The study aimed to evaluate the inclusion effects of
Stryphnodendron rotundifolium
(barbatimão) extracts in substitution of the lasalocid sodium on the ingestive behaviour, intake, ruminal ...parameters, and digestibility of feedlot lambs. Twenty-four pantaneiro lambs were used, with an average age of 150 ± 4.59 days and an initial body weight of 21.2 ± 3.63 kg. The lambs were distributed in three treatments in an experimental design with randomized blocks. The treatments correspond to the additive supplements: LAS (0.019 g of lasalocid sodium/lamb/d); DGB (1.50 g of barbatimão dried ground bark/lamb/d); DHE (0.30 g of barbatimão dry hydroalcoholic extract/lamb/d). The DHE increased 59.74 min in the time spent for ingestion per day, resulting in an efficiency reduction of dry matter (DM) ingestion (127 g of DM/h of feed). There was a reduction of 1.8 mg/dL in the ammoniacal nitrogen concentration with extract supplementation compared to LAS. The DGB reduced total volatile fatty acids by 48.9% compared to the control treatment. The inclusion of barbatimão extracts (DGB and DHE) reduced 12.05% of ruminal butyrate content. The supplementation of barbatimão extracts replacing lasalocid sodium in the diet of feedlot lambs did not affect intake and caused small changes on ingestive behaviour.

Filamentous cyanobacteria are morphologically complex, with several representative species amenable to routine genetic manipulation, making them excellent model organisms for the study of development. Furthermore, two of the developmental alternatives, nitrogen-fixing heterocysts and motile hormogonia, are essential to establish nitrogen-fixing symbioses with plant partners. These symbioses are integral to global nitrogen cycles and could be artificially recreated with crop plants to serve as biofertilizers, but to achieve this goal, detailed understanding and manipulation of the hormogonium and heterocyst gene regulatory networks may be necessary. To test the hypothesis that the administration of 2.5 mg of estradiol benzoate (EB) followed by 1500 mg of long acting progesterone (LA P4) causes similar uterine changes and molecular dynamics in ...anovulatory mares to those observed in cyclic ones, we evaluated the changes of estrogen (ERα and ERβ) and progesterone receptors (PR) in anestrous, transitional and cyclic mares by RT-qPCR and immunohistochemistry. In addition, we evaluated uterine edema, tonus and estrogens and progesterone plasma profile. Endometrial biopsies were taken from anestrous and transitional mares immediately before EB injection, 48 h after EB administration and five days after LA P4 was given. In cyclic mares, biopsies were collected at estrus and at five days after ovulation. Similar estrogen peaks were achieved after the injection of the single EB dose between treated and cyclic groups, as well as maximum uterine edema. Uterine tone was increased to diestrus levels after administration of 1500 mg of LA P4. Changes in relative abundance of transcripts for PR, ERα and ERβ when progesterone stimulated endometrium was compared to estrogen stimulated endometrium were similar between cyclic and non-cyclic treated mares. However, apparent decreased PR in the endometrial glandular epithelium was not observed in non-cyclic mares five days after LA P4 administration as observed at five days after ovulation in cyclic mares. The protocol produced similar endometrial edema, uterine tonus and changes in relative abundance of PR, ERα and ERβ transcripts to those observed in cyclic mares during late estrus and early diestrus, as well as similar estradiol and estrogen conjugate plasma concentrations.

Most mosquito and midge species use hearing during acoustic mating behaviors. For frog-biting species, however, hearing plays an important role beyond mating as females rely on anuran calls to obtain ...blood meals. Despite the extensive work examining hearing in mosquito species that use sound in mating contexts, our understanding of how mosquitoes hear frog calls is limited. Here, we directly investigated the mechanisms underlying detection of frog calls by a mosquito species specialized on eavesdropping on anuran mating signals: Uranotaenia lowii. Behavioral, biomechanical and neurophysiological analyses revealed that the antenna of this frog-biting species can detect frog calls by relying on neural and mechanical responses comparable to those of non-frog-biting species. Our findings show that in Ur. lowii, contrary to most species, males do not use sound for mating, but females use hearing to locate their anuran host. We also show that the response of the antennae of this frog-biting species resembles that of the antenna of species that use hearing for mating. Finally, we discuss our data considering how mosquitoes may have evolved the ability to tap into the communication system of frogs.
