This research demonstrates, a facile approach to fabricate the nano ZnO system in an unique combination of surfactant-polyol-assembly (SPA) acting as a caging agent restricting the ZnO crystallite size in nano-regime. This SPA is suitable for health and hygiene products and such optimized technique is among the very few researches exploring the impact of embedding low concentrations of nano ZnO system into the matrix of sodium salt of long chain fatty acids (soap bar) and liquid cleansing personal care products. The fabricated nano ZnO in SPA and infused products were systematically characterized using various advanced and appropriate techniques. The hexagonal wurtzite structure of nano ZnO-SPA is evaluated based on XRD pattern which also exhibit an average crystallite size as 20.18 nm and high specific surface area as 52.99 m2/g. The SEM-supported morphological assessment confirms the formation of agglomerates of ultrafine ZnO rods and spherical particles. Novel nano ZnO having wideband gap energy (3.66 eV) embedded in soap bar act as a UV-blocker preventing the oxidation of unsaturated long chain fatty acids. Soap bar without ZnO experienced degradation and reduction in whiteness to 17.85% whereas 2.5 mg/g nano ZnO infused soap shows the reduction to 7.9% which clearly reflects the increased photostability of soap bar. The antibacterial efficacy of nano ZnO-SPA and infused products are investigated against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) by Zone of Inhibition (ZOI) and European standard EN:1276. Infused products exhibited high antibacterial efficacy up to 4.43 log reduction equivalent to >99.99% germ kill.
The pathways for biosynthesis of glycogen in bacteria and starch in plants are evolutionarily and biochemically related. They are regulated primarily by ADP–glucose pyrophosphorylase, which evolved ...to satisfy metabolic requirements of a particular organism. Despite the importance of these two pathways, little is known about the mechanism that controls pyrophosphorylase activity or the location of its allosteric sites. Here, we report pyruvate-bound crystal structures of ADP–glucose pyrophosphorylase from the bacterium Agrobacterium tumefaciens, identifying a previously elusive activator site for the enzyme. We found that the tetrameric enzyme binds two molecules of pyruvate in a planar conformation. Each binding site is located in a crevice between the C-terminal domains of two subunits where they stack via a distinct β-helix region. Pyruvate interacts with the side chain of Lys-43 and with the peptide backbone of Ser-328 and Gly-329 from both subunits. These structural insights led to the design of two variants with altered regulatory properties. In one variant (K43A), the allosteric effect was absent, whereas in the other (G329D), the introduced Asp mimicked the presence of pyruvate. The latter generated an enzyme that was preactivated and insensitive to further activation by pyruvate. Our study furnishes a deeper understanding of how glycogen biosynthesis is regulated in bacteria and the mechanism by which transgenic plants increased their starch production. These insights will facilitate rational approaches to enzyme engineering for starch production in crops of agricultural interest and will promote further study of allosteric signal transmission and molecular evolution in this important enzyme family.

The second-order polynomial models were developed using response surface methodology to understand effect of input variables on the responses in terms of,—juice recovery (%), TSS (°Bx) and clarity (%T at 590 nm). Interaction of incubation temperature with other input variables was observed to affect all the responses. Optimum values for input variables could be obtained with statistically valid models (

