Nitrogen (N) is a key macronutrient representing a limiting factor for plant growth and development and affects productivity in wheat. In this study, durum wheat response to N chronic starvation ...during grain filling was investigated through a transcriptomic approach in roots, leaves/stems, flag leaf and spikes of cv. Svevo. Nitrogen stress negatively influenced plant height, tillering, flag leaf area, spike and seed traits, and total N content. RNA-seq data revealed 4,626 differentially expressed genes (DEGs). Most transcriptomic changes were observed in roots, with 3,270 DEGs, while 963 were found in leaves/stems, 470 in flag leaf, and 355 in spike tissues. A total of 799 gene ontology (GO) terms were identified, 180 and 619 among the upregulated and downregulated genes, respectively. Among the most addressed GO categories, N compound metabolism, carbon metabolism, and photosynthesis were mostly represented. Interesting DEGs, such as N transporters, genes involved in N assimilation, along with transcription factors, protein kinases and other genes related to stress were highlighted. These results provide valuable information about the transcriptomic response to chronic N stress in durum wheat, which could be useful for future improvement of N use efficiency.
With over 20,000 species, Asteraceae is the second largest plant family. High-throughput sequencing of nuclear and chloroplast genomes has allowed for a better understanding of the evolutionary ...relationships within large plant families. Here, the globe artichoke chloroplast (cp) genome was obtained by a combination of whole-genome and BAC clone high-throughput sequencing. The artichoke cp genome is 152,529 bp in length, consisting of two single-copy regions separated by a pair of inverted repeats (IRs) of 25,155 bp, representing the longest IRs found in the Asteraceae family so far. The large (LSC) and the small (SSC) single-copy regions span 83,578 bp and 18,641 bp, respectively. The artichoke cp sequence was compared to the other eight Asteraceae complete cp genomes available, revealing an IR expansion at the SSC/IR boundary. This expansion consists of 17 bp of the ndhF gene generating an overlap between the ndhF and ycf1 genes. A total of 127 cp simple sequence repeats (cpSSRs) were identified in the artichoke cp genome, potentially suitable for future population studies in the Cynara genus. Parsimony-informative regions were evaluated and allowed to place a Cynara species within the Asteraceae family tree. The eight most informative coding regions were also considered and tested for "specific barcode" purpose in the Asteraceae family. Our results highlight the usefulness of cp genome sequencing in exploring plant genome diversity and retrieving reliable molecular resources for phylogenetic and evolutionary studies, as well as for specific barcodes in plants.

Recent studies have demonstrated that the immunomodulatory drugs (IMiDs) lead to the degradation of the transcription factors Ikaros and Aiolos. However, why their loss subsequently leads to multiple ...myeloma (MM) cell death remains unclear. Using CRISPR-Cas9 genome editing, we have deleted IKZF1/Ikaros and IKZF3/Aiolos in human MM cell lines to gain further insight into their downstream gene regulatory networks. Inactivation of either factor alone recapitulates the cell intrinsic action of the IMiDs, resulting in cell cycle arrest and induction of apoptosis. Furthermore, evaluation of the transcriptional changes resulting from their loss demonstrates striking overlap with lenalidomide treatment. This was not dependent on reduction of the IRF4-MYC “axis,” as neither protein was consistently downregulated, despite cell death occurring, and overexpression of either factor failed to rescue for Ikaros loss. Importantly, Ikaros and Aiolos repress the expression of interferon-stimulated genes (ISGs), including CD38, and their loss led to the activation of an interferon-like response, contributing to MM cell death. Ikaros/Aiolos repressed CD38 expression through interaction with the nucleosome remodeling and deacetylase complex in MM. IMiD-induced loss of Ikaros or treatment with interferon resulted in an upregulation of CD38 surface expression on MM cells, priming for daratumumab-induced NK cell-mediated antibody-dependent cellular cytotoxicity. These results give further insight into the mechanism of action of the IMiDs and provide mechanistic rationale for combination with anti-CD38 monoclonal antibodies.

