The copper(I) catalyzed azide–alkyne cycloaddition ‘click’ reaction yields a specific product under mild conditions and in some of the most chemically complex environments. This reaction has been ...used extensively to tag DNA, proteins, glycans and only recently RNA. Click reactions in aqueous buffer typically include a ligand for Cu(I), however we find that acetonitrile as a minor co-solvent can serve this role. Here we investigate the click labeling of RNA and DNA in aqueous buffer to determine the relationship between the stoichoimetry of Cu(I) and the acetonitrile co-solvent that affects nucleic acid stability. We find that very low concentrations of acetonitrile perform equally well and obviate the need for any additional Cu(I) stabilizing ligand. These pseudo-ligandless reaction conditions are optimal for nucleic acids click conjugations.
Hemiparasitic plants commonly known as mistletoe (muérdago in Spanish) in the families Santalaceae and Loranthaceae are common in various kinds of plants or trees, and many hemiparasitic plants are ...used for medicinal purposes in various parts of the world. The objective of the present work, carried out in Psittacanthus linearis (suelda con suelda), a representative species in the seasonally dry forest (SDF) from the north of Perú, was to study aspects of in vitro tissue culture, carry out preliminary phytochemical analysis, and assess antibacterial activity. Seeds of individuals of P. linearis, which used Prosopis pallida (algarrobo) as host plant, were collected and used to induce in vitro seed germination, clonal propagation, callus induction and organogenesis. Stems, leaves and fruits of individuals of P. linearis were dried, powdered, and subjected to ethanol extraction. Posteriorly the extract was first recovered with ethanol and the remnant with chloroform, which formed the ethanolic and chloroformic fraction. A preliminary phytochemical screening was performed and preliminary antibacterial studies with Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa were carried out and their results are discussed. This is the first report about in vitro tissue culture, phytochemical analysis and antibacterial activity of P. linearis. The results may have important implications for understanding physiological and biochemical interactions between host and hemiparasitic species as well as P. linearis with P. pallida and other SDF species.
Introducción: al ser el hombre un ser bio-psico-social, cobra un gran valor el estudio de la salud mental. Objetivo: determinar el estado de salud mental en pacientes del Consultorio Médico 59 del ...Policlínico Universitario Luis Augusto Turcios Lima. Método: estudio observacional, descriptivo, longitudinal y retrospectivo en los. La población de estudio la constituyeron 282 familias, con una muestra de 181 familias (445 pacientes)Resultados: el 54,31 % de los pacientes fueron del sexo femenino, el 17,27 % se encontró en el rango etario de 50 a 59 años y el 33,25 % presentó como nivel de escolaridad el preuniversitario terminado. 82 pacientes son jubilados o peritados, 67 son estudiantes, 30 son amas de casa y 11 refieren no trabajar. El 12,81 % de los pacientes son cuentapropistas. Las enfermedades somáticas predominantes fueron la hipertensión arterial (34,16 %) y la diabetes mellitus (12,58 %). El 0,90 % de los pacientes presentó epilepsia, el 0,67 % depresiones y el 0,67 esquizofrenias o psicosis. El 5,84 % de la población presentó el hábito de fumar y el 0,90 % presentó alcoholismo.Conclusiones: existe una población envejecida, con un elevado nivel escolar. Los volúmenes de desempleo estuvieron condicionados mayormente por los retiros y la presencia de un alto número de estudiantes. La hipertensión arterial y diabetes mellitus constituyen factores de riesgo para el desarrollo de demencias. La epilepsia y depresiones estuvieron presentes en la población de estudio.
RESUMEN Introducción: las onicomicosis son infecciones fúngicas de la lámina ungueal y tejidos adyacentes. Objetivo: determinar la eficacia del tratamiento alternativo con oleozón tópico en pacientes ...con onicomicosis de los consultorios 24 y 25 del Policlínico Universitario “Luis Augusto Turcios Lima” de la provincia de Pinar del Río, de 2017-2018. MétodoS: se realizó un estudio observacional, descriptivo y de corte transversal a los pacientes diagnosticados con onicomicosis en los consultorios 24 y 25 del Policlínico “Luis Augusto Turcios Lima” durante el 2017-2018, el universo estuvo conformado por 90 pacientes con el diagnostico de esta enfermedad, se trabajó con la totalidad de ellos; se estratificó en tres grupos, grupo A (ketoconazol tópico más fluconazol tableta), grupo B (oleozón tópico) y grupo C (fluconazol tableta más oleozón tópico). Resultados: predominó el grupo etáreo de 60-69 (31,1 %) y el sexo masculino (65,6 %), predominaron los pacientes que presentaron cambios de coloración en las uñas (32 %), en el grupo A el 56,7 % de los pacientes presentaron mejoría entre tres y seis meses, el grupo B el 93,3 % en el mismo periodo que el grupo A y en el grupo C el 100 % se curaron en el mismo periodo que los demás grupos. Conclusiones: predominó el grupo de 60-69 años de edad, el sexo masculino fue el más afectado. El signo más frecuente fue el cambio de coloración y el síntoma el dolor; el tratamiento combinado fue el más efectivo.
