Emerging infectious diseases (EIDs) pose a risk to human welfare, both directly and indirectly, by affecting managed livestock and wildlife that provide valuable resources and ecosystem services, ...such as the pollination of crops. Honeybees (Apis mellifera), the prevailing managed insect crop pollinator, suffer from a range of emerging and exotic high-impact pathogens, and population maintenance requires active management by beekeepers to control them. Wild pollinators such as bumblebees (Bombus spp.) are in global decline, one cause of which may be pathogen spillover from managed pollinators like honeybees or commercial colonies of bumblebees. Here we use a combination of infection experiments and landscape-scale field data to show that honeybee EIDs are indeed widespread infectious agents within the pollinator assemblage. The prevalence of deformed wing virus (DWV) and the exotic parasite Nosema ceranae in honeybees and bumblebees is linked; as honeybees have higher DWV prevalence, and sympatric bumblebees and honeybees are infected by the same DWV strains, Apis is the likely source of at least one major EID in wild pollinators. Lessons learned from vertebrates highlight the need for increased pathogen control in managed bee species to maintain wild pollinators, as declines in native pollinators may be caused by interspecies pathogen transmission originating from managed pollinators.

Projections of climate change impacts on crop yields are inherently uncertain1. Uncertainty is often quantified when projecting future greenhouse gas emissions and their influence on climate2. ...However, multi-model uncertainty analysis of crop responses to climate change is rare because systematic and objective comparisons among process-based crop simulation models1, 3 are difficult4. Here we present the largest standardized model intercomparison for climate change impacts so far. We found that individual crop models are able to simulate measured wheat grain yields accurately under a range of environments, particularly if the input information is sufficient. However, simulated climate change impacts vary across models owing to differences in model structures and parameter values. A greater proportion of the uncertainty in climate change impact projections was due to variations among crop models than to variations among downscaled general circulation models. Uncertainties in simulated impacts increased with CO2 concentrations and associated warming. These impact uncertainties can be reduced by improving temperature and CO2 relationships in models and better quantified through use of multi-model ensembles. Less uncertainty in describing how climate change may affect agricultural productivity will aid adaptation strategy development and policymaking.
Supermassive black holes have powerful gravitational fields with strong gradients that can destroy stars that get too close, producing a bright flare in ultraviolet and X-ray spectral regions from ...stellar debris that forms an accretion disk around the black hole. The aftermath of this process may have been seen several times over the past two decades in the form of sparsely sampled, slowly fading emission from distant galaxies, but the onset of the stellar disruption event has not hitherto been observed. Here we report observations of a bright X-ray flare from the extragalactic transient Swift J164449.3+573451. This source increased in brightness in the X-ray band by a factor of at least 10,000 since 1990 and by a factor of at least 100 since early 2010. We conclude that we have captured the onset of relativistic jet activity from a supermassive black hole. A companion paper comes to similar conclusions on the basis of radio observations. This event is probably due to the tidal disruption of a star falling into a supermassive black hole, but the detailed behaviour differs from current theoretical models of such events.

Autophagy is a cell-protective and degradative process that recycles damaged and long-lived cellular components. Cancer cells are thought to take advantage of autophagy to help them to cope with the ...stress of tumorigenesis; thus targeting autophagy is an attractive therapeutic approach. However, there are currently no specific inhibitors of autophagy. ULK1, a serine/threonine protein kinase, is essential for the initial stages of autophagy, and here we report that two compounds, MRT67307 and MRT68921, potently inhibit ULK1 and ULK2 in vitro and block autophagy in cells. Using a drug-resistant ULK1 mutant, we show that the autophagy-inhibiting capacity of the compounds is specifically through ULK1. ULK1 inhibition results in accumulation of stalled early autophagosomal structures, indicating a role for ULK1 in the maturation of autophagosomes as well as initiation.
Background: Autophagy is an intracellular lysosomal degradation pathway implicated in many diseases, but there are currently no specific autophagy inhibitors.
Results: Small molecule inhibition of ULK1, the upstream autophagy initiating kinase, blocks autophagosome initiation and maturation.
Conclusion: ULK1 plays a role in autophagosome maturation as well as initiation.
Significance: ULK1 can be targeted to block autophagy for disease therapy.
In an age of advancing endoscopic and lithotripsy technologies, the management of urolithiasis poses a unique opportunity to advance compassionate veterinary care, not only for patients with ...urolithiasis but for those with other urinary diseases as well. The following are consensus‐derived, research and experience‐supported, patient‐centered recommendations for the treatment and prevention of uroliths in dogs and cats utilizing contemporary strategies. Ultimately, we hope that these recommendations will serve as a foundation for ongoing and future clinical research and inspiration for innovative problem solving.
The very large collection area of ground-based gamma-ray telescopes gives them a substantial advantage over balloon or satellite based instruments in the detection of very-high-energy (>600 GeV) ...cosmic-ray electrons. Here we present the electron spectrum derived from data taken with the High Energy Stereoscopic System (H.E.S.S.) of imaging atmospheric Cherenkov telescopes. In this measurement, the first of this type, we are able to extend the measurement of the electron spectrum beyond the range accessible to direct measurements. We find evidence for a substantial steepening in the energy spectrum above 600 GeV compared to lower energies.