Disseminated Candida albicans infection results in high morbidity and mortality despite treatment with existing antifungal drugs. Recent studies suggest that modulating the host immune response can ...improve survival, but specific host targets for accomplishing this goal remain to be identified. The extracellular matrix protein thrombospondin-1 is released at sites of tissue injury and modulates several immune functions, but its role in C. albicans pathogenesis has not been investigated. Here, we show that mice lacking thrombospondin-1 have an advantage in surviving disseminated candidiasis and more efficiently clear the initial colonization from kidneys despite exhibiting fewer infiltrating leukocytes. By examining local and systemic cytokine responses to C. albicans and other standard inflammatory stimuli, we identify a crucial function of phagocytes in this enhanced resistance. Subcutaneous air pouch and systemic candidiasis models demonstrated that endogenous thrombospondin-1 enhances the early innate immune response against C. albicans and promotes activation of inflammatory macrophages (inducible nitric oxide synthase⁺, IL-6(high), TNF-α(high), IL-10(low)), release of the chemokines MIP-2, JE, MIP-1α, and RANTES, and CXCR2-driven polymorphonuclear leukocytes recruitment. However, thrombospondin-1 inhibited the phagocytic capacity of inflammatory leukocytes in vivo and in vitro, resulting in increased fungal burden in the kidney and increased mortality in wild type mice. Thus, thrombospondin-1 enhances the pathogenesis of disseminated candidiasis by creating an imbalance in the host immune response that ultimately leads to reduced phagocytic function, impaired fungal clearance, and increased mortality. Conversely, inhibitors of thrombospondin-1 may be useful drugs to improve patient recovery from disseminated candidiasis.

The Indo-Pacific lionfish Pterois volitans is an invasive species that was first recorded in the Guanahacabibes National Park (GNP), a marine protected area in western Cuba, in 2009. In order to ...determine the invasion progression of this species, we studied lionfish abundance, size, and diet at 6 sites in the GNP between 2010 and 2014. The species’ density, biomass, and length increased over this period, probably due to the abundance of food and shelter in the GNP. Analysis of stomach contents indicated that lionfish fed primarily on fish and crustaceans; main prey were teleosts, predominantly Gobiidae, Pomacentridae, Mullidae, Labridae, Scaridae, and Grammatidae. This example of a rapid increase in an unmanaged population at the onset of invasion provides information that can be used to design a management program targeting lionfish.
The purpose of this study was to assess bulk tank milk sampling strategies for estimating total bacterial count (TBC). Nine large dairies in Wisconsin that produced and shipped at least 1 milk load ...per day were selected for this study. Total bacteria count was performed for each milk load produced during a 13-mo period. The milk shipment frequency was once (n=3), twice (n=4), or 3 times daily (n=2 farms). A threshold of 8,000cfu/mL was used to define increased TBC. The proportion of increased TBC (TBCref) during the study period was defined as the reference probability of an increased TBC for each farm. The number of milk loads that would need to be tested to estimate TBCref precisely (TBCref ± 0.05) in selected time periods (month, quarter, 6mo, or a year) was calculated assuming independence among TBC measurements. Sampling simulations (systematic or simple random sampling) were used to assess the validity of the independence assumption and compare different sampling schedules (every second, every third, or every seventh milk load) used for estimating TBCref in a 13-mo or 30-d TBC series. The number of milk loads tested to estimate TBCref depended on the time period of interest. For farms with daily milk shipments, at least 94% of all milk loads produced would need to be tested to estimate TBCref during a 30-d period. In contrast, when the period of interest was a year, reductions of up to 88% in the number of milk loads tested could be achieved. As the probability of an increased TBC departed from 0.50 toward 1 or 0, fewer samples were needed to estimate TBCref. A sampling schedule based on TBC performed on every second milk load resulted in 100% of 5,000 random samples (taken from the 13-mo TBC series) within the range of TBCref ± 0.05, indicating that sampling half of the milk loads would precisely estimate TBCref. Results of this study suggest that dairy consultants and processors can adjust the frequency of testing of milk loads depending on the goal of the milk quality monitoring program.