De marzo a junio de 2018 se estudió el bosque estacionalmente seco en Cerro Tres Puntas de Pilasca (Salas-Motupe, Lambayeque, Perú) con la finalidad de obtener información sobre su estructura y ...composición florística. En un transecto lineal de 11.4 ha, aproximadamente, se evaluaron especímenes de plantas leñosas con DAP ≥ 5.0 cm. Se registraron 410 individuos que representan a 17 especies, 17 géneros y 10 familias. Los taxones más abundantes fueron Vachellia macracantha (Fabaceae 154 individuos) y Celtis iguanaea (Cannabaceae 55 individuos) y las familias mejor representadas corespondieron a Fabaceae, con siete especies y Malvaceae, con dos. El área basal total fue de 343.86 m2 ha-1, en la que destacan Ficus obtusifolia con 139.23 m2 ha-1 y Beilschmiedia sulcata con 120.90 m2 ha-1. El Índice de Valor de Importancia alcanzó los mayores valores en F. obtusifolia (49.34), V. macracantha (46.75) y C. ruizii (41.57); en tanto que el Índice de Valor de la Familia fue más alto para Fabaceae, con 111.86, seguida de Moraceae (46.74) y Lauraceae (45.33). Si bien, los resultados muestran que este bosque es una versión precaria de los existentes en la región, todavía se identificó la presencia de individuos de F. obtusifolia entre 35 - 39.9 m de altura y DAP de 210 a 219.9 cm. El presente estudio contribuirá a desarrollar un programa de conservación, y de reforestación eficaz para mejorar el bosque del Cerro Tres Puntas, ubicado en una de las áreas más pobres del país.
The study of isotopic variations of endogenous and toxic metals in fluids and tissues is a recent research topic with an outstanding potential in biomedical and toxicological investigations. Most of ...the analyses have been performed so far in bulk samples, which can make the interpretation of results entangled, since different sources of stress or the alteration of different metabolic processes can lead to similar variations in the isotopic compositions of the elements in bulk samples. The downscaling of the isotopic analysis of elements at the sub-cellular level, is considered as a more promising alternative. Here we present for the first time the accurate determination of Cu isotopic ratios in four main protein fractions from lysates of neuron-like human cells exposed in vitro to 10 mu M of natural uranium for seven days. These protein fractions were isolated by Size Exclusion Chromatography and analysed by Multi-Collector Inductively Coupled Plasma Mass Spectrometry to determine the Cu isotopic variations in each protein fraction with regard to the original cell lysate. Values obtained, expressed as delta Cu-65, were -0.03 +/- 0.14 % (U-c, k = 2), -0.55 +/- 0.20 % (U-c, k = 2), -0.32 +/- 0.21 % (U-c, k = 2) and + 0.84 +/- 0.21 % (U-c, k = 2) for the four fractions, satisfying the mass balance. The results obtained in this preliminary study pave the way for dedicated analytical developments to identify new specific disease biomarkers, to gain insight into stress-induced altered metabolic processes, as well as to decipher metabolic pathways of toxic elements.
El síndrome de Goodpasture es una vasculitis poco frecuente y con alta mortalidad, que puede afectar tanto a capilares glomerulares como alveolares, en presencia de anticuerpos anti-MBG. Se presenta ...el caso de un paciente masculino de 51 años, que al examen físico se observó temperatura corporal de 39°C, disnea, crepitantes pulmonares y edema de miembros inferiores. Se solicitaron exámenes de laboratorio reportando anemia, proteinuria, hematuria,leucocituria y cilindruria. Al examen serológico, se presentó positividad para anticuerpos anti-MBG, ANCA, antifosfolípidos. Biopsia renal con cambios mesangiales, incremento difuso de matriz, fibrosis intersticial y atrofia multifocal de túbulos, más capilaritis en biopsia pulmonar. El paciente fue diagnosticado con síndrome de Goodpasture, asociado a síndrome Antifosfolípido y vasculitis ANCA. Los anticuerpos antifosfolípidos complican el pronóstico del síndrome de Goodpasture al causar trastornos trombofílicos. Por consiguiente, se inició el tratamiento con metilprednisolona, ciclofosfamida y tratamiento de mantenimiento con micofenolato mofetil + ácido acetilsalicílico, logrando revertir la curva de mortalidad y alcanzando remisión completa de la enfermedad.

The use of Metal-Enhanced Fluorescence (MEF) phenomenon in fluorescence-based bioassays affords for increased sensitivity to be realized by incorporating metal nanoparticles onto planar surfaces. The ...close-range interactions of metal-fluorophores result in increased fluorescence emission from the bioassays, which in turn affords for the detection of target biomolecules at lower concentrations. Moreover, the use of silver nanoparticles increases the photostability of fluorophores improving the detectability of fluorescence emission under prolonged use of excitation light. Although numerous reports on MEF-based biosensing applications exist, the contribution of protein coverage on Silver Island Films (SIFs) on the increased fluorescence emission was never investigated. This work presents our findings on the quantitative comparison of protein surface coverage on SIFs and blank glass slides. In this regard, identical protein bioassay for a model protein (biotinylated bovine serum albumin, b-BSA) on these surfaces is constructed and the relative extent of protein surface coverage on SIFs and blank glass slides was determined using radio-labeled biomolecules. It was found that the total scintillation counts on SIFs and blank glass slides were similar for BSA concentrations ranging from 1 μM to 1 pM, which implies that increased fluorescence in MEF-based biosensing applications is only due to metal-fluorophore interactions.
