Saharan dust inputs affect present day ecosystems and biogeochemical cycles at a global scale. Previous Saharan dust input reconstructions have been mainly based on marine records from the African ...margin, nevertheless dust reaching western-central Europe is mainly transported by high-altitude atmospheric currents and requires high altitude records for its reconstruction. The organic and inorganic geochemical study of sediments from a southern Iberia alpine lacustrine record has provided an exceptional reconstruction of Saharan dust impact and regional climatic variations during the Holocene. After the last deglaciation, results indicate that Saharan dust reached Western Europe in a stepwise fashion from 7.0 to 6.0cal.kyrBP and increased since then until present, promoting major geochemical changes in the lacustrine system. Effective humidity reconstruction indicates wetter conditions during the early Holocene and progressive aridification during middle–late Holocene time, boosting abrupt changes in the lacustrine system. Cyclostratigraphic analyses and transport mechanisms both point to solar irradiance and aridity as major triggering factors for dust supply over Western Europe during the Holocene.

Base isolation is an efficient strategy for protecting structures, especially in countries with high seismic risk, such as Chile. This paper presents the conceptual model, mathematical model, experimental validation and numerical analysis of a roller type base isolation device that aims to solve problems of limited tensile strength (compared to its compressive strength) and lateral instability of all types of rubber bearing isolators when faced with elevated axial load. The conceptual model describes the device’s components and operation. The mathematical model establishes its constitutive law based on the equilibrium equations formulated considering large lateral displacements. Experimental tests were run on a shake-table with a load frame to simulate the isolator’s interaction with the superstructure, considering a combination of the device’s design parameters, in order to identify their effect. In the numerical analysis, six simple frame buildings were modelled and subjected to a seismic record using the proposed roller isolator. Error parameters were obtained between the numerical predictions and the experimental results in each loading and unloading cycle, varying between 1.6% and 5.1% for dissipated energy and 4.0% to 17.7% for the magnitude of force. The proposed device worked as a seismic isolator, reducing the structure’s response in a magnitude order in relation to the building fixed on its base.
TRIM29 (ATDC) exhibits a contextual function in cancer, but seems to exert a tumor-suppressor role in breast cancer. Here, we show that TRIM29 is often silenced in primary breast tumors and cultured ...tumor cells as a result of aberrant gene hypermethylation. RNAi-mediated silencing of TRIM29 in breast tumor cells increased their motility, invasiveness, and proliferation in a manner associated with increased expression of mesenchymal markers (N-cadherin and vimentin), decreased expression of epithelial markers (E-cadherin and EpCAM), and increased expression and activity of the oncogenic transcription factor TWIST1, an important driver of the epithelial-mesenchymal transition (EMT). Functional investigations revealed an inverse relationship in the expression of TRIM29 and TWIST1, suggesting the existence of a negative regulatory feedback loop. In support of this relationship, we found that TWIST1 inhibited TRIM29 promoter activity through direct binding to a region containing a cluster of consensus E-box elements, arguing that TWIST1 transcriptionally represses TRIM29 expression. Analysis of a public breast cancer gene-expression database indicated that reduced TRIM29 expression was associated with reduced relapse-free survival, increased tumor size, grade, and metastatic characteristics. Taken together, our results suggest that TRIM29 acts as a tumor suppressor in breast cancer through its ability to inhibit TWIST1 and suppress EMT.
SAET (Shoreline Analysis and Extraction Tool) is a novel open-source tool to enable the completely automatic detection of shoreline position changes using the optical imagery acquired by the ...Sentinel-2 and Landsat 8 and 9 satellites. SAET has been developed within the ECFAS (European Coastal Flood Awareness System) project, which is intended to be the first European service for coastal flood forecasting, management, and recovery analysis. The tool is developed to characterise the shoreline response associated with punctual events such as coastal storms as well as any other phenomenon. For a given beach segment, SAET facilitates the selection of the satellite images closest in time to the analysed events that offer an adequate cloud coverage level for analysing the shoreline change. Subsequently, the tool automatically downloads the images from their official repositories, pre-processes them and extracts the shoreline position with sub-pixel accuracy. In order to do so, an initial approximate definition of the shoreline is carried out at the pixel level using a water index thresholding, followed by an accurate extraction operating on the shortwave infrared bands to produce a sub-pixel line in vector formats (points and lines). The tool offers different settings to be adapted to the different coastal environments and beach typologies. Its main advantages refer to its autonomy, its efficiency in extracting complete satellite scenes, its flexibility in adapting to different environments and conditions, and its high subpixel accuracy. This work presents an accuracy assessment on a long Mediterranean sandy beach of SDSs extracted from L8 and S2 imagery against coincident alongshore reference lines, showing an accuracy of about 3 m RMSE. At the same time, the work shows an example of the usage of SAET for characterising the response to Storm Gloria (January 2020) on the beaches of Valencia (E Spain). SAET provides an efficient and completely automatic workflow that leads to accurate SDSs while only relying on publicly available information. The tool appears to be a useful extraction tool for beach monitoring, both for public administrations and individual users.