Individuals with type 2 diabetes mellitus are at increased risk for heart failure (HF), particularly those with coexisting atherosclerotic cardiovascular disease and/or kidney disease. Some but not ...all dipeptidyl peptidase-4 inhibitors have been associated with increased HF risk. We performed secondary analyses of HF and related outcomes with the dipeptidyl peptidase-4 inhibitor linagliptin versus placebo in CARMELINA (The Cardiovascular and Renal Microvascular Outcome Study With Linagliptin), a cardiovascular outcomes trial that enrolled participants with type 2 diabetes mellitus and atherosclerotic cardiovascular disease and/or kidney disease.
Participants in 27 countries with type 2 diabetes mellitus and concomitant atherosclerotic cardiovascular disease and/or kidney disease were randomized 1:1 to receive once daily oral linagliptin 5 mg or placebo, on top of standard of care. All hospitalization for HF (hHF), cardiovascular outcomes, and deaths were prospectively captured and centrally adjudicated. In prespecified and post hoc analyses of HF and related events, Cox proportional hazards models adjusting for region and baseline history of HF were used. Recurrent hHF events were analyzed using a negative binomial model. In a subset of participants with left ventricular ejection fraction captured within the year before randomization, HF-related outcomes were assessed in subgroups stratified by left ventricular ejection fraction > or ≤50%.
CARMELINA enrolled 6979 participants (mean age, 65.9 years; estimated glomerular filtration rate, mL/min per 1.73m
; hemoglobin A1c, 8.0%; 62.9% men; diabetes mellitus duration, 14.8 years), including 1873 (26.8%) with a history of HF at baseline. Median follow-up was 2.2 years. Linagliptin versus placebo did not affect the incidence of hHF (209/3494 6.0% versus 226/3485 6.5%, respectively; hazard ratio HR, 0.90; 95% CI, 0.74-1.08), the composite of cardiovascular death/hHF (HR, 0.94; 95% CI, 0.82-1.08), or risk for recurrent hHF events (326 versus 359 events, respectively; rate ratio, 0.94; 95% CI, 0.75-1.20). There was no heterogeneity of linagliptin effects on hHF by history of HF at baseline, baseline estimated glomerular filtration rate or urine albumin-creatinine ratio, or prerandomization left ventricular ejection fraction.
In a large, international cardiovascular outcome trial in participants with type 2 diabetes mellitus and concomitant atherosclerotic cardiovascular disease and/or kidney disease, linagliptin did not affect the risk of hHF or other selected HF-related outcomes, including among participants with and without a history of HF, across the spectrum of kidney disease, and independent of previous left ventricular ejection fraction.

Many previous studies have shown that the fidelity of three-dimensional cardiovascular flow simulations depends strongly on inflow and outflow boundary conditions that accurately describe the ...characteristics of the larger vascular network. These boundary conditions are generally based on lower-dimensional models that represent the upstream or downstream flow behavior in some aggregated fashion. However, the parameters of these models are patient-specific, and no clear technique exists for determining them. In this work, an ensemble Kalman filter (EnKF) is implemented for the purpose of estimating parameters in cardiovascular models through the assimilation of specific patients’ clinical measurements. Two types of models are studied: a fully zero-dimensional model of the right heart and pulmonary circulation, and a coupled 0D–1D model of the lower leg. Model parameters are estimated using measurements from both healthy and hypertensive patients, and demonstrate that the EnKF is able to generate distinct parameter sets whose model predictions produce features unique to each measurement set. Attention is also given toward the quality of model predictions made in the absence of direct clinical counterparts, as well as techniques to improve filter robustness against shrinking ensemble covariance.
