Objectives: To identified the ability of Actinomycetes isolates to inhibit the growth of the bacterium Pseudomonas aeruginosa ATCC 27853. The samples in this experiment were from Kebun Raya Bogor that had been rejuvenated on Starch Casein Agar (SCA). Methods: Six dilution series 10-1; 10-2; 10-3; 10-4; 10-5; 10-6 Actinomycetes isolates were used to observe the inhibition zone of P.aeruginosa growth on Mueller Hinton Agar (MHA) media by diffusion method. Results: The effective incubation time occurred at 24 hours, and then it resulted in the average clear zone diameter of 14.70 mm, 10.57 mm, 8.53 mm, 8.47 mm, 6.97 mm, and 5.30 mm. The results of the One – Way Anova test with p-value = 0.000 (p < 0.005) showed some differences at each concentration to inhibit the growth of P.aeruginosa ATCC 27853 at 24 hours incubation period. Conclusion: The most effective concentration of Actinomycetes isolates that can potentially be antibacterial was the concentration of 10-1 with potential solid inhibitory power.Keywords: Actinomycetes, antibacterial, Pseudomonas aeruginosa
COVID-19 is caused by a coronavirus and was first identified in Wuhan, China. Clinical symptoms of COVID-19 patients show a significant decrease in lymphocytes. Decreased lymphocyte count is a factor ...that increases severity in COVID-19 disease. COVID-19 patients who died reported a very low number of lymphocytes compared to COVID-19 patients who recovered. Increasing lymphocytes to normal levels again, especially in the initial phases of infection, is one of the keys to healing COVID-19 patients. The immune system can be used as a strategy for COVID-19 treatment. Still, immunomodulators must be given at the right time at the beginning of infection, with the right dose, not to cause deterioration in the patient. Immunomodulator administration in the final stage can cause severity in COVID-19 disease due to increased cytokine storm. Therefore, Immunodulator screening, which increases lymphocyte counts, is essential for COVID-19 treatment strategies in the initial stage. Lymphocytes are easily isolated from human blood and consist of NK (Natural Killer) cells, T cells (for cell-mediated, adaptive cytotoxic immunity), and B cells (for humoral, antibody-driven adaptive immunity). Lymphocytes can be isolated from the blood and cultured in vitro. Animal intervention studies showed that immunomodulatory substances could be used for immunomodulator screening candidates for COVID-19 in initial stage therapy. Some candidates for active ingredients that can be used as immunomodulators are honey. Honey has been shown to increase mouse T cell proliferation. This systematic review shows that honey can increase lymphocyte proliferation and have an anti-inflammatory function by lowering IL-6 levels in several in vivo studies.
Tobacco leaf contains antibacterial secondary metabolite compounds, such as phenol, alkaloids, and essential oils. This study compares the potential antibacterial effects of Indonesian tobacco leaf ...extracted using the heat reflux method (producing an extract) and pyrolysis method (providing a bio-oil). The tobacco leaf extract was challenged against Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853. The bio-oil from the pyrolysis method showed inhibitory Kirby Bauer zones higher than those of the extract from heat reflux method, with the maximum results in the pyrolysis method indicating zones of 6.35 mm (S. aureus), 5.90 mm (E. faecalis), 3.97 mm (E. coli), and 5.025 mm (P. aeruginosa). Further study analyzed the effectiveness of the disc and well diffusion antibacterial test methods for measuring the antibacterial effect of bio-oils against Escherichia coli ATCC 25922 and Pseudomonas aeruginosa ATCC 27853. The bio-oil used in the well diffusion test method showed the most significant antibacterial effectiveness. It showed the biggest inhibition zone, with a maximum of 11.65 mm and 8.90 mm for E. coli and P. aeruginosa. Our results showed Nicotiana tabacum var. Virginia Bio Oil from Ponorogo (Indonesia) is a strong potential antimicrobial, especially using well diffusion test.