To examine the effects of caffeinated coffee consumption on intraocular pressure (IOP), ocular perfusion pressure (OPP), and ocular pulse amplitude (OPA) in those with or at risk for primary ...open-angle glaucoma (POAG).
We conducted a prospective, double-masked, crossover, randomized controlled trial with 106 subjects: 22 with high tension POAG, 18 with normal tension POAG, 20 with ocular hypertension, 21 POAG suspects, and 25 healthy participants. Subjects ingested either 237 ml of caffeinated (182 mg caffeine) or decaffeinated (4 mg caffeine) coffee for the first visit and the alternate beverage for the second visit. Blood pressure (BP) and pascal dynamic contour tonometer measurements of IOP, OPA, and heart rate were measured before and at 60 and 90 min after coffee ingestion per visit. OPP was calculated from BP and IOP measurements. Results were analysed using paired t-tests. Multivariable models assessed determinants of IOP, OPP, and OPA changes.
There were no significant differences in baseline IOP, OPP, and OPA between the caffeinated and decaffeinated visits. After caffeinated as compared with decaffeinated coffee ingestion, mean mm Hg changes (± SD) in IOP, OPP, and OPA were as follows: 0.99 (± 1.52, P<0.0001), 1.57 (± 6.40, P=0.0129), and 0.23 (± 0.52, P<0.0001) at 60 min, respectively; and 1.06 (± 1.67, P<0.0001), 1.26 (± 6.23, P=0.0398), and 0.18 (± 0.52, P=0.0006) at 90 min, respectively. Regression analyses revealed sporadic and inconsistent associations with IOP, OPP, and OPA changes.
Consuming one cup of caffeinated coffee (182 mg caffeine) statistically increases, but likely does not clinically impact, IOP and OPP in those with or at risk for POAG.
Filoviruses, including Ebola virus, are unusual in being filamentous animal viruses. Structural data on the arrangement, stoichiometry and organisation of the component molecules of filoviruses has ...until now been lacking, partially due to the need to work under level 4 biological containment. The present study provides unique insights into the structure of this deadly pathogen.
We have investigated the structure of Ebola virus using a combination of cryo-electron microscopy, cryo-electron tomography, sub-tomogram averaging, and single particle image processing. Here we report the three-dimensional structure and architecture of Ebola virus and establish that multiple copies of the RNA genome can be packaged to produce polyploid virus particles, through an extreme degree of length polymorphism. We show that the helical Ebola virus inner nucleocapsid containing RNA and nucleoprotein is stabilized by an outer layer of VP24-VP35 bridges. Elucidation of the structure of the membrane-associated glycoprotein in its native state indicates that the putative receptor-binding site is occluded within the molecule, while a major neutralizing epitope is exposed on its surface proximal to the viral envelope. The matrix protein VP40 forms a regular lattice within the envelope, although its contacts with the nucleocapsid are irregular.
The results of this study demonstrate a modular organization in Ebola virus that accommodates a well-ordered, symmetrical nucleocapsid within a flexible, tubular membrane envelope.

The upcoming launch of the Operational Land Imager (OLI) will start the next era of the Landsat program. However, the Automated Cloud-Cover Assessment (CCA) (ACCA) algorithm used on Landsat 7 ...requires a thermal band and is thus not suited for OLI. There will be a thermal instrument on the Landsat Data Continuity Mission (LDCM)-the Thermal Infrared Sensor-which may not be available during all OLI collections. This illustrates a need for CCA for LDCM in the absence of thermal data. To research possibilities for full-resolution OLI cloud assessment, a global data set of 207 Landsat 7 scenes with manually generated cloud masks was created. It was used to evaluate the ACCA algorithm, showing that the algorithm correctly classified 79.9% of a standard test subset of 3.95 10 9 pixels. The data set was also used to develop and validate two successor algorithms for use with OLI data-one derived from an off-the-shelf machine learning package and one based on ACCA but enhanced by a simple neural network. These comprehensive CCA algorithms were shown to correctly classify pixels as cloudy or clear 88.5% and 89.7% of the time, respectively.