Ribosomal S6 kinases (S6Ks) are critical regulators of cell growth, homeostasis, and survival, with dysregulation of these kinases found to be associated with various malignancies. While S6K1 has ...been extensively studied, S6K2 has been neglected despite its clear involvement in cancer progression. Protein arginine methylation is a widespread post-translational modification regulating many biological processes in mammalian cells. Here, we report that p54-S6K2 is asymmetrically dimethylated at Arg-475 and Arg-477, two residues conserved amongst mammalian S6K2s and several AT-hook-containing proteins. We demonstrate that this methylation event results from the association of S6K2 with the methyltransferases PRMT1, PRMT3, and PRMT6 in vitro and in vivo and leads to nuclear the localisation of S6K2 that is essential to the pro-survival effects of this kinase to starvation-induced cell death. Taken together, our findings highlight a novel post-translational modification regulating the function of p54-S6K2 that may be particularly relevant to cancer progression where general Arg-methylation is often elevated.
Symptoms of depressive disorders such as anhedonia and despair can be a product of an aberrant adaptation to stress conditions. Chronic unpredictable stress model (CUS) can generate an increase in ...the activity of the hypothalamic-pituitary-adrenal axis (HPA) and induce a reduction of neurotrophin signaling and the proliferation of neural progenitors in the adult dentate gyrus, together with increased oxidative stress. Levels of the endocannabinoid anandamide (AEA) seem to affect these depression-by-stress-related features and could be modulated by fatty acid amide hydrolase (FAAH). We aimed to evaluate the effects of FAAH inhibitor, URB597, on depressive-like behavior and neural proliferation of mice subjected to a model of CUS. URB597 was administered intraperitoneally at a dose of 0.2 mg/kg for 14 days after CUS. Depressive-like behaviors, anhedonia, and despair were evaluated in the splash and forced swimming tests, respectively. Alterations at the HPA axis level were analyzed using the relative weight of adrenal glands and serum corticosterone levels. Oxidative stress and brain-derived neurotrophic factor (BDNF) were also evaluated. Fluorescence immunohistochemistry tests were performed for the immunoreactivity of BrdU and Sox2 colabeling for comparison of neural precursors. The administration of URB597 was able to reverse the depressive-like behavior generated in mice after the model. Likewise, other physiological responses associated with CUS were reduced in the treated group, among them, increase in the relative weight of the adrenal glands, increased oxidative stress, and decreased BDNF and number of neural precursors. Most of these auspicious responses to enzyme inhibitor administration were blocked by employing a cannabinoid receptor antagonist. In conclusion, the chronic inhibition of FAAH generated an antidepressant effect, promoting neural progenitor proliferation and BDNF expression, while reducing adrenal gland weight and oxidative stress in mice under the CUS model.

La valutazione preliminare dei pazienti e l’apprezzamento del rischio di ipossia e di ...inalazione sono essenziali per lo sviluppo di protocolli di anestesia individualizzati. L’adattamento dei trattamenti antiaggreganti e anticoagulanti consente, a seconda del tipo di procedura endoscopica, di ridurre il rischio di sanguinamento. La somministrazione controllata di terapie volte all’ottenimento di un livello ottimale di sedazione associata a un’oculata gestione delle vie aeree consente di garantire la qualità e la sicurezza degli esami programmati. Sono disponibili per il professionista dell’anestesia diverse opzioni di trattamento con ipnotici e oppioidi per eseguire una sedazione appropriata. Conoscere le particolarità di ogni atto endoscopico interventistico (colangiopancreatografia retrograda endoscopica CPRE, dissecazione sottomucosa) permette di anticipare le difficoltà tecniche legate all’anestesia. La fornitura di strumenti tecnici di monitoraggio (capnografi adattati alla ventilazione spontanea, indice bispettrale), di somministrazione (sedazione intravenosa a obiettivo di concentrazione SIVOC) e di ossigenazione (ossigenazione nasale ad alto flusso) garantisce la modernizzazione di questo settore in continua evoluzione.