In the present study, biofilm formation was quantified in UTI isolates of Pseudomonas aeruginosa (n = 22) using the crystal violet assay and was categorized into; strong (n = 16), weak (n = 4), and ...moderate (n = 2) biofilm producers. Further experiments were done using strong (n = 4) and weak (n = 4) biofilm producers. Biofilm formation was greater in Luria broth followed by natural urine and artificial urine on silicone and silicone-coated latex. Cell adhesion and twitching motility were greater in strong biofilm producers. The presence of thick biofilm with an increased number of dead and total number of cells of strong biofilm producers was observed using CLSM. The concentrations of exopolymeric substances (eDNA, protein, and pel polysaccharide) were high in strong biofilm producers. FEG-SEM visualization of biofilm produced by strong biofilm producers showed more cells encased in thick biofilm matrix than weak ones. Overall results provide evidence for increased cell adhesion and twitching motility in strong biofilm producers.
Aggregation of misfolded α-synuclein (α-syn) is a key characteristic feature of Parkinson's disease (PD) and related synucleinopathies. The nature of these aggregates and their contribution to ...cellular dysfunction is still not clearly elucidated. We employed mass spectrometry-based total and phospho-proteomics to characterize the underlying molecular and biological changes due to α-syn aggregation using the M83 mouse primary neuronal model of PD. We identified gross changes in the proteome that coincided with the formation of large Lewy body-like α-syn aggregates in these neurons. We used protein-protein interaction (PPI)-based network analysis to identify key protein clusters modulating specific biological pathways that may be dysregulated and identified several mechanisms that regulate protein homeostasis (proteostasis). The observed changes in the proteome may include both homeostatic compensation and dysregulation due to α-syn aggregation and a greater understanding of both processes and their role in α-syn-related proteostasis may lead to improved therapeutic options for patients with PD and related disorders.
Colorectal Cancer (CRC) is one of the late complications observed in patients suffering from inflammatory bowel diseases (IBD). Carcinogenesis is promoted by persistent chronic inflammation occurring ...in IBD. Understanding the mechanisms involved is essential in order to ameliorate inflammation and prevent CRC. Thrombospondin 1 (TSP-1) is a multidomain glycoprotein with important roles in angiogenesis. The effects of TSP-1 in colonic tumor formation and growth were analyzed in a model of inflammation-induced carcinogenesis. WT and TSP-1 deficient mice (TSP-1-/-) of the C57BL/6 strain received a single injection of azoxymethane (AOM) and multiple cycles of dextran sodium sulfate (DSS) to induce chronic inflammation-related cancers. Proliferation and angiogenesis were histologically analyzed in tumors. The intestinal transcriptome was also analyzed using a gene microarray approach. When the area containing tumors was compared with the entire colonic area of each mouse, the tumor burden was decreased in AOM/DSS-treated TSP-1-/- versus wild type (WT) mice. However, these lesions displayed more angiogenesis and proliferation rates when compared with the WT tumors. AOM-DSS treatment of TSP-1-/- mice resulted in significant deregulation of genes involved in transcription, canonical Wnt signaling, transport, defense response, regulation of epithelial cell proliferation and metabolism. Microarray analyses of these tumors showed down-regulation of 18 microRNAs in TSP-1-/- tumors. These results contribute new insights on the controversial role of TSP-1 in cancer and offer a better understanding of the genetics and pathogenesis of CRC.

This article explores a strategic approach to fabricating nano scale silver (100–2000 ppm) doped zinc oxide (ZnOS) in a unique Surfactant-Polyol-Assembly (SPA) that acts as a caging agent, ...restricting the growth of particles. This highly suitable approach for health and hygiene products is among the few works of literature exploring the fabrication and infusion of nano ZnOS to elevate the germicidal efficacy of liquid cleansing products significantly. XRD reveals a hexagonal wurtzite structure having an average crystallite size of 15.14 nm, total crystallinity of 97.62%, and a high specific surface area of 70.68 m2/g. Surface morphological analysis using SEM exhibits monodisperse spherical particles. UV-Vis spectra indicate Burstein-Moss-effect with blue-shift of UV absorbance edge, increase in bandgap energy from 3.55 to 3.64 eV with higher doping. Significant germicidal efficacy of nano ZnOS and infused products having p values < 0.05 compared to AR grade ZnO are established against Gram-positive and Gram-negative pathogens using zone-of-inhibition (ZOI) and European standard EN 1276. Infused products with 0.2% of ZnOS exhibit ZOI up to 13.25 mm with a log reduction of 2.13, while nano ZnOS 2000 ppm shows ZOI up to 11.75 mm with a log reduction of 5.41, which corresponds to >99.999% germ kill.

Down syndrome (DS) children have a unique genetic susceptibility to develop leukemia, in particular, acute megakaryocytic leukemia (AMkL) associated with somatic GATA1 mutations. The study of this ...genetic susceptibility with the use of DS as a model of leukemogenesis has broad applicability to the understanding of leukemia in children overall. On the basis of the role of GATA1 mutations in DS AMkL, we analyzed the mutational spectrum of GATA1 mutations to begin elucidating possible mechanisms by which these sequence alterations arise. Mutational analysis revealed a predominance of small insertion/deletion, duplication, and base substitution mutations, including G:C>T:A, G:C>A:T, and A:T>G:C. This mutational spectrum points to potential oxidative stress and aberrant folate metabolism secondary to genes on chromosome 21 (eg, cystathionine-β-synthase, superoxide dismutase) as potential causes of GATA1 mutations. Furthermore, DNA repair capacity evaluated in DS and non-DS patient samples provided evidence that the base excision repair pathway is compromised in DS tissues, suggesting that inability to repair DNA damage also may play a critical role in the unique susceptibility of DS children to develop leukemia. A model of leukemogenesis in DS is proposed in which mutagenesis is driven by cystathionine-β-synthase overexpression and altered folate homeostasis that becomes fixed as the ability to repair DNA damage is compromised.