Bumblebees are major pollinators of crops and wildflowers in northern temperate regions. Knowledge of their ecology is vital for the design of effective management and conservation strategies but key ...aspects remain poorly understood. Here we employed microsatellite markers to estimate and compare foraging range and nest density among four UK species: Bombus terrestris, Bombus pascuorum, Bombus lapidarius, and Bombus pratorum. Workers were sampled along a 1.5‐km linear transect across arable farmland. Eight or nine polymorphic microsatellite markers were then used to identify putative sisters. In accordance with previous studies, minimum estimated maximum foraging range was greatest for B. terrestris (758 m) and least for B. pascuorum (449 m). The estimate for B. lapidarius was similar to B. pascuorum (450 m), while that of B. pratorum was intermediate (674 m). Since the area of forage available to bees increases as the square of foraging range, these differences correspond to a threefold variation in the area used by bumblebee nests of different species. Possible explanations for these differences are discussed. Estimates for nest density at the times of sampling were 29, 68, 117, and 26/km2 for B. terrestris, B. pascuorum, B. lapidarius and B. pratorum, respectively. These data suggest that even among the most common British bumblebee species, significant differences in fundamental aspects of their ecology exist, a finding that should be reflected in management and conservation strategies.
The African Monsoon Multidisciplinary Analysis (AMMA) is a major international campaign investigating far‐reaching aspects of the African monsoon, climate and the hydrological cycle. A special ...observing period was established for the dry season (SOP0) with a focus on aerosol and radiation measurements. SOP0 took place during January and February 2006 and involved several ground‐based measurement sites across west Africa. These were augmented by aircraft measurements made by the Facility for Airborne Atmospheric Measurements (FAAM) aircraft during the Dust and Biomass‐burning Experiment (DABEX), measurements from an ultralight aircraft, and dedicated modeling efforts. We provide an overview of these measurement and modeling studies together with an analysis of the meteorological conditions that determined the aerosol transport and link the results together to provide a balanced synthesis. The biomass burning aerosol was significantly more absorbing than that measured in other areas and, unlike industrial areas, the ratio of excess carbon monoxide to organic carbon was invariant, which may be owing to interaction between the organic carbon and mineral dust aerosol. The mineral dust aerosol in situ filter measurements close to Niamey reveals very little absorption, while other measurements and remote sensing inversions suggest significantly more absorption. The influence of both mineral dust and biomass burning aerosol on the radiation budget is significant throughout the period, implying that meteorological models should include their radiative effects for accurate weather forecasts and climate simulations. Generally, the operational meteorological models that simulate the production and transport of mineral dust show skill at lead times of 5 days or more. Climate models that need to accurately simulate the vertical profiles of both anthropogenic and natural aerosols to accurately represent the direct and indirect effects of aerosols appear to do a reasonable job, although the magnitude of the aerosol scattering is strongly dependent upon the emission data set.
Vaccination and infection promote the formation, tissue distribution, and clonal evolution of B cells, which encode humoral immune memory. We evaluated pediatric and adult blood and deceased adult ...organ donor tissues to determine convergent antigen-specific antibody genes of similar sequences shared between individuals. B cell memory varied for different pathogens. Polysaccharide antigen-specific clones were not exclusive to the spleen. Adults had higher clone frequencies and greater class switching in lymphoid tissues than blood, while pediatric blood had abundant class-switched convergent clones. Consistent with reported serology, prepandemic children had class-switched convergent clones to severe acute respiratory syndrome coronavirus 2 with weak cross-reactivity to other coronaviruses, while adult blood or tissues showed few such clones. These results highlight the prominence of early childhood B cell clonal expansions and cross-reactivity for future responses to novel pathogens.
We present a systematic temporal and spectral study of all Swift-X-ray Telescope observations of gamma-ray burst (GRB) afterglows discovered between 2005 January and 2007 December. After constructing ...and fitting all light curves and spectra to power-law models, we classify the components of each afterglow in terms of the canonical X-ray afterglow and test them against the closure relations of the forward shock models for a variety of parameter combinations. The closure relations are used to identify potential jet breaks with characteristics including the uniform jet model with and without lateral spreading and energy injection, and a power-law structured jet model, all with a range of parameters. With this technique, we survey the X-ray afterglows with strong evidence for jet breaks (~12% of our sample), and reveal cases of potential jet breaks that do not appear plainly from the light curve alone (another ~30%), leading to insight into the missing jet break problem. Those X-ray light curves that do not show breaks or have breaks that are not consistent with one of the jet models are explored to place limits on the times of unseen jet breaks. The distribution of jet break times ranges from a few hours to a few weeks with a median of ~1 day, similar to what was found pre-Swift. On average, Swift GRBs have lower isotropic equivalent gamma -ray energies, which in turn result in lower collimation corrected gamma -ray energies than those of pre-Swift GRBs. Finally, we explore the implications for GRB jet geometry and energetics.
