We evaluated whether ELISPOT assay can predict tuberculosis (TB) development in kidney‐transplantation (KT) recipients with a negative tuberculin skin test (TST). All adult patients admitted to a KT ...institute between June 2008 and December 2009 were enrolled; TB development after KT was observed between June 2008 and December 2010. Isoniazid (INH) was given to those patients with positive TST or clinical risk factors for latent TB infection (LTBI). ELISPOT assay was performed on all patients, and TB development after KT was observed by a researcher blinded to the results of ELISPOT. A total of 312 KT recipients including 242 (78%) living‐donor KT were enrolled. Of the 312 patients, 40 (13%) had positive TST or clinical risk factors for LTBI and received INH; none developed TB after KT. Of the remaining 272 patients, 4 (6%) of 71 with positive ELISPOT assay developed TB after KT, whereas none of the 201 patients with negative (n = 171) or indeterminate ELISPOTs (n = 30) developed TB after KT (rate difference between positive and negative/indeterminate ELISPOT, 3.3 per 100 person‐years 95% CI 1.4–5.1, p<0.001). Positive ELISPOT results predict subsequent development of TB in KT recipients in whom LTBI cannot be detected by TST or who lack clinical risk factors for LTBI.
Positive results of an interferon‐gamma releasing assay anticipate the subsequent development of tuberculosis in kidney transplant recipients in whom latent tuberculosis infection cannot be detected by tuberculin skin test or who lack clinical risk factors for latent tuberculosis infection. We present a foliation-focused critical review of the boundary conditions and dynamics of 4D gravitational theories. A general coordinate transformation introduces a new foliation and ...changes the hypersurface on which a natural boundary condition is imposed; in this sense gauge transformations must be viewed as changing the boundary conditions. The issue of a gauge invariant boundary condition is nontrivial and has been extensively studied in the literature. We turn around the difficulty in obtaining such a boundary condition (and subtleties observed in the main body) and take it as one of the indications of an enlarged Hilbert space so as to include the states satisfying different boundary conditions. Through the systematical reduction procedure we obtain, up to some peculiarities, the explicit form of the reduced Lagrangian that describes the dynamics of the physical states. We examine the new insights offered by the 3D Lagrangian on BMS-type symmetry and black hole information. In particular we confirm that the boundary dynamics is an indispensable part of the system information.
In acute myeloid leukemia (AML), about 25-30% of patients harbor a constitutively active receptor tyrosine kinase (RTK) FLT3 encoded by a FLT3 allele harboring internal tandem duplication (FLT3-ITD) ...mutation. The presence of FLT3-ITD correlates with poor prognosis in AML and it makes FLT3 an attractive therapeutic target in AML. Unfortunately, to date small-molecule inhibitors of FLT3 have resulted in only partial and transient clinical responses with residual leukemic blasts resistant to FLT3 inhibitors detected in blood or bone marrow. In this study, we investigated whether the RTK Axl is responsible for resistance of FLT3-ITD(+) AML cells to PKC412 and AC220, FLT3 inhibitors currently under clinical trials for FLT3-ITD(+) AML patients. Upon treatment with PKC412 or AC220, phosphorylation of Axl was significantly enhanced in the FLT3-ITD(+) MV4-11 AML cell line and in primary blasts from a FLT3-ITD(+) AML patient. Consistently, a PKC412-resistant AML cell line and PKC412-resistant primary blasts from FLT3-ITD(+) AML patients had significantly higher levels of constitutively phosphorylated Axl and total Axl when compared with a PKC412-sensitive AML cell line and PKC412-sensitive primary blasts from FLT3-ITD(+) AML patients. We also found that resistance of AML cells against the FLT3 inhibitor PKC412 and AC220 was substantially diminished by the inhibition of Axl via a small-molecule inhibitor TP-0903, a soluble receptor Axl fusion protein Axl-Fc or knockdown of Axl gene expression by shRNA. Collectively, our study suggests that Axl is required for resistance of FLT3-ITD(+) AML cells against the FLT3 inhibitor PKC412 and AC220, and that inhibition of Axl activation may overcome resistance to FLT3-targeted therapy in FLT3-ITD(+) AML.
We extend the foliation-based quantization scheme of Park (2015 Eur. Phys. J. C 75 459) to arbitrary asymptotically flat backgrounds including those that are time- and position- dependent. One of the ...ingredients to accomplishing the extension is imposition of a Neumann-type boundary condition. The quantization procedure, especially the gauge-fixing-induced reduction, provides a new insight into the black hole information paradox. The hypersurface degrees of freedom in the asymptotic region-whose dynamics should be responsible for part of the 'hair'-as well as transitions among various excitations play a central role in the global formulation of the information and proposed solution of the information paradox. In retrospect, the quantization scheme reveals the origin of the difficulty of the information problem: the problem's ties with the quantization of gravity and subtle boundary dynamics as well as the multilayered techniques required for its setup and study. We also comment on the implications of the asymptotic symmetries for the present quantization framework.
The RENO experiment has observed the disappearance of reactor electron antineutrinos, consistent with neutrino oscillations, with a significance of 4.9 standard deviations. Antineutrinos from six 2.8 ... GW(th) reactors at the Yonggwang Nuclear Power Plant in Korea, are detected by two identical detectors located at 294 and 1383 m, respectively, from the reactor array center. In the 229 d data-taking period between 11 August 2011 and 26 March 2012, the far (near) detector observed 17102 (154088) electron antineutrino candidate events with a background fraction of 5.5% (2.7%). The ratio of observed to expected numbers of antineutrinos in the far detector is 0.920±0.009(stat)±0.014(syst). From this deficit, we determine sin(2)2θ(13)=0.113±0.013(stat)±0.019(syst) based on a rate-only analysis.
Extending the renormalizability proposal of the physical sector of 4D Einstein gravity, we have recently proposed renormalizability of the 3D physical sector of gravity-matter systems. The main goal ...of the present work is to conduct systematic one-loop renormalization of a gravity-matter system by applying our foliation-based quantization scheme. In this work we explicitly carry out renormalization of a gravity-scalar system with a Higgs-type potential. With the fluctuation part of the scalar field gauged away, the system becomes renormalizable through a metric field redefinition. We use dimensional regularization throughout. One of the salient aspects of our analysis is how the graviton propagator acquires the “mass” term. One-loop calculations lead to renormalization of the cosmological and Newton constants. We discuss other implications of our results as well: time-varying vacuum energy density and masses of the elementary particles as well as the potential relevance of Neumann boundary condition for black hole information.

The recent manufacturing trend toward mass customization and further personalization of products requires factories to be smarter than ever before in order to: (1) quickly respond to customer ...requirements, (2) resiliently retool machinery and adjust operational parameters for unforeseen system failures and product quality problems, and (3) retrofit old systems with upcoming new technologies. Furthermore, product lifecycles are becoming shorter due to unbounded and unpredictable customer requirements, thereby requiring reconfigurable and versatile manufacturing systems that underpin the basic building blocks of smart factories. This study introduces a modular factory testbed, emphasizing transformability and modularity under a distributed shop-floor control architecture. The main technologies and methods, being developed and verified through the testbed, are presented from the four aspects of rapid factory transformation: self-layout recognition, rapid workstation and robot reprogramming, inter-layer information sharing, and configurable software for shop-floor monitoring.
