Hemoglobin (Hgb) forms tetramers (dimerized α–β dimers), which enhance its globular stability and may also facilitate small gas molecule transport, as shown by recent all-atom Newtonian solvated ...simulations. Hydropathic bioinformatic thermodynamic scaling enables close comparisons of hemoglobin dimers with myoglobin and neuroglobin, and reveals many nonlocal wave-like features of strained Hgb structures at the coarse-grained amino acid level. The thermodynamic analysis employs two hydropathic scales, one describing abrupt first-order unfolding transitions, the other continuous second-order transitions. Small molecule exchange at hemes is a first-order process. Wave-like collective tetrameric features appropriate to ligand absorption and release, seen in optical experiments (short times), are identified thermodynamically at long times. Strain fields localized near hemes interfere with extended strain fields associated with dimer interfacial misfit, resulting in novel wavelength dependent dimer correlation function Fano antiresonances.
Improvements in the welfare of animals in the intensive production industries are increasingly being demanded by the public. Scientific methods of welfare improvement have been developed and are ...beginning to be used on farms, including those provided by precision livestock farming. The number of welfare challenges that animals are facing in the livestock production industries is growing rapidly, and farmers are a key component in attempts to improve welfare because their livelihood is at stake. The challenges include climate change, which not only exposes animals to heat stress but also potentially reduces forage and water availability for livestock production systems. Heat-stressed animals have reduced welfare, and it is important to farmers that they convert feed to products for human consumption less efficiently, their immune system is compromised, and both the quality of the products and the animals' reproduction are adversely affected. Livestock farmers are also facing escalating feed and fertiliser costs, both of which may jeopardise feed availability for the animals. The availability of skilled labour to work in livestock industries is increasingly limited, with rural migration to cities and the succession of older farmers uncertain. In future, high-energy and protein feeds are unlikely to be available in large quantities when required for the expanding human population. It is expected that livestock farming will increasingly be confined to marginal land offering low-quality pasture, which will favour ruminant livestock, at the expense of pigs and poultry unable to readily digest coarse fibre in plants. Farmers also face disease challenges to their animals' welfare, as the development of antibiotic resistance in microbes has heralded an era when we can no longer rely on antibiotics to control disease or improve the feed conversion efficiency of livestock. Farmers can use medicinal plants, pro-, pre- and synbiotics and good husbandry to help maintain a high standard of health in their animals. Loss of biodiversity in livestock breeds reduces the availability of less productive genotypes that survive better on nutrient-poor diets than animals selected for high productivity. Farmers have a range of options to help address these challenges, including changing to less intensive diets, diversification from livestock farming to other enterprises, such as cereal and pseudocereal crops, silvopastoral systems and using less highly selected breeds. These options may not always produce good animal welfare, but they will help to give farm animals a better life.

China is the world's biggest livestock producer, and has a rapidly expanding intensive livestock production in response to growing demand. The large size of the country and geographical dispersion of ...the livestock production systems means that animals are often transported long distances to slaughter. This study investigated perceptions of animal welfare issues by stakeholders in the Chinese transport and slaughter industry using utility scores and adaptive conjoint analysis. An initial workshop for experts in this field identified key concerns; these were then included in a questionnaire, which was distributed electronically to stakeholders. Stakeholders, particularly those with higher levels of education, were most concerned about the absence of pre-slaughter stunning and failure to maintain unconsciousness throughout the slaughter process. For all livestock species electrical stunning was considered the best method of stunning and blunt trauma the worst; for cattle and sheep stunning using a penetrating captive bolt was considered preferable to the use a percussive captive bolt. Other concerns considered very important were journey quality and livestock workers' experience and attitudes. Heat stress and closed-sided vehicles were of greater concern than cold stress. Loading facilities and journey length were considered of intermediate importance, while lairage and methods for catching chickens were of least concern. The importance of some welfare concerns, e.g. livestock having to remain standing during a journey, was more commonly recognised by stakeholders who reported a high level of knowledge and experience. Therefore, these welfare issues could be a focus for future training activities. Compared to respondents directly involved in livestock transport, respondents involved in teaching and researching within livestock production rated the presented animal welfare issues as more important. These results can be used to guide development of training programmes, animal welfare research, and certification and regulatory control to target challenges to animal welfare in livestock transport and slaughter in China.