In the present paper, the problem of controlling the unsteady flow separation over an aerofoil, using plasma actuators, is addressed. Although a plasma flow is essentially a two-phase flow, it has been proved that a local force term may be successfully considered within a flow solver in order to mimic the effect of a local acceleration (see 63). By exploiting the Chimera Overlapping Grids technique to allow the inclusion of one or more actuators, accurate numerical simulations have been carried out with the in-house Finite Volume solver Xnavis. The effects of varying both number and position of the actuator/sensor pairs along the aerofoil are investigated by focusing on the dynamic control of the boundary layer separation. The capabilities of the proposed control strategy are also examined by showing how the practical flow separation control problem can be formulated as a simple output regulation problem, so that a simple control strategy may be used. Several numerical simulations of incompressible flows around a pitching NACA0012 at Reynolds Re=20,000 illustrate the effectiveness of the proposed approach, in the presence of time-varying angles of attack and complex non-linear dynamics, which are neglected in the control design. The present work extends the former paper of Broglia et al. 2, where the control algorithm was applied to the simple case of single input/single output regulation and implemented in a Finite Elements numerical code; in that work the boundary layer control of an airfoil at fixed angle of attack was addressed. In the present paper a multi input/multi output approach is theoretically developed and numerically implemented. Robust and fast flow reattachment is achieved, along with both stabilisation and increase/reduction of the lift/drag, respectively. A major advantage of the presented method is that the chosen controlled outputs can be easily measured in realistic applications.
The transcription factor interferon regulatory factor 4 (IRF4) is critical for the development, maintenance, and function of plasma cells. The mechanism by which IRF4 exerts its action in mature ...plasma cells has been elusive due to the death of all such cells upon IRF4 loss. While we identify apoptosis as a critical pathway for the death of plasma cells caused by IRF4 loss, we also determine that IRF4 did not regulate the intrinsic apoptotic pathway directly. By using an inducible IRF4 deletion system in the presence of the overexpression of anti-apoptotic BCL2, we identify genes whose expression is coordinated by IRF4 and that in turn specify plasma cell identity and mitochondrial homeostasis.

Plasma cells can provide lifelong immunity by the continued production of high-affinity, antigen-specific antibody. Low et al. reveal that the transcription factor IRF4 acts within established plasma cells to prevent apoptosis and maintain cellular identity and mitochondrial homeostasis.
Since January 2008, the U.S. Department of Interior / U.S. Geological Survey have been providing free terrain-corrected (Level 1T) Landsat Enhanced Thematic Mapper Plus (ETM+) data via the Internet, ...currently for acquisitions with less than 40% cloud cover. With this rich dataset, temporally composited, mosaics of the conterminous United States (CONUS) were generated on a monthly, seasonal, and annual basis using 6521 ETM+ acquisitions from December 2007 to November 2008. The composited mosaics are designed to provide consistent Landsat data that can be used to derive land cover and geo-physical and bio-physical products for detailed regional assessments of land-cover dynamics and to study Earth system functioning. The data layers in the composited mosaics are defined at 30
m and include top of atmosphere (TOA) reflectance, TOA brightness temperature, TOA normalized difference vegetation index (NDVI), the date each composited pixel was acquired on, per-band radiometric saturation status, cloud mask values, and the number of acquisitions considered in the compositing period. Reduced spatial resolution browse imagery, and top of atmosphere 30
m reflectance time series extracted from the monthly composites, capture the expected land surface phenological change, and illustrate the potential of the composited mosaic data for terrestrial monitoring at high spatial resolution. The composited mosaics are available in 501 tiles of 5000

m pixels in the Albers equal area projection and are downloadable at
