Raphaël Nkaka est un historien rwandais, né en 1958, actuellement professeur à l’University of Rwanda et auteur en 2013 d’une thèse de référence sur l’histoire du racisme et des politiques raciales ...au Rwanda du début du xxe siècle au génocide des Tutsi en 1994. Dans cet entretien, il relate d’abord son parcours scolaire et universitaire, entre 1977 et 1987 avant qu’il ne devienne professeur dans l’enseignement secondaire, à une époque marquée à la fois par le profond renouvellement de l’historiographie rwandaise et par l’importance des discriminations et des violences ethnoraciales au Rwanda. Il revient ensuite sur ses activités postérieures au génocide, comme enseignant et chercheur à l’université, où il est engagé dès 1994, et comme producteur d’émissions radiophoniques. Sa trajectoire biographique permet ainsi de revenir sur de multiples questionnements concernant l’histoire du Rwanda, mais aussi, plus généralement, l’histoire de l’Afrique contemporaine et son écriture.
In a systematic sequencing screen of synaptic genes on the X chromosome, we have identified an autistic female without mental retardation (MR) who carries a de novo frameshift Ile367SerfsX6 mutation ...in Interleukin-1 Receptor Accessory Protein-Like 1 (IL1RAPL1), a gene implicated in calcium-regulated vesicle release and dendrite differentiation. We showed that the function of the resulting truncated IL1RAPL1 protein is severely altered in hippocampal neurons, by measuring its effect on neurite outgrowth activity. We also sequenced the coding region of the close related member IL1RAPL2 and of NCS-1/FREQ, which physically interacts with IL1RAPL1, in a cohort of subjects with autism. The screening failed to identify non-synonymous variant in IL1RAPL2, whereas a rare missense (R102Q) in NCS-1/FREQ was identified in one autistic patient. Furthermore, we identified by comparative genomic hybridization a large intragenic deletion of exons 3–7 of IL1RAPL1 in three brothers with autism and/or MR. This deletion causes a frameshift and the introduction of a premature stop codon, Ala28GlufsX15, at the very beginning of the protein. All together, our results indicate that mutations in IL1RAPL1 cause a spectrum of neurological impairments ranging from MR to high functioning autism.
Contre le présupposé selon lequel il n’y aurait pas d’archives au Rwanda – présupposé accolé du reste à un grand nombre d’États d’Afrique subsaharienne –, cet article vise à mettre en évidence la ...richesse du patrimoine archivistique rwandais et l’utilisation qui en est faite par les chercheurs, notamment lorsqu’il s’agit d’analyser les processus historiques ayant conduit au génocide des Tutsi en 1994. Quand bien même ils ne font pas l’objet de politiques de versements ou d’inventaires systématiques, de même qu’ils peuvent être conservés dans des conditions engageant leur préservation, de tels fonds existent, dispersés partout dans le pays, notamment dans les administrations locales, et permettent d’écrire une histoire soucieuse des spécificités locales et de la complexité des dynamiques historiques, dans ses continuités et ses discontinuités. L’article revient également sur l’histoire des Archives nationales, depuis leur création en 1979 jusqu’à leur actuelle restructuration, dans un double mouvement de réorganisation institutionnelle et de numérisation. La création depuis une dizaine d’années de fonds archivistiques reconstitués, spécifiquement consacrés au génocide, pose des questions aussi bien en ce qui concerne les évolutions de l’historiographie que les politiques archivistiques elles-mêmes.
Objective
Asparagine‐linked glycosylation 13 (ALG13) deficiencies have been repeatedly described in the literature with the clinical phenotype of a developmental and epileptic encephalopathy (DEE). ...Most cases were females carrying the recurrent ALG13 de novo variant, p.(Asn107Ser), with normal transferrin electrophoresis.
Methods
We delineate the phenotypic spectrum of 38 individuals, 37 girls and one boy, 16 of them novel and 22 published, with the most common pathogenic ALG13 variant p.(Asn107Ser) and additionally report the phenotype of three individuals carrying other likely pathogenic ALG13 variants.
Results
The phenotypic spectrum often comprised pharmacoresistant epilepsy with epileptic spasms, mostly with onset within the first 6 months of life and with spasm persistence in one‐half of the cases. Tonic seizures were the most prevalent additional seizure type. Electroencephalography showed hypsarrhythmia and at a later stage of the disease in one‐third of all cases paroxysms of fast activity with electrodecrement. ALG13‐related DEE was usually associated with severe to profound developmental delay; ambulation was acquired by one‐third of the cases, whereas purposeful hand use was sparse or completely absent. Hand stereotypies and dyskinetic movements including dystonia or choreoathetosis were relatively frequent. Verbal communication skills were absent or poor, and eye contact and pursuit were often impaired.
Significance
X‐linked ALG13‐related DEE usually manifests as West syndrome with severe to profound developmental delay. It is predominantly caused by the recurrent de novo missense variant p.(Asn107Ser). Comprehensive functional studies will be able to prove or disprove an association with congenital disorder of glycosylation.
Vingt-sept ans après le génocide des Tutsi du Rwanda, cet article revient sur certains des travaux en sciences sociales consacrés à cet évènement majeur de la fin du XXe siècle, afin d’en proposer un ...bilan historiographique. Sont notamment analysées les recherches consacrées aux décennies précédant le génocide, pour tenter d’inscrire celui-ci dans sa profondeur historique sans en faire l’aboutissement inexorable des évolutions sociales et politiques du pays depuis la période coloniale et les luttes politiques de l’indépendance. Sur l’évènement lui-même, l’article expose les différentes grilles de lecture qui en ont été proposées, entre celles qui insistent surtout sur son arrière-plan idéologique dans le cadre d’un racisme qui se traduit par de multiples pratiques de cruauté pendant les massacres, et celles qui pointent davantage les structures sociales et politiques qui conditionnent la mobilisation des groupes de tueurs.
As a result of exome-based sequencing work performed by the DDD study, de novo variants in CNOT3 have emerged as a newly recognised cause of a developmental disorder. This paper describes molecular ...and clinical details of 16 probands with developmental disorders and de novo CNOT3 variants. It is the first such description of the developmental phenotype associated with CNOT3 variants. Eight of these cases were discovered as part of the DDD study, while the other eight were found as a result of large-scale sequencing work performed by other groups. A highly specific phenotype was not recognised in these 16 cases. The most consistent phenotypic features seen in subjects with de novo variants in CNOT3 were hypotonia, relatively small stature, developmental delay, behavioural problems and intellectual disability. There is no easily recognisable facial phenotype, but some common dysmorphic features such as anteverted nares, thin upper lip and low set eyebrows were shared among some of the probands. Haploinsufficiency appears to be the most likely mechanism of action, with eight cases found to have protein-truncating variants. Of the other eight cases (all missense variants), three share an amino acid substitution at the same position which may therefore represent an important functional domain.
Tramadol is a weak opioid analgesic indicated for the treatment of moderate to severe pain. Tramadol intoxication can be lethal, and this drug is frequently involved in voluntary overdose. ...Classically, tramadol intoxication is associated with neurological and respiratory side effects. In contrast, cardiac effects are poorly documented in the literature. We report a case of severe tramadol intoxication, with plasma concentration 20 times the toxic threshold, complicated by refractory cardiogenic shock, successfully treated by extra corporeal life support (ECLS) with a favorable cardiac outcome and ECLS weaning at day 10. Seizure, clonus, and nonreactive mydriasis were present during 4 days, and complete awakening was delayed to day 15. Poisoning caused by high doses of tramadol can lead to refractory cardiogenic shock, and ECLS can be considered as effective rescue therapy in this context.