Globally, priority areas for biodiversity are relatively well known, yet few detailed plans exist to direct conservation action within them, despite urgent need. Madagascar, like other globally ...recognized biodiversity hot spots, has complex spatial patterns of endemism that differ among taxonomic groups, creating challenges for the selection of within-country priorities. We show, in an analysis of wide taxonomic and geographic breadth and high spatial resolution, that multitaxonomic rather than single-taxon approaches are critical for identifying areas likely to promote the persistence of most species. Our conservation prioritization, facilitated by newly available techniques, identifies optimal expansion sites for the Madagascar government's current goal of tripling the land area under protection. Our findings further suggest that high-resolution multitaxonomic approaches to prioritization may be necessary to ensure protection for biodiversity in other global hot spots.
ABSTRACT
We present an H i study of the galaxy group LGG 351 using Widefield ASKAP L-band Legacy All-sky Blind Survey (WALLABY) early science data observed with the Australian Square Kilometre Array ...Pathfinder (ASKAP). LGG 351 resides behind the M 83 group at a velocity range (cz) of ∼3500–4800 km s−1 within the rich Hydra-Centaurus overdensity region. We detect 40 sources with the discovery of a tidally interacting galaxy pair and two new H i sources that are not presented in previous optical catalogues. 23 out of 40 sources have new redshifts derived from the new H i data. This study is the largest WALLABY sub-sample to date and also allows us to further validate the performance of ASKAP and the data reduction pipeline askapsoft. Extended H i emission is seen in six galaxies indicating interaction within the group, although no H i debris is found. We also detect H i in a known ultra-faint dwarf galaxy (dw 1328−29), which demonstrates that it is not a satellite of the M 83 group as previously thought. In conjunction with multiwavelength data, we find that our galaxies follow the atomic gas fraction and baryonic Tully–Fisher scaling relations derived from the GALEX Arecibo SDSS Survey. In addition, majority of our galaxies fall within the star formation main sequence indicating inefficiency of gas removal processes in this loose galaxy group.
Growing prosperity, but also disease outbreaks, natural disasters, and consumer preferences are changing global meat consumption. We investigated the 2000-2019 trends in 35 countries monitored by the ...Food and Agriculture Organization and the Organisation for Economic Co-operation and Development. We also tested relationships with Gross Domestic Product (GDP). Several countries appeared to be reaching peak consumption of some meats, and three (New Zealand, Canada, and Switzerland) have reached this. Poultry consumption increased over time in most countries, and beef and mutton/lamb consumption decreased in many. Using cluster analysis, we divided countries into two clusters: one in which increases in GDP per capita matched increases in meat consumption; and a second one of nine countries, for which there was no association between per capita change in GDP and meat consumption. There was evidence of a tipping point around USD 40,000 of GDP per capita, after which increases in economic well-being do not lead to increased meat consumption.

We take a transcriptomics approach to build the ...first reproducible multi-tissue RNA expression signature by gene-chip profiling tissue from sedentary normal subjects who reached 65 years of age in good health.
One hundred and fifty probe-sets form an accurate classifier of young versus older muscle tissue and this healthy ageing RNA classifier performed consistently in independent cohorts of human muscle, skin and brain tissue (n = 594, AUC = 0.83-0.96) and thus represents a biomarker for biological age. Using the Uppsala Longitudinal Study of Adult Men birth-cohort (n = 108) we demonstrate that the RNA classifier is insensitive to confounding lifestyle biomarkers, while greater gene score at age 70 years is independently associated with better renal function at age 82 years and longevity. The gene score is 'up-regulated' in healthy human hippocampus with age, and when applied to blood RNA profiles from two large independent age-matched dementia case-control data sets (n = 717) the healthy controls have significantly greater gene scores than those with cognitive impairment. Alone, or when combined with our previously described prototype Alzheimer disease (AD) RNA 'disease signature', the healthy ageing RNA classifier is diagnostic for AD.
