We used radiotelemetry to evaluate seasonal survival rates and mortality factors for a critically endangered island endemic, the Vancouver Island marmot (Marmota vancouverensis Swarth, 1911). ...Recovery of radio transmitters and marmot remains suggested that predation was the major cause of mortality, accounting for at least 24 of 29 (83%) known-fate deaths recorded since radiotelemetry efforts began in 1992. Wolves (Canis lupus L., 1758) and cougars (Puma concolor (L., 1771)) apparently accounted for 17 deaths (59%). Three marmots (10%) were killed by golden eagles (Aquila chrysaetos (L., 1758)), four (14%) were killed by unknown predators that probably included all of the above species, two (7%) died from unknown causes, and three (10%) died during hibernation in a single burrow. We concluded that predation was the proximate cause of recent declines in wild Vancouver Island marmot populations, that losses were highly concentrated in late summer, and that previous studies exaggerated the importance of winter mortality. We suggest that high predation rates were associated with forestry and altered predator abundance and hunting patterns.

Abstract only O-GlcNAcylation is a dynamic, reversible posttranslational modification (PTM) that regulates a multitude of biological processes. Fluctuations in O-GlcNAC of various calcium handling ...proteins impact their functionality in cardiomyocytes. Here, we show for the first time that TRPC6, a nonselective receptor-operated cation channel and mediator of hypertrophy and fibrosis, is constitutively O-GlcNAcylated in the ankyrin repeat domain (AR4), at Ser 14, Thr 70, and Thr 221 within the N-terminal cytoplasmic segment. Of these, only substitution of Thr 221 with alanine (T221A) results in a change of function, notably a hyperactive TRPC6 channel with 5X greater increase in consequent NFAT promoter activity, which is a marker of TRPC6 calcium signaling. Patch-clamp analysis of T221A mutant channels found a 75-80% increased conductance compared to WT. Myocardial injection of T221A in homozygous TRPC6 KO mice by AAV-9 mediated gene transfer results in systolic dysfunction, hypertrophy, and cardiac fibrosis, by loss of OGlcNAc modification at site T221. T221 is highly conserved across species and found in the AR4 domain, which forms the core structure of TRPC6 intracellular domain. Mutating the site in its closest homologs, TRPC3 and TRPC7, also activates channel activity. T221 O-GlcNAcylation also protects the nascent protein from premature proteasomal degradation. Molecular modeling from the crystal structure of human TRPC6 predicts that OGlcNACylation stabilizes electrostatic interactions with the 193-203 loop near AR4, and loop connecting AR4 to the linker helix 1 (LH1) at S199, E200, and E246. Mutating these sites to alanine also increases TRPC6-NFAT signaling similar to what was observed in the T221A mutant. In summary, this study highlights that O-GlcNAcylation of TRPC6 is an important PTM needed to stabilize channel function, and its decline results in gain-of-function related diseases.
Following habitat alteration of fragmentation, competition, parasitism, and predation from species that live in the new habitats may reduce the survival and reproductive success of species living in ...the original habitats. Negative influences from species that live outside the remnant patches are expected to be greater in small rather than in large remnant patches because more "external" species are expected to move through the centers of small remnant patches. We surveyed birds within remnant patches of old-growth montane forests on Vancouver Island, Canada, (1) to evaluate whether the richness and abundance of non-old-growth bird species were larger at the center of small rather than large patches and (2) to evaluate whether the opposite was true of old-growth bird species. More non-old-growth bird species were present at the center of small remnant patches of old growth than in large old-growth patches. We found no relationship, however, between patch size and richness or abundance of old-growth bird species at the center of remnant patches of old growth. This was true for old-growth species with open, cup-shaped nests and cavity nests. Old-growth birds may have been affected less in our study area than in other areas because they evolved within heterogeneous montane forests and interacted with non-old-growth species throughout their evolutionary histories or because the contrast between old-growth forests and logged areas was less than that between the forests and agricultural/urban areas that were surveyed in other studies.

Labor productivity per se generally is not measured in the restaurant industry. Labor cost as a percentage of sales is the measure usually used to assess productivity, and it consists of 3 variables: ...1. the average check, 2. the average hourly wage, and 3. labor productivity, or the number of customers served per hour of direct labor. By distilling labor cost into these 3 components, restaurant managers can enhance their efforts to control costs and can better identify the effect that labor productivity alone has on profit. Managers also can gain greater insight into and control over a restaurant's average hourly wage and labor cost per customer by measuring productivity by job classification. Labor productivity is the only variable of labor cost not directly affected by demographic or geographic factors and, therefore, affords the greatest opportunity to achieve cost reductions.
Photosynthetic microorganisms typically have multiple isoforms of the electron transfer protein ferredoxin, although we know little about their exact functions. Surprisingly, a Chlamydomonas ...reinhardtii mutant null for the ferredoxin-5 gene (FDX5) completely ceased growth in the dark, with both photosynthetic and respiratory functions severely compromised; growth in the light was unaffected. Thylakoid membranes in dark-maintained fdx5 mutant cells became severely disorganized concomitant with a marked decrease in the ratio of monogalactosyldiacylglycerol to digalactosyldiacylglycerol, major lipids in photosynthetic membranes, and the accumulation of triacylglycerol. Furthermore, FDX5 was shown to physically interact with the fatty acid desaturases CrΔ4FAD and CrFAD6, likely donating electrons for the desaturation of fatty acids that stabilize monogalactosyldiacylglycerol. Our results suggest that in photosynthetic organisms, specific redox reactions sustain dark metabolism, with little impact on daytime growth, likely reflecting the tailoring of electron carriers to unique intracellular metabolic circuits under these two very distinct redox conditions.