Covid-19 patients were first detected in Depok, Cinere, West Java, and increased on June ...20, 2020. There have been 45,029 positive cases of Covid-19 on June 2021. Gandul Cinere village is located bordered by South Jakarta. Jakarta has a relatively high number of Covid-19 positive patients. Many people lost their jobs during the pandemic, including in the Jakarta and surrounding areas. Anis Baswedan, the governor, expects the unemployment rate due to the Covid-19 Pandemic in the DKI Jakarta area will reach 10 percent in 2021. This shows that economic problems cannot be ruled out easily. Therefore, efforts are needed to improve the economy of UMKM (Usaha Mikro Kecil dan Menengah) to further strengthen Indonesia's economy, especially in the DKI Jakarta and surrounding areas. Red Ginger (Zingiber officinale var Rubrum) and lemongrass (Cymbopogon citratus) are easy to find in Indonesia. Palm Sugar Ginger Lemongrass (MI JAREN) instant drink is an excellent herbal product to be studied and marketed in the Gandul village community. This instant herbal drink-making training was provided online, along with how to package it. The activity results showed that society in Kampung KB Gandul has good knowledge, only still requires guidance for the marketing, especially online. It is hoped that this community service activity can improve the economy in Kampung KB Gandul while also keeping the people of Kampung KB Gandul to avoid COVID-19 infection.

The addition of six histidine amino acid residues at its C-terminal is intended for the protein isolation process using the His-tag antibody.
Methods
The study aimed to investigate the optimization of the EGFP polyarginine protein expression in
Saccharomyces cerevisiae in sufficient quantities for the protein isolation stage. This study also analyzed EGFP expression without polyarginine to analyze the polyarginine addition effect on expression processes. Protein expression was qualitatively measured by looking at expression fluorescence and protein levels of EGFP and EGFP - PolyR proteins.
Results
The addition of a PolyR group to the C-terminal of EGFP carrying C-terminal 6×His-tag showed similar fluorescence expression levels compare to EGFP without addition of PolyR as well. Moreover, yeast with plasmid insertion showed decrease
S. cerevisiae growth curve but still preserving the fluorescence EGFP.
Conclusions
The expression of the EGFP modified protein in
S. cerevisiae is not affected by the addition of arginine.

Saccharomyces cerevisiae INVSc1 at relatively low levels. The lyticase incubation time modification and administration of 3-5 kDa microfilter to concentrate increased the yield of isolated protein.
Conclusions: The sufficient amount of protein isolation in
S. cerevisiae can be achieved by using lyticase and sonicators combination for the lysis process.

Therefore, a continuous understanding of health protocols is needed. PHBSis part of the health protocol that must be run in this New Normal era. Continued knowledgeprovision is required in order to prevent the transmission of Covid-19, especially in theGandul Village area, Cinere, which borders the red zone area of Covid-19. Health cadreswho are the vanguard need assistance and guidance in understanding and applying PHBS ineveryday life. The methods used were online counseling, online discussion, online mentoring,and pre-/post-test implementation. The result showed increased knowledge about Clean andHealthy Living Behavior after given counseling and discussion with the respondent.
Breast cancer is one type of cancer that ranks first in Asia and in Indonesia with a relative frequency of 18.6%. Breast cancer also occupies the top 5 positions for causes of cancer deaths. Many ...breast cancer sufferers are found clinically with advanced stage symptoms, so they need special attention. In fact, breast cancer can be overcome if the patient begins to detect and treat it early. Early efforts to carry out breast cancer screening are with the BSE program or self breast examination. Counseling was carried out at Posbindu Anggrek Bulan and Posbindu Delima Senja. The implementation of this service activity uses a method of counseling, video playback and simulation that is very good for providing knowledge and skills related to health education to the community. There is an increase in knowledge of the material given through the pretest and posttest. Self-awareness skills as breast cancer prevention efforts were carried out well by participants. The results of community service activities are expected to get a lot of information about breast cancer as well as overall female reproductive health information.
