), and fluorides were also shown to increase with increasing zeolite mass. Lead removal efficiencies using natural zeolite were 75 and 98% under 20 and 40 min, respectively. Removal of As(III) using modified zeolite mass was 91% within contact time of 10 min. Adsorption of fluoride on modified zeolite was 80% within 5 min. Adsorption of lead, As(III), and fluorides indicated promising potential of natural zeolites from Uganda for treating polluted water.
Africa faces immense food and health insecurity challenges, a problem partly attributed to food loss and waste during postharvest handling and distribution. In the context of research to meet the ...sustainable development goals, this project specifically addressed the postharvest loss of the ripe indigenous eggplant (
lam) fruit called "Igba Yinrin" by Yoruba in South-West Nigeria, which is usually discarded in farms. The study was carried out on ripe and unripe fruits to better understand their value by comparing their effects in diabetes treatment.
The study sought to assess the effects of a diet including ripe or unripe mature eggplant fruits in the sucrose-induced diabetic-like fruit fly. Bioactive compounds were identified and quantified with HPLC-UV, while the antioxidant vitamin (A, C, E), carotenoid, and mineral (Na, K, Ca, Mg, Fe, P, and Zn) content was analyzed in the fruits. Extracts were used to investigate their in vitro anti-inflammatory properties on cyclooxygenases (COX 1 and 2), 5-lipoxygenase (5-LOX), and anti-diabetes enzymes α-amylase and α-glucosidase, while extract-supplemented diets (0.25-1% concentration) were fed to the fruit flies for 14 days.
Interestingly, the results showed that the ripe fruits had a significantly (
< 0.05) higher total phenol and flavonoid content, as well as a higher content of vitamins, carotenoids, and minerals, than the unripe fruits. The in vivo activities of antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), and glutathione transferase (GST) and the total thiol level increased, while the blood glucose, reactive oxygen species (ROS), and malondialdehyde (MDA) levels decreased in
(fruit fly). An in silico docking analysis showed strong binding affinity of the above-mentioned enzymes under investigation with the ligands hesperidin, naringin, and myricetin, which are bioactive compounds contained in the examined extracts.
There was no significant difference in the biological effects of the ripe and unripe fruit extracts on inflammatory and anti-diabetes enzyme activities, which means that the ripe fruit, usually discarded, could serve as a sustainable alternative source of food nutrients.
This study explores the critical determinants impacting labor productivity in brickwork operations within the construction industry—a matter of academic and practical significance, particularly in ...the era of increasing human–robot collaboration. Through an extensive literature review on construction labor productivity, this study identifies factors affecting brickwork productivity. Data were collected from active construction sites during brick wall construction through on-site measurements and participatory observation, and the relative importance of these factors is determined using Principal Component Analysis (PCA)-factor analysis. The validity of the analysis is established through the Kaiser–Meyer–Olkin (KMO) test and Bartlett’s test of sphericity, with a KMO value of 0.544 and significance at the 0.05 significance level. The analysis reveals four principal components explaining 75.96% of the total variance. Notably, this study identifies the Euclidean distances for the top factors: weather (0.980), number of helpers (0.965), mason competency (0.934), and number of masons (0.772). Additionally, correlation coefficients were observed: wall area had the highest correlation (0.998), followed by wall length (0.853) and height (0.776). Interestingly, high correlations did not necessarily translate to high factor importance. These identified factors can serve as a foundation for predictive modeling algorithms for estimating production rates and as a guideline for optimizing labor in construction planning and scheduling, particularly in the context of human–robot collaboration.