We identify a novel and statistically robust multi-tissue RNA signature of human healthy ageing that can act as a diagnostic of future health, using only a peripheral blood sample. This RNA signature has great potential to assist research aimed at finding treatments for and/or management of AD and other ageing-related conditions.
The anti-apoptotic protein MCL-1 is a key regulator of cancer cell survival and a known resistance factor for small-molecule BCL-2 family inhibitors such as ABT-263 (navitoclax), making it an ...attractive therapeutic target. However, directly inhibiting this target requires the disruption of high-affinity protein-protein interactions, and therefore designing small molecules potent enough to inhibit MCL-1 in cells has proven extremely challenging. Here, we describe a series of indole-2-carboxylic acids, exemplified by the compound A-1210477, that bind to MCL-1 selectively and with sufficient affinity to disrupt MCL-1-BIM complexes in living cells. A-1210477 induces the hallmarks of intrinsic apoptosis and demonstrates single agent killing of multiple myeloma and non-small cell lung cancer cell lines demonstrated to be MCL-1 dependent by BH3 profiling or siRNA rescue experiments. As predicted, A-1210477 synergizes with the BCL-2/BCL-XL inhibitor navitoclax to kill a variety of cancer cell lines. This work represents the first description of small-molecule MCL-1 inhibitors with sufficient potency to induce clear on-target cellular activity. It also demonstrates the utility of these molecules as chemical tools for dissecting the basic biology of MCL-1 and the promise of small-molecule MCL-1 inhibitors as potential therapeutics for the treatment of cancer.
Geological investigations planned for the Europa Clipper mission will examine the formation, evolution, and expression of geomorphic structures found on the surface. Understanding geologic features, ...their formation, and any recent activity are key inputs in constraining Europa’s potential for habitability. In addition to providing information about the moon’s habitability, the geologic study of Europa is compelling in and of itself. Here we provide a high-level, cross-instrument, and cross-discipline overview of the geologic investigations planned within the Europa Clipper mission. Europa’s fascinating collection of ice-focused geology provides an unparalleled opportunity to investigate the dynamics of icy shells, ice-ocean exchange processes, and global-scale tectonic and tidal stresses. We present an overview of what is currently known about the geology of Europa, from global to local scales, highlighting outstanding issues and open questions, and detailing how the Europa Clipper mission will address them. We describe the mission’s strategy for searching for and characterizing current activity in the form of possible active plumes, thermal anomalies, evidence for surface changes, and extremely fresh surface exposures. The complementary and synergistic nature of the data sets from the various instruments and their integration will be key to significantly advancing our understanding of Europa’s geology.
Opinion on the role of protein in promoting athletic performance is divided along the lines of how much aerobic-based versus resistance-based activity the athlete undertakes. Athletes seeking to gain ...muscle mass and strength are likely to consume higher amounts of dietary protein than their endurance-trained counterparts. The main belief behind the large quantities of dietary protein consumption in resistance-trained athletes is that it is needed to generate more muscle protein. Athletes may require protein for more than just alleviation of the risk for deficiency, inherent in the dietary guidelines, but also to aid in an elevated level of functioning and possibly adaptation to the exercise stimulus. It does appear, however, that there is a good rationale for recommending to athletes protein intakes that are higher than the RDA. Our consensus opinion is that leucine, and possibly the other branched-chain amino acids, occupy a position of prominence in stimulating muscle protein synthesis; that protein intakes in the range of 1.3-1.8 g · kg
